Effect of a blend of magnesium oxide on Equine Squamous Gastric Disease in young trotter horses under training

  • Background: Equine squamous gastric disease (ESGD), as part of the equine gastric ulcer syndrome (EGUS), are common in racing horses. The use of buffering feed supplements to treat and/or prevent gastric ulcers is an option to control this condition. Objective: The purpose of this study was to evaluate the effect of a 30-day supplementation with a blend of magnesium oxide (MgO) on ESGD scores in trotters under training. Methods: Forty-two young trotters were submitted to a gastroscopic evaluation to assess their ESGD score and were randomly assigned in a group supplemented with MgO or in a control group. After 30 days, a second evaluation by gastroscopy was performed. The effect of the MgO supplementation was assessed by comparing the evolution of the ESGD score in supplemented and control groups between day 0 and day 30. Results: The results confirm the high prevalence of EGUS in young Trotters. The supplementation significantly decreased the ESGD scoring in the supplemented group whereas the control group remain unchanged. Conclusion: The oral MgO supplementation was efficient to control ESGD in the population studied.

제주마에서 Myostatin 유전자 변이 특성 구명 (Characterization of Myostatin Gene Variants in Jeju Horses)

  • 제주마는 한국의 말 품종으로 제주도에서 오랜 기간 자생하여 왔다. 제주마의 기원은 몽골마에서 유래한 것으로 추정되었다. 그러나 최근 연구에서 제주마와 몽골 토종마를 비교 한 결과, 유전적으로 가까우나 제주마가 몽고마와 독립적으로 진화해 온 것으로 보고되었다. 제주마는 경주마로 이용되어 경주 능력이 주요한 경제 형질로 활용되고 있다. Myostatin (MSTN) 유전자는 다양한 포유류에서 골격근량에 영향을 미치는 것으로 연구되었다. Thoroughbred에서 MSTN 유전자 내 존재하는 변이가 경주 능력 및 스태미너에 영향을 미치는 것으로 알려져 있다. 우리는 제주마 1,433두를 포함하여 여러 말 품종의 MSTN 유전자내 변이 빈도를 비교하였다. MSTN 유전자의 g.66493737 변이 중 장거리 적성 유전자형(TT)의 경우 제주마에서 빈도가 0.826으로 한라마(0.285) 및 Thoroughbred (0.252)보다 높은 것으로 확인되었다. 또한 다른 연구를 참고하여 8종 말에서 g.66493737 빈도를 비교하였다. 그 결과, 경주마로 활용되는 품종은 CC형의 빈도가 높았으나, TT형의 빈도는 낮은 것으로 확인되었다. 반대로 승용마 및 역용마 등의 품종은 TT형의 빈도가 높은 것으로 확인되었다. 제주마의 경주 거리(400 m, 800 m, 900 m, 1,000 m, 1,110 m, 1,200 m)별로 유전자형과 도착 기록을 비교 분석했다. 그 결과 1,000 m 이하의 경주에서는 CT형이 TT형보다 더 빠른 도달 기록을 보였다. 그러나 1,110 m 이상의 경주에서는 거의 동일한 결과가 확인되었다. 본 연구에서는 제주마의 MSTN 유전자 변이가 경주 거리 적성과 관련이 있을 수 있음을 시사하였다. 이러한 결과는 추후 연구를 통해 제주마에서 경주 거리 적성 및 경주 능력과 연관된 마커 개발을 위한 자료로 활용이 가능할 것으로 사료된다.

개체식별 및 친자판정을 위한 말의 적혈구항원형에 관한 연구 (Genetic Studies of Redcell Types for Individual Identification and Parentage Verification in Horse Breeds.)

  • 제주마의 기원 및 혈통보존을 위한 기초자료를 마련할 목적으로 국내에서 사육중인 210두(제주마 73두, 교잡마 118두, 몽고마 19두)의 말을 대상으로 적혈구항원형의 표현형 분포와 유전자 빈도를 조사한 결과를 요약하면 다음과 같다. 제주마, 교잡마, 그리고 몽고마 적혈구항원형의 표현형은 각각 Aa (27.4%, 63.6%, 63.2%), Ca (97.3%, 94.9%, 89.5%), K- (97.3%, 99.2%, 84.2%), Pa (39.7%, 44.9%, 42.1%), 그리고 Ua (71.2%, 70.3%, 63.2%)에서 가장 높은 빈도를 나타내었고 D 시스템의 유전자형과 Q 시스템의 표현형은 각각 Dbcm/dghm (12.3%), Dbcm/cgm (14.4%), Dcgm/dghm (15.8%)와 Qc (56.2%), Qabc (36.4%), Qc (31.6%)에서 높은 빈도로 관찰되었다. 적혈구항원형의 유전자 빈도의 경우 제주마는 A- (0.287), Ca (0.827), Ddghm (0.226), K- (0.985), Pa (0.358), Qc (0.494), U- (0.529), 교잡마는 Aa (0.529), Ca (0.776), Dbcm (0.306), K- (0.995), P-(0.531), Q- (0.504), U- (0.548), 몽고마는 Aa (0.421), Ca (0.895), Ddghm (0.421), K- (0.842), Pa (0.447), Qc (0.448), Ua (0.632) 대립유전자가 가장 높은 빈도를 보였으며 대립유전자 Dcfgk 와 D- 는 모든 말에서 관찰되지 않았다.

