The properties and extracting conditions of juice preperation from Schizandra nigra Max. (흑오미자(Schizandra nigra Max) 즙액의 추출조건과 추출물의 특성)

  • To determine the properties for juice preperation of Black Omija (Schizandra nigra Max.) and Omija (Schizandra chinensis), yield of extraction, chromaticity and lightness, pH and soluble solid of the extract were investigated. The rate of extract yield from Schizandra nigra was highest when extracted for 3 hours at 80$^{\circ}C$ in 20% ethanol solution. For the desirable chromaticity coordinates, the optimum extraction time and temperature of Schizandra nigra extract were 3 hours at 80$^{\circ}C$. The lightness of the extract was low of the value when extraction time and temperature was long and high. The sugar content of the extract of S. nigra was 2.0­2.6% Brix, lower than that of S. chinensis, but the difference was insignificant. The pH of the extract from S. nigra was 0.1­0.2 higher than that of S. chinensis. Although the pH of the extract from S. nigra was a little low when extracted by water, the pH range was enough to maintain the stability of color of extract from the S. chinensis.

Effect of Silkpeptide on Physicochemical Properties of Bread Dough (실크펩티드 첨가한 빵반죽의 이화학적 특성)

  • Physicochemical properties of bread dough added with silkpeptide were investigated. Protein content of silkpeptide was 90.83%. In amino acid analysis, glycine content was highest at 18,760.04 mg%. Alanine, serine, and tyrosine contents were much higher in silkpeptide flour than wheat flour. Mixed silkpeptide showed low lightness and redness values and high yellowness. Farinograph water absorption decreased as silkpeptide content increased. Both arrival and development times of silkpeptide-added dough were longer than those of wheat flour, As silkpeptide content increased, degree of weakness increased, Maximum viscosity of amylograph decreased gradually with addition of silkpeptide, while gelatinization temperature was not affected. Extensograph showed extensibility and resistance to extension of dough increased, while ratio of resistence to extensibility highly increased with increasing amount of silkpeptide. Silkpeptide added to bread dough showed oxidation effect, indication that it could be used as natural additive for improving bread dough quality.

The Effects of Germination Conditions on GABA and the Nutritional Components of Barley (발아조건에 따른 보리의 GABA 함량 및 영양성분 변화)

  • Steeping and germination conditions were investigated in order to produce barley containing a high ${\gamma}$-aminobutyric acid (GABA) content, and the changes in GABA and nutritional components before and after germination were also evaluated in this study. Water absorption rates of three barleys increased alongside both steeping time and steeping temperature. The highest GABA contents of the three barleys, 10.4-14.1 mg/100 g, were obtained from the steeping condition of $25^{\circ}C$ for 24 hr. The GABA contents of germinated barleys ranged from 14.3 to 20.9 mg/100 g, increasing by 3.9 to 14.6 times compared with raw barley. The crude lipid, crude ash and total mineral contents were slightly decreased after germination. The major fatty acids of the three barleys before and after germination were linoleic and palmitic acids. ${\beta}$-glucan contents of three barleys were slightly decreased after germination.

Anti-inflammatory Effects of Lemon Myrtle (Backhousia citriodora) Leaf Extracts in LPS-induced RAW 264.7 Cells (LPS로 유도된 RAW 264.7 세포에 대한 레몬 머틀 잎 추출물의 항염증 효과)

  • Lemon myrtle (Backhousia citriodora) has been identified as one of the plants that are likely to undergo important commercial exploitation. This study was carried out to investigate the anti-inflammatory activities and nitric oxide synthase (iNOS) expression of hot water (LMW) and 80% ethanol (LME) extracts from lemon myrtle leaf in lipopolysaccharide-induced (LPS) RAW 264.7 cells. The total phenol content of LMW and LME was 207.44 and $331.54{\mu}g$ tannic acid equivalents (TAE)/mg, respectively (p<0.01). DPPH radical scavenging activities of LMW and LME were remarkably increased in a dose-dependent manner, and were about 90.69% and 92.50% at 0.5 mg/ml, respectively. Superoxide dismutase (SOD) activities of LMW and LME were 106.22% and 103.58% at 1 mg/ml, respectively. The highest activity (91.03%) of nitrite-scavenging was observed for LME at 1 mg/ml at pH 1.2, while the activity for LMW was about 81.03% under the same conditions (p<0.05). Anti-inflammatory effect was examined in LPS stimulated RAW 264.7 cells. Nitric oxide (NO) production were reduced to 35.41% and 78.39% by addition of LMW and LME at 0.5 mg/ml, respectively (p<0.05). LMW and LME reduced protein expression of inducible nitric oxide synthase (iNOS) in a dose-dependent manner (p<0.05). These results, we conclude that lemon myrtle may be a highly valuable natural product owing to its high-quality functional components as well as its anti-oxidant and anti-inflammatory activities.

