Study of Histochemical Changes of Mouse Ovary Under X-ray Irradiation (방사선 조사선량에 따른 생쥐 난소의 조직화학적 변화)

  • This research investigated the morphological changes of folliculus ovary according to the dose of the X-rays when mice were exposed to X-rays, in terms of histochemical reaction. Dyeing methods such as Masson's trichrome method and reticular fiber method were used. The morphological changes of growing folliculus ovary got distinct as the dose of X-rays increased. Especially, the nuclei of granular cells showed manifest condensation and the changes of the transparent zone were distinct. As a result of histochemical reaction according to Masson's trichrome method and reticular fiber method, the changed granular cells, the deformed basilar membrane of folliculus ovary and the abnormal arrangement of the reticular fiber were observed.

  • Histochemical observations on the effect of gold alloys crown margin in the periodontal membrane were caried out by use of rabbits. The animal were setted with gold alloys crown on the incisor teeth. The cervical margins were given extence into gingival sockets. The animals were sacrificed at the 30 days experimented period. Specimens of tissue were obtained from the mandibular incisor teeth with jaw. All tissues were fixed immediately in 10% neutral formalin solution. Different histochemical staining methods for the determination of fibrous components were the Mallory-Heidenhain Bielschowsky-Gomori, Gomori's aldehyde fuchsin. Periodic acid-Schiff reaction and Hematoxylin eosin stanin method. The results were as follows. The periodontal membrane is composed primarily of collagenous fibers and fibrous component run functional arrangement but in lower gold content groups, horizontal fiber groups were irregularly run. Elastic fibers were obligue run in experimental groups. There were no significant differences in reticularfibers in distribution.

Identification of Root Age by Histochemical Staining of Secretory Duct Layers in Ginseng (인삼 분비도관의 조직화학적 염색에 의한 연근판별)

  • Identification of the age of ginseng root is very important in commercial market as well as in research field. However, any criterion abut it has not been clearly established yet. We studied to find a clear morphological key for identification of ginseng root\\`s age using the histochemical staining method. Fresh sections of 3, 4, 5, and 6 year old roots were stained with 1% nile blue, observed under the light microscopy, and compared each other. The number of secretory duct layers(SDL) is a useful key to confirm the age of ginseng root as follow; three-year-old root has two, four-year-old one has three, fie-year-old one has four, and six-year-old one has five resin duct layers on each cortical region of roots. Secretory ducts are thought to be formed by the vascular cambium every year. Unlike the surrounding parenchyma cells, secretory epithelial cells lack starch grains in the cytoplasm.

  • The effect of pyridoxine on the incisors were histochemically studied for the purpose of identifying the phosphoric acid. The microscopic observations were carried out by means of phosphoric acid reaction(Serraet Queiroz-Lopes method), mucoprotein reaction (Clara method), periodic acid-Schiff reaction (McManus method), methylen blue stain and hematoxylin-eosin stain. In the experimental result, it was found that phosphoric acid reactions of ameloblast, Tomes' process and preenamel were remarkably increased after pyridoxine administration.

Purification and Characterization of Glutathione Peroxidase Isolated from Rat Erythrocyte and Histochemical Study of its Localization in Liver of White Rat (흰쥐 적혈구에 있는 Glutathione Peroxidase의 순화 및 성질과 간에서의 용작부위에 대한 조직화학적 연구)

  • A glutathione peroxidase from white rat (Wistar strain)erythrocytes was partially purified and characterized. In addition, localization of this enzyme in the liver was studied by histochemical method. A glutathione peroxidase was purified approximately 33.5-folds by ammonium sulfate precipitation, Sephadex filtration column and DEAE-Sephadex column chromatography. The optimum temperature of the crude glutathione peroxidase was $40^\\circC$, and the optimum pH was 7.5. This crude glutathione peroxidase was most stable at $30^\\circC$ and the values of Km and Vmax were calculated to be 8.5mM and 15.6 $\\mu$moles/min for glutathione, and 40 $\\mu$M and 10.5 $\\mu$moles/min for hydrogen peroxide, respectively. The molecular weight of this enzyme was estimated by Sephadex G-200 gel filtration to be approximately 90, 000. By electron microscopic examination, histochemical reaction products were microbodies that were prominent in the peripheral parts of the lobule. The reaction products exhibited round shapes, the diameter of which varied $0.2\\sim0.7 \\muM$ and their boundary membranes were not distint.

