Differential Expression of Isoflavone Biosynthetic Genes in Soybean During Germination (콩 발아기간 중 isoflavone 생합성 유전자 발현 변이)

  • Soybean isoflavones are essential secondary metabolites synthesized through the phenylpropanoid pathway, and they play vital roles in human health. Isoflavone content is a complex quantitative trait controlled by multiple genes, and the genetic mechanisms underlying isoflavone biosynthesis remain largely unknown. Therefore, the present study analyzed the content of isoflavone and expression of six key genes involved in its biosynthesis (i.e., CHS6, HID, IF7GT, IF7MaT, GmIMaT1, and GmIMaT3) during soybean seed germination. Isoflavone content was quantified using high-performance liquid chromatography, and isoflavone biosynthetic gene expression was analyzed using quantitative real-time PCR. Two cultivars, namely 'Daepung2ho' and 'Pungsannamulkong', which are high- and low-isoflavone cultivars, respectively, were used. Isoflavone accumulation gradually increased with the progression of the germination period. As such, malonyl glucosides accounted for over 80% of the total content, whereas acetyl glucosides were present at trace amounts. Transcriptional analysis of isoflavone biosynthetic genes demonstrated expression patterns parallel to isoflavone content; however, there was no clear correlation between isoflavone content and gene expression. Moreover, most isoflavone biosynthetic genes showed different expression patterns depending on the individual gene or genotypes. Among the tested genes, HID showed consistently higher expression, except at 3 days after germination, and its expression was upregulated in 'Daepung2ho' but downregulated in 'Pungsannamulkong'. In addition, all tested genes exhibited different expression patterns between cotyledons and hypocotyls and responded differently to the germination period. These findings suggest that the expression levels of isoflavone biosynthetic genes are not consistent with the germination period and appear to be genotype-dependent.

A Study on the Image of a Nurse in Korean Modern Novels (한국 현대소설에 나타난 간호사 이미지 연구)

  • This study analyzed how the literary works embodying nurses accommodate various discourses about nurses and reproduce them in literature. The subject of analysis is a Korean modern novel featuring a nurse, and 29 works were selected from 1927 to 2016. The analysis method is the content analysis of the novel among the qualitative research methods. The unit of analysis adopts a narrative or dialogue that deals with the image of a nurse in a novel as a unit of analysis, The image determining factors of previous studies were integrated and categorized into 4 types and considered. As a result of analyzing the image of a nurse in the novel, First, traditional image types include Lee Kwang-soo 『Love』, Kim Eui-jung 『Doctor Han』, Jo Jung-rae 『Han River』, Gong Ji-young's 「Field of Stars」, Baek Min-seok 「Poor Little Hans」 Second, social image types include Kang Kyung-ae 「Dark」, Kim Kyung-wook 「Heaven's Gate」, Choi Jeong-hee 「Cheonmaek」 Third, professional image types include Lee Cheong-jun 「Mr. Jo Man-deuk」, 「Discharge」, Choi In-hoon 『The Square』, Kim Yeon-soo 「The Night in the Tunnel Where I Listened to Jusaeng Tudipini」, Jeong Se-lang 『Public health teacher Ahn Eun-young』and Fourth, personal image types include Choi In-ho 「Apprentice Patient」, Kim Jeong-han 「The Third Ward」, Eun Hee-kyung 『Minor League』, Hoon Kim 「Hwajang」, Ha Seong-ran 「The Joy of Eating」, Kim Ji-yeon 「Hippocrates Love Song」, Park Kyung-ri 「Era of Distrust」, Jeong Mi-kyung 「The Lady of Arsenal」 typed as. Through the image of the nurse in the novel, the implications of the novel for human care were discussed.

Analysis of Volatile Organic Compounds in Sediments Using HS-GC/MS - Confirmation of Matrix Effects in External and Internal Standard Methods - (HS-GC/MS를 이용한 퇴적물 중 휘발성유기화합물 분석 - 외부 및 내부표준방법에서 매질영향 확인 -)

  • Volatile Organic Compounds (VOCs) in sediments, which can cause human health problems, have been monitored in Korea since 2014. Measured VOC concentrations can be affected by matrix type and the volatility of target substances. In this study, (1) VOCs volatility and the influence of matrix interference were confirmed, and (2) internal standards (IS) method was applied to improve analytical method. For these purposes, method detection limit (MDL), calibration linearity, precision and accuracy of VOCs were compared in various matrices using the IS. Some of VOCs in sediments showed different peak areas and reduced rates compared to water matrix. It was suggested that adsorption properties of sediments hindered the migration to vapor during heat pretreatment in headspace method. A calibration curve was created in clean sand. Recovery rates for the calibration curve method and IS applying method were 64.1~83.1% and 99.1~119.3%, respectively. Relative standard deviations ranged from 11.1% to 21.6% for the calibration curve method and those for IS ranged 4.7% to 13.7%. In case of real sediment, calibration curve and 1,2-Dichlorobenzene-d4 (ODCB) among IS were not suitable. The average recovery rate of Fluorobenzene (FBZ) increased by 56.4% and Relative Standard Deviation (RSD) by 4.7%. However, the recovery rate was increased in the samples with large values of igniting intensity. This study confirmed that influence of the matrix of VOCs in sediment, and addition of IS materials improved precision and accuracy. Although IS corrects volatilization and adsorption, it is recommended that more than two types of IS should be added rather than single.