우수 마 선택을 위한 최신 전략 (Recent Strategy for Superior Horses)

  • 말은 인류에 의해 상대적으로 일찍 가축화된 종 중 하나로써, 경주능력, 강건성 및 항병성 등과 같은 능력을 위해 인공적으로 선택되었다. 그 결과, 현재 경주마로 많이 쓰이고 있는 서러브레드의 게놈은 운동 능력에 특화된 유전자형을 많이 갖고 있다. 최근 NGS 기술의 도래와 함께 전장게놈을 대상으로 경주마의 우수한 유전형질을 찾는 연구가 유전체학의 관점에서 진행되고 있다. 그 결과 말의 게놈에 대해서도 GWAS (Genome-wide Association study)가 적용되고 있고, 우수 경주능력을 나타내는 유전자 마커가 발굴되고 있다. 아울러, 특정 샘플의 전장 전사체를 NGS 기법으로 분석할 수 있는 RNA-Seq 기법 역시 활용되고 있는데, 이를 통하여 각 개체별, 운동 전후, 한 개체의 조직별 특정 유전자의 발현 양상과 함께 전사체의 서열 등을 확인할 수 있다. DNA 서열의 변화 없이 유전자 발현을 조절하는 강력한 인자로써 DNA methylation이 주목받고 있다. 말의 게놈에 있어서도 운동 특이적 또는 개체 특이적 DNA methylation 패턴을 보여 주었고, 이는 우수 개체 선정을 위한 마커 개발에 좋은 단서를 제공해 줄 것이다. 유전자 발현을 억제하는 miRNA와, 포유동물의 유전체 내 절반 정도를 차지하고 있는 이동성 유전인자는 기능유전체 연구에 있어서 중요한 인자들이다. 이들은 인간의 게놈에서 많이 연구가 되어 왔으나, 말에서의 연구는 현재 미미한 실정이다. 하지만, 현재까지 말에서 되어 있는 위의 두 인자에 대한 연구 현황을 알아보고, 차후 우수 마 선별 연구에 적용될 가능성을 제시하였다. 기능유전체 및 후성유전체 분석 기법이 발전함에 따라 말에서도 본 연구에서 소개된 여러 가지 분석 기법이 적용되고, 우수한 경주마를 선정하는 데 많은 도움을 줄 것으로 기대하고 있다. 이에 현재까지의 우수한 경주마를 선택하기 위한 많은 연구들 및, 말 연구에 대한 앞으로의 발전 가능성에 대해 고찰하고 토의하였다.

Changes in Salivary Cortisol Concentration in Horses during Different Types of Exercise

  • This study aimed to estimate the change of stress level in horses based on cortisol concentration levels in their saliva. A total of 61 horses were divided into the following three groups: i) tourist riding experience (TR, n = 23); ii) resting group (RR, n = 14); and iii) horse-riding education (ER, n = 24). The saliva samples of TR and ER groups were taken using plain cotton Salivettes four times a day: at 07:00 (basal), 11:00 (Exercise 1, after 1-hour exercise in the morning), 14:00 (Exercise 2, after 1-hour exercise in the afternoon), and 16:00 (Exercise 3, after 1-hour exercise in the afternoon). The saliva samples of RR were measured at the same time. The samples were analyzed using the SAS program general linear model procedure. In a percentage relative to the base value, cortisol levels in Exercise 3 were confirmed to decrease in all groups as compared to the basal value percentage in the following sequence: ER>TR>RR. The highest peak was confirmed in Exercise 2 (approximately 131%) of RR group and the lowest peak appeared in Exercise 3 (approximately 52%) of ER group. Therefore, resting without any particular exercise can also increase the stress level of horses. Thus, it is better to exercise, as exercise can reduce the stress level, even in cases when riders are clumsy or lack appropriate horse-riding experience. The results of the present study are useful to equestrian center owners and educational riding instructors in that they provide a meaningful insight into a better horse management.

