이 연구는 우리나라 공공도서관 어린이 서비스의 현재 수준을 조망해 보기 위하여 전국의 공공도서관을 대상으로 현황을 조사하여 분석하였다. 설문조사를 통해 전국 공공도서관 중 171개관을 대상으로 분석하였으며, 도서관의 소재 지역과 예산 규모에 따라 영역을 구분하여 살펴보았다. 어린이 서비스를 시설 및 공간, 자료, 인력, 이용자봉사, 운영 일반 및 강좌 5개 영역으로 나누어 다양한 측면들을 조사하고 분석하였다. 분석결과 어린이 서비스의 수준과 내용에서 도서관 소재 지역과 규모에 따라 다양한 편차가 나타나는 것을 알 수 있으며, 가장 주목할 만한 것은 전체적으로 공공도서관 서비스에서 어린이 서비스가 차지하는 비중에 비해 투여되는 자원의 규모는 상대적으로 낮은 수준에 놓여 있다는 사실이다.

This paper tries to a n.0, pply the concept and process of marketing which have been developed in profit sector to library management. Since the end of 60's certain marketing researchers, among them Kotler, Levy, and Shapiro have advanced the theseses that marketing is not just a business organization as well. Recently libraries have been interested in markeing also. Marketing is a concept of sensitively serving and satisfying human needs through voluntary exchanges of value. Library is a value exchange system in which library service is exchanged with community's patronage. In order for library user to involve in the value exchange system voluntarily, library should analyze user's needs and offer products satisfying the needs. For doing this, library should understand marketing. In this paper, author introduces the marketing concepts and process, tries to show how to a n.0, pply the key concepts and process to public library management. The needs of marketing in library sector, the effectiveness and barriers in a n.0, pplying marketing to library also discussed.

본 연구는 정보전문가의 교수활동 지원을 위한 참고정보서비스 실태를 사서교사의 참고정보서비스 참여를 중심으로 분석하기 위해 수행되었다. 조사방법은 전국사서교사협의회 회원교(212개)와 전국에서 계약직 사서가 가장 많이 배치된 경기도의 2개 도시 학교도서관(115개) 사서전수(327명)를 대상으로 2006년 12월 1일부터 12월 31일까지 31일간 3차에 걸쳐 데이터를 수집하였으며, 총 254개(회수율 77.7%)의 질문지가 분석에 사용되었다. 그 결과 사서교사와 사서들은 독립변인(성별, 학교급. 근무경력, 고용구분)에 따라 교수활동지원을 위한 참고정보서비스 참여도에 차이가 있었다. 이에 본 연구에서는 이 같은 문제를 해결하는 방안으로 문헌정보학 교과과정과 직무연수 등을 통해 교수활동 지원을 위한 참고정보서비스 교육을 강화할 것과 교수활동에 사서교사와 사서가 참여하여 참고정보서비스를 강화할 수 있는 제도적 장치를 마련할 것 등을 제언하였다.

A human bacterial artificial chromosome (BAC) library was constructed with high molecular weight DNA extracted from the blood of a male Korean. This Korean BAC library contains 100,224 clones of insert size ranging from 70 to 150 kb, with an average size of 86 kb, corresponding to a 2.9-fold redundancy of the genome. The average insert size was determined from 288 randomly selected BAC clones that were well distributed among all the chromosomes. We developed a pooling system and three-step PCR screen for the Korean BAC library to isolate desired BAC clones, and we confirmed its utility using primer pairs designed for one of the clones. The Korean BAC library and screening pools will allow PCR-based screening of the Korean genome for any gene of interest. We also determined the allele types of HLA-DRA and HLA-DRB3 of clone KB55453, located in the HLA class II region on chromosome 6p21.3. The HLA-DRA and DRB3 genes in this clone were identified as the DRA*010202 and DRB3*01010201 types, respectively. The haplotype found in this library will provide useful information in future human disease studies.

