Cerebral Infarction Model in Rat on Magnetic Resonance Imaging (흰 쥐의 뇌경색 병변에 대한 자기공명영상)

  • It is important to study using experimental animals for research about stroke. Magnetic Resonance Imaging(MRI) is avaluable diagnosticmethods for stroke diagnosis. The purpose of this research is to know the Magnetic Resonance Imaging(MRI) and histopathological characteristics findings after induction of photothrombotic cerebral infarction in rat brain. Male Sprague-Dawley rats were anesthetized, Rose Bengal dye(20 mg/kg) was intravenously injected. The right sensonrimotor cortex of rat brain was exposed to cold light of 7 mm diameter at a position of 1 mm anterior and 3.5 mm lateral to bregma for 20 min. The post-infarction effects were monitored by T1 weighted and T2 weighted images of brain MRI. Histopathological changes were observed after Hematoxylin & Eosin staining. The lesion appeared clearly high signal intensity area on T2 weighted images(the major axis $7.04{\pm}0.11$ mm, the minor axis $3.08{\pm}0.04$ mm) and also H&E staining was same result. In conclusion, MRI was avaluable diagnostic methods for diagnosis and serial changes of stroke.

Effect of Oral Administration of Processed Sulphur on Hepatotoxicity (법제 유황 경구투여가 간독성에 미치는 영향)

  • This study was to evaluate the effects of oral administration of Processed Sulphur on Hepatotoxicity. Processed Sulphur was administered orally to rats for 28 days. We measured the body and liver weight index, heamtological and biomedical parameters. We also observed the histopathological changes of liver in rats. No significant differences in body weight, liver weight index, heamtological and biomedical parameters and histopathological changes of hepatocyte between control and Processed Sulphur fed group were found. Our data indicate that hepatotoxicity was not caused by oral medication of Processed Sulphur up to 60mg/200g/day for 28 days in rats. Therefore, Processed Sulphur appears to be safe and non-toxic in these studies and a no-observed adverse effect level (NOAEL) in rats is established at 60mg/200g/day. The data could provide satisfactory preclinical evidence of safety to launch clinical trial on standardized formulation of mineral extracts.

Single-dose and 4-week repeated dose Toxicity of Aconitum Sinomontanum Nakai Pharmacopuncture: An Experimental Study

  • Background: This study aimed to assess the toxicity of Aconitum sinomontanum Nakai (ASN) pharmacopuncture. Methods: To investigate the toxicity of ASN pharmacopuncture, single and 4-week repeated dose toxicity experiments were conducted on BALB/c mice. In the single-dose toxicity experiment, mice were assigned 1 of 4 groups (5 males, 5 females per group). Then, 31.25, 62.5, and 125 mg/kg of ASN pharmacopuncture were administered to the mice in the experimental groups at acupoint ST36, while 0.2 mL of normal saline was administered to the control group at ST36. After a 4-week repeated dose regimen, the mice were assigned into 4 groups (5 males, 5 females per group). Then, 15.625, 31.25, and 62.5 mg/kg of ASN pharmacopuncture at ST36 were administered to the mice in the experimental groups, while 0.2 mL of normal saline was administered to the control group at ST36. Mortality, morbidity, general body and organ weight changes (after 4 weeks repeated dose), serum hematological and biochemical values, and histopathological changes in the liver and kidney were observed. Results: In both single and 4-week repeated dose toxicity experiments, no deaths or symptoms occurred in any of the groups. There were no significant differences between groups in terms of body and organ weights, serum hematological and biochemical values, and specific organ histopathological changes. Conclusion: ASN pharmacopuncture injection did not demonstrate significant toxicity in BALB/c mice compared with the control group, with a no-observed-adverse-effect level for a single dose of >125 mg/kg, and for 4 weeks repeated dose it was more than 62.5 mg/kg/day.

