Expression of Human KCNE1 Gene in Zebrafish (Zebrafish에서 인간 KCNE1 유전자 발현에 관한 연구)

  • This study was aimed to produce a transgenic zebrafish expressing the human KCNE1 gene. Initially, the entire CDS of the human KCNE1 gene was amplified from a human genomic DNA sample by polymerase chain reaction using a primer set engineered with restriction enzyme sites (EcoRI, BamHI) at the 5' end of each primer. The resultant 402 bp KCNE1 amplicon flanked by EcoR1 and BamH1 was obtained and subsequently cloned into a plasmid vector pPB-CMVp-EF1-GreenPuro. The integrity of the cloned CDS sequence was confirmed by DNA sequencing analysis. Next, the recombinant vector containing the human KCNE1 (pPB-CMVp-hKCNE1-EF1-GreenPuro) was introduced into fertilized eggs of zebrafish by microinjection. Successful expression of the recombinant vector in the eggs was confirmed by the expression of the fluorescence protein encoded in the vector. Finally, in order to assure that the stable expression of the human KCNE1 gene occurred in the transgenic animal, RNAs were extracted from the animal and the presence of KCNE1 transcripts was confirmed by RT-PCT as well as DNA sequencing analysis. The study provides a methodology to construct a useful transgenic animal model applicable to the development of diagnostic technologies for gene therapy of LQTS (Long QT Syndrome) as well as tools for cloning of useful genes in fish.

Comparative Study on the Nutritional Value of Pidan and Salted Duck Egg

  • Pidan and salted duck eggs are of nutritional rich alternative duck egg products which are predominantly consumed in China, Thailand, South Korea and other Chinese migrated countries. Both eggs are rich in proteins, lipids, unsaturated fatty acids and minerals. A Pidan whole egg contains 13.1% of protein, 10.7% of fat, 2.25% of carbohydrate and 2.3% of ash, whereas the salted duck egg contains 14% of protein, 16.6% of fat, 4.1% of carbohydrate and 7.5% of ash. The fresh duck egg contains a range of 9.30-11.80% of protein, 11.40-13.52% of fat, 1.50-1.74% of sugar and 1.10-1.17% of ash. Proteins, lipids, and ash contents are found to be greatly enhanced during the pickling and salting process of pidan and salted duck eggs. However, the alkaline induced aggregation of pidan leads to degradation and subsequent generation of free peptides and amino acids. Very few amino acids are found to be lost during the pickling and storage. However, no such losses of amino acids are reported in salted duck eggs during the salting process of 14 d. Phospholipids and cholesterol contents are lower in pidan oil and salted duck egg yolk oil. Thus, the pidan and salted duck eggs are nutritionally rich alternatives of duck egg products which will benefit the human health during consumption.

Identification of Microorganisms from Eggs in Hypermarket in the Northern Gyeonggi Area (경기 북부 일부 지역 대형 마트 유통계란에 오염된 미생물의 분리)

  • Microorganisms or their toxins can be transferred to eggs and cause food poisoning in humans. Therefore, this study was conducted to detect microbial contamination of eggs and to identify microorganisms in any contaminated eggs. Four different brands of eggs were collected from hypermarkets in the northern Gyeonggi area. The total bacterial counts on the shells of the eggs varied greatly between brands. In addition, various bacterial species including Klebsiella pneumoniae, Pseudomonas mendocina, Alcaligenes xylosoxidans, Alcaligenes faecalis, and Enterobacter cloacae were identified on eggshells. Furthermore, mean of total bacterial counts of four brands was $3.4{\times}10^4 cfu/m{\ell}$ and E. coli was detected on the eggshell of one brand egg. However, Salmonella was not identified on all brands of collected eggs. We also demonstrated that the E. coli isolated from the eggshell was not pathogenic based on the absence of pathogen-specific gene expression patterns. Taken together, the result of this study indicate that strict quality control and improved distribution controls are required to decrease microbial contamination and improve human health.

Enhanced Green Fluorescent Protein Gene under the Regulation of Human Oct4 Promoter as a Marker to Identify Reprogramming of Human Fibroblasts

  • Recent studies on nuclear transfer and induced pluripotent stem cells have demonstrated that differentiated somatic cells can be returned to the undifferentiated state by reversing their developmental process. These epigenetically reprogrammed somatic cells may again be differentiated into various cell types, and used for cell replacement therapies through autologous transplantation to treat many degenerative diseases. To date, however, reprogramming of somatic cells into undifferentiated cells has been extremely inefficient. Hence, reliable markers to identify the event of reprogramming would assist effective selection of reprogrammed cells. In this study, a transgene construct encoding enhanced green fluorescent protein (EGFP) under the regulation of human Oct4 promoter was developed as a reporter for the reprogramming of somatic cells. Microinjection of the transgene construct into pronuclei of fertilized mouse eggs resulted in the emission of green fluorescence, suggesting that the undifferentiated cytoplasmic environment provided by fertilized eggs induces the expression of EGFP. Next, the transgene construct was introduced into human embryonic fibroblasts, and the nuclei from these cells were transferred into enucleated porcine oocytes. Along with their in vitro development, nuclear transfer embryos emitted green fluorescence, suggesting the reprogramming of donor nuclei in nuclear transfer embryos. The results of the present study demonstrate that expression of the transgene under the regulation of human Oct4 promoter coincides with epigenetic reprogramming, and may be used as a convenient marker that non-invasively reflects reprogramming of somatic cells.

