  • hCG로 면역화된 생쥐의 비장세포와 골수종양세포(SP 2/0 Ag14)를 융합하여 hCG에 대한 단일클론항체를 생산하는 잡종세포(hybridoma)를 얻었다. 생산된 면역글로부린 type과 titer 그리고 면역분석법 이용시 감도 등을 조사함으로써 그 특성을 조사하였다. 배지나 복수내에 존재하는 항체를 순수분리 정제하기 위하여 gel-filtration, DEAE-ion exchange chromatography, 그리고 affinity chromatography 원리에 의한 방법 등을 사용하여 전기영동(SDS-PAGE)으로 순수도를 조사함으로 상호 비교하였다. 또한 hCG의 정량을 위하여 항체를 plastic tube에 피복시킨 것과 화학발광체로 표지된 항체를 이용한 소위 two-site immunochemiluminometric assay(ICMA)를 개발하여 생산된 항체의 이용가능성을 제시하였다.

Human chorionic gonadotropin therapy in hypogonadic severe-oligozoospermic men and its effect on semen parameters

  • Objective: This study aimed to evaluate whether human chorionic gonadotropin (hCG) therapy is beneficial for improving semen parameters and clinical hypogonadism symptoms in hypogonadic oligozoospermic or severe oligozoospermic men with low or borderline testosterone levels. Methods: A weekly dose of 250 ㎍ (equivalent to approximately 6,500 IU) of hCG was administered subcutaneously for 3-6 months to 56 hypogonadic oligozoospermic or severe oligozoospermic men. Semen, biochemical, and genetic analyses were performed before the start of treatment followed by analyzing semen parameters every 3 months after the start of therapy. We grouped participants into responders and non-responders depending on positive changes in semen parameters. Results: Out of 56 men, 47 (83.93%) responded, while 9 (16.07%) did not. Upon statistical analysis, it was found that age did not affect the overall outcomes (p=0.292); however, men with higher body mass index (BMI; 28.09±3.48 kg/m2) showed better outcomes than those with low BMI (25.33±3.06 kg/m2) (p=0.042). The duration of therapy (in months) was higher in non-responders than in responders (p=0.020). We found significant improvements in sperm concentration (p=0.006) and count (p=0.005) after 3 months of therapy. Sperm motility and progressive motility were also found to be higher in responders, but did not show statistically significant changes. Conclusion: We conclude that hCG therapy can be beneficial in men with hypogonadic oligozoospermia or severe oligozoospermia.

Estrus Behavior and Superovulatory Response in Black Bengal Goats (Capra hircus) Following Administration of Prostaglandin and Gonadotropins

  • The present study was conducted to explore the possibilities of estrus induction and superovulation in a native Indian breed of goats called 'Black Bengal'. Forty-two adult non-pregnant females were divided in two groups, of which 18 goats were subjected to a superovulatory treatment comprising of equine chorionic gonadotropin (eCG), Prostaglandin (PGF2$\alpha$) and human chorionic gonadotropin (hCG) to induce superovulation. The remaining 24 goats received no treatment and served as controls for the parameter under study as well as recipients for embryo transfer studies. The average duration of estrus was found to be significantly increased in treated goats (34.2${\pm}$3.4 h) compared to controls 3.0${\pm}$2.4 h). The average duration between PGF administration and occurrence of estrus was 2.0${\pm}$5.2 h. After mid ventral laparotomy, superovulatory responses indicated a significant increase in the number of follicles, which was 8.27${\pm}$0.37 in the treatment group compared to 4.16${\pm}$0.17 in the control group. The number of corpora lutea was also significantly increased in treated animals compared to control (2.90${\pm}$0.86 vs. 0.74${\pm}$0.04) respectively per ovary per goat.

가토의 수정란이식에 관한 연구 I. PMSG와 HCG투여에 따른 난소반응 (Studies on Embryo Transfer in Rabbit I. Ovarian response to PMSG and hCG administration)

  • As a preliminary experiment to establish the process of embryo transfer in rabbit, present sutdies were carried out with 75 mature Japanese of ovary to pregnant mare's serum gonadotropin(PMSG) and human chorionic gonadotropin(hCG) and collection rate of embryos at various times after hCG injection. Female rabbits were superovulated using 50∼100IU hCG or 75∼100IU PMSG and 50∼751IU hCG injected 83hrs apart. The results obtained were as follows: 1. The average number of growth follicles obtained from all of rabbits treated with hCG or PMSG-hCG was 28.1. PMSG-hCG treatment group (30.9) was clearly increased more than hCG treatment group (16.7). 2. In ovulation score, PMSG-hCG treatment group (21.0) was increased more than hCG treatment group (7.9), showing the same trends in the growth of follicles. 3. The ovulation rate per follicles developed was higher in the rabbits treated with 100 IU PMSG and 75 IU hCG (18.9%) than that from the other groups. 4. The oviduct score (72.9%) was inclined to higher than that from uteri (57.1%) in score of embryo collection.