디지털 도서관 담론이 학교도서관 분야에도 예외 없이 형성되고 있다. 이런 상황에서 현재의 디지털 학교도서관 담론이 가지고 있는 몇 가지 맥락적 오류를 검토하여 그 논리적 모순을 지적하고 나아가 우리 학교도서관이 온전히 그 기능을 회복하는 길을 그 본질적 원리에 천착하여 탐색한다.

Nebulin is a (Mr 600∼900 kDa) large actin-binding protein specific to skeletal muscle and thought to act as a molecular template that regulates the length of thin filaments. Cardiac muscles of higher vertebrates have been shown earlier to lack nebulin. Recently, full-length nebulin mRNA transcripts have been detected in heart muscle, but at lower levels than in skeletal muscle. Nebulin expression also was detected in the kidney, eye, and otic canal, suggesting that nebulin isoforms may also be expressed in these organs. We have searched for nebulin isoforms in brain of human using PCR and Northern blot. Here, we provide evidence that nebulin mRNA transcripts are expressed in brain. Seven nebulin isoforms (B, C, D, E, F, G and H form) are obtained in human skeletal muscle and four isoforms (B, C, G and H form) in human brain cDNA library. We cloned the 1.3 kb of nebulin fragment from human adult brain library by PCR. The identity of the PCR product was confirmed by sequence analysis. The partial brain nebulin sequence was 99% identical to the skeletal muscle cDNA as determined by Blast alignment. It contains two simple-repeats HR1, HR2 and linker-repeats exon l35∼143 except exon 140. It was different from skeletal muscle B form, which contain HR1 and HR8. These data suggest that nebulin isoform diversity occurs even more extensively than previously known, likely contributing to the distinct thin filament architecture of different striated muscles.

사람 성장호르몬 수용체(hGHR) cDNA는 PCR방법에 의하여 fagment로서 보고되어진 바 있으나, liver cDNA로 부터 전장을 cloning한 보고는 없는 실정으로 본 연구에서는 기능을 가진 약 4.6kbp의 cDNA hGHR을 cloning 하는데 성공하였다. 먼저 cloning하기 위하여 human liver mRNA와 human breast cancer tissue로부터 회수한 mRNA를 RT-PCR방법에 의하여 human cDNA library와 cloning에 필요한 probe를 제작하였다. human library mRNA는 GT-PCR방법에 의하여 증폭하여 증폭되어진 산물은 λZAP Vector를 이용하여 cDNA library를 구축하였고,screeing을 위하여 임 보고 되어진 hGHR fragment native sequence를 기초로 N-terminal부분의 primer를 설계하여 950bp의 probe를 얻는데 성공하였다. 이 probe를 이용하여 준비된 human liver cDNA library로부터 2.5$\times$10 6개의 plaque로부터 6개의 positive clone을 획득하였고, 이들중 poly Asignal인 "AATAAA"를 포함하고 있는 가장 긴 약 3.8kbp의 clone을 sequencing한 결과 open reading frame을 포함하고 있었으나, 5'부분의 결손되어 있었다. 그리하여 이 부분은 human breast cancer tissue로 부터 회수한 mRNA를 RT-PCR에 의하여 증폭하였고, sequencing결과 이미 보고되어진 native hGHR와 비교한 결과 하나의 nucleotide가 silent mutation으로 판명되었다.한편 human liver cDNA library로부터 cloning한 3.8cp의 positive clone의 5'end의 결손된 부분에 silent mutation된 PCR 산물을 연결함으로써 native hGHR와 유사한 cDNA hGHR subcloning에 성공하였다. 이러한 cDNA hGHR의 clone이 function을 가지고 있는지를 검토하기 위하여 eukaryotic 발현 vector인 pCXN2에 의거 ligation한 후 chinese hamster ovary cell[CHO-KI]에 transfect를 실시하였다. Dexamethasone은 첨가하지 않고 hGH만의 존재하에서 이들 cell을 배양시키고 cell menbrane에서 발현 여부를 판정키 위하여 hGHR monocloual antibody를 사용하여 flow cytometery해석을 실시하는 한편 125I-hGH binding assay에 의하여 hGH binding activity를 측정하였다. 최종적으로 GH signal transduction의 target genedf으로 알려져 있는 serine protease inhibitor 2.1(Spi 2.1) gene의 promotor activity를 검토한 결과 hGHR을 transfect한 CHO Cell에 있어서 hGH의 농도에 의존적으로 증가되었다. 따라서 본 실험에서 cloning한 cDNA hGHR는 native hGHR와 같은 기능을 가지는 것으로 판명되었다.것으로 판명되었다.

