Hepatitis C virus (HCV) has been considered as a mojor causative agent of post-transfusion related non-A, non-B hepatitis. In this study, the cDNA sequence of NS4 region of HCV (HCV-S) obtained from a Korean patient's plasms was determined. Comparative nucleotide sequence analysis between to type II. 67.2% homology to type III, and 66.4% homology to type IV. The putative amino acid sequence homologies to types I, II, III, and IV were 82.8-84.7%, 92.5-95.1%. 72.5, and 71.1%, respectively. This data strongly suggests that HCV-S should be classified as type II. Significant similarities of hydrophobicity profiles and putative transmembranous domains were found in HCV-S and four major prototypes, indicating that the protein structure is similar in spite of the heterogeneities of intertype homologies at the level of the psrimary nucleotide and amino acid sequences.
We develop a framework to construct geometric representations of finite groups G through the correspondence between real toric spaces $X^{\mathbb{R}}$ and simplicial complexes with characteristic matrices. We give a combinatorial description of the G-module structure of the homology of $X^{\mathbb{R}}$. As applications, we make explicit computations of the Weyl group representations on the homology of real toric varieties associated to the Weyl chambers of type A and B, which show an interesting connection to the topology of posets. We also realize a certain kind of Foulkes representation geometrically as the homology of real toric varieties.
Phospholipase D2 (PLD2) has been implicated in the tyrosine kinase-mediated signaling pathways, but the regulation events are yet to be identified. Herein, we demonstrate that pleckstrin homology (PH) domain of PLD2 (PLD2-PH) exerts an antitumorigenic effect via the suppression of PLD2 and focal adhesion kinase (FAK). The kinase domain of FAK interacts with PLD2-PH and induces tyrosine phosphorylation and activation of PLD2. Furthermore, PLD2 increased tyrosine phosphorylation of FAK. However, ectopic expression of the PLD2-PH competes for binding to FAK and reduces the interaction between PLD2 and FAK, thereby suppressing FAK-induced PLD activation and tyrosine phosphorylation of FAK. The PLD2-PH suppressed the migration and invasion of glioblastoma cells, as well as tumor formation in a xenograft mouse model. This study uncovers a novel role of PLD2-PH as a negative regulator of PLD2 and FAK.
The follicle stimulating hormone receptor (FSHR) is a glycoprotein hormone, that belongs to the GPCR superfamily. FSHR plays a major role in reproduction. The aberrant activation of FHS receptor leads to infertility and several reproductive disorders. The recently recognized roles of the FSHR in diverse extragonadal tissues is also closely related to Alzheimer's disease and cancers. Analysing the structural characteristics of the receptor is important in understanding the pathophysiology of diseases associated with the receptor. In this present study, homology modelling of FSHR-TM domain was developed using four different templates. Totally 20 models were developed using single template-based approach and selected three based on the validation of RC plot, RMSD, ProSA, QMEAN and ERRAT values. The developed models would be useful for further research on the structural characteristics and binding characteristics of the FSHR-TM domain.
P2X receptors are ATP-activated ion channels in the plasma membrane. P2X receptors have a role in a diverse range of disorders, making them a valuable therapeutic target. Hence, the present investigation employed homology modelling of the P2X receptor based on the crystal structure of 5SVJ, 6AH4, 5YVE and 5SVL. Twenty models, using both single- and multiple template-based methods, were developed, and the best model was chosen based on the validation result. We observed that a strategy based on multiple templates provided greater accuracy. Future studies involving binding site and docking analysis can make use of the produced structures.
The gasdermins are a family of recently identified pore-forming effector proteins that cause membrane permeabilization and pyroptosis, a lytic pro-inflammatory type of cell death. A role in the regulation of cell proliferation and/or differentiation is suggested by the differentiation status-specific expression of gasdermin proteins in epithelial tissues. One of the GSDM protein is Gasdermin A (GSDMA), which decreased in stomach and esophageal cancers, suggesting a tumor suppressor role. GSDMA receptor antagonists have been researched as potential treatments for inflammatory diseases and baldness. GSDMA's significance in a wide range of disorders makes it an important therapeutic target. As a result, homology modelling of the GSDMA receptor was undertaken in the current study using the crystal structures of Mus musculus (GSDMA3), Human gasdermin D (GSDMD), and Murine gasdermin D (murine GSDMD). The best model was chosen based on the validation results after 20 models were developed utilising single template-based approaches. The generated structures can be used for further binding site and docking studies in the future.