First Blindness Cases of Horses Infected with Setaria Digitata (Nematoda: Filarioidea) in the Republic of Korea

  • Ocular setariases of cattle were reported but those of equine hosts have never been reported in the Republic of Korea (Korea). We found motile worms in the aqueous humor of 15 horses (Equus spp.) from 12 localities in southern parts of Korea between January 2004 and November 2017. After the affected animals were properly restrained under sedation and local anesthesia, 10 ml disposable syringe with a 16-gauge needle was inserted into the anterior chamber of the affected eye to successfully remove the parasites. The male worm that was found in 7 of the cases showed a pair of lateral appendages near the posterior terminal end of the body. The papillar arrangement was 3 pairs of precloacal, a pair of adcloacal, and 3 pairs of postcloacal papillae, plus a central papilla just in front of the cloaca. The female worms found in the eyes of 8 horses were characterized by the tapering posterior terminal end of the body with a smooth knob. Worms were all identified as Setaria digitata (von Linstow, 1906) by the morphologic characteristics using light and electron microscopic observations. This is the first blindness cases of 15 horses infected with S. digitata (Nematoda: Filarioidea) in Korea.

Recent Advances in Artificial Insemination (AI) in Horses: Stallion Management, Processing and Preservation of Semen and Insemination Techniques

  • The efficiency of artificial insemination (AI) for horses remains unsatisfactory. It is mainly because each process of AI causes a detrimental effect on semen quality. To sustain quality of semen properly, several factors including libido of stallions and sperm damage during sperm processing and preservation should be considered. Stallions with decent libido produce a high ratio of sperm to seminal plasma in their ejaculates, which is the ideal semen composition for maintaining sperm quality. Thus, to maximize the fertility rate upon AI, stallions should be appropriately managed to enhance their libido. Seminal plasma should have a positive effect on horse fertility in the case of natural breeding, whereas the effects of seminal plasma on both sperm viability and quality in the context of AI remain controversial. Centrifugation of semen is performed during semen processing to remove seminal plasma and to isolate fine quality sperm from semen. However, the centrifugation process can also result in sperm loss and damage. To solve this problem, several different centrifugation techniques such as Cushion Fluid along with dual and single Androcoll-E$^{TM}$ were developed to minimize loss of sperm and to damage at the bottom of the pellet. Most recently, a new technique without centrifugation was developed with the purpose of separating sperm from semen. AI techniques have been advanced to deliver sperm to optimal region of female reproductive tract at perfect timing. Recombinant equine luteinizing hormone (reLH) and low dose insemination techniques have been developed to maximize both fertility rate and the efficiency of AI. Horse breeders should consider that the entire AI procedure should be optimized for each stallion due to variation in individual horses for a uniformed AI protocol.

Thoroughbred Horse Single Nucleotide Polymorphism and Expression Database: HSDB

  • Genetics is important for breeding and selection of horses but there is a lack of well-established horse-related browsers or databases. In order to better understand horses, more variants and other integrated information are needed. Thus, we construct a horse genomic variants database including expression and other information. Horse Single Nucleotide Polymorphism and Expression Database (HSDB) (http://snugenome2.snu.ac.kr/HSDB) provides the number of unexplored genomic variants still remaining to be identified in the horse genome including rare variants by using population genome sequences of eighteen horses and RNA-seq of four horses. The identified single nucleotide polymorphisms (SNPs) were confirmed by comparing them with SNP chip data and variants of RNA-seq, which showed a concordance level of 99.02% and 96.6%, respectively. Moreover, the database provides the genomic variants with their corresponding transcriptional profiles from the same individuals to help understand the functional aspects of these variants. The database will contribute to genetic improvement and breeding strategies of Thoroughbreds.

Genetic Diversity of mtDNA D-loop and Maternal Origin of Three Chinese Native Horse Breeds

  • In order to protect the genetic resource of native horse breeds, the genetic diversity of mitochondrial DNA (mtDNA) D-loop of three native horse breeds in western China were investigated. Forty-three 600 bp mtDNA D-loop sequences were analyzed by PCR and sequencing techniques, 33 unique haplotypes with 70 polymorphic sites were detected in these horses, which account for 11.67% of 600 bp sequence analyzed, showing the abundant genetic diversity of the three native horse breeds in western China. The Neighbour-Joining (NJ) phylogenetic tree based on 247 bp of 43 D-loop sequences demonstrated the presence of seven major lineages (A to G), indicating that the three native horse breeds in western China originated from multiple maternal origins. Consistent with the front, the NJ phylogenetic tree based on 600 bp of mtDNA D-loop sequences of 43 Chinese western native horses and 81 sequences of six horse breeds from GenBank indicated that the three horse breeds had distributed into the seven major lineages (A to G). The structure of the phylogenic tree is often blurred because the variation in a short segment of the mitochondrial genome is often accompanied by high levels of recurrent mutations. Consequently, longer D-loop sequences are helpful in achieving a higher level of molecular resolution in horses.
