신형 TL 선량계인 $CaSO_4:Dy,P$를 이용한 Hp(10) 측정용 배지의 필터체계 설계 (Design of a Badge Filter System for Measurement of Hp(10) with the New Type of TL Dosimeter $CaSO_4:Dy,P$)

  • 한국원자력연구소에서 개발한 $CaSO_4:Dy,P$를 이용하여 ICRU에서 권고하는 Hp(10)을 평가하고자 하였다. $CaSO_4:Dy,P$는 광자에 내하여 높은 에너지 의존성을 지니므로 Hp(10) 평가를 위해서는 필터물질을 이용하여 모든 에너지에 대하여 반응도를 적절히 보상하여야 한다. ISO에서 권고하는 열형광선량계의 성능요건을 만족하는 필터설계를 위해 몬테칼로 코드를 이용한 이론적 계산과 검증 실험을 수행하였다. 완성된 필터 하에서 $CaSO_4:Dy,P$ 소자는 $20{\sim}662keV$에 이르는 광자 에너지 범위에서 $0.75{\sim}1.0$의 상대반응도를 보였다. 특히 직경이 큰 후방필터를 설계함으로써 전방필터의 두께를 감소하고 필터 조합을 단순화하였으며 테이퍼를 도입하여 선량계의 방향의존성을 대폭 개선할 수 있었다.

A549 폐 상피세포에서 합토글로빈에 의한 염증반응 조절 (Inflammatory Regulation by Haptoglobin in A549 Cells)

  • 합토글로빈(Hp)은 조직손상이나 감염 등의 염증시에 혈중 농도가 증가하는 급성기반응단백질로서 주로 간에서 생합성되지만 염증을 동반한 폐에서도 발현됨이 보고되었다. 폐에서 합성되는 Hp이 염증반응에 어떤 영향을 주는지를 조사하고자, Hp을 과발현하는 폐상피세포를 사용하여 COX-2 및 염증관련 cytokine들의 발현을 조사하였다. Stable transfection 또는 transient transfection된 A549 세포에서 Hp이 잘 발현되고 있음을 확인한 후 COX-2의 발현을 측정한 결과, Hp의 과발현에 의해 COX-2 합성이 현저히 증가함을 확인할 수 있었다. 또한 Hp을 과발현하는 A549세포에 LPS ($1{\mu}g/ml$) 또는 $IL-1{\beta}$ (100 U/ml)를 각각 24시간 동안 처치하였을 때, Hp에 의한 COX-2 발현 증가는 LPS또는 $IL-1{\beta}$자극에 의해 협동적으로 증가하였다. ACP-based PCR 방법으로 염증관련 cytokine들의 발현을 측정한 결과, Hp에 의해 SPARC의 발현이 현저히 저하되는 반면에 IL-4 및 S100A1 유전자 발현은 약간 증가함을 알 수 있었다. 이러한 결과들은 폐상피세포에서 Hp이 염증반응을 활성화시키는 기전으로 작용함을 시사한다.

Spectroscopic Studies on Interaction of Protoberberines with the Deoxyoligonucleotide d(GCCGTCGTTTTACA)2

  • The topoisomerase II poisoning effect of certain protoberberine alkaloids is associated with anti-cancer activity. Structure-activity relationships of protoberberine analogues substituted on the ring protons reveal that substitution at the C19 position is an important determinant of biological activity. In this study, the effects of substituent modification at the C19 position on the interaction of protoberberines with DNA are determined using UV and NMR spectroscopy. The line broadening effect on aliphatic resonances, chemical shift changes of the imino protons of HP14 upon berberine and berberrubine binding to HP14, and the rate of the exchange process between protoberberine analogs bound indicate that berberrubine binds HP14 more specifically than berberine. In addition, the free HP14 is altered by the substituent at the 19-position. UV spectra of berberrubine have shown a hypochromic effect together with a slight red shift, which are usually regarded as characteristics of DNA intercalation. These results are consistent with our previous report that the berberrubine is partially intercalated with HP14 with molar ratio 1 : 1, whereas a non-specific interaction is predominant between the berberine and HP14.