Human Embryo Research and Tort Liability (배아연구와 불법행위책임)

  • Recently, many nations said "yes" to human embryonic stem cell research, signing an executive order to permit funding for the research in the mame of achieving health and life of humankind. Human Embryo Research is permitted by our Bioethics & Biosafety Act. But, illegal research cannot be divorced from civil liability since it requires the destruction of eggs of fertilized eggs and personal rights of embryo-creator. After all, though we allow to do research embryo, we should control the capacity of abuse of embryo research for embryo-creator. If research violate the law(Bioethics & Biosafety Act or Civil Law, etc), it comes to a delict by pecuniary loss and non-pecuniary loss. When it comes to pecuniary loss, Human Embryo is not body but special property. Supreme Court maintained a stance that mental suffering is generally deemed as compensable for damages for the loss of property where a person's property right is invaded by a tort or non-performance of obligation. Thus, where mental suffering occurs, which cannot be compensated by recovery of property losses, the situation must be a special circumstance and the injured could claim consolation money for such losses only if the offender knew or would have known of such special circumstances(Supreme Court Decision 96Da31574 delivered on Nov, 26, 1996, etc.). That is to say, Supreme Court regards mental suffering through person's property right invaded by a tort as damages that have arisen through special circumstances. According to Civil law article 393 (2), the injured could claim consolation money for such losses only if only if the offender had foreseen or could have foreseen such circumstances. Also our court will solve through damages for non-pecuniary loss by complementary function of consolation money in that pecuniary loss could be difficult to valuate.

Human Cases of Fascioliasis in Fujian Province, China

  • Fascioliasis is a foodborne zoonotic parasitic disease. We report 4 cases occurring in the same family, in whom diagnosis of acute fascioliasis was established after series of tests. One case was hospitalized with fever, eosinophilia, and hepatic lesions. MRI showed hypodense changes in both liver lobes. The remaining 3 cases presented with the symptom of stomachache only. Stool analysis was positive for Fasciola eggs in 2 adult patients. The immunological test and molecular identification of eggs were confirmed at the National Institute of Parasitic Diseases, Chinese Center for Disease Control and Prevention, Shanghai, China. The results of serological detection were positive in all the 4 patients. DNA sequencing of PCR products of the eggs demonstrated 100% homology with ITS and cox1 of Fasciola hepatica. The conditions of the patients were not improved by broad-spectrum anti-parasitic drugs until administration of triclabendazole.

Two Cases of Human Infection by Adult of Spirometra erinacei (만손 열두조충 성충에 의한 인체감염 2례)

  • Two cases of human infection by adult Spirometra erinacei were proved by collection of worms in Korea. The patients were 24 and 21-year old males residing in a mountainous village in Kangwon-do. They had the clinical complaints of abdominal pain and epigastric discomfort, revealing the diphyllobothriid eggs, $53~59{\times}37-42{\mu}m$ in size, in their stools. They were treated with 500 mg atabrine and purgated with magnesium salts, and 3~4 hours later several chains of tapeworm segments were recovered from the diarrheal stools. The recovered worms revealed the morphological characteristics of spiral-form or coiled uteri, separated vaginal opening from the cirrus sac, incorporated seminal vesicle into the cirrus sac, distribution of testes at the junction between proglottides, asymmetrical eggs with one pointed eggs, etc. and were identified as Spirometra erinacei (Rudolphi, 1819). The cases had the history of eating raw flesh of the snakes and these are considered the source of infection.

Two Helminthic Cases of Human Mummy Remains from Joseon-Period Graves in Korea

  • Our previous research on coprolite specimens from the mummies of Joseon Dynasty (1392-1910 CE) has revealed various species of parasite eggs. Herein, we added 2 new helminthic cases of human remains from Joseon-period graves in the Republic of Korea (Korea). The organic materials precipitated on the hip bones of 2 half-mummied cases (Goryeong and Gwangmyeong cases) were collected, rehydrated, and examined by a microscope. In the sample from Goryeong-gun (gun=County), ova of Trichuris trichiura, Clonorchis sinensis, and Metagonimus spp. were detected, and eggs of T. trichiura and A. lumbricoides were found from the sample of Gwangmyeong-si (si=City). By adding this outcome to the existing data pool, we confirm our previous estimates of Joseon-period parasite infection rates. The overall rates of A. lumbricoides, T. trichiura, and C. sinensis decreased dramatically from Joseon to the modern period. In Goryeong mummy specimen, we also found Metagonimus spp. eggs that has rarely been detected in archaeological samples so far.