Development of recombinant human chorionic gonadotropin (hCG) using high-density culture technique of suspension-adapted chinese hamster ovary (CHO) cells

  • Human chorionic gonadotropin (hCG) is a heterodimeric glycoprotein hormone consisting of non-covalently linked two subunits, the ${\alpha}$ and ${\beta}$ subunit. It has been used as a infertility drug for ovulation to mimic luteinizing hormone $(LH).^{1)}$ A stable cell line was established by transfection of Rc/CMV-i-dhfr-hCG, expression vector containing hCG ${\alpha}-$ and ${\beta}-genes$, into dihydrofolate reductase-deficient CHO cells and subesquent methotrexate-mediated gene amplification. Anchorage-dependent CHO cells were adapted into a serum-free and/or animal component-free suspension medium through gradual serum weaning for the hCG production. The established cell line showed typical morphological characteristics and growth profile of CHO cells, and could produce FSH with passage-to-passage consistency. The high density perfusion culture of the CHO cells was carried out in Celligen Plus bioreactor equipped with a spin-filter as a internal cell retention device. The cell density reached up to $>1x10^{7}$ cells/ml in less than 7 days and a perfusion-control strategy based on cellular consumption rates of glucose was $established.^{2)}$ Biologically active recombinant hCG was purified by a series of chromatographic steps including anion exchange chromatography and hydrophobic interaction chromatography to homogeneity. The highly purified recombinant hCG was characterized for physicochemical, immunological and biological properties.

Improving the meiotic competence of small antral follicle-derived porcine oocytes by using dibutyryl-cAMP and melatonin

  • Objective: We increased the nuclear maturation rate of antral follicle derived oocytes by using a pre-in vitro maturation (IVM) culture system and improved the developmental potential of these porcine pathenotes by supplementing with melatonin. Furthermore, we investigated the expression patterns of genes involved in cumulus expansion (HAS2, PTGS2, TNFAIP6, and PTX3) derived from small and medium antral follicles before and after oocyte maturation. Methods: Only the cumulus oocyte-complexes (COCs) derived from small antral follicles were induced with [Pre-SF(+)hCG] or without [Pre-SF(-)hCG] the addition of human chorionic gonadotropin (hCG) during the last 7 h of the pre-IVM period before undergoing the regular culture system. The mature oocytes were investigated on embryonic development after parthenogenetic activation (PA). Melatonin (10-7 M) was supplemented during in vitro culture (IVC) to improve the developmental potential of these porcine pathenotes. Results: A pre-IVM culture system with hCG added during the last 7 h of the pre-IVM period [Pre-SF(+)hCG] effectively supported small antral follicle-derived oocytes and increased their nuclear maturation rate. The oocytes derived from medium antral follicles exhibited the highest nuclear maturation rate in a regular culture system. Compared with oocytes cultured in a regular culture system, those cultured in the pre-IVM culture system exhibited considerable overexpression of HAS2, PTGS2, and TNFAIP6. Porcine embryos treated with melatonin during IVC exhibited markedly improved quality and developmental competence after PA. Notably, melatonin supplementation during the IVM period can reduce and increase the levels of intracellular reactive oxygen species (ROS) and glutathione (GSH), respectively. Conclusion: Our findings indicate that the Pre-SF(+)hCG culture system increases the nuclear maturation rate of small antral follicle-derived oocytes and the expression of genes involved in cumulus expansion. Melatonin supplementation during IVC may improve the quality and increase the blastocyst formation rate of porcine embryos. In addition, it can reduce and increase the levels of ROS and GSH, respectively, in mature oocytes, thus affecting subsequent embryos.

저성선자극호르몬 성선저하증 (Hypogonadotropic Hypogonadism)으로 진단된 남성불임 환자에서 성선자극호르몬 (Gonadotropin) 치료가 정자형성 및 임신에 미치는 영향 (Outcome of Gonadotropin Therapy for Infertile Men with Hypogonadotropic Hypogonadism)

  • 목 적: 저성선자극호르몬 성선저하증은 남성불임의 흔치 않은 원인이다. 저자들은 성선자극호르몬 특히 인간융 모성선자극호르몬 (Human chorioinc gonadotropin: hCG)/인간폐경성선자극호르몬 (Human menopausal gonadotropin: hMG) 치료가 정자형성 및 임신에 미치는 영향에 대해 알아보았다. 연구방법: 2001년 11월부터 2007년 3월까지 불임을 주소로 내원하여 저성선자극호르몬 성선저하증으로 진단되어 성선자극호르몬 (hCG/hMG) 치료를 받은 10명의 진료 기록을 후향적으로 분석하였다. 치료 후 임신 여부를 알아보았으며, 치료 전 고환의 용적에 따라 10 cc 미만인 군 (n=4)과 10 cc 이상인 군 (n=6)으로 나누어 치료 전후의 정액지표와 혈중 FSH, LH 및 testosterone 등의 호르몬 검사를 시행하여 비교하였다. 결 과: 10명의 환자 중 7명 (70%)에서 임신에 성공하였으며 치료 후 혈중 FSH, testosterone 수치가 의미있게 증가하였다. 고환 용적이 큰 군에서 치료 후 정액량, 정자수, 운동성 및 testosterone이 유의하게 증가하였다. 결 론: 불임을 주소로 온 환자에게 흔치는 않지만 면밀한 검사를 통해 저성선자극호르몬 성선저하증을 진단할 수 있어야 하며, hCG/hMG 병합요법은 자연임신 뿐만 아니라 최근의 보조생식술과 연계하여 충분히 성공적인 치료에 도달할 수 있다.

