Amperometric Determination of Histamine using Immobilized Enzyme Reactors with Different Carriers (담체 고정화 효소 반응기를 이용한 Histamine의 전기화학적 측정)

  • Histamine is a kind of primary biogenic amine arising from the decarboxylation of the amino acid L-histidine. The toxicology of histamine and its occurrence and formation in foods are especially emphasized in fermented foods. In this study, the biosensor for detection of histamine with functionalized multi-walled carbon nanotubes (MWCNT) was developed. We also searched for an appropriate insoluble substrate to immobilize the enzyme. The developed biosensor showed a detection limit of $0.1{\mu}M$ hydrogen peroxide. The enzyme reactor was prepared with diamine oxidase immobilized on insoluble carriers including CNBr-activated sepharose 4B, calcium alginate, and controlled pore size glass beads. The coupling efficiency of CNBr-activated sepharose 4B, calcium alginate, and controlled pore size glass beads were 48.5%, 40.3%, and 51.0%, respectively. In addition, the response currents on histamine with each immobilized enzyme reactor prepared with CNBr-activated sepharose 4B, calcium alginate, and controlled pore size glass beads were 120 nA, 110 nA, and 140 nA at $100{\mu}M$ of histamine concentration, respectively. Therefore, it is suggested that controlled pore size glass beads are the best carriers for immobilizing diamine oxidase to detect histamine in this biosensor.

Free Amino Acid Composition of Tap Root in Panax species (종별 동체 유리아미노산)

  • Free amino acid (FAA) compositions in the central part (pith-xylem : P-X) and the outer part (phloem-cortex : P-C) of root were investigated for P ginseng (P.g), p. quinque-folium (P.q) an, B P nutoginsen (P.n) by an amino acid analyzer. Total free amlno acids content (TFAA) was highest in p.맥 and lowest in p.n. The TFAA of P-Xs were higher than those of P-Cs in these Panax species except p.n. The higher the TFAA in P-X, the higher the ratio of TFAA in P-X to that in P-C. Seven- teen free amino acids and ammonia were identified, and four unknown peaks appeared before the usual amino acids eluted. The total aspartic acid equivalent of these unknown peaks was corresponded to 77% of known TFAA in P-C of p.n, 17% in P. n, and 7% in p.q. The pattern of unknown peaks of p.g was different from p.q and similar to P.n. In all samples six major amino acids and ammonia accounted for 90~95% of TFAA. Arginine was comprised from 29% (P.n) to 43% (P.g) by amole as amino acid and from 50 to 71% by amole as nitrogen (N amole) in TFAA. Ammonia was the second abundant one by amole and the third by Npmole. Histidine was the second by Npmole. Praline was one of major FAA in p.q. Pattern similarity of FAA composition (excluding Arg and Am) by simple correlation was closer between P-C of p.g and P-X of p.q than between both P-Xs and quite different between the P-X of p.g and that of p.n. The pattern similarities of major FAA percent abundance excluding Arg and Am were significant only between P-X and P-C of the same species. Arginine content (amole) had positive correlation (r=0.859, p=0.05) with Arg/Am among species. Ammonia content was higher than arginine in p.n. Tryptophan content was greatest in p.n among species and higher than lysine only in p.n. The ratios of TFAA to N(W/W) were in the range of 3.89~4.14 for TFAA and 3.61~3.92 for TFAA plus ammonia.

  • To utilize deteriorated sweet persimmon(Diospyros kaki T.) effectively, this study was investigated about the effective (Dying method of the powder. Chemical components, minerals, fatty acids and amino acids of hot-air dried and freeze dried conditions were determined Sample conditions used on analysis as follow; pretreatments of hot-air dry and freeze dry were soft, soft+peel, mixer, mixer+peel. The contents of crude protein, crude lipid, crude ash of hot-air dry were 0.9-1.1%,15.0-39.0% and 2.3-3.3%, respectively. And those of freeze dry were 1.3-2.2%, 27.-49.0% and 2.5%, respectively. Potassium, magnesium, phosphorus, calcium and sodium content in hot-air dry and freeze dry were high. Other minerals were less than 3.00ppm in all conditions. The major fatty acid contents were detected capric acid(C$\^$10:0/), lauric acid(C$\_$l2:0)/, tridecanoic acid(C$\_$13:00/), palmitic acid(C$\_$l6:00/), palmitoleic acid(C$\_$l6:1/), oleic acid(C$\_$18:1), linoleic acid(C$\_$18:2/), linolenic acid(C$\_$18:3/). The essential amino acids such as aspartic acid, threonine, serine, glutamic acid, glycine, alanine, valine, methirmine, isoleucine, leucine, tryosine, phenylalanine, histidine, lysine, arginine in freeze dry were contained richly. On the basis of chemical analysis, hot-air dry method will have to supply the additional different method, because simple hot-air dry method was shorten the dry time but had many problems. Therefore, the effective drying method considering changed color and nutrition was shown freeze dry method.