Effect of Different Processes on Texture of Fermented Cucumber Pickles (열처리조건이 오이지의 질감에 미치는 영향)

  • Effects of different processes or texture of cucumber pickles fermented in 7% NaCl solution were studied. Cucumbers were treated with different processes: control; preheating in 7% NaCl, 0.04M $CaCl_2$ solution ($60^{\circ}C$, 1hr); combination of preheating and post microwave heating (5 min, midium high level, 2450 Mhz); preheating in the solution which was adjusted to pH 8.5. In each process the pH and total acidity of brine solution, total pectin contents in alcohol insoluble solids (AIS), hot water soluble pectin (HWSP) contents and firmness of cucumber pickles were determined at 0, 5, 10, 15 and 25 days of storage periods. Total pectin contents and the firmness of pickles were decreased during 25 days of storage. Control group showed the lowest value and post microwave heating group marked the highest value in total pectin contents and firmness retention of cucumber pickles. The results of this study indicated that total pectin content seems to correlate with the firmness of cucumber pickles. However, HWSP content did not show any correlation with firmness of cucumber pickles. Post microwave heating method could be used to retain firmness of fermented cucumber pickles.

Antimicrobial Activity of Solvent Fraction from Cornus officianalis (산수유의 용매분획별 항균활성)

  • To develop natural food preservatives, ethanol and water extracts were prepared from the cornus (Cornus of officianalis) and antimicrobial activities were examined against 10 microoganisms which were food borne pathogens and/or food poisoning microoganisms, food-related bacteria and yeasts. Ethanol extract exhibited antimicrobial activity for the microoganisms tested, except lactic acid bacteria and yeast. Especially, minimum inhibitory concentrations (MIC) of the ethanol extracts were determined as 0.25 mg/mL against bacteria and 2 mg/mL against target lactic bacteria and yeasts. Antimicrobial activity of the ethanol extracts were not destroyed by the heating at 121$^{\circ}C$ for 15 min and not affected by pH. The ethanol extract of cornus exhibiting high antimicrobial activities were fractionated in the other of diethylether and butanol fractions to test antimicrobial activity The antimicrobial activity adjust bacteria test was highest in the ethanol fraction.

α-Glucosidase Inhibitory Effect of Vicine and α-Eleostearic Acid from the Seeds of Momordica charantia (여주씨에서 분리한 Vicine과 α-Eleostearic Acid의 α-glucosidase 저해효과)

  • BACKGROUND: Momordica charantia (MC) have been used in various parts of the world to treat diabetes. Despite the highest effects of phytochemicals than any other part of the plant, the main components of seed of MC grown in Korea and their properties have not been studied extensively. METHODS AND RESULTS: The seeds of MC were extracted into five different polarity solvents: 30%, 50%, 70%, 95% ethanol and distilled water. The 95% ethanol extract showed the most potent inhibition ($IC_{50}=88.7{\mu}g/mL$) against ${\alpha}$-glucosidase. To investigate the compounds responsible for this effect, activity guided fractionation of MC seeds by chromatography yielded two phytochemicals which were confirmed as vicine (1) and ${\alpha}$-eleostearic acid (2) based on their NMR and ESI-MS spectroscopic data. Among them, ${\alpha}$-eleostearic acid (2) possessed potent ${\alpha}$-glucosidase inhibitory activities with $IC_{50}$ values from $32.4{\mu}g/mL$. CONCLUSION: Collectively, the results from this present study strongly suggest that both extract and ${\alpha}$-eleostearic acid (2) have potent ${\alpha}$-glucosidase inhibitory activity. Furthermore, ${\alpha}$-eleostearic acid (2) as the most active ${\alpha}$-glucosidase inhibitor was proven to be present in high quantities in the MC seeds by a HPLC chromatogram.