하프팬케익 그래프의 다양한 성질 분석 (Analysis of Various Characteristics of the Half Pancake Graph)

  • The Pancake graph is node symmetric and useful interconnection network in the field of data sorting algorithm. The Half Pancake graph is a new interconnection network that reduces the degree of the Pancake graph by approximately half and improves the network cost of the Pancake graph. In this paper, we analyze topological properties of the Half Pancake graph $HP_n$. Fist, we prove that $HP_n$ has maximally fault tolerance and recursive scalability. In addition, we show that in $HP_n$, there are isomorphic graphs of low-dimensional $HP_n$. Also, we propose that the Bubblesort $B_n$ can be embedded into Half Pancake $HP_n$ with dilation 5, expansion 1. These results mean that various algorithms designed for the Pancake graph and the Bubble sort graph can be executed on $HP_n$ efficiently.

뇌로의 악물송달(I) - 사람, 랫트 및 가토에 있어서의 할로페리돌의 약물속도론 연구

  • 할로페리돌(HP)은 정신분열증 환자에 널리 사용되는 Dopamine D$_2$-receptor의 antagonist인 antipsychotic drug이다. 이 약물의 혈장농도와 임상 반응사이의 'curvilinear'한 상관성 존재여부와 여기에 대한 대사체(reduced haloperidol, RH)의 영향에 대해 논란은 많지만, 본 연구 팀에서도 위의 상관성이 존재하며 또한 여기에 RH가 영향을 미칠 것으로 보고한바 있다. 따라서 본 연구에서는 앞의 결과에 대한 기전을 밝히고, 궁극적으로 효율적인 뇌송달 시스템의 개발가능성을 검토하기위한 1차적 인구로 HP의 약물속도론적 연구를 사람, 랫트 및 가토에서 실시하여 그특성을 비교하였다. 사람경우는 13명의 정신분열증 초기환자를 대상으로 경구(20mg HP, 5명) 및 주사(10mg HP, 8명) 투여한 후, 또한 랫트 및 가토의 경우는 마리당 5mg의 HP 및 RH를 각각 정맥주사한 후 경시적으로 혈장중 HP 및 RH의 농도를 측정하여 체내동태 특성을 검토하였다.

Buckling analysis of arbitrary point-supported plates using new hp-cloud shape functions

  • Considering stress singularities at point support locations, buckling solutions for plates with arbitrary number of point supports are hard to obtain. Thus, new Hp-Cloud shape functions with Kronecker delta property (HPCK) were developed in the present paper to examine elastic buckling of point-supported thin plates in various shapes. Having the Kronecker delta property, this specific Hp-Cloud shape functions were constructed through selecting particular quantities for influence radii of nodal points as well as proposing appropriate enrichment functions. Since the given quantities for influence radii of nodal points could bring about poor quality of interpolation for plates with sharp corners, the radii were increased and the method of Lagrange multiplier was used for the purpose of applying boundary conditions. To demonstrate the capability of the new Hp-Cloud shape functions in the domain of analyzing plates in different geometry shapes, various test cases were correspondingly investigated and the obtained findings were compared with those available in the related literature. Such results concerning these new Hp-Cloud shape functions revealed a significant consistency with those reported by other researchers.

