Baicalein induced Apoptosis of Rat Hepatic Stellate Cells

  • Baicalein (5,6,7-trihydroxyflavone), a flavonoid originated from the root of Chinese medicinal herb Scutellaria baicalensis, has been shown to exert anti-inflammatory and antioxidant effects and hepatic stellate cells play an important role in the pathogenesis or hepatic fibrosis. In this study, we investigated apoptosis stimulation by baicalein in activated rat hepatic stellate cells (T-HSC/C1-6). Transformed rat hepatic stellate cells (T-HSC/C1-6) were treated with baicalein(100uM, 70uM, 40uM). Apoptosis was determined by detection of DNA fragmentation in gel electrophoresis, morphological alternations by flow cytometry and quantification of phosphatidylserine externalization by Annexin V labeling.   • In this study, mechanical properties of high strength cement composites (HSC) containing recycled glass powder (GP) after heating were investigated. As a result, at 100Mpa, as the heating temperature increased, the compressive strength increased while the elastic modulus decreased . At 140Mpa, after heating at 300℃, the spalling occurred excluding GP0, and it is believed to be due to the high density of HSC.

Characterization of intracellular Ca2+ mobilization in gefitinib-resistant oral squamous carcinoma cells HSC-3 and -4

  • Oral squamous cell carcinoma (OSCC) metastasis is characterized by distant metastasis and local recurrence. Combined chemotherapy with cisplatin and 5-fluorouracil is routinely used to treat patients with OSCC, and the combined use of gefitinib with cytotoxic drugs has been reported to enhance the sensitivity of cancer cells in vitro. However, the development of drug resistance because of prolonged chemotherapy is inevitable, leading to a poor prognosis. Therefore, understanding alterations in signaling pathways and gene expression is crucial for overcoming the development of drug resistance. However, the altered characterization of Ca2+ signaling in drug-resistant OSCC cells remains unclear. In this study, we investigated alterations in intracellular Ca2+ ([Ca2+]i) mobilization upon the development of gefitinib resistance in human tongue squamous carcinoma cell line (HSC)-3 and HSC-4 using ratiometric analysis. This study demonstrated the presence of altered epidermal growth factor- and purinergic agonist-mediated [Ca2+]i mobilization in gefitinib-resistant OSCC cells. Moreover, Ca2+ content in the endoplasmic reticulum, store-operated calcium entry, and lysosomal Ca2+ release through the transient receptor potential mucolipin 1, were confirmed to be significantly reduced upon the development of apoptosis resistance. Consistent with [Ca2+]i mobilization, we identified modified expression levels of Ca2+ signaling-related genes in gefitinib-resistant cells. Taken together, we propose that the regulation of [Ca2+]i mobilization and related gene expression can be a new strategy to overcome drug resistance in patients with cancer.

  • Hematopoiesis is regulated by crosstalk between long-term repopulating hematopoietic stem cells (LT-HSCs) and supporting niche cells in the bone marrow (BM). Here, we describe the role of KAI1, which is mainly expressed on LT-HSCs and rarely on other hematopoietic stem-progenitor cells (HSPCs), in niche-mediated LT-HSC maintenance. KAI1 activates TGF-β1/Smad3 signal in LT-HSCs, leading to the induction of CDK inhibitors and inhibition of the cell cycle. The KAI1-binding partner DARC is expressed on macrophages and stabilizes KAI1 on LT-HSCs, promoting their quiescence. Conversely, when DARC+ BM macrophages were absent, the level of surface KAI1 on LT-HSCs decreases, leading to cell-cycle entry, proliferation, and differentiation. Thus, KAI1 acts as a functional surface marker of LT-HSCs that regulates dormancy through interaction with DARC-expressing macrophages in the BM stem cell niche. Recently, we showed very special and rare macrophages expressing α-SMA+ COX2+ & DARC+ induce not only dormancy of LT-HSC through interaction of KAI1-DARC but also protect HSCs by down-regulating ROS through COX2 signaling. In the near future, the strategy to combine KAI1-positive LT-HSCs and α-SMA/Cox2/DARC triple-positive macrophages will improve the efficacy of stem cell transplantation after the ablative chemo-therapy for hematological disorders including leukemia.

Strength Properties of High-Strength Concrete Exposed at High Temperature (고온을 받은 고강도 콘크리트의 강도특성)

  • A review is presented of experimental studies on the strength performance of concrete exposed at short-term and rapid heating as in a fire and after cooling. Emphasis is placed on concretes with high original compressive strengths, that is, high-strength concrete(HSC). The compressive strength-temperature relationships from the reviewed test programs are distinguished by the test methods used in obtaining the data(unstressed, unstressed residual strength, and stressed tests) and by the aggregate types(normal or lightweight), The compressive strength properties of HSC vary differently with temperature than those of NSC. HSC have higher rates of strength loss than lower strength concrete in the temperature range of between 20$^{\circ}C$ to about 400$^{\circ}C$. These difference become less significant at temperatures above 400$^{\circ}C$ compressive strengths of HSC at 800$^{\circ}C$ decrease to about 30 % of the original room temperature strength. A comparison of lest results with current code provisions on the effects of elevated temperatures on concrete compressive strength and elastic modulus shows that the CEN Eurocodes and the CEB provisions are unconservative.

