Placenta is a special organ that contains many nutrients such as growth factors, minerals, and bioactive peptides. Dipeptides of glycine and leucine are major components of porcine placenta extracts (PPE) that has been used as an alternative of human placenta extracts. In this study, we investigated whether major peptides of PPE, Glycyl-L-Leucine (Gly-Leu), L-Leucyl-Glycine (Leu-Gly), and L-Leucyl-L-Leucine (Leu-Leu), affect skin hydration and elasticity in vitro and in vivo. We found that Gly-Leu and Leu-Gly dipeptides induced the expression of transglutaminase 1 in normal human epidermal keratinocytes (NHEKs) whereas Leu-Leu dipeptides did not. Treatment with Gly-Leu or Leu-Gly significantly increased hyaluronan (HA) synthesis in NHEKs and the upregulation of hyaluronan synthase 2 (HAS2) mRNA level was confirmed. In addition, elastase activity was inhibited in NHEKs treated with Gly-Leu or Leu-Gly dipeptides. Oral administration of Gly-Leu or Leu-Gly dipeptides increased skin hydration and elasticity in UVB-irradiated hairless mice. The significant upregulation of HA in UVB-irradiated hairless mice was observed in response to oral administration of Gly-Leu or Leu-Gly. These results suggest that the major dipeptides of porcine placenta, Gly-Leu and Leu-Gly, are potentially active ingredients for skin moisturization formulations.
Background: Cancer-associated fibroblasts (CAFs) are abundant in tumor microenvironments and interact with cancer cells to promote tumor proliferation in oral squamous cell carcinoma (OSCC). Cathepsin D (CTSD) is a soluble lysosomal aspartic endopeptidase involved in tumor proliferation and angiogenesis. In this preliminary study, we observed CTSD expression in OSCC and CAFs, postulating that CTSD might act as a bridge between OSCC and CAFs. Methods: Human epidermal keratinocytes (HEKs), OSCC, and immortalized human normal oral fibroblasts (hTERT-hNOFs) were used in this study. Additionally, we used hTERT-hNOFs transfected with an empty vector, WT (wild-type)-YAP (Yes-associated protein), and YAPS127A (YAP serine 127 to alanine). YAP127A hTERT-hNOFs activated fibroblasts similar to CAFs. To identify CTSD expression between OSCC and CAFs, conditioned medium (CM) was collected from each cell. Protein expression of CTSD was identified by western blotting. Results: To identify the expression of CTSD in fibroblasts stimulated by OSCC, we treated fibroblasts with CM from HEK and OSCC. Results indicated that hTERT-hNOFs with OSCC CM showed a weakly increased expression of CTSD compared to stimulation by HEK CM. This indicates that CAFs, YAPS127 hTRET-hNOFs, overexpress CTSD protein. HEK cells showed no CTSD expression, regardless of treatment with fibroblast CM, whereas OSCC highly expressed CTSD proteins compared with the CTSD expression in HEK cells. We also found that CTSD expression was unaffected by changes in transforming growth factor-β levels. Conclusion: This study proposes that CTSD might have potential as an interacting executor between OSCC and CAFs. Further studies are needed to investigate the role of CTSD in tumor and stromal cells.