Histological analysis of five organs in streptozotocin-induced diabetic rats (Streptozotocin 유도 당뇨 흰쥐에서 주요 장부 간 조직학적 변화 비교 연구)

  • Objectives : This study was conduct to compare of histological changes on four target organs which related with diabetes in streptozotocin-induced diabetic rats. Methods : Diabetes was induced in male Sprague-Dawley rats by consecutive injection of streptozotocin (STZ) at different doses of 30, 40 and 50 mg/kg for 5 days. After 4 weeks, all rats were sacrificed, five different organs such as pancreas, liver, kidney, and lung were isolated and observed their histological changes by hematoxylin and eosin (H&E), Periodic acid-Schiff (PAS) and Masson's trichrome staining. The changes of body weight, blood glucose, and food and water intake were also measured. Results : The multiple administration of STZ was induced diabetes in rats with hyperglycemia, decrease of body weight, increase water and food intake, and histopathological changes of target organs, compared with those of normal rats in both dose-dependent and time-dependent manner. In histological analysis, pancreas was showed decrease of the islet numbers with beta-cell loss. Kidney showed morphological damage with glomerulus hypertrophy, and also lung was showed bronchial epithelial damage with inflammatory cells infiltration. In liver, the portal vein and hepatic artery could not observed, and showed inflammatory cell infiltration with liver fibrosis. Conclusions : These results suggest that the increase of the capacity of STZ, each of the more chronic disease, it can be seen that the damage was deep. Thus, evaluate the resulting drug appropriate depending on the purpose of the model is expected to be selected.

Korean Red Ginseng enhances cardiac hemodynamics on doxorubicin-induced toxicity in rats

  • Background: Korean Red Ginseng (KRG) has been known to possess many ginsenosides. These ginsenosides are used for curing cardiovascular problems. The present study show the protective potential of KRG against doxorubicin (DOX)-induced myocardial dysfunction, by assessing electrocardiographic, hemodynamic, and biochemical parameters and histopathological findings. Methods: Animals were fed a standard chow and adjusted to their environment for 3 days before the experiments. Next, the rats were equally divided into five groups (n = 9, each group). The animals were administered with KRG (250 and 500 mg/kg) for 10 days and injected with DOX (20 mg/kg, subcutaneously, twice at a 24-h interval) on the 8th and 9th day. Electrocardiography and echocardiography were performed to study hemodynamics. Plasma levels of superoxide dismutase, catalase, glutathione peroxidase, and malondialdehyde were measured. In addition, the dose of troponin I and activity of myeloperoxidase in serum and cardiac tissue were analyzed, and the histopathological findings were evaluated using light microscopy. Results: Administration of KRG at a dose of 250 and 500 mg/kg recovered electrocardiographic changes, ejection fraction, fractional shortening, left ventricular systolic pressure, the maximal rate of change in left ventricle contraction (-dP/dtmax), and left ventricle relaxation (-dP/dtmax). In addition, KRG treatment significantly normalized the oxidative stress markers in plasma, dose dependently. In addition, the values of troponin I and myeloperoxidase were ameliorated by KRG treatment, dose dependently. And, KRG treatment showed better histopathological findings when compared with the DOX control group. Conclusion: These mean that KRG mitigates myocardial damage by modulating the hemodynamics, histopathological abnormality, and oxidative stress related to DOX-induced cardiomyopathy in rats. The results of the present study show protective effects of KRG on cardiac toxicity.

Histopathological Effects of Tissue Adhesives on Experimental Peripheral Nerve Transection Model in Rats

  • Objective : Our aim was to evaluate the histopathological effects of tissue adhesives on peripheral nerve regeneration after experimental sciatic nerve transection in rats and to search whether these tissue adhesives may possess a therapeutic potential in peripheral nerve injuries. Methods : This experimental study was performed using 42 female Wistar-Albino rats distributed in 6 groups subsequent to transection of right sciatic nerves. Group I underwent external circumferential neurolysis; Group II received suture repair; Group III had local polymeric hydrogel based tissue adhesive administration; Group IV received suture repair and polymeric hydrogel based tissue adhesive application together; Group V had gelatin based tissue adhesive application and Group VI had suture repair and gelatin based tissue adhesive together. After a 6-week follow-up period, biopsies were obtained from site of neural injury and groups were compared with respect to histopathological scoring based on inflammatory, degenerative, necrotic and fibrotic changes. Results : There were remarkable differences between control group and study groups with respect to inflammation (p=0.001), degeneration (p=0.002), necrosis (p=0.007), fibrosis (p<0.001) and vascularity (p=0.001). Histopathological scores were similar between study groups and the only noteworthy difference was that Group V displayed a lower score for necrosis and higher score in terms of vascularization. Conclusion : Our results imply that tissue adhesives can be useful in repair of peripheral nerve injuries by decreasing the surgical trauma and shortening the duration of intervention. Results with gelatin based tissue adhesive are especially promising since more intense vascularity was observed in tissue after application. However, trials on larger series with longer durations of follow-up are essential for reaching more reliable conclusions.