1985년부터 1993년까지 장원도 일대에서 채집한 20균주와 외부에서 분양 받은 6균주를 대상으로 교배실험, PCR-RFLP 분석, 그리고 RAPD분석을 통해 상호 유연 관계를 알고자 하였다. 교배실험에서는 A. gallica 6균주, A. ostoyae 5균주, A. tabescens 2균주만이 친화성을 나타내었다. A8(KNU-250)균주는 A. gallica와 A. ostoyae에 모두 친화성을 나타내어 배양적 특징과 유사한 결과가 나타났다. rDNA의 IGS영역의 PCR-RFLP분석에서 A. mellea와 A. ostoyae 각 균주들 간의 유사도는 1.000으로 100%의 상동성을 보여주었다. A. tabescens는 $0.919{\sim}0.974$로 높은 유사성을 나타냈다. A. gallica는 $0.619{\sim}1.000$의 유사도로 나타났다. A19와 A22균주는 고립된 한 집단으로 나타났으며 유사도는 1.000으로 100% 상동성을 보였고, 다른 집단과의 유사도가 $0.051{\sim}0.108$로 10% 미만의 상동성을 나타내고 있다. RAPD분석을 통한 A. mellea의 유사도는 $0.983{\sim}1.000$으로 상동성이 높게 나타났으며, A. ostoyae도 $0.994{\sim}1.000$으로 높은 상동성을 보여 주었다. A. tabescens의 균주간의 유사도는 $0.594{\sim}0.953$으로 나타났으며, A. gallica는 $0.280{\sim}0.733$의 유사도를 나타내 균주간의 변이가 나타난 것으로 생각된다. 특히, A19, A22균주가 다른 집단과 거리가 먼 한 집단으로 나타났는데 그들 간의 유사도는 0.921로 92%의 상동성을 보였고, 다른 4개 집단과의 유사도가 $0.012{\sim}0.069$로 7% 미만의 상동성을 나타내고 있다. 본 실험에서 분류된 Armillaria속균은 5종으로 4종은 각각 이미 알려진 종으로 A. mellea, A. ostoyae, A. tabescens, A. gallica로 동정되었고 남은 1종은 미기록종이라고 생각된다.
우리나라 토양으로부터 분리된 (주와 김 2009) poly (butylene succinate-co-butylene adipate; PBSA) 분해균 Burkholderia cepacia PBSA-7, Bacillus licheniformis PBSA-8 및 Burkholderia sp. PBSA-9에서 PBSA 분해효소를 암호화하는 유전자를 분석하였다. PCR을 수행하여 B.cepacia PBSA-7과 Burkholderia sp. PBSA-9는 약 1.5 Kb, B. licheniformis PBSA-8은 약 600 bp의 lipase 유전자(lip A) 절편을 가지는 것을 확인하였다. 세 균주 모두에서 유전자의 염기서열 내 lipase box인 Gly-X1-Ser-X2-Gly와 Ala-X1-Ser-X2-Gly가 존재하였다. B. cepacia PBSA-7의 PBSA 분해효소 유전자는 family I-1 lipases와 36~40%, family I-2 lipases와는 82~92%의 높은 유전적 상동성을 보였다. 또한 B. licheniformis PBSA-8의 PBSA 분해효소 유전자는 subfamily 1-4 lipases와 64~65%의 유전적 상동성을 나타내었으나, subfamily 1-5에 속하는 lipase들과는 거의 유전적 상동성이 없는 것으로 나타났다. Burkholderia sp. PBSA-9의 PBSA 분해효소 유전자도 family I-1 lipases와 35~37%, family I-2 lipases와는 83~94%로 높은 유전적 상동성을 보여 B. cepacia PBSA-7의 PBSA 분해효소 유전자와 유사한 결과를 보였다.
