Note Biological Control of Fusarium Wilt in Tomato by Plant Growth-Promoting Yeasts and Rhizobacteria

  • Three plant growth-promoting yeasts and two rhizobacteria were tested for controlling tomato wilt caused by Fusarium oxysporum f. sp. lycopersici under green-house and field conditions. Under greenhouse and field conditions, all treatments were significantly reduced disease severity of tomato wilt relative to the infected control. The highest disease reductions in pots (75.0, 67.4%) and field (52.5, 42.4%) were achieved by Azospirillum brasilense and Bacillus subtilis compared to infected control. Under field condition all treatments produced the highest tomato yield compared to the control plants inoculated with the pathogen.

Effects of Shading and Mixing Ratio of Recycling Spawn bed Logs on the Yield of Gastrodia elata Blume (차광정도와 접종목 흔용이 천마의 수량에 미치는 영향)

  • To establish the economical cultivation method of Gastrodia elata Blume by controlling the shading rate and the mixing ratio of recycling spawn bed logs, a serial experiment was carried out. When G. elata was grown in the field with no shading, the yield of spawn tuber was higher than that of tuber of G. elata. The yield of G. elata was high in the rainproof green house covered with one layer of $95\%$ shading net, in the rainproof green house covered with two layers of $95\%$ shading net, and in the field with no shading, in that order. In the case of mixing ratio of recycling spawn bed logs, the yield was high in the fresh logs, in the mixing ratio of 2:1, and the ratio of 1:1, in that order. The income of the rainproof green house covered with one layer of $ 95\%$ shading net which brought the best yield was $81\%$ in the mixing ratio of 2:1 over the fresh logs.

Somatic embryogenesis and in vitro plant regeneration from various explants of the halophyte Leymus chinensis (Trin.)

  • The halophyte Leymus chinensis (Trin.) is a perennial rhizome grass (tribe Gramineae) that is widely distributed throughout China, Mongolia and Siberia. This study was conducted to investigate an optimal condition for plant regeneration from mature seeds, leaf base segments, and root segments in L. chinensis. Plant growth regulators affecting embryogenic callus induction and plant regeneration were investigated by four-factor-three-level [L9 (34)] orthogonal test in this study. The effects of explants types (mature seeds, leaf base segments and root segments), callus types, medium types were examined in this study. Wild type (WT) and Jisheng No. 1 plants (JS) were used for primary callus induction. A clear explants difference was seen during callus induction; mature seeds were considered as the preferred explants; and the highest frequency of callus induction was obtained in Medium 6 using mature seeds as explants in WT. Plant regeneration ability was evaluated by frequencies of green callus forming, shooting, rooting, and shooting with roots. Effect of α-naphthalene acetic acid (NAA) on shoot regeneration was remarkable with the highest frequency of 70.8% in WT after 2-month culture. The medium with 0.2- 0.5 mg/L NAA was found to have the highest shoot induction. All regenerated shoots were successfully rooted when transferred on half-strength Murashige and Skoog (MS) basal medium. The acclimatized plantlets were grown to mature with flowering and seeds setting in green house conditions.

Effect of Chelating Agents on the Grwoth of Chinese cabbage and Availability of Nutrients in Plastic Film House Soils (시설재배지에서 킬레이트제 처리가 양분 유효도와 배추생육에 미치는 영향)

  • This study is conducted to evaluate the effects of chelating agents for improving plant growth and reusing accumulated nutrients in soils of plastic film house. Two experiments were carried out at follows: i) The incubation test was conducted using soils treated with 0, 300 mM of EDTA and DTPA to examine the availability of nutrients. ii) For the pot test, chinese cabbages were cultivated in soils with 0, 0.1, 0.5, 1, and 5 mM of EDTA and DTPA to examine the impacts of plant growth response. The application of chelating agents increased ther availability of soil nutrients in the following order: DTPA > EDTA > control. Inorganic concentration of chinese cabbages in DTPA treatments consderably increased in nitrogen, phosphate, iron and aluminium contents than that of the other treatments. The optimal concentration of DTPA for vigorous plant growth as 0.5 mM. Thus, DTPA was more effective than other chelating agents for healty growth of cabbages and the availability of nutrients accumulated in plastic film house.

Effect of Cotton Leaf Mosaic Disease on Morphology, Yield and Fibre Characteristics of Upland Cotton in Pakistan

  • The effect of cotton leaf mosaic disease on morphology, yield and fibre characteristics was examined for a susceptible cotton candidate variety CRIS-168. Plants inoculated at most susceptible growth stage (six week) under screen house showed severe mosaic symptoms. There was a significant reduction in plant height and yield. Cotton leaf mosaic disease was found to produce severe effects on plant morphology with 24.1% reduction in plant height, 25% in internode length and 37.5% in number of sympodia on main stem. However no changes were observed against number of monopodial branches per plant. Inoculated plants showed 82% decrease in yield/plant, 80% in number of boll set/ plant, 12.1% in boll weight, 12.8% in lint weight, 10.8% in seed weight, and 6.8% in seed index. Cotton leaf mosaic disease also showed effects on fibre characteristics with 0.8% decrease in GOT and 1.6% in fibre length. In contrast, uniformity ratio, fibre fineness and maturity index was increased by 20.5%, 14.4% and 0.9%, respectively.