The Inducible form of Heat Shock Protein 70 (Hsp70) is Expressed in the Rat Cerebellar Synapses in Normal Condition (흰쥐 소뇌 정상 연접에서 열충격단백질70(HSP70)의 표현)

  • Heat shock protein 70 (HSP70) is a multigene family composed of constitutively expressed members(Hsc70) and stress-inducible members (Hsp70). In the mammalian nervous system, a considerable amount of HSPs is also synthesized under normal conditions suggesting that they play an important role in the metabolism of unstressed cells. In this study we examined the expression of Hsp70 in the synapses of rat cerebellar neurons. Immunohistochemistry using specific antibodies revealed that both Hsp70 and Hsc70 are expressed in the cerebellar tissue, with strongest expression in Purkinje cells followed by granule cells. Neurons in deep cerebellar nuclei were also intensely stained by Hsp70 antibody. Immunocytochemical stainings of cultured cerebellar cells showed that Hsp70 is expressed in both Purkinje and granule cells. The expression was punctate in the soma and along dendritic trees, and the punctae were colocalized with those of PSD95, a postsynaptic marker. Immunoblotting also indicates that Hsp70 is associated with the postsynaptic density fraction. Taken together, our results indicate that the Hsp70 is expressed in cerebellar neurons in normal conditions, and that some are localized in the synapses.

Inhibitory Effect of Injinchunggan-tang(Yinchenqinggan-tang) on Fibrogenesis of Rat Hepatic Stellate Cells (인진청간탕(茵蔯淸肝湯)이 간성상세포의 섬유화 억제에 미치는 영향)

  • Objectives : This study was performed to investigate the anti-fibrogenic effect of Injinchunggan-tang on cultured rat hepatic stellate cells. Materials and Methods : Hepatic stellate cells(HSC-T6) were treated with various concentrations of Injinchunggan-tang extract for 24, 48, and 72 hours. The extraction was done with distilled water. After the treatment, cell viability, proliferation, procollagen levels and the mRNA of the TIMP-1, TIMP-2, and ASMA were measured by using MTT assay. BrdU assay, procollagen type I C-peptide EIA kit and RT-PCR. Results : The proliferation, mRNA expression and synthesis of collagen of the hepatic stellate cells were inhibited by Injinchunggan-tang treatment in a dose-dependent manner. This indicates the prescription has inhibitory effect on fibrogenesis of the liver by regulating the fibrogenesis associated genes in transcription. Cell viability was inhibited in time- and dose-dependent manners. It seemed that the drug should be used with sufficient dose to acquire treatment effect. Conclusion : These results suggest that Injinchunggan-tang is beneficial in the treatment of cirrhotic patients as well as for the patients with chronic hepatitis.

  • Variations of temperature and velocity fields in a Hele-Shaw convection cell (HSC) were investigated using a holographic interferometry and 2-D PIV system with varying Rayleigh number. To measure quasi-steady variation of temperature field, two different measurement methods of holographic interferometry, double-exposure method and real-time method, were employed. In the double-exposure method, unwanted waves were eliminated effectively using a digital image processing technique. The reconstructed images are clear, but transient flow cannot be reconstructed clearly. On the other hand, transient convective flow can be reconstructed well using the real-time method. However, the fringe patterns reconstructed by the real-time method contain more noises, compared with the double-exposure method. Experimental results show a steady flow pattern at low Rayleigh numbers and a time-dependent periodic flow structure at high Rayleigh numbers. The periodic flow pattern at high Rayleigh numbers obtained by the real-time holographic interferometer method is in a good agreement with the PIV results.

Rheological Characteristics of Fiber-Reinforced High-Strength AFR Concrete (섬유보강 고강도 내화콘크리트의 레올로지 특성 분석)

  • The fiber(NY, PP) known to the effective material on improvement of the fire-resistance of HSC(high strength concrete) has a difference for fluidity according to the variation of a length and contents of fiber. In this study, to analyze the effect of a length and contents of the fiber on the fluidity of HSC and fheological characteristics, we calculated a viscosity of mortar by mini slump-flow, simple V-lot and viscometer. With the test results, the fluidity characteristic showed a moderate difference by a length and contents of the fiber, but showed a significant difference by increase of the fiber contents. ${\ast}$ AFR Concrete (Advenced Fire Resistant Concrete)
