  • This study was to investigate the effects of maternal diabetes on the lung tissue of the fetal rat using lectin histochemistry and electron microscope technique. Maternal diabetes was induced by intraperitoneal injection of streptozotocin (75 mg/kg the body weight) into pregnant Sprague-Dawley rats on the 7th day of gestation. Fetuses of streptozotocin induced diabetic rats exhibited delayed lung maturation and reduced air space. In lectin histochemistry, the binding of Maclura pomifera (MPA) to fetal lungs from diabetic mothers was reduced, but no significant changes in the bindings of Concanavalin A (Con A), Wheat germ agglutinin (WGA), Ricinus communis I (RCA I) and Griffonia simplicifolia (GSI-$B_4$) were noted. Because the MPA has affinity to terminal N-acetyl-D-galactosamine residues constantly linked O-glycosidically to serine or threonine, the present findings may indicates that maternal diabetes interfere with the processes of O-linked glycosylation in fetal rat lung.

Histology and lectin histochemistry in the vomeronasal organ of Korean native cattle, Bos taurus coreanae

  • The vomeronasal organ (VNO) is critical for reproduction and social behavior in ruminants, including cattle. The present study examined the structure of the VNO and its epithelial cells in neonatal and adult Korean native cattle (Hanwoo), Bos taurus coreanae, using immunohistochemistry and lectin histochemistry. Histologically, the VNO comprised two types of epithelia: medial vomeronasal sensory (VSE) and lateral vomeronasal non-sensory epithelia (VNSE). Numerous blood vessels and nerve bundles were observed within the vomeronasal cartilage encased lamina propria. Immunohistochemistry revealed high expression level of protein gene product9.5 and moderate expression level of olfactory marker protein in the neuroreceptor cells of the VSE and occasionally in some ciliated cells of the VNSE in both neonates and adults. The properties of the glycoconjugates in the VNO were investigated using 21 lectins, most of which were expressed at varied intensities in the VSE and VNSE, as well as in the lamina propria. Several lectins exhibited variations in their intensities and localization between neonatal and adult VNOs. This study is the first descriptive lectin histochemical examination of the VNO of Korean native cattle with a focus on lectin histochemistry, confirming that the VNO of Korean native cattle is differentiated during postnatal development.

A Study on the Epidermal Structure and Mucosubstance Histochemistry of Spotty Belly, Devil Stinger, Stone Fish, Cubed Snailfish, and Japanese Bluefish

  • Structure and mucosubstance histochemistry in the epidermis of five teleostean species, i. e., spotty belly, Agrammus agrammus, devil stinger, Inimicus japonicus, stone fish, Erosa erosa, cubed snailfish, Liparis tessellatus, and Japanese bluefish, Scombrops boops were investigated. The epidermis of five species studied is composed of three layers: superficial, middle, and basal layer. The superficial layer is comprised of rather flattened cells. Mucous cells, the type commonly found in fishes are completely lacking in the epidermis of devil stinger, stone fish, and cubed snailfish. The epidermis of devil stinger and stone fish have multicellular glands which do not have mucosubstances. The skin surface of them is covered with mucous layer. The superficial cells in the epidermis of devil stinger, stone fish, and cubed snailfish are mucus-secreting cells. The composition and the amount of the mucosubstances vary in species and body regions. The mucous layer on the skin surface and superficial epidermal cells of devil stinger contain a mixture of neutral and acidic(sulfated and nonsulfated) mucosubstances. In stone fish, the mucous layer has acidic(sulfated and non-sulfated) mucin and the superficial epidermal cells contain neutral mucin. In cubed snailfish, the type of epidermal mucosubstances is identified as a mixture of neutral and acidic(non-sulfated) mucin. The mucous cells of the epidermis in spotty belly and Japanese bluefish contain neutral mucin.

메기 송과체의 신경세포에 관한 조직화학 및 전자현미경적 연구 (Histochemical and Electron Microscopic Study on the Nerve Cells of the Pineal Body of Catfish, Parasilurus asotus)

  • This study was carried out to clarify the neuronal organization in pineal body of catfish. The pineal body was observed by acetylcholinesterase histochemistry and electron microscopy. The neuronal types observed in the pineal body of catfish were bipolar and multipolar cells. Multipolar cells were found throughout the pineal end vesicle and whole stalk, but bipolar cells only in the end vesicle and distal stalk. The pineal tract was formed by the long axons of these neurons. The neuronal clusters(pineal ganglia) were also observed in the end vesicle. In summary, the type of neurons in catfish pineal was different from that of other species, and the neuronal distribution differed depending on the region of pineal body. These results reflect the interspecific and regional differences of the pineal organization of fishes.

  • 본 연구는 인삼에 존재하는 calcium oxalate 결정체의 분포 위치, 형태 및 미세구조에 관하여 밝히고자 광학현미경, 조직화학적 방법 및 주사 전자현미경적 방법 등을 사용하여 연구 하였다. 인삼의 결정체는 calcium oxalate의 성분으로 구성되며, 결정체는 몸통뿌리, 지근, 뇌두, 줄기, 장엽병, 소엽병, 꽃대 등 대부분의 기관에서 관찰되었다 특히 지상부는 잎에서,지하부는 뇌두에서 가장 많은 결정체가 관찰되었다. 인삼에 존재하는 결정체는 silver nitrate-rubeanic acid를 사용한 조직화학적 반응을 실시한 결과 calcium oxalate치 성분으로 확인되었다. 또한 주사전자현미경으로 관찰한 결과 삼각뿔 형태의 결정체가 다수 모여서 형성된 유형, 프리즘 형태의 소결정이 모여서 형성된 유형 및 얇은 판상의 소결정이 모여서 형성된 유형 등 3가지 형태로 구분 할 수 있었다.

