Antiinflammatory Activity of the Medicinal Plant Geum Japonicum

  • G. japonicum is a perennial hem and the flowering plant has been used as a diuretic and an astringent in Japan and China. However, little information is available about the anti-inflammatory action of G. japonicum. Therefore, the objective of this study was to investigate the antiinflammatory action of fractions from G. japonicum methanol extract. Inhibition of NO production was observed when cells were cotreated with fractions of G. japonicum and lipopolysaccharide. We observed that ethyl acetate fraction of G. japonicum inhibited NO production by LPS-activated RAW 264.7 cells, and that the suppression induced by ethyl acetate fraction of G. japonicum was associated with antioxidant activity and direct NO clearance. In addition, only ethyl acetate fraction of G. japonicum inhibited stimulated $PGE_2,\;TNF-\alpha,\;IL-1\beta$ production, whereas water and methyl chloride fractions showed no such effects. The ethyl acetate fraction of G. japonicum methanol extract showed a remarkable scavenging activity on the 1,1-diphenyl-2 picrylhydrazyl radical. Based on the results, ethyl acetate fraction of G. japonicum may be useful source as natural antioxidants and antiinflammation. Therefore, the results obtained from this study provide an alternative protective mechanism of ethyl acetate fraction of G. japonicum and provide information on the potential use of ethyl acetate fraction of G. japonicum in chemoprevention or pathogenic conditions related to overproduction of NO and $PGE_2$. However, the mechanism of the inflammatory effect must be evaluated through various parameters for induction of NO production.

  • In this study, the neutralization and dechlorination process of MWI(Municipal Waste Incinerator) fly ash with $H_2SO_4$ are investigated to recover HCI, which is delivered from the reaction of chloride in the ash and sulfuric acid. The coarse crystalline gypsum and fine impurity containing heavy metal are also separated by 500# wet screening followed by recrystallization of the dechlorinated ash mainly made of $CaSO_4$. As a results, Using 100g MWI fly ash and 85g cone. sulfuric acid as raw material, 52.6g hydrochloric acid with 35% assay and 116.9g crystalline gypsum with 98% or more assay are recovered. In this process, 7.85g fine impurity containing heavy metal and 2.65g coarse impurity are also separated.

Oxidative stress-induced aberrant G9a activation disturbs RE-1-containing neuron-specific genes expression, leading to degeneration in human SH-SY5Y neuroblastoma cells

  • Oxidative stress-induced neurodegeneration is one of several etiologies underlying neurodegenerative disease. In the present study, we investigated the functional role of histone methyltransferase G9a in oxidative stress-induced degeneration in human SH-SY5Y neuroblastoma cells. Cell viability significantly decreased on H2O2 treatment; however, treatment with the G9a inhibitor BIX01294 partially attenuated this effect. The expression of neuron-specific genes also decreased in H2O2-treated cells; however, it recovered on G9a inhibition. H2O2-treated cells showed high levels of H3K9me2 (histone H3 demethylated at the lysine 9 residue), which is produced by G9a activation; BIX01294 treatment reduced aberrant activation of G9a. H3K9me2 occupancy of the RE-1 site in neuron-specific genes was significantly increased in H2O2-treated cells, whereas it was decreased in BIX01294-treated cells. The differentiation of H2O2-treated cells also recovered on G9a inhibition by BIX01294. Consistent results were observed when used another G9a inhibitor UCN0321. These results demonstrate that oxidative stress induces aberrant activation of G9a, which disturbs the expression of neuron-specific genes and progressively mediates neuronal cell death. Moreover, a G9a inhibitor can lessen aberrant G9a activity and prevent neuronal damage. G9a inhibition may therefore contribute to the prevention of oxidative stress-induced neurodegeneration.

Effect of Stevia rebaudiana on the Bioactive Compounds from Agarwood Leaf (Aquilaria spp.) by Lactic Fermentation and Spray Drying

  • Agarwood (Aquilaria spp) has high economic value. However, essential oil production from agarwood is a time-consuming process. Additionally, agarwood leaves have not been utilized even though they contain various bioactive ingredients. In this study, agarwood leaves were fermented using Lactobacillus plantarum ATCC 8014 with or without Stevia (4, 8, and 12%; v/v). The fermented fluid was mixed with maltodextrin (15%; w/v) and subjected to spray drying (inlet temperature, 120℃; outlet temperature, 65-70℃). The contents of polyphenols, polysaccharides, saponins, and flavonoids and the viability of L. plantarum were determined. Fermentation enhanced the levels of bioactive compounds. The contents of polyphenol (69.19 ± 4.05 mg GAE/g of sample), polysaccharide (20.75 ± 0.98 mg GE/g of sample), saponin (305.23 ± 4.21 mg OAE/g of sample), and flavonoid (7.86 ± 0.72 mg QE/g of sample), and the viability of L. plantarum (8.72 ± 0.17 log CFU/ml) were markedly upregulated in the samples containing Stevia (12%; v/v). This indicated that the supplementation of Stevia during fermentation decreases the fermentation time (9 h), upregulates bioactive compound production in agarwood leaves, enhances microencapsulation during spray drying, and increases the viability of L. plantarum under simulated gastric digestion conditions.

  • G009는 단백다당체로서 분자량 0.5-200만 사이에 광범위하게 분포되어 있으며 구성당은 glucose를 주로하여 galactose, mannose 등으로 구성되었고, 아미노산은 glycine, leucine, alanine 이외 기타 아미노산으로 구성되어 있다. 담도결찰 쥐에서 G009투여군은 대조군에 비하여 간섬유화 유발물질로 추정되는 혈중 Procolagen type III peptide의 감소시켰고 간조직중 총 collagen의 양도 감소시켰다. 또한 조직학적 검사 결과로도 간섬유화의 억제가 뚜렷하였다. 각종 간손상 유발물질에 대하여도 G009투여군은 대조군에 대하여 정상군과 동일한 수준으로 GOT, GPT, triglyceride수치를 보였으며 간조직도 정상군에 가까운 결과를 보였다.

  • 도심지역의 중금속 배출원은 매우 다양하며, 배출 중금속은 물질별 노출 수준, 노출기간에 따라 건강유해영향을 일으킬 수 있다. 따라서 직업적 노출이 없는 도심 지역의 일반인의 혈중 중금속의 모니터링은 독성학적 측면에서 중요하다. 본 연구는 서울 도심지역의 직업적 노출이 없는 성인 남녀 144명(20∼75세)을 대상으로 중금속(납, 크롬, 니켈, 카드뮴)의 혈액 내 축적된 정도를 평가하였다. 채취한 혈액의 분석은 GF-AAS를 사용하였다. 물질별 혈중 중금속 농도는 납 63.2 $\mu\textrm{g}$/L, 크롬 0.12 $\mu\textrm{g}$/L, 니켈 $\mu\textrm{g}$/L, 카드뮴 1.43 $\mu\textrm{g}$/L로 각각 조사되었다.

  • This note discusses the inter-loss time ofan M/G/1/1 queuing system. The inter-loss time is defined as the time duration between two consecutive losses of arriving customers. In this study, we present the explicit Laplace transform of the inter-loss time distribution of an M/G/1/1 queuing system.

  • In this paper, as a generalization of symmetric bi-derivations and symmetric bi-f-derivations of a lattice, we introduce the notion of symmetric bi-(f, g)-derivations of a lattice. Also, we define the isotone symmetric bi-(f, g)-derivation and obtain some interesting results about isotone. Using the notion of $Fix_a(L)$ and KerD, we give some characterization of symmetric bi-(f, g)-derivations in a lattice.