STUDIES ON THE IDENTIFICATION OF Pleioblastus Simoni IN KOREAN BAMBOOS (Pleioblastus Sinomi Nakai (해장죽)의 동정의 관한 연구)

  • Studies on the anatomical observation of Bamboos distributed in East-South Asia for systematic botany were scarecely reported except for the morphological studies on Bamboos, which was investigated by Dr. Hiroshi Usui, present author and others. Recently it has been certain that it could be hardly classify some species of bamboo family without this work. For this reason the author reported the anatomical studies on Korean bamboos in 1959, and furthermore the author intended to identify the two species of Pleioblastum Simoni Nakai in Korean bamboos by studying the characteristics of internal structure and qualitative assay of free amino acid containg in Pleioblastus Simoni Nakai(1), (2). In view of fact that there is some differences between two species, it is likely to be identifiable completely different species one another. Furthermore, a lot of characteristics surveyed by the author were as follow: It was reassumed that two species cultivated under same conditions for three years were different alternately in the species of Pleioblastus Simoni Nakai. At the same time, these studies could clarify some evolutional processes of bamboo family. On the other hand, it was found that two species had not only the obvious difference in aerenchyma, the size of thickwalled parenchyma and bulliform cell, but also the internode of Pleioblastus Simoni Nakai(1) is longer than the other, the speed of growing is more rapid, the leaf of former is narrow and that of latter is wide. The free amino acids containg in each species were found as 18 kinds, especially Pleioblastus Simoni Nakai (2) had not Histidine in spite of containg in Pleioblastus Simoni Nakia(1). From the characteristics and the experiments described above, it seemed to the author that Pleioblasus Simoni Nakai (1) which was growing at the region of Ulsan in Korea.

Effect of Alcohols Toward the Transphosphatidylation Activity in Phospholipase D Catalyzed Reaction (포스포리파제 D 촉매반응에서 포스파티딜 전달반응 활성에 미치는 알코올의 영향)

  • In the presence of alcohol, phospholipase D (PLD) is known to perform transphosphatidylation activity, during which the overall reaction rate of PLD increased. To elucidate the reaction mechanism of transphosphatidylation further, we investigated rate constants of transphosphatidylation reaction of the purified ${\alpha}$-type PLD from cabbage in the presence of various alcohols. The second-oder rate constants of PLD transphosphatidylation showed a large increase with the primary alcohols examined as expected. In the case of butanol we observed the second-oder rate constant of $33.33{\pm}1.33M^{-1}sec^{-1}$. This second-order rate constant of transphosphatidylation was as 400 times greater as the second-order hydrolysis rate constant of $0.078M^{-1}sec^{-1}$ which was adjusted for the water concentration. A linear free energy relationship between the $pK_a$ of alcohol and transphosphatidylation rate gives a Br${\o}$nsted slope of ${\beta}_{nu}$ = 0.12 ${\pm}$ 0.03. This small ${\beta}_{nu}$ value implicates that the transition state of break down of phosphatidyl-enzyme intermediate (E-P) is likely dissociative. Finally, a reaction mechanism of cabbage PLD is suggested on the basis of our results presented here and the histidine residue known to be located in the active site of cabbage PLD.

Physicochemical Composition of Ramie Leaves (Boehmeria nivea L.) (모시잎의 이화학적 성분)

  • We quantitated the major chemical components of ramie leaves (Boehmeria nivea L.) powder. The proximate compositions (all w/w) was 5.42% moisture, 28.15% crude protein, 6.95% crude fat, 15.27% crude ash, and 54.79% carbohydrate, respectively. The total, insoluble, and soluble dietary fiber contents were $39.66{\pm}1.84g/100g$, $20.32{\pm}2.02g/100g$, and $19.34{\pm}2.84g/100g$, respectively. The major free sugars were glucose, galactose and lactose. Seventeen amino acids were isolated. Essential amino acids constituted 44.65% of the total. When free amino acid levels were evaluated, 25 kinds of components were detected, of which 18.15% were essential. Only caproic acid methyl ester and pentadecanoic acid were detected when fatty acid examined. Composition of Amino Acids in Domestic and Foreign Garlic Cultivars (국내외 재배종 마늘의 아미노산 조성)

  • Correlation and similarity of garlic cultivars was analyzed by measuring the composition of twenty amino acids contents in the bulb and callus from twelve garlic cultivars which were collected from Korea and foreign countries. Arginine and asparagine occupied more than 78% of total amino acids both in bulb and callus followed by glutamic acid, lysine, aspartic acid, valine, glycine, histidine and ornithine in order. Based on correlation coefficients of amino acid composition in the bulb, twelve garlic cultivars were classified into three major groups. Danyang, Euiseong, Seosan, Jeju, Mongol and Turkey cultivars belong to group-I, Namdo, China, Mexico and Nepal cultivars are in group-II, and group-III includes Philippine and Daeseo cultivars. Based on amino acid composition in callus, group-I includes Mongol, Euiseong, Danyang and Seosan cultivars, group-II includes Jeju, Daeseo, China and Namdo cultivars, group-III includes Napal, Mexico, Philippine and Turkey cultivars. Composition of amino acid contained in both callus and bulb is a clear standard to identify northern-type garlic cultivars. Especially the composition of amino acids in callus is more distinctive standard of classification between northern and southern type garlic cultivars than that in bulbs.