진피 상층부 모세혈관은 젊은 사람에 비해 노인에서 그 수와 크기가 감소되어 있어 피부에서 모세혈관이 차지하는 비율이 나이에 따라 급격히 낮아진다. 연교는 물푸레나무과에 속하는 개나리 열매를 건조한 것으로 주로 염증성 또는 항균성 질환에 오랫동안 사용되어져온 약재로서 지금까지 피부 혈관신생과 관련된 효능은 보고되지 않았다. 따라서 본 연구에서는 연교 추출물이 혈관신생과 관련된 인자들에 미치는 영향을 피부 각질형성 세포주를 이용하여 조사하고자 하였다. 우리는 먼저 연교 추출물이 혈관신생과 관련된 인자들의 발현에 어떤 영향을 주는지 알아보고자 각질형성세포에 연교 추출물을 처리하고 혈관신생과 관련된 55개 단백질의 발현을 분석하였다. 발현 변화를 보인 인자들 중 혈관내피세포 성장인자(VEGF, vascular endothelial growth factor)는 강력한 혈관신생 촉진인자로서 연교 추출물에 의해 유의하게 발현이 증가되었다. 따라서 연교 추출물이 VEGF 생성에 미치는 영향에 대해 자세히 알아보고자 연교 추출물을 농도별로 세포에 처리하고 단백질 발현과 mRNA 발현 변화를 조사한 결과, 연교 추출물은 VEGF의 유전자 수준뿐만 아니라 단백질 수준의 발현을 2배이상 농도 의존적으로 증가시켰다. 다음으로 연교 추출물에 의한 VEGF 발현 증가에 관여하는 신호전달 기전을 밝히고자 MAPK 활성을 살펴본 결과, 연교 추출물을 세포에 처리하면 5 min 내 p38 MAPK의 활성이 관찰되었으며, 특이적 억제제 전처리를 통해 p38 MAPK 활성을 억제하면 연교 추출물을 처리하더라도 VEGF의 유전자 및 단백질 발현이 완전히 억제됨을 확인하였다. 이러한 결과로부터 연교는 피부 각질형성세포에 작용하여 p38 MAPK 활성을 통해 VEGF 생성을 유도함을 알 수 있었고, 피부에서 노화에 따른 표피아래 모세혈관 손상을 개선하는데 도움을 줄 수 있는 후보 물질로서 연교의 새로운 효능을 제안한다.
Chronic exposure to ultraviolet (UV) radiation, regarded as a major cause of extrinsic aging or photoaging characterized by wrinkle formation and skin dehydration, exerts adverse effects on skin by causing the overproduction of reactive oxygen species. Agastache rugosa Kuntze, known as Korean mint, possesses a wide spectrum of biological properties including anti-oxidation, anti-inflammation, and anti-atherosclerosis. Previous studies have reported that A. rugosa protected human keratinocytes against UVB irradiation by restoring the anti-oxidant defense system. However, the anti-photoaging effect of A. rugosa extract (ARE) in animal models has not yet been evaluated. ARE was orally administered to hairless mice at doses of 100 or 250 mg/kg/day along with UVB exposure for 12 weeks. ARE histologically improved UVB-induced wrinkle formation, epidermal thickening, erythema, and hyperpigmentation. In addition, ARE recovered skin moisture by improving skin hydration and transepidermal water loss (TEWL). Along with this, ARE increased hyaluronic acid levels by upregulating HA synthase genes. ARE markedly increased the density of collagen and the amounts of hydroxypoline via two pathways. First, ARE significantly downregulated the mRNA expression of matrix metalloproteinases responsible for collagen degradation by inactivating the mitogen-activated protein kinase/activator protein 1 pathway. Second, ARE stimulated the transforming growth factor beta/Smad signaling, consequently raising the mRNA levels of collagen-related genes. In addition, ARE not only increased the mRNA expression of anti-oxidant enzymes but also decreased inflammatory cytokines by blocking the protein expression of nuclear factor kappa B. Collectively, our findings suggest that A. rugosa may be a potential preventive and therapeutic agent for photoaging.

Ginsenosides are active constituents of ginseng (Panax ginseng) that have possible anti-aging, physiological and pharmacological activities, such as anti-cancer and anti-inflammatory effects. Although the ginseng root is generally used more often than the aerial parts for medicinal purposes, the flowers also contain numerous ginsenosides, including Rb2, Rc, Rd, Re and Rg1. Therefore, an extract from the flowers of the P. ginseng could have the pharmacological efficacy of bioactive compounds including ginsenosides. The high hydrostatic pressure extraction (HHPE) is a method that is used for the efficient extraction of bioactive compounds from plant materials. In this study, we compared the yield of ginsenosides from ginseng flowers under different conditions of extraction pressure and time of HHPE. The results indicate that the total yield of the ginsenosides improved as the pressure increased from 0.1 to 80 MPa and treatment duration increased to 24 hours. In addition, the ginsenoside extracts from HHPE at 80 MPa, which possessed a higher total ginsenoside concentration, decreased the viability of the primary human epidermal keratinocytes (HEKs) significantly than the ginsenoside extracts from HHPE at 0.1 MPa. Collectively, we found that the method of HHPE that was performed for 24 hours at 80 MPa showed the highest yield of ginsenosides from the flowers of P. ginseng. In addition, our study provides a foundation for the efficient extraction of ginsenosides, which had a potent bioactivity, from flowers of P. ginseng through HHPE.