신생아 Hirschsprung's Disease에서 Barium Enema와 Acetylcholinesterase 조직화검사법을 이용한 진단적 접근 (Diagnostic Strategy Using Barium Enema and Rectal Suction Biopsy with Acetylcholinesterase Histochemistry in Neonates with Suspious Hirschsprung's Disease)

  • To investigate the diagnostic accuracy and applicability of barium enema(BE) and rectal suction biopsy with acetyl cholinesterase(AChE) histochemistry in the diagnosis of neonatal Hirschsprung's disease(HD), we retrospectively reviewed the findings of BE and AChE staining in 96 neonates with suspected HD during a 10-year period from January 1991 to December 2000. Sixty-nine cases of HD(58 males and 11 females) and 27 cases of non-HD are included in this study. In regard to BE, HD was based on definite transitional zone, suspicious HD on reversed rectosigmoid index(RSI <1), and non-HD on normal RSI(RSI>1). The histochemical criterion used for the diagnosis of HD was that of Chow et al(1977), i.e., the presence of many coarse discrete cholinergic nerve fibers in the muscularis mucosae and in the immediately subjacent submucosa regardless of infiltration of cholinergic nerve fibers in the lamina propria. Of 66 neonates with HD who underwent BE, transitional zone was identified in 33 cases(50 %) and reversed RSI in 19 cases(21 %), microcolon in 4 cases and normal finding in 10 cases(15 %) while of 27 neonates with non-HD, there was normal finding in 16 cases and reversed RSI in 9 cases(41 %). Thus diagnostic accuracy based on transitional zone was 64 %. The positive predictive value of reversed RSI for the diagnosis of HD was 68 %. Of 42 neonates with HD who underwent AChE histochemistry, there were 41 AChE-positive reactions and one AChE-negative reaction in a neonate with total colonic aganglionosis, while of 27 cases of non-HD, there were one equivocal AChE-positive reaction and 26 AChE-negative reactions. Thus AChE histochemical study showed a 97 % diagnostic accuracy with a 98 % sensitivity and a 96 % specificity. In conclusion, we believe that BE is valuable as a first diagnostic step since about 80 % of neonates with HD show significant radiologic findings such as a transitional zone or reversed RSI. Lectin histochemistry of the olfactory mucosa of Korean native cattle, Bos taurus coreanae

  • Background: The olfactory mucosa (OM) is crucial for odorant perception in the main olfactory system. The terminal carbohydrates of glycoconjugates influence chemoreception in the olfactory epithelium (OE). Objectives: The histological characteristics and glycoconjugate composition of the OM of Korean native cattle (Hanwoo, Bos taurus coreae) were examined to characterize their morphology and possible functions during postnatal development. Methods: The OM of neonate and adult Korean native cattle was evaluated using histological, immunohistochemical, and lectin histochemical methods. Results: Histologically, the OM in both neonates and adults consists of the olfactory epithelium and the lamina propria. Additionally, using periodic acid Schiff and Alcian blue (pH 2.5), the mucus specificity of the Bowman's gland duct and acini in the lamina propria was determined. Immunohistochemistry demonstrated that mature and immature olfactory sensory neurons of OEs express the olfactory marker protein and growth associated protein-43, respectively. Lectin histochemistry indicated that numerous glycoconjugates, including as N-acetylglucosamine, mannose, galactose, N-acetylgalactosamine, complex type N-glycan, and fucose groups, were expressed at varied levels in the different cell types in the OMs of neonates and adults at varying levels. According to our observations, the cattle possessed a well-developed olfactory system, and the expression patterns of glycoconjugates in neonatal and adult OMs varied considerably. Conclusions: This is the first study to describe the morphological assessment of the OM of Korean native cattle with a focus on lectin histochemistry. The findings suggest that glycoconjugates may play a role in olfactory chemoreception, and that their labeling properties may be closely related to OM development and maturity.

Histochemistry of Mucosubstances on the Pedal Sole of Five Abalone Species

  • The present study describes histochemical properties of the mucosubstances in the pedal sole of abalone, in relation to two main additional functions known to date. Only neutral mucosubstances were demonstrated in the lateral epithelium of the pedal sole of the abalone, however, two distinct types of cells, one containing neutral and the other containing acidic mucosubstances, were identified in the ventral epithelium. The epithelial mucocytes distributed throughout both epithelial and subepithelial glands located in the ventral subepithelial region showed a mixture of neutral and acidic mucosubstances. However, acidic mucosubstances were found in larger amounts than neutral ones in epithelial mucocytes, and vice versa in subepithelial glands. Among these acidic mucosubstances, sulphated substances were dominant. Lesser amounts of carboxylated substances also occurred together with low levels of neuramic and sialic acid. Collectively, the notable morphological differences between abalone and other gastropoda were observed in the ventral surface of the foot, which showed a tall columnar epithelium, peripherally concentrated mucocytes, and subepithelial glands. Mucosubstances are presumably produced by these cells and may also play a role in clinging to some substances to support abalones in addition to the main functions of lubrication and protection.