Enzyme-Histochemical Study of Philtral Orbicularis Oris Muscle Fiber Types in Korean Male Cadaver (한국인 성인남성 사체에서 시행한 인중 구륜근 섬유들의 효소-조직화학적 분석)

  • The orbicularis oris muscle (OOM) is a very important muscle that originate from the second branchial arch and is innervated by the facial nerve. The aim of this study was to elucidate distribution types of two muscle fibers that composing OOM by using enzyme-histochemical examinations and tried to make a basis for a clinical application. The fresh frozen tissues from the superior and inferior portions of the OOM were taken from post mortem 65-year-old Korean male adult. Total five different sagittal sections were used on the midline of the philtrum, the middle portion of lower lip, the mouth corner, and each midlateral side of upper and lower mouth. We used enzyme-histochemical staining such as Periodic Acid-Schiff (PAS), Succinic Dehydrogenase (SDHase), reduced Nicotinamide Adenine Dinucleotide-Tetrazolium Reductase (NADH-TR), Adenosine Triphosphatase (ATPase) in pH 9.4, 4.6 and 4.3, and Modified Gomori Trichrome. There were about 30.24 % type 1 muscle fiber and 65.40 % type 2 muscle fiber in the midline of the philtrum (p < 0.05). Enzyme-histochemical staining is very useful and innovative method to elucidate characteristics of muscle fibers. We expect that chiloplasty and reconstruction of the lip portions for cleft lip patients, based on these results, are better to recovery function and aesthetic. However, we have some problems as an intramuscular variability and the inter-individual variation etc. Therefore we have to make progress these studies continuously to overcome these problems.

Pathological Studies on Experimental Poisoning of Organic Phosphate Insecticide (유기인제 중독에 관한 병리학적 연구)

  • The authors believe that farm livestok will be greatly affected by the marked increasing use of organic phosphate. This study was carried out to observe the clinical signs and histopathological changes of mouse, guinea pig, hamster and rabbit that were orally administered with diazinon used usually as agricultural insecticide, and cholinesterase (ChE) activity was histochemically examined in the liver, heart, kidney, adrenal gland, duodenum and salivary gland of these experimental animals administered with diazinon. The results obtained were as follows: 1. Clinical signs such as dullness, severe salivation, ataxia, dyspnea, irregular slight convulsion and inappetance and as the histopathological changes cloudy swelling, congestion and hemorrhage of parenchymal organs, catarrh or local necrsois of the gastrointestinal tract, congestion or hemorrhage of the other organs were observed. Especially, hemorrhage of adrenal glands (rabbit, guinea pig) and pulmonary congestion and hemorrhage were necessarily constant. 2. In the histochemical study, ChE activity appeared intensely in the liver, heart, medulla of adrenal glands and salivary glands (submaxillary and parotid) of control animals, but ChE activity was negative or markedly decreased in experimental animals administered with diazinon. There was no marked difference between the control and experimental animals in ChE activity of the kidney. 3. Histochemical observation of ChE activity was helpful to explain the clinical signs and histopathological changes and was regarded as a diagnostic method for organic phosphate poisoning.

Morphological and Histochemical Study on the Salivary Gland of Korean Slug(Incilaria fruhstorferi) (한국산 산민달팽이 (Incilaria fruhstorferi)의 타액선의 형태 및 조직화학적인 연구)

  • The results of observation on the salivary gland and salivary secretory duct of Korean slug, Incilaria fruhstorferi, in histochemical method are as follows. It is observed that there are six kinds of gland cells(Type-A, B, C, E and F) making up the salivary gland of Korean slug. Of those, type-A gland cell is observed as an acid mucous cell, and type-B, C, D, F gland cells as neutral mucous cells. But in type-E gland cell, while the membrane surrounding granules exhibit alcianophilia, granules show no reaction. The salivary secretory duct composing the salivary gland of Korean slug is composed of supporting epithelial cell and four kinds of gland cells(type-A, E, F and G), of which type-A, E, F gland cells compose both the acinus of salivary gland and endothelial tissue of salivary secretory duct, and are secreted into lumen through salivary secretory duct. But, type-G gland cell is observed only in the endothelium of salivary secretory duct and mucous granules are observed as neutral mucopolysaccharide.

  • By means of Alizarin zirconium stain method the fluoric reaction which showed in the case of deficient hard tissue was observed. Among the hard tissue. the fluoric density appesrs in the graduation that the largest is primary calcified matrix and least is secondany calcified matrix.
