Platelet Derived Growth Factor-B and Human Epidermal Growth Factor Receptor-2 Polymorphisms in Gall Bladder Cancer

  • Gall bladder cancer (GBC) is a gastro-intestinal cancer with high prevalence among north Indian women. Platelet derived growth factor-B (PDGFB) and human epidermal growth factor receptor-2 (HER2) may play roles in the etiology of GBC through the inflammation-hyperplasia-dysplasia-carcinoma pathway. To study the association of PDGFB and HER2 polymorphisms with risk of GBC, 200 cases and 300 controls were considered. PDGFB +286A>G and +1135A>C polymorphisms were investigated with an amplification refractory mutation system and the HER2 $Ile^{655}Val$ polymorphism by restriction fragment length polymorphism. Significant risk associations for PDGFB +286 GG (OR=5.25) and PDGFB +1135 CC (OR=3.19) genotypes were observed for GBC. Gender wise stratification revealed susceptibility for recessive models of PDGFB +1135A>C (OR=3.00) and HER2 $Ile^{655}Val$ (OR=2.52) polymorphisms among female GBC cases. GBC cases with gall stones were predisposed to homozygous +286 GG and +1135 CC genotypes. Significant risk associations were found for ACIle (OR=1.48), GAVal (OR=1.70), GAIle (OR=2.00) haplotypes with GBC cases and GCIle haplotype with female GBC cases (OR=10.37, P=<0.0001). Pair-wise linkage disequilibrium revealed negative associations among variant alleles. On multi-dimensional reduction analysis, a three factor model revealed significant gene-gene interaction for PDGFB +286A>G, PDGFB +1135A>C and HER2 Ile165Val SNPs with GBC. Protein-protein interaction showed significant association of PDGFB and HER2 with the epidermal growth factor receptor signaling pathway.

  • 저온 플라즈마를 발생시키는 대기압 마이크로-플라즈마 젯(Micro-plasma jet)을 이용하여 플라즈마와 세포와의 상호작용에 대한 연구를 진행하였다. 실험에 사용된 세포는 인체의 방광암 세포(Human bladder carcinoma cell, EJ)이며, 플라즈마 처리 후 bioassay를 통하여 세포 예정사 효과를 확인하였다. 수십 kHz (low frequency)의 펄스파 전압을 인가하여 발생시킨 플라즈마는 형성 기체로 헬륨을 사용하였고, 광 방출 분광법으로 산소의 첨가량에 따른 활성 종들의 변화를 비교해 보았다. 플라즈마 처리 후에는 DAPI staining을 통하여 세포 예정사에서 형성되는 apoptotic body를 확인하였고, 세포막 외부로 이동하는 Phosphatidic Serin (PS)과 결합하는 Annexin-V assay를 통하여 apoptosis rate를 측정하였다. 이를 바탕으로 암세포에 미치는 플라즈마 활성종의 영향을 분석하였다.

The High Expressed Serum Soluble Neural Cell Adhesion Molecule, a High Risk Factor Indicating Hepatic Encephalopathy in Hepatocelular Carcinoma Patients

  • Objective: To investigate whether the expression of serum soluble neural cell adhesion molecule (sNCAM) is associated with hepatic encephalopathy (HE) in hepatocelular carcinoma (HCC) patients. Materials and Methods: The Oncomine Cancer Microarray database was used to determine the clinical relevance of NCAM expression in different kinds of human cancers. Sera from 75 HCC cases enrolled in this study were assessed for expression of sNCAM by enzyme linked immunosorbent assay (ELISA). Results: Dependent on the Oncomine Cancer Microarray database analysis, NCAM was down regulated in 10 different kinds of cancer, like bladder cancer, brain and central nervous system cancer, while up-regulated in lung cancer, uterine corpus leiomyoma and sarcoma, compared to normal groups. Puzzlingly, NCAM expression demonstrated no significant difference between normal and HCC groups. However, we found by quantitative ELISA that the level of sNCAM in sera from HCC patients with HE ($347.4{\pm}151.9ng/ml$) was significantly more up-regulated than that in HCC patients without HE ($260.3{\pm}104.2ng/ml$), the p-value being 0.008. sNCAM may be an important risk factor of HE in HCC patients, the correlation coefficients was 0.278 (P<0.05) on rank correlation analysis. Conclusions: This study highlights that up-regulated level of serum sNCAM is associated with HE in HCC patients and suggests that the high expression can be used as an indicator.