Antibody phage display provides a powerful and efficient tool for the discovery and development of monoclonal antibodies for therapeutic and other applications. Antibody clones from synthetic libraries with optimized design features have several distinct advantages that include high stability, high levels of expression, and ease of downstream optimization and engineering. In this study, a fully synthetic human scFv library with six diversified CDRs was constructed by polymerase chain reaction assembly of overlapping oligonucleotides. In order to maximize the functional diversity of the library, a ${\beta}$-lactamase selection strategy was employed in which the assembled scFv gene repertoire was fused to the 5'-end of the ${\beta}$-lactamase gene, and in-frame scFv clones were enriched by carbenicillin selection. A final library with an estimated total diversity of $7.6{\times}10^9$, greater than 70% functional diversity, and diversification of all six CDRs was obtained after insertion of fully randomized CDR-H3 sequences into this proofread repertoire. The performance of the library was validated using a number of target antigens, against which multiple unique scFv sequences with dissociation constants in the nanomolar range were isolated.

본 연구에서는 도서관에 대한 전문적인 지식과 공간구성에 대한 기획, 디자인 및 건축에 대한 기본 지식을 갖춘 인재 양성을 위해, 자기설계전공 기반의 도서관공간기획전공을 개발하고 이에 대한 교육목표, 인재상, 전공역량 등을 제시하고자 하였다. 이를 위해 관련 문헌분석, 전문가 의견수렴, 그리고 수요자 설문조사를 수행하였으며, 그 결과는 다음과 같다. 첫째, 도서관공간기획전공에 요구되는 인재상은 공간기획인재, 디자인적사고인재, 창의적실무인재로 파악되었다. 둘째, 인재상별 전공역량은 도서관환경 분석 능력, 공간콘셉트도출 능력, 이용자 요구 분석능력, 이해관계 조정 능력, 공간구성능력, 공간디자인 이해 능력, 실무 적용 능력으로 총 7개의 역량을 도출하였다. 이를 기반으로 향후 도서관공간기획전공의 교육목표, 인재상, 인재상에 부합한 전공역량을 실현할 수 있는 교과목이 개발되어야 하며, 도서관공간기획전공이 자기설계 기반의 전공으로 학생들이 실패하지 않고 적응할 수 있도록 이에 대한 운영 방안이 설계되어야 할 것으로 보인다.

본 연구는 도서관을 움직이는 핵심요소인 시설 (설치에 대한 법률적 근거), 경영(전담부서), 인적자원, 예산을 중심으로 교육청 소속 도서관의 미래 발전을 위한 정책적 과제를 점검하고 이에 대한 해결책을 제시하고자 한다. 본 연구를 위해서 국가법령정보센터와 국가도서관통계시스템을 활용하여 관련 자료를 수집 분석하였다. 연구 결과, 교육청 소속 도서관의 행정기구가 달라서 도서관기능을 수행하는 기관의 명칭이 다양하게 나타나며, 전담부서의 도서관업무는 보조업무로서 정책의 우선순위에서 배제될 가능성이 있는 것으로 판단되어 이에 대한 개선이 절실하다. 또한 소수직렬 승진기회를 제공하기 위한 사무관의 총 정원의 확대와 전체 예산대비 도서관 투입예산의 증가가 반드시 필요한 것으로 분석되었다.