Nephrotoxicity of Acetaminophen and Gentamicin in Combination in Rats

  • Acetaminophen (APAP) and gentamicin are widely used for many patients, but little in-formation is available regarding the combined effects of APAP and gentamicin. This study was aimed to investigate the potent nephrotoxicity following combined-treatment with APAP and gentamicin. Serum biochemical parameters and histopathological changes in the kidney were observed in female SD rats after continuous daily treatment with either 600 mg/kg/day APAP, and/or 300 mg/kg/day gentamicin for 3 days, and compared with saline sham-treated control animals. APAP and gentamicin combination-treated rats exhibited inconsistent increasing tendency in blood urea nitrogen (BUN) by 96 hours after the last treatment, compared to control or the animals treated with each drug. The relative kidney weights were also increased. Histopathological findings of kidneys revealed that necrosis of proximal convoluted tubules were higher in rats treated with APAP and gentamicin combination than the rats treated with each drug alone. These results suggest that combination use of both drugs have more severe nephrotoxicity than treating each drug alone.

  • Endocrine disrupting chemicals probably cause the cytological or/and morphological changes of germinal cells in gonad. Accordingly, this study was aimed to make sure that the effect of hormone-mimicking chemicals on gonad morphology such as decrease of germinal cells, inhibition of cellular maturation and change in the ratio of germinal cells in the different developmental phase can be observed by histopathological procedures and can be a useful bio-indicator for the evaluation of endocrine disruption by environmental chemicals. In this experiment, female Japanese medaka were exposured to quercetin, a phytoestrogen, at the concentration of 100 $\mu\textrm{g}$/L. quercetin showed the significant decrease in the number and rate of vitellogenic follicular oocytes in the treated group for 4 and 6 weeks. The weak development of yolk could be also observed. We could conclude that quercetin has anti-estrogenic or androgen-like potency by exerting the inhibition effect on oogenesis in fish female- gonad. From the result of this study, the applied methods and techniques can be evaluated to be a useful biomonitoring means for water pollution, expecting a good result of the subsequent study on apoptosis.

  • Acute toxicities of DWH-01 (Ranitidine : Bismuth : Sucralfate= 1.5: 2 : 6) were investigated in Sprague-Dawley rats. Seven days after oral administration of DWH-01 with different doses (10 g/kg, 5 g/kg, 2.5 g/kg, 1.25 g/kg, 0.625 g/kg), we examined numbers of deaths, general signs, weight measurement and histopathological examination for both sexes of rats. Summaried results are: 1) No deaths were occurred, 2) There were no pathological and clinical differences compared with control group, 3) No significant changes of body weights were observed, and 4) In histopathological examinations of organs and tissues, there was some hemorrhage in a lung tissue of low dose group for male and female respectively, but it was thought to be caused by environmental factor. The results suggest that toxicity of DWH-01 is low and its $LD_{50}$ is over 10 g/kg in Sprague-Dawley rats.

Anti-Allergic Effect of Ulmus davidiana Cortex on Contact Dermatitis Induced by Dinitrofluoro-Benzene in Mice

  • Objective: The root bark of Ulmus davidiana var. Japonica (Ulmi Radicis cortex, URC) is a medicinal herb used for promoting diuresis and treating dampness. In Korea, URC has long been used as an efficacious therapy for inflammation, burns, frostbite and skin diseases such as eczema and psoriasis. Methods: In the present study, we used 1-fluoro-2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis (CD) mouse model to investigate the anti-allergic and the anti-inflammatory effects of URC on skin lesion, histopathological changes and specific antibody production. Results: URC treatment, 10 mg/mL, effectively inhibited skin lesions induced by repeated paintings with DNFB. In the histopathological observation, topical application of URC inhibited spongiosis. In addition, URC lowered the production levels of total immunoglobulin and IgG2a in serum. Conclusion: These data indicate that URC has an anti-inflammatory effect that produces an improvement of skin lesions in CD mice.