두경부 편평세포암종에서 상피성장인자수용체의 발현과 세포주기에 관한 연구 (EXPRESSION OF THE EPIDERMAL GROWTH FACTOR RECEPTOR AND CELL CYCLE ANALYSIS IN THE HEAD AND NECK SQUAMOUS CELL CARCINOMAS)

  • Growth factors and the receptors play an important role in the regulation of the growth and development of mammalian cells. In particular, epidermal growth factor is a polypeptide with potent mitogenic activity that stimulates proliferation of various normal and neoplastic cells through the interaction with its specific receptor(EGFR). EGFR has been described as a parameter of poor prognosis in many human neoplasms such as breast, bladder, and vulvar cancers. The objectives of this study are the evaluation of the expression of EGFR and cell cycle analysis in the head and neck squamous cell carcinomas(SCC), and the evaluation of the correlation between clinico-patholgic features and expression of EGFR and S-phase fraction. 37 head and neck squamous cell carcinoma specimens were evaluated for expression of EGFR by Western blot analysis and S-phase fraction by cell cycle analysis using the flow cytometry. The obtained results were as follows : 1. The expressions of EGFR were observed in 20 specimens(54%) among 37 head and neck SCC specimens. In case of oral SCC, 15 specimens(56%) out of 27 specimens were observed, and in case of nasopharyngeal SCC 5 specimens(50%) out of 10 specimens. 2. There was no correlation between clinical features(location, stage) of head and neck SCC and expression of EGFR (p>0.05). 3. There was a significant correlation between histo-pathological differentiation of head and neck SCC and expression of EGFR (p<0.02). 4. There was a significant correlation between expression of EGFR and S-phase fraction of cell cycle in the head and neck SCC (p<0.05). The above results suggest that expression of EGFR and S-phase fraction of cell cycle are adjunctive prognostic marker in the head and neck squamous cell carcinomas.

한국인 구강 편평세포암에서 Glutathione S-transferase와 CYP1A1 유전자의 다형성 (GENETIC POLYMORPHISMS OF THE GLUTATHIONE S-TRANSFERASE AND CYP1A1 GENES IN KOREAN ORAL SQUAMOUS CELL CARCINOMA)

  • Many chemical compopunds are converted into reactive electrophilic metabolites by the oxidative(Phase I) enzymes, which are mainly cytochrome P-450 enzyme(CYPs). Phase II conjugating enzymes, such as glutathione S-transferase(GST), usually act as inactivation of enzymes. Genetic polymorphisms have been found to be associated with increased susceptibility to cancer of the lung, bladder, breast and colorectal. Many of the polymorphic genes of carcinogen metabolism show considerably different type of cancer among different ethnic groups as well as individuals within the same group. The aim of this study is (1) to establish the frequencies of genetic polymorphisms of GSTM1 and CYP1A1 in Korean oral squamous cell carcinoma(SCC), (2) to associate oral SCC with the risk of these genetic polymorphisms. The genetic polymorphisms of the GSTM1 and the CYP1A1 genes among 50 Korean oral SCC were analyzed using polymerase chain reaction(PCR). The results suggest that the homozygote and the mutant type of CYP1A1 MspI polymorphisms may be associated with genetic susceptibility to oral SCC in Korean. A combination of the GSTM1 null type with the homozygote(m1/m1), and the mutant(m2/m2) type of CYP1A1 MspI polymorphisms showed a relatively high risk of oral SCC in Korean. In the smoking group, the GSTM1 wild genotype may be the high risk factor of oral SCC in Korean. These data coincide with the hypothesis which states that different susceptibility to cancer of genetic polymorphisms exist among different ethnic group and different types of human cancer.