In Vitro Plant Regeneration from Stolen Node Explant in Eremochloa Ophiuroides (Munro) Hack

  • In vitro shoot regeneration and multiple shoot induction has been obtained from the stolen node explants in Eremochloa ophiuroides (Munro) Hack. The highest number of shoots ($10.66{\pm}0.21$) was observed from initial explants after one month culture duration on Murashige and Skoog (MS) medium containing 6-benzyladenine (BA: 0.5 mg/l). First generation shoot was excised and sub-cultured on the same fresh media for further multiplication of shoots. An enhanced number of second round shoots ($15.33{\pm}0.21$) was obtained compared to the initial culture media containing BA (0.5 mg/l). The number of shoots/stolon node was higher among all the concentrations of BA than kinetin (KN). In vitro regenerated shoots were successfully rooted in the phytohormone free MS medium. Plantlets generated with roots were transferred to pots containing compound mixture of soil and kept in green house conditions. Acclimatized plants showed 100% survival rate with normal morphology in green house conditions. The present study demonstrates the effect of explant and different plant growth regulators towards in vitro response in E. ophiuroides. Moreover, the study reveals the effect of cytokinin on induction of shoot number per stolen node explant in E. ophiuroides.

Bacterial Soft rot of Kalanchoe blossfeldiana by Erwinia herbicola in Korea (Erwinia herbicola 의한 Kalanchoe blossfeldiana세균성무름병)

  • A new bacterial disease was found on leaves of Kalanchoe blossfeldiana plant grown under vinyl-house condition in winter of 1998 in Taejeon. the first symptoms of the disease are the appearance of the water-soaked and light brown spots. Later they become soft rot with brown color. Causal bacteria were isolated from diseased tissues and the same symptoms as the natural infection were developed on Kalanchoe blossfeldiana leaves by needle-prick inoculation. The causal bacterium was identified Erwinia hervicola by its bacteriological characteristics. This is the first reported of this bacterium to occur on kalanchoe blossfeldiana plant in Korea. Therefore, we proposed to name the diseases as \"bacterial soft rot of Kalanchoe blossfeldiana\" by E. herbiocla.

Effects of Some Heavy Metals(Al, Cd, Hg, and Pb) on ATP Content in Plant Leaves (식물엽의 ATP함량에 미치는 중금속(Al, Cd, Hg 및 Pb)의 영향)

  • The present study was carried out to estimate the comparative effects of hydroponic heavy metals (Al, Cd, Hg, and Pb) on ATP content in plant leaves grown with Hoagland solution under green house condition. The two plants, kidneybean(Phaseolus vulgaris L.) and buckwheat(Fagopyrum esculentum M nch), showed similar inhibitory effect of heavy metals on ATP content in order of Hg, Cd, Pb, and Al. But the overall inhibitory effect was greater in kidneybean than in buckwheat. The affinity of heavy metals, in vitro, toward the enzyme (luciferin-luciferase) is in order of Hg, Al, Cd, and Pb, similar to that toward ATP. The results showed that the inhibitory effect of heavy metals on ATP hydrolysis is mainly due to the coordination of heavy metals with enzyme than ATP.

  • Growth status, plant nutrition and soil chemical properties were investigated for lettuce, spring onion and red pepper grown under vinyl house near Yesan. Low Ca and Mg with high K in soil resulted in Ca deficiency with slight Mg insufficiency in Korean lettuce. In this soil pH was low and EC was extremely low. Spring onion(Allium wakegi) can not emerge or showed poor growth(50%) due to high EC(above 0.5m mho/cm) and low pH (below 6.2). Red pepper plant showed wilt disease probably due to low soil pH.

Improvement of Landfarming Applicability from Analysis of Case Studies (토양경작법의 사례 분석을 통한 적용방안 개선)

  • Considering six screen matrix to select an optimum remediation method for the Kunsan military base contaminated with petroleum oil, the following order was obtained: landfarming > biopile > soil washing > thermal desorption = incineration. When the landfarming method was applied for the remediation of 2,250 $m^3$ soil contaminated with petroleum oil ranging from 500 to 2,404 mg/kg as TPH, contamination level decreased below target concentration 450 mg/kg after 20~42 days depending on the initial contamination. From the evaluation of case studies of landfarming, it is suggested that ratty-truss or single-arch structure is suitable in the landfarming plant for the treatment of large-scale contaminated soil requiring long period of remediation. But, vinyl-house structure is suitable in the landfarming plant for the treatment of small-scale contaminated soil requiring short period of remediation. Therefore vinyl-house structure is recommended in the remediation of contaminated soil less than 5,000 $m^3$ requiring within 1 year of remediation period but ratty-truss or single-arch structure is recommended for the remediation of contaminated soil more than 5,000 $m^3$.