A study on the Effect of Calcium Chloride Admixture on strengths of Concrete (혼화제인 염화칼슘이 콘크리트 강도에 미치는 영향에 관한 연구)

  • In many cold weather concrete constructison jobs calcium chloride $CaCl_2$ can be used safely as an accelerating admixture. For producing satisfactory concrete during cold weather calcium chloride is used to develop the level of strength required in a shorter period by obtaining higher early strength, the resulting in crease in heat of hydration. In this paper, to get adequated data and information of the effect on strength of concrete in using calcium chloride as an accelerating admixture, Portland cement (Type I), High-early-strength cement(Type II) and Pozzolans cement with certain 1.5 percentage of calcium chloride by weight of the cement were tested. As the result of this experiment, followings were founded: 1. At the 1.5 percent of calcium chloride cement ratio, the early strength was accelerated to the highest level, and some 1.5 percent of calcium chloride cement ratio was suitable for the stabilization of the concrete structures. 2. For Some 50 percent of Water Cement ratio was suitable, making good Concrete in the Cold weather by admixture of Calicum Chloide. 3. The concrete of Pozzorans cement in early strength was weak but that in later rised by degree. 4. As abtaining higher early strength the curing period can be reduced, but the finishing work should be done as early as possible.

Synthesis of High Affinity Anion Exchanger Using Ultrafine Fibrous PPmb Nonwoven Fabric by Co60 Irradiation Method (방사선 조사에 의한 초극세 폴리프로필렌 섬유부직포를 이용한 고효율 음이온교환체의 합성)

  • The aminated polypropylene melt blown ion exchange fibers were synthesized with acrylic acid monomer onto polypropylene melt blown fibers by radiation-induced polymerization and subsequent amination. Degree of grafting was increased with increasing the acrylic acid monomer concentration and total dose. The highest degree of grafting was obtained 140% at a monomer concentration of 20 v/v% acrylic acid and total dose of 4 kGy. Optimum condition of Mohr's salt was 5.0 $\times10^{-3}$ M. Degree of amination was increased with increasing degree of grafting. Water content was about 1.5 times higher than that of trunk polymer. The maximum ion-exchange capacity was 7.3 meq/g which was 2$\sim$3 times higher than a commercial ion exchange fiber. The average pore size was decreased and BET surface area was increased in order of PPmb, PPmb- g- AAc and APPmb- g- AAc. The average pore size and BET surface area of synthesised fibers were $366.1\;{\AA},\;3.71m^2/g,\;143.3\;{\AA},\;4.94m^2/g,\;40.97\;{\AA},\;8.98m^2/g$, respectively.

Fruit Quality of 'Wonhwang' Pear Trees with Low-pesticides and In Vitro Regrowth of Stem Cuttings as Affected by Time of Defoliation (시기별 적엽이 저농약 '원황'배의 과실품질과 삽수의 기내 재생장에 미치는 영향)

  • The study was conducted on the effects of time of defoliation on fruit quality of pear (Pyrus pyrifolia Nakai) trees, managing with low-pesticides, and regrowth of stem cuttings in vitro. Treatments included for 40% of uniform defoliation at early-August, end-August, and early-September, as well as control (no defoliation). Defoliation at early-September and control increased growth of water sprouts as well as concentrations of carbohydrates, total nitrogen, and free sugar in one-year old shoots. Defoliation at early-September and control increased fruit yield and mean fruit weight, with high soluble solids content and fruit surface color of $a^*$ observed for both defoliation at end-August and early-September. Defoliation at early-August increased rates of electrolyte leakage in stem cuttings at $-18^{\circ}C$ in vitro. There were no significantly different for germination rates of the cuttings between the treatments at -18 and $-21^{\circ}C$ in vitro, with the highest germination of the cuttings observed for defoliation at early-September and control at $-27^{\circ}C$. Therefore, orchard management should be performed to be minimized for defoliation of the spur leaves until end-August, causing from precipitation and pests.
