An Analysis on Shapes of the Gaps between Comics frames and Frames of 'The Three Bonds And The Morals in Human Relations' (삼강행실도의 만화칸과 칸 사이 형태 분석)

  • 'The Three Bonds And The Morals in Human Relations' was published in Joseon Dynasty period in 1434, relatively early days of comic history in the world. The gaps between comics frames have symbolic meaning as comics frames have in the area of symbols of comics. After comics frames appeared, it was likely that the advent of the gaps between frames had its own value in the aspect of comics history. Historical backgrounds of domestic comics would be built on firm base led by studying on the symbolic meaning of the gaps between comics frames. Two forms of the gaps between frames were shown in 'The Three Bonds And The Morals in Human Relations.' The first form is the gap between cloud frames. The second is the gap between cloud frame and background frame. Although a little difference is found in the shape and display, two forms of the gaps between frames in 'The Three Bonds And The Morals in Human Relations' have a similarity to modern comics. This study is to demonstrate valid background for the domestic history of comics by means of analyzing the shapes of the gap between comics frames in 'The Three Bonds And The Morals in Human Relations.' The objective of this study is to prove the meaning of the shapes of the gaps between comics frames 'The Three Bonds And The Morals in Human Relations', focusing the gaps between frames among symbols of comics.

Ectopic Overexpression of COTE1 Promotes Cellular Invasion of Hepatocellular Carcinoma

  • Family with sequence similarity 189, member B (FAM189B), alias COTE1, a putative oncogene selected by microarray, for the first time was here found to be significantly up-regulated in hepatocellular carcinoma (HCC) specimens and HCC cell lines. mRNA expression of COTE1 in HCC samples and cell lines was detected by reverse transcription-polymerase chain reaction (RT-PCR) and real-time PCR, while protein expression of COTE1 in HCC tissues was assessed by immunohistochemistry. In addition, invasion of HCC cells was observed after overexpressing or silencing COTE1. In the total of 48 paired HCC specimens, compared with the adjacent non-cancer tissues, the expression of COTE1 was up-regulated in 31 (p<0.01). In HCC cell lines, COTE1 expression was significantly higher than in normal human adult liver (p<0.01). Overexpression of COTE1 enhanced HCC-derived LM6 and MHCC-L cellular invasion in vitro. In contrast, COTE1 knockdown via RNAi markedly suppressed these phenotypes, as documented in LM3 and MHCC-H HCC cells. Mechanistic analyses indicated that COTE1 could physically associate with WW domain oxidoreductase (WWOX), a tumor suppressor. COTE1 may be closely correlated with invasion of hepatocellular carcinoma (HCC) cells and thus may serve as an effective target for gene therapy.

Computational Identification and Comparative Analysis of Secreted and Transmembrane Proteins in Six Burkholderia Species

  • As a step towards discovering novel pathogenesis-related proteins, we performed a genome scale computational identification and characterization of secreted and transmembrane (TM) proteins, which are mainly responsible for bacteria-host interactions and interactions with other bacteria, in the genomes of six representative Burkholderia species. The species comprised plant pathogens (B. glumae BGR1, B. gladioli BSR3), human pathogens (B. pseudomallei K96243, B. cepacia LO6), and plant-growth promoting endophytes (Burkholderia sp. KJ006, B. phytofirmans PsJN). The proportions of putative classically secreted proteins (CSPs) and TM proteins among the species were relatively high, up to approximately 20%. Lower proportions of putative type 3 non-classically secreted proteins (T3NCSPs) (~10%) and unclassified non-classically secreted proteins (NCSPs) (~5%) were observed. The numbers of TM proteins among the three clusters (plant pathogens, human pathogens, and endophytes) were different, while the distribution of these proteins according to the number of TM domains was conserved in which TM proteins possessing 1, 2, 4, or 12 TM domains were the dominant groups in all species. In addition, we observed conservation in the protein size distribution of the secreted protein groups among the species. There were species-specific differences in the functional characteristics of these proteins in the various groups of CSPs, T3NCSPs, and unclassified NCSPs. Furthermore, we assigned the complete sets of the conserved and unique NCSP candidates of the collected Burkholderia species using sequence similarity searching. This study could provide new insights into the relationship among plant-pathogenic, humanpathogenic, and endophytic bacteria.