Comparative Morphology of Eggs of Heterophyids and Clonorchis sinensis Causing Human Infections in Korea (한국의 인체기대 이형흡충류 및 간흡충 충란의 비교형태학적 검토)

  • In order to provide some clues for differential diagnosis of trematode infections in fecal examination, the comparative morphology of eggs of 5 kinds of heterophyid flukes (Metagonimus yokogawai, Heterophyes heterophyes nocens, Heterophyopsis continua, Stellantchasmus falcatus and Pygidiopsis summa) and Clonorchis sinensis was studied. The eggs were obtained from distal portion of uteri of worms which were recovered from men after treatment. The characteristic shape and appearance of each kind of eggs were observed in detail under light microscope, and their length and width measured and compared one another. The results are as follows: 1. Eggs of C. sinensis are elongated ovoidal in shape with attenuated anterior end, 25.3~33. 2 (28. 3 in average) ${\mu}m$ long and 14.2~17.4(5.9) ${\mu}m$ wide with length/width ratio of 1.60~2.00 (1.78). They differ from all heterophyid eggs in that they have prominent wrinkling (muskmelon pattern) at their shell surface. 2. P. summa eggs are ovoid to pyriform in shape and characterized by the smallest size of all kinds examined, 19.8~22.9 (21.6) ${\mu}m$ long and 11.1~13.4 (12.1) ${\mu}m$ wide and the ratio 1.63~1.99 (1.78). 3. Eggs of S. falcatus are elongated ovoidal and most slender form, 25.3~29.2 (27.2) ${\mu}m$ long and 11.1~13.4 (12.5) ${\mu}m$ wide with the ratio of 2.00~2.57 (2.17). 4. Eggs of M. yokogawai are ellipsoid to elliptical in shape with round both ends, 26.9~31.6 (28.5) ${\mu}m$ long and 14.2~18.2 (16.8)${\mu}m$ wide with the ratio of 1.48~2.11 (1.70). 5. H. continua eggs are oval in shape, sometimes similar to M. yokogawai or H. h. nocen$ eggs, however, the relative breadth is broadest among all kinds, with maximum width at posterior half portion. They are 23.7~27.7 (25.0) ${\mu}m$ long, 15.8~18.9 (16.4) ${\mu}m$ wide with the ratio of 1. 33~1.75 (1.53). 6. Eggs of H. h. nocens are ellipsoid to ovoid in shape but sometimes more slender than M. yokogawai and have slightly pointed both ends. They are 23.7~29.2 (25.7) p.m long, 14.2~15.8 (15.4) ${\mu}m$ wide, and the ratio 1.50~2.06 (1.67). From the results, it is concluded that eggs, of 5 kinds of heterophyids and C. sinensis can be morphologically differentiated one another, however, careful observation and measurement on sufficient number of eggs are needed.

A Survey of Parasitic Eggs in Soils and Vegetables in a Korean Rural Area (일부농촌지역의 토양 및 채소류에서의 기생충란 조사)

  • The law prohibits use of human excreta without proper treatment since 1968 in Korea, but the rate of infection of parasitic diseases is high among all ages of people. An examination of parasitic eggs in soil and vegetables was performed from April to September, 1974, soil samples were collected near toilet, at gardens, at vegetable farms with excreta used and not used, and strawberry farms in Shindong myon, Chunseong-gun, Kangwon province, and vegetables collected were cabbage, young radish, lettuce, Welshonion, spinach and strawberry. Main findings are as follows 1. Positive samples of Ascaris lumbricoides are 18 out of 25(64%) in the soils near toilet, 9 out of 28(3.2%) in the garden soils and 8 out of 18(44%) in the soils of vegetable farms. 2. Trichocephalus trichiurus and Enterobius vermicularis were detected in 8 soil samples respectively Trichostrongylus orientalis 7(25%) and Ancylostoma duodenale 3(11%) out of in the soils near toilets. 3. Parasitic egg negatives are 62(55%) out of 112 soil samples. 4. Ascaris lumbricoides is found in 11 roots(48%) of cabbage and young radish out of 23 respectively, 14 lettuce(34%) out of 41,4 Welsh onion(40%) out of 10,3 spinach(33%) out of 9, and 2 strawberry(11%) out of 18. 5. Ancylostoma duodenale positives are 2 roots of cabbage(9%) out of 23, 4 young radish (17%) out of 23, and 7 lettuce(17%) out of 41, and negatives of parasitic eggs on vegetables are 62 roots(51%) out of 124 samples.