Structure Based Protein Engineering of Aldehyde Dehydrogenase from Azospirillum brasilense to Enhance Enzyme Activity against Unnatural 3-Hydroxypropionaldehyde

  • 3-Hydroxypropionic acid (3HP) is a platform chemical and can be converted into other valuable C3-based chemicals. Because a large amount of glycerol is produced as a by-product in the biodiesel industry, glycerol is an attractive carbon source in the biological production of 3HP. Although eight 3HP-producing aldehyde dehydrogenases (ALDHs) have been reported so far, the low conversion rate from 3-hydroxypropionaldehyde (3HPA) to 3HP using these enzymes is still a bottleneck for the production of 3HP. In this study, we elucidated the substrate binding modes of the eight 3HP-producing ALDHs through bioinformatic and structural analysis of these enzymes and selected protein engineering targets for developing enzymes with enhanced enzymatic activity against 3HPA. Among ten AbKGSADH variants we tested, three variants with replacement at the Arg281 site of AbKGSADH showed enhanced enzymatic activities. In particular, the AbKGSADHR281Y variant exhibited improved catalytic efficiency by 2.5-fold compared with the wild type.

Nomogram Models for Distinguishing Intraductal Carcinoma of the Prostate From Prostatic Acinar Adenocarcinoma Based on Multiparametric Magnetic Resonance Imaging

  • Objective: To compare multiparametric magnetic resonance imaging (MRI) features of intraductal carcinoma of the prostate (IDC-P) with those of prostatic acinar adenocarcinoma (PAC) and develop prediction models to distinguish IDC-P from PAC and IDC-P with a high proportion (IDC ≥ 10%, hpIDC-P) from IDC-P with a low proportion (IDC < 10%, lpIDC-P) and PAC. Materials and Methods: One hundred and six patients with hpIDC-P, 105 with lpIDC-P and 168 with PAC, who underwent pretreatment multiparametric MRI between January 2015 and December 2020 were included in this study. Imaging parameters, including invasiveness and metastasis, were evaluated and compared between the PAC and IDC-P groups as well as between the hpIDC-P and lpIDC-P subgroups. Nomograms for distinguishing IDC-P from PAC, and hpIDC-P from lpIDC-P and PAC, were made using multivariable logistic regression analysis. The discrimination performance of the models was assessed using the receiver operating characteristic area under the curve (ROC-AUC) in the sample, where the models were derived from without an independent validation sample. Results: The tumor diameter was larger and invasive and metastatic features were more common in the IDC-P than in the PAC group (P < 0.001). The distribution of extraprostatic extension (EPE) and pelvic lymphadenopathy was even greater, and the apparent diffusion coefficient (ADC) ratio was lower in the hpIDC-P than in the lpIDC-P group (P < 0.05). The ROC-AUCs of the stepwise models based solely on imaging features for distinguishing IDC-P from PAC and hpIDC-P from lpIDC-P and PAC were 0.797 (95% confidence interval, 0.750-0.843) and 0.777 (0.727-0.827), respectively. Conclusion: IDC-P was more likely to be larger, more invasive, and more metastatic, with obviously restricted diffusion. EPE, pelvic lymphadenopathy, and a lower ADC ratio were more likely to occur in hpIDC-P, and were also the most useful variables in both nomograms for predicting IDC-P and hpIDC-P.

hp-DISCONTINUOUS GALERKIN METHODS FOR THE LOTKA-MCKENDRICK EQUATION: A NUMERICAL STUDY

  • The Lotka-McKendrick model which describes the evolution of a single population is developed from the well known Malthus model. In this paper, we introduce the Lotka-McKendrick model. We approximate the solution to the model using hp-discontinuous Galerkin finite element method. The numerical results show that the presented hp-discontinuous Galerkin method is very efficient in case that the solution has a sharp decay.

  • Let A be an expansive dilation on ℝn, and p(·) : ℝn → (0, ∞) be a variable exponent function satisfying the globally log-Hölder continuous condition. Let Hp(·)A (ℝn) be the variable anisotropic Hardy space defined via the non-tangential grand maximal function. In this paper, the author obtains the boundedness of anisotropic convolutional ��-type Calderón-Zygmund operators from Hp(·)A (ℝn) to Lp(·) (ℝn) or from Hp(·)A (ℝn) to itself. In addition, the author also obtains the duality between Hp(·)A (ℝn) and the anisotropic Campanato spaces with variable exponents.