Study on the Characteristic of Physicochemical Quality of Oolong herbs tea by Extraction Conditions (추출조건에 따른 우롱차의 이화학적 품질특성에 관한 연구)

  • The extraction condition and quality attributes components in Oolong tea were investigated. Extraction yield was high in $80{\sim}85^{\circ}C$ above 50% ethanol solution. Sucrose content most high among the free sugar ranged $37.2{\sim}55.0\;mg/100\;g$, while arabinose was the least. Organic acids in ethanolic extracts were furmaric, citric and malic acid. Free amino acids were 15 kinds and contents of proline, tyrosine and glutamic acid were comparatively high. Tannin content extracted from water and 25% ethanol solution were 38.6 and $38.5\;{\mu}g/100\;g$, it decreased as ethanol concentration increase. Caffeine content did not changed as extraction conditions. Ascorbic acid content was $6.5\;{\mu}g/g$ when extracted from 25% ethanol solution, it decreased as ethanol concentration increase. Bitter and astringent taste affected to overall preference of Oolong tea. Tea manufactured from 25% ethanol solution extracts recorded most high organoleptic score than any other extraction condition.

Characterization and Cloning of a Phytase from Escherichia coli WC7. (Escherichia coli WC7가 생산하는 Phytase의 효소특성과 그 유전자의 클로닝)

  • Phytase from Escherichia coli WC7 was purified from cell extracts and its molecular mass was estimated to be 45 kDa by SDS-PAGE. Its optimum temperature and pH for phytate hydrolysis was 6$0^{\circ}C$ and pH 5.0, respectively. The enzyme was stable up to 6$0^{\circ}C$ and over broad pH range (pH 2-12). The enzyme had higher affinity for sodium phytate than p-nitrophenylphosphate (pNPP). That is, the apparent Km value for sodium phytate and pNPP were $0.15\pm$0.02 mM and 2.82$\pm$0.05 mM, respectively. The gene encoding the phytase was cloned in E. coli XL1-Blue. Sequence analysis showed an open reading frame of 1241 Up encoding a signal peptide (22 aa) and a mature enzyme (410 aa). WC7 phytase was expressed up to 17.5 U/ml in the transformed E. coli XL1-Blue/pUEP, which was 23-fold higher than the activity from wild strain.

Development of Growing Pig Diets for Friendly Environment by Using Phytase (인분해효소를 이용한 환경친화성 육성돈사료개발)

  • 본 연구는 육성돈에 있어 인분해효소인 phytase의 첨가가 환경친화성사료개발을 위한 기초자료로 사용하기 위해 영양소 이용률을 측정하였다. 대조구는 옥수수-대두박의 육성돈 사료이고, 처리구는 대조구 사료에 Phytase의 첨가 수준을 500 unit 및 1,000 unit을 첨가한 구로 3×3 Latin Square 방법으로 실시하였다. 건물과 질소에 대한 소화율에서는 각 처리구간에 유의적인 차이가 없었다(P>0.05). 회분, 칼슘 및 인의 소화율의 경우 phytase 첨가구가 대조구에 비해 높은 수준을 보였다(P<0.05). 돼지가 섭취한 인의 양은 phytase 첨가구가 대조구에 비해 증가하였다. 또한 인의 흡수량에서 phytase 첨가구가 대조구에 비해 높았다(P<0.05) 분과 뇨의 총 인 배설량은 유의적인 차이를 나타내지 않았다. 칼슘의 섭취량은 처리구가 대조구에 비해 높았다. 칼슘의 분 배설량은 각 처리구간에 별 다른 차이를 보이지 않았으나(P>0.05), 뇨의 배설량은 처리구가 대조구보다 적은 양이 배설되었다. 소화된 아미노산 중 cystine, isoleucine, tyrosine, phenylalanine은 처리구가 대조구에 비해 소화율이 현저히 증가된 것을 알 수 있었다(P<0.05). Asparagine, threonine, serine, methionine, leucine의 경우 phytase를 500 unit 첨가한 구가 높은 소화율을 나타내었다(P<0.05). 한편 lysine, histidine, arginine 등의 다른 아미노산은 처리구가 대조구보다 약간의 소화율 증가를 보였으나 통계적 유의차는 나타나지 않았다(P>0.05). 결론적으로 인분해효소 phytase를 사료내 첨가하므로서 분중 영양소를 줄일 수 있는 환경친화성 사료를 개발 가능하다고 사료된다. 체결홈이 형성된 체결부를 갖는 안경집과 이것에 결합되는 체결구가 마련되어 개폐되는 커버가 형성된 케이스로 구성된다.로 금형설계 및 제작기간을 단축하고자 한다.ere demonstrated in rats after i.c.v., intraperitoneally and orally administration, respectively.