Acid treatment effects on the contents of quercetin glycosides and aglycone in red onion powder (산처리에 의한 적양파 분말의 quercetin 배당체와 aglycone의 농도변화)

  • Flavonoids are bioactive plant metabolites that have a range of beneficial effects on human health. Quercetin 4'-glycoside (Q4'G), quercetin 3,4'-diglycoside (Q3,4'G), and quercetin aglycone (QA) are the main flavonoids found in onions. QA, in particular, is likely to have a greater biological effect than glycosides. To develop an onion extract with high quercetin content, the optimal extraction conditions for red onion powder containing the outer layer of the onion were determined. The effects of acid treatment on the concentration of quercetin glycosides and QA were evaluated. The flavonoids of red onion powder were optimally extracted under 60-70% ethanol at 70℃ for 2 h. The deglycosylation of Q3,4'G and an increase in Q4'G content occurred within 6 h of 0.2% acetic acid treatment. The QA content and deglycosylation of Q4'G eventually peaked at 24 h. In addition, QA content and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity were highly correlated, with a correlation coefficient of 0.90.

  • The COVID-19 pandemic has affected many aspects of human life. The pandemic not only caused millions of fatalities and problems but also changed public sentiment and behavior. Owing to the magnitude of this pandemic, governments worldwide adopted full lockdown measures that attracted much discussion on social media platforms. To investigate the effects of these lockdown measures, this study performed sentiment analysis and latent Dirichlet allocation topic modeling on textual data from Twitter published during the three lockdown waves in Malaysia between 2020 and 2021. Three lockdown measures were identified, the related data for the first two weeks of each lockdown were collected and analysed to understand the public sentiment. The changes between these lockdowns were identified, and the latent topics were highlighted. Most of the public sentiment focused on the first lockdown as reflected in the large number of latent topics generated during this period. The overall sentiment for each lockdown was mostly positive, followed by neutral and then negative. Topic modelling results identified staying at home, quarantine and lockdown as the main aspects of discussion for the first lockdown, whilst importance of health measures and government efforts were the main aspects for the second and third lockdowns. Governments may utilise these findings to understand public sentiment and to formulate precautionary measures that can assure the safety of their citizens and tend to their most pressing problems. These results also highlight the importance of positive messaging during difficult times, establishing digital interventions and formulating new policies to improve the reaction of the public to emergency situations.

Assessment of the Freshwater Comprehensive Soundness in the Geum River Basin - Focusing on Ecological Soundness and Water Welfare - (금강유역의 하천 종합건전성 평가 - 생태적 건전성 및 물 복지를 중심으로 -)

  • Freshwater soundness can be broadly divided into the health of river ecosystems and social services for human water use, that is, water welfare. In this study the ecological soundness and water welfare index of rivers were calculated, and the freshwater comprehensive soundness was analyzed using the quadrant model for 14 basins in the Geum River basin. As for the ecological soundness of the river, the area upstream of the dam (based on Daecheong Dam) was evaluated as 'good', and the area downstream of the dam was evaluated as 'bad'. On the other hand as for water welfare, the area downstream of the dam with a large population was in a relatively 'good' condition, and the area upstream of the dam had a relatively 'poor' level of water welfare. In terms of freshwater comprehensive soundness, it was found that the basins of Mujunamdaecheon, Bocheongcheon and Daecheongdam showed good ecological soundness and water welfare, and the Geumganggongju, Nonsancheon and Geumganhagueon basins were all poor. The Gapcheon basin showed good results in 2014 due to continuous investment and systematic management. Therefore, it is necessary to find ways to utilize it for other watersheds through benchmarking.

An Analysis of Indoor Air Quality and Risk Assessment for One-room Housing around the University in the Post-Corona Era (포스트 코로나 시대의 대학교 주변 원룸형 주택에 대한 실내 공기질 분석 및 위해성 평가)