본 연구에서는 기능성 미백물질인 나이아신아마이드(NI)의 피부 투과율을 개선시키기 위해 NI를 고형지질 나노입자(solid lipid nanoparticles, SLNs))에 봉입하고 피부 투과율을 평가하였다. NI는 이중 에멀션 가온용융 유화법으로 SLNs 내에 효과적으로 봉입할 수 있었으며, 평균 입자 크기는 263.30 ~ 436.93 nm이었고 제타전위는 -34.77 ~ -57.60 mV인 안정한 입자를 제조하였다. NI의 피부 투과 연구를 위해 인체 표피 조직에서 유래된 피부각질세포로 만든 3차원적 인공피부(SkinEthicTM RHE)을 사용하였다. NI의 피부 투과 및 침적 실험 결과 모든 SLNs 제형이 NI의 피부 투과 및 침적율을 개선시켰으며, SLNs을 적용하였을 때가 그렇지 않을 때 보다 투과율은 약 5.4 ~ 7.6 배, 침적율은 9.5 ~ 20.8 배 개선되었을을 확인하였다. 따라서, 본 연구에서 제조된 고형지질나노입자는 기능성 미백물질인 나이아신아마이드의 피부 투과율을 개선하기에 충분한 결과를 보여 주었다.
The skin, the largest organ in the body, undergoes age-related changes influenced by both intrinsic and extrinsic factors. The primary external factor is photoaging which causes hyperpigmentation, uneven skin surface, deep wrinkles, and markedly enlarged capillaries. In the human dermis, it decreases fibroblast function, resulting in a lack of collagen structure and also decreases keratinocyte function, which compromises the strength of the protective barrier. In this study, we found that treatment with γ-aminobutyric acid (GABA) had no toxicity to skin fibroblasts and GABA enhanced their migration ability, which can accelerate skin wound healing. UVB radiation was found to significantly induce the production of matrix metalloproteinase 1 (MMP-1), but treatment with GABA resulted in the inhibition of MMP-1 production. We also investigated the enhancement of filaggrin and aquaporin 3 in keratinocytes after treatment with GABA, showing that GABA can effectively improve skin moisturization. In vivo experiments showed that oral administration of GABA significantly improved skin wrinkles and epidermal thickness. After the intake of GABA, there was a significant decrease observed in the increase of skin thickness measured by calipers and erythema. Additionally, the decrease in skin moisture and elasticity in hairless mice exposed to UVB radiation was also significantly restored. Overall, this study demonstrates the potential of GABA as functional food material for improving skin aging and moisturizing.
본 연구는 인간 각질형성 세포주를 이용하여 피부 표면의 산성화를 조절하는 물질을 발굴하고, 이를 통해 각질층의 보습 능력 및 피부 장벽 기능에 미치는 영향을 조사하기 위해 수행되었다. 꿀풀은 아프리카 북서부 및 북미에 널리 분포하는 허브 중 하나로 세포사멸, 항산화 및 항염에 대한 효능이 연구되어 왔다. 하지만 NHE1의 발현 조절 및 피부 장벽기능 회복에 대한 연구는 진행되지 않았다. 꿀풀의 피부 pH 조절 효과를 확인하기 위하여 꿀풀의 활성 성분 분석 결과 로즈마린산과 카페인산이 검출되었다. 꿀풀과 이것으로부터 검출된 성분인 카페인산은 인간 각질형성 세포주인 HaCaT 세포에 고농도(100 ㎍/mL 또는 100 µM)를 처리한 결과 독성을 보이지 않았다. 노화된 피부에서는 각질층의 산성 pH를 유지하기 위한 나트륨-수소 이온 교환 펌프(NHE1)의 발현감소가 나타나는 것으로 알려져 있으며, 이러한 이유로 노화 피부에서 나타나는 피부 장벽 기능 회복의 저하에 중요한 원인으로 작용할 것으로 추측된다. 꿀풀 추출물 및 카페인산은 각질형성 세포에서 NHE1의 발현을 증가시켰으며, 자연 보습 인자 전구체인 필라그린과 세라마이드 합성 효소인 serine plmitoyl transferase (SPT)의 발현을 증가시켰다. 또한, 직접적인 pH 조절 효과를 확인하기 위해 각질형성 세포 내/외 pH 수치를 측정한 결과 꿀풀과 카페인산은 세포 외부 pH를 감소시켰다. 위 결과들을 종합해 볼 때, 꿀풀과 카페인산은 NHE1 조절을 통해 피부 pH를 조절할 수 있고, 자연 보습 인자(NMF)와 세라마이드 합성을 증가시켜 피부 장벽 기능 회복을 도울 수 있을 것으로 추측된다. 이러한 결과는 꿀풀과 카페인산이 피부 건강에 긍정적인 영향을 미칠 수 있다는 가능성을 보여주며, 이를 활용한 새로운 피부 보호 제품 개발에 대한 토대가 될 수 있다.