A Review Study on Movement System Impairment Syndromes (동태손상(動態損傷) 증후군(症候群)에 관한 연구 현황 고찰)

  • Objectives: Sahrmann, an American physical therapist, has developed a set of movement-related diagnoses on musculoskeletal pain syndromes (MPS). We reviewed articles for studies of objectification and quantification of Oriental medicine diagnosis system about MPS. Methods: The authors reviewed a total of 12 studies found on PubMed to obtain movement system impairment (MSI)-based classification categories. Results: The MSI system has been developed for objectification and quantification of physical therapists' MPS diagnoses. The MSI system of classification is based on the basic premise that loss of precise movement is the result of specific repetitive movements and positions in everyday life. The MSI system defines 8 categories in the shoulder region, 5 in the lumbar region, and 7 in the knee region. Treatment involves (1) educating the person about the specific directions of alignment and movement that appear to be contributing to the musculoskeletal problem, (2) modifying the direction - specific alignment and movement patterns during daily activities, and (3) exercises to address the impairments. Conclusions: The authors propose that MSI studies could help to advance an Oriental medical diagnosis system on musculoskeletal pain syndromes, because MSI shares similarity with Oriental medicine in terms of holism, views of constant motion, and mi-byung (subhealth) treatment.

Human Sensibility Ergonomics Makeup Recommendation System using Context Sensor Information (상황 센서정보를 이용한 감성공학적 메이크업 추천 시스템)

  • It is important for the strategy of cosmetic sales to investigate the sensibility and the preference degree in the environment that the makeup style has been changed focusing on the consumer center. We proposed the human sensibility ergonomics makeup recommendation system (MakeupRS) using the context sensor information applying the collaborative filtering technique as one of methods in the makeup style development centered on the consumer's sensibility and the preference. In the collaborative filtering technique, the Pearson correlation coefficient applying to the case amplification is used to calculate similarity weights between the users. To investigate the sensibility according to the effect of makeup styles, the makeup styles were analyzed in terms of 6 style factors, such as, the foundation, the color lens, the eye shadow, the eye lash, the cheek brusher, and the lipstick. Ultimately, this paper suggests empirical application to verify the adequacy and the validity with the human sensibility ergonomics makeup recommendation system.

Cloning and Expression of $\beta$-Glucuronidase from Lactobacillus brevis in E. coli and Application in Bioconversion of Baicalin and Wogonoside

  • The $\beta$-glucuronidase (GUS) gene from Lactobacillus brevis RO1 was cloned and expressed in Escherichia coli GMS407. The GUS gene was composed of 1,812 bp, encoding a 603-amino-acid protein belonging to glycosyl hydrolase family 2 with three conserved domains. The amino acid similarity was higher than 70% with the $\beta$-glucuronidases of various microorganisms, yet less than 58% with the $\beta$-glucuronidase of L. gasseri ADH. Overexpression and purification of the GUS was performed in $\beta$-glucuronidase-deficient E. coli GMS407. The purified GUS protein was 71 kDa and showed 1,284 U/mg of specific activity at optimum conditions of pH 5.0 and $37^{\circ}C$. At $37^{\circ}C$, the GUS remained stable for 80 min at pH values ranging from 5.0 to 8.0. The purified enzyme exhibited a half-life of 1 h at $60^{\circ}C$ and more than 2 h at $50^{\circ}C$. When the purified GUS was applied to transform baicalin and wogonoside into their corresponding aglycones, $150\;{\mu}M$ of baicalin and $125\;{\mu}M$ of wogonoside were completely transformed into baicalein and wogonin, respectively, within 3 h.

Exopolysaccharide Produced by Pediococcus acidilactici M76 Isolated from the Korean Traditional Rice Wine, Makgeolli