Effects of 17β-estradiol, Interleukin-1β, and Human Chorionic Gonadotropin on Activity and mRNA Expression of Plasminogen Activators in Porcine Endometrial Cells

  • This study aimed to investigate changes in the activity and mRNA expression of plasminogen activators (PAs) induced by $17{\beta}$-estradiol ($E_2$), human chorionic gonadotropin (hCG), and interleukin-$1{\beta}$ ($IL-1{\beta}$) in porcine endometrial cells. Endometrial cells were isolated from the epithelium and cultured to 80% confluence. They were then treated for 24 h with $E_2$ (0.2, 2, 20, and 200 ng/mL), $IL-1{\beta}$ (0.1, 1, 10, and 100 ng/mL), and hCG (0.5, 1, 1.5 and 2 IU/mL). mRNA expressions of urokinase-type (uPA) and tissue-type (tPA) PAs were analyzed using reverse transcription PCR, and activities were measured using a PA activity assay. mRNA expressions of uPA and tPA increased with $E_2$ treatment; however, this was not significant. Similarly, treatment with hCG did not influence the mRNA expressions of PAs. Interestingly, treatment with 0.1 ng/mL $IL-1{\beta}$ significantly reduced the mRNA expression of uPA, but did not affect that of tPA. Treatment with 2, 20, and 200 ng/mL $E_2$ increased PA activity compared with the control group; treatment with 0.1 and 1 ng/mL $IL-1{\beta}$ significantly increased PA activity compared with the other $IL-1{\beta}$ treatment groups, whereas treatment with 10 and 100 ng/mL $IL-1{\beta}$ decreased. Treatment with 2 IU/mL hCG increased PA activity compared with the other treatment groups, although there were no significant differences between the hCG and control groups. In conclusion, the activity and mRNA expression of PAs were differently regulated by the hormone/cytokine and its concentration in porcine endometrial cells. Therefore, understanding PA regulatory mechanisms may help to improve the reproductive potential of domestic animals.

COOH-Terminal Animo Acids of Tethered-Buman Glycoprotein Bormone $\alpha$-Subunit Play an Important Role for Secretion

  • Human chorionic gonadotropin (hCG) is a member of the glycoprotein hormone family which includes FSH. hCG TSH. These hormone family is characterized by a heterodimeric structure composed a common $\alpha$-subunit noncovalently linked to a hormone specific $\beta$-subunit. To determine u and $\beta$ -subunits can be synthesized as a single polypeptide chain (tethered-hCG) and also display biological activity, the tethered-hCC and -FSH molecule by fusing the carboxyl terminus of the hCG $\beta$-subunit to the amino terminus of the $\alpha$-subunit was constructed. To determine the importance of $\alpha$ COOH -terminal amino acid, we also deleted the $\alpha$ COOH-terminal amino acids. The expressing vectors were transfected into CHO-K 1 cells. The tethered-wthCG and -wtFSH was efficiently secreted. The $\alpha$ Δ83hCG and $\alpha$ Δ 83FSH mutants had no secretion. These results are the first conclusive evidence that COOH-terminal amino acids are very important for secretion in human glycoprotein hormone $\alpha$-subunit. These results demonstrated that the $\alpha$ Δ83hCG and $\alpha$ Δ 83FSH mutants could be play a pivotal role in the secretion of tethered-molecule.

Does blastomere biopsy in preimplantation genetic diagnosis affect early serum ${\beta}$-hCG levels?

  • Objective: To determine whether the serum ${\beta}$-human chorionic gonadotropin (hCG) profile following preimplantation genetic diagnosis (PGD) is lower than that of intracytoplasmic sperm injection (ICSI) cycles. Methods: A total of 129 PGD cycles and 1,161 age-matched ICSI cycles, which resulted in pregnancy (serum ${\beta}-hCG{\geq}5$ mIU/mL) on post-ovulation day (POD) 12 were included. We compared the mean serum ${\beta}$-hCG levels on POD 12, 14, 21, and 28, doubling time of serum hCG, and created a cut-off value for predicting a singleton pregnancy in each group. Results: The mean serum ${\beta}$-hCG concentration of the PGD group was significantly lower than that of the control group on POD 12, 14, and 21. The doubling time of serum ${\beta}$-hCG at each time interval showed no significant difference. The cut-off-value of serum ${\beta}$-hCG for predicting a single viable pregnancy was 32.5 mIU/mL on POD 12 and 113.5 mIU/mL on POD 14 for the PGD group, which was lower than that for the control group. Conclusion: Blastomere biopsy may decrease the ${\beta}$-hCG-producing activity of the trophoblasts, especially in early pregnancy. Setting a lower cut-off value of serum ${\beta}$-hCG for predicting pregnancy outcomes in PGD may be needed.