밀착연접(tight junction)은 사람의 표피에서 관찰되는 세포간 이음(intercellular junction) 중 하나로서, 표피 과립세포층의 세포간격에 위치하여 인접한 세포들을 접합시키고 전해질과 수분의 이동을 조절하는 장벽기능을 비롯한 다양한 생물학적 기능을 수행한다. 이러한 밀착연접의 대표적 단백성분인 occludin은 그 발현도가 밀착연접의 표피-생물학적 기능성과 높은 상관관계가 있는 것으로도 알려져 있다. 본 연구에서는 신체 여러 부위의 정상피부에서 occludin의 발현도 또는 발현정도의 상이성 여부를 알아보기 위하여 occludin에 대한 항체를 이용한 간접 면역형광검사를 시행하여 그 결과를 비교 검토하고자 하였다. 신체 7개 부위별로 각 4개씩의 정상 피부조직을 채취하여 항 occludin 항체를 이용한 간접 면역형광검사를 시행한 결과, 양성반응을 보이는 최종 희석배수의 역수(reciprocal of end-point titer)로 표시한 부위별 occludin 항원의 발현도는 두피에서 300, 얼굴에서 600, 목에서 300, 팔에서 200, 배에서 600, 그리고 등과 다리에서는 모두 400으로 관찰되어 신체 부위별 occludin의 발현도에는 통계학적으로 유의한 차이가 있음을 알 수 있었다(p=0.001). 본 연구에서 관찰된 신체 부위별 occludin의 발현도의 차이는 밀착연접의 부위별 발현의 차등을 시사하는 소견으로서, 표피 피부장벽의 생물학적 기능의 가능한 부위별 차등성과도 관계가 있을 것으로 사료된다.
인체 피부 림프계는 간질액을 배출하고 면역 시스템을 활성화하는 중요한 역할을 한다. 자외선과 자연적인 노화와 같은 환경 요인들은 종종 이러한 림프관의 구조적 변화를 일으키며, 이로 인해 피부 기능 장애를 발생시키기도 한다. 그러나 이러한 연구를 위한 동물 실험 대체 방안이 없기 때문에 여전히 연구를 진행하기엔 적합하지 않은 제한 사항들이 존재한다. 인체 피부 림프계를 더 잘 이해하고 림프관 형성에 관련된 분자 및 생리학적 변화를 조사하기 위해, 생체 모방 미세유체 플랫폼인 'skin-lymph-on-a-chip'을 제작하여, 새로운 체외 인체 피부 림프 모델을 개발하였다. 간단히 말해, 이 플랫폼은 체외에서 분화된 일차 정상 인간 표피 각질형성세포(NHEKs)와 피부 림프 내피세포(HDLECs)를 공동 배양하는 것을 의미한다. 약 500 시간 동안 자연 발효를 통해 확보하고 집약된 인삼 뿌리 추출물인 LymphanaxTM의 림프관 형성 효과를 평가하기 위해 해당 시스템에 적용하였고, 분자 수준 요인들의 변화는 딥러닝 기반 알고리즘을 사용하여 분석하였다. 결론적으로, LymphanaxTM는 skin-lymph-on-a-chip에서 건강한 림프관 형성을 촉진하였고, 그 성분들이 칩내에서 분화된 NHEKs를 거의 침투하지 않는 결과를 통해, HDELCs에 간접적으로 영향을 미쳤음을 확인하였다. 전반적으로, 이 연구는 기존과 차별화된 체외 인체 피부 림프모델 시스템의 확보와 더불어 이를 통한 LymphanaxTM의 림프 활성화 효과에 대한 새로운 관점을 제공한다.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