  • This work is aimed to increase knowledge of the functional exopolysaccharide (EPS) from lactic acid bacteria (LAB) in makgeolli, a Korean fermented rice wine. Among LAB strains isolated from makgeolli, strain M76 was selected as a functional strain producing a bioactive EPS, based on its antioxidative activity on the DPPH radical. The 16S rRNA gene sequencing analysis showed a high sequence similarity (99.0%) with P. acidilactici, but had different biochemical properties with the already known P. acidilactici type strains in the aspect of carbohydrates utilization. The obtained P. acidilactici M76 produced a soluble EPS above 2 g/l. One-step chromatography using gel filtration after ethanol precipitation from the supernatant of P. acidilactici M76 was enough to obtain purified EPS with a single peak, showing a molecular mass of approximately 67 kDa. Componential and structural analyses of EPS by TLC, HPLC, and FT-IR indicated that the EPS is a glucan, consisting of glucose units. The purified EPS had antioxidant activity on the DPPH radical of 45.8% at a concentration of 1 mg/ml. The purified EPS also showed proliferative effect on the pancreatic RIN-m5F cell line and remarkable protection activity on alloxan-induced cytotoxicity. This potent antioxidant and antidiabetic EPS by LAB in makgeolli may contribute to understanding the functionality of makgeolli.

Antifungal Activity of Bacillus sp. BCNU 2002 against the Human Pathogens (인체 병원성 진균에 대한 Bacillus sp. BCNU 2002의 항진균 효과)

  • An endospore-forming, rod-shaped bacterium was isolated from forest soil samples collected at the Taebaek mountain of Gangwon province, Korea, and taxonomically characterized by physiological, biochemical and phylogenetic methods. Its 16S rRNA sequences showed the maximum similarity of 97% with B. amyloliquefaciens. In addition, the isolate BCNU 2002 was determined to have the ability to produce enzymes such as amylase, protease, gelatinase and catalase. The in vitro antifungal activity of Bacillus sp. BCNU 2002 was also examined against human pathogenic fungi such as Aspergillus niger, Candida albicans, Epidermophyton floccosum, Saccharomyces cerevisiae, Trichophyton mentagrophytes and Trichophyton rubrum. A maximum production level of antifungal substances of Bacillus sp. BCNU 2002 was achieved under aerobic incubation at $28^{\circ}C$ for 7 days in LB broth. BCNU 2002 showed strong antifungal activities against T. mentagrophytes and T. rubrum with the range of percentage inhibition from 56.25 to 63.23%. It was also confirmed that ethylacetate extract of cultured broth showed a strong antifungal activity against A. niger, C. albicans, S. cerevisiae and T. rubrum by agar diffusion method. The peptide fraction also exhibited broad antifungal spectrum against various pathogenic fungi. The minimum inhibitory concentration values for active extracts ranged between 125 ${\mu}g$/mL and 1000 ${\mu}g$/mL.

A Wavelet-Based Watermarking Scheme of Digital Image Using ROI Method (ROI를 이용한 웨이브렛 기반 디지털 영상의 워터마킹 기법)

  • General watermarking techniques tend not to consider intrinsic characteristics of image, so that watermarks are embeded to entire images. In this paper, we present a watermarking algorithm based on wavelet domain, and the watermark is embedded into large coefficients in region of interest(ROI) being based on principle of multi-threshold watermark coding(MTWC) for robust watermark insertion. We try to accomplish both image duality and robustness using human visual system(HVS). The watermarks are embedded in middle frequency bands because the distortion degree of watermarked images appears to be less than lower frequency bands, and the embedded watermarks in the middle bands showed high extraction ratios after some distortion. The watermarks are consisted of pseudo random sequences and detected using Cox's similarity mesurement.

Establishment of Highly Tumorigenic Human Gastric Carcinoma Cell Lines from Xenograft Tumors in Mice

  • Patient's primary tumor-derived tumor cell lines likely represent ideal tools for human tumor biology in vitro and in vivo. Here, we describe eight human gastric carcinoma cell lines derived from established tumors in vivo upon subcutaneous transplantation of primary gastric carcinoma specimens in BALB/c nude mice. These xenografted gastric tumor cell lines (GTX) displayed close similarity with primary gastric tumor tissues in their in vivo growth pattern and genomic alterations. GTX-085 cells were resistant to cisplatin, while GTX-087 was the most sensitive cell line. GTX-085 was the only cell line showing a metastatic potential. Epithelial cell adhesion molecule (EPCAM) expression was especially strong in all tissue samples, as well as in cell cultures. GTX-139, the largest tumor graft obtained after injection, displayed distinct expression of CD44v6, fibroblast growth factor receptor 2 (FGFR2), and prominin 1 (PROM1, also known as CD133). In summary, we established eight xenograft gastric cancer cell lines from gastric cancer patient tissues, with their histological and molecular features consistent with those of the primary tumors. The established GTX cell lines will enable future studies of their responses to various treatments for gastric cancer.