  • In this study, in order to grasp the current situation of indoor environmental pollution and indoor ventilation in one-room around the university in the post-corona era, we analyzed the experimental data and conducted a questionnaire survey on university students. By analyzing the content, the effects of formaldehyde, dust and other pollution on the human body, which are usually not easily detectable, are digitized and more easily taken into account. Among the experimental results, the concentration of VOC and HCHO, gas pollutants among indoor pollutants, exceeded the recommended criteria of the Ministry of Environment in most studio apartments. Overall, the average CO2 concentration was lower than the Ministry of Environment's maintenance standard (1000ppm), but it was relatively high in summer and winter, and it is believed to be caused by cooling and heating in an enclosed space. The levels of PM2.5 and PM10, particulate pollutants, increased in November and December, and it is believed that ventilation defects due to degradation in external temperature. There was no clear difference between the two types, and there was a very high correlation between PM2.5 and PM10, HCHO and VOC. It was found that temperature was closely correlated with all sources except CO2, and humidity was closely correlated with all sources except PM2.5 and PM10. Health risk assessment was conducted for formaldehyde. The average ECR of studio R2 in May was 3.91E-4, and the ECR figure in September was 3.65E-4, which was very high compared to other residential spaces. The R2 level was calculated as 4 people per 10,000 people in the lifetime risk of cancer of residents, exceeding the allowable risk. R8 also showed higher ECR results than other spaces after R2, especially in October, 2.01E-4, six times higher than R7 measured in October, and 1.87E-4 in July, four times higher than R9.

A Study on the Job Recognition and Educational Needs of Care Workers according to the Types of Working Place in the Urban Area (일 도시 지역 근무지 형태에 따른 요양보호사의 직무인식과 교육요구도 분석)

  • Purpose: This study is designed to provide care workers with basic data on the development of job training programs for enhancing professionalism of senior care services by identifying and analyzing job recognition and education requirements according to the type of work place. Methods: Data were collected from April to July 2019 from 177 care workers working at the Elderly Care Facilities, Elderly Housing Welfare Facilities and Rehabilitation Facilities in the B Metropolitan City. It was analyzed using the SPSS Win 21.0 Statistical Program. Results: The results of job recognition of care worker according to the type of work place shows as follows. As for health care welfare facilities for senior citizens and residential welfare facilities for the elderly was the highest level for physical change and mobile care protection belonging to basic care protection technologies. For the rehabilitation and welfare facility, human rights and prevention of abuse among senior citizens which is belonging to occupational ethics and attitudes of care workers showed the highest. Conclusion: It is required for differentiated job training to enhance capacity for care workers according to the type of work place, and it is needed to developed systematic and specific customized hands-on training to perform, not for simple knowledge.

Spatial Gap-filling of GK-2A/AMI Hourly AOD Products Using Meteorological Data and Machine Learning (기상모델자료와 기계학습을 이용한 GK-2A/AMI Hourly AOD 산출물의 결측화소 복원)

  • Since aerosols adversely affect human health, such as deteriorating air quality, quantitative observation of the distribution and characteristics of aerosols is essential. Recently, satellite-based Aerosol Optical Depth (AOD) data is used in various studies as periodic and quantitative information acquisition means on the global scale, but optical sensor-based satellite AOD images are missing in some areas with cloud conditions. In this study, we produced gap-free GeoKompsat 2A (GK-2A) Advanced Meteorological Imager (AMI) AOD hourly images after generating a Random Forest based gap-filling model using grid meteorological and geographic elements as input variables. The accuracy of the model is Mean Bias Error (MBE) of -0.002 and Root Mean Square Error (RMSE) of 0.145, which is higher than the target accuracy of the original data and considering that the target object is an atmospheric variable with Correlation Coefficient (CC) of 0.714, it is a model with sufficient explanatory power. The high temporal resolution of geostationary satellites is suitable for diurnal variation observation and is an important model for other research such as input for atmospheric correction, estimation of ground PM, analysis of small fires or pollutants.

Genome characterization and mutation analysis of human influenza A virus in Thailand

  • The influenza A viruses have high mutation rates and cause a serious health problem worldwide. Therefore, this study focused on genome characterization of the viruses isolated from Thai patients based on the next-generation sequencing technology. The nasal swabs were collected from patients with influenza-like illness in Thailand during 2017-2018. Then, the influenza A viruses were detected by reverse transcription-quantitative polymerase chain reaction and isolated by MDCK cells. The viral genomes were amplified and sequenced by Illumina MiSeq platform. Whole genome sequences were used for characterization, phylogenetic construction, mutation analysis and nucleotide diversity of the viruses. The result revealed that 90 samples were positive for the viruses including 44 of A/H1N1 and 46 of A/H3N2. Among these, 43 samples were successfully isolated and then the viral genomes of 25 samples were completely amplified. Finally, 17 whole genomes of the viruses (A/H1N1, n=12 and A/H3N2, n=5) were successfully sequenced with an average of 232,578 mapped reads and 1,720 genome coverage per sample. Phylogenetic analysis demonstrated that the A/H1N1 viruses were distinguishable from the recommended vaccine strains. However, the A/H3N2 viruses from this study were closely related to the recommended vaccine strains. The nonsynonymous mutations were found in all genes of both viruses, especially in hemagglutinin (HA) and neuraminidase (NA) genes. The nucleotide diversity analysis revealed negative selection in the PB1, PA, HA, and NA genes of the A/H1N1 viruses. High-throughput data in this study allow for genetic characterization of circulating influenza viruses which would be crucial for preparation against pandemic and epidemic outbreaks in the future.