인간 암세포인 AGS와 T24에서의 apoptosis 유도에 미치는 Bacillus subtilis 혈전용해효소 BK-17의 영향 (Effect of a Fibrinolytic Enzyme (BK-17) from Bacillus subtilis on Apoptosis Induction in AGS and T24 Human Carcinoma Cells)

  • 인간 암세포의 생육에 미치는 혈전용해효소(BK-1)의 영향을 조사하기 위해, 세포증식, 생존력, 형태변화 및 apoptosis 유도 등을 포함한 여러 가지 생화학적 실험을 하였다. 그 결과, AGS 인간 위장 암세포 및 T24 인간 방광 암세포상에의 BK-17 처리는 그 암세포들의 생존력 및 생율을 농도의존적 방법으로 감소시켰다. 현미경 관찰은, BK-17 처리에 의한 항 생육 효과는 막 수축, 세포의 rounding up, apoptotic bodies와 같은 형태학적 변화를 나타내었다. 특히, RT-PCR과 Western blotting data는, BK-17 처리가 항 apoptosis Bcl-2 군들 특히 Bcl-2, and $Bcl-X_L$의 down-regulation 그리고 AGS 세포에서, apoptosis 촉진 매개체 Bax와 Bad의 up-regulation를 유도했다는 것을 보여주었다. BK-17에 의해 유도된 AGS 세포의 apoptosis는 caspase-3, caspase-8 그리고 caspase-9의 단백질가수분해 활성과 관련이 있었다. 이상의 결과를 볼 때, BK-17은 apoptotic cell death의 유도와 밀접한 관련이 있다는 것을 보여주고 있다.

인체 암세포에서 genistein에 의한 cyclooxygenase-2 및 telomerase의 활성 저하 (Genistein-induced Growth Inhibition was Associated with Inhibition of Cyclooxygenase-2 and Telomerase Activity in Human Cancer Cells.)

  • 본 연구에서는 대두의 대표적인 생리활성 물질인 genistein의 처리에 따른 암세포의 증식억제에서 telomerase 및 COX-2 활성의 변화 연관성을 조사하였다. 이를 위하여 4가지 종류의 암세포주를 사용하였으며, genistein 처리에 의하여 암세포들의 증식억제에서 백혈병 세포인 U937 세포의 감수성이 감장 높게 나타났으며, genistein 처리에 따라 telomere 조절인자들의 발현이 대부분 억제되었으며, telomerase의 활성도 매우 유의적으로 감소되었다. 또한 genistein처리 농도가 증가함에 따라 COX-2의 발현이 전사 및 번역 수준에서 모두 감소되었으며 이에 따른 $PGE_2$의 생성 역시 현저하게 감소되었으나, COX-1의 발현에는 큰 변화가 없었다. 이러한 결과들은 genistein의 항암 활성을 이해하는 귀중한 자료로서 활용될 것으로 생각된다.

유방암세포에서 구절초 추출물의 암전이 억제 효과 (Chrysanthemum zawadskii var. latilobum Extracts Inhibits of TPA-induced Invasion by Reducing MMP-9 Expression Via the Suppression of NF-${\kappa}B$ Activation in MCF-7 Human Breast Carcinoma Cells)

  • Chrysanthemum zawadskii Herbich var. latilobum Kitamura (Compositae), colloquially known "Gujulcho" in Korea, has been used in traditional medicine for the treatment of various diseases, including cough, common cold, bladder-related disorders, gastroenteric disorders, hypertension, and inflammatory diseases, such as pneumonia, bronchitis, pharyngitis, and rheumatoid arthritis (RA) However, the effect of Chrysanthemum zawadskii var. latilobum on breast cancer invasion is unknown. In this study, we investigated the inhibitory effect of Chrysanthemum zawadskii var. latilobum extract (CZE) on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced matrix metalloproteinase-9 (MMP-9) expression and cell invasion, as well as the molecular mechanisms involved in MCF-7 cells. CZE were not cytotoxic up to 100 ${\mu}g/ml$ concentration in the MCF-7 cell line. CZE decreased MMP-9 expression. TPA substantially increased NF-${\kappa}B$ DNA binding activity. Pre-treatment with CZE inhibited TPA-stimulated NF-${\kappa}B$ binding activity and NF-${\kappa}B$ related protein expression. To identify invasion ability of MCF-7 cells decreased by CZE, we used martrigel invasion assay. As a result, it is significantly decreased cell invasion. These results indicate that CZE-mediated inhibition of TPA-induced MMP-9 expression and cell invasion involves the suppression of the NF-${\kappa}B$ pathway in MCF-7 cells. Chrysanthemum zawadskii var. latilobum may have potential value in restricting breast cancer metastasis.
