Doxorubicin Productivity Improvement by the Recombinant Streptomyces peucetius with High-Copy Regulatory Genes Cultured in the Optimized Media Composition

  • Doxorubicin is a clinically important anticancer polyketide compound that is typically produced by Streptomyces peucetius var. caesius. To improve doxorubicin productivity by S. peucetius, a doxorubicin pathway-specific regulatory gene, dnrI, was cloned into a high-copy-number plasmid containing a catechol promoter system. The S. peucetius containing the recombinant plasmid exhibited approximately 9.5-fold higher doxorubicin productivity compared with the wild-type S. peucetius. The doxorubicin productivity by this recombinant S. peucetius strain was further improved through the optimization of culture media composition. Based on the Fractional Factorial Design (FFD), cornstarch, $K_2HPO_4$, and $MgSO_4$ were identified to be the key factors influencing doxorubicin productivity. The Response Surface Method (RSM) results based on 20 independent culture conditions with varying amounts of key factors predicted the highest theoretical doxorubicin productivity of 11.1 mg/l with corn starch of 46.33 g/l, $K_2HPO_4$ of 4.63 g/l, and $MgSO_4$ of 9.26 g/l. The doxorubicin productivity of the recombinant S. peucetius strain with the RSM-based optimized culture condition was experimentally verified to be 11.46 mg/l, which was approximately 30.8-fold higher productivity compared with the wild-type S. peucetius without culture media optimization.

Managing maritime automobile terminals: an approach toward decision-support model for higher productivity

  • The article describes actions and strategies to obtain higher productivity on maritime automobile terminals. The main focus is on elaboration of efficient and effective organizational structure to model and implement short-term, mid-term and long-term strategies. In addition, with an empiric approach we combined the analyses of current findings in important scientific papers and our acknowledgments in practical research of north Adriatic maritime automobile terminals. The main goal is to propose actions towards increasing system's productivity. Based on our research of the north Adriatic maritime automobile terminals and with Lambert's model an in-deep analysis of limiting factors, user's expectations and possibilities for productivity increase has been performed. Moreover, with our acknowledgments a three-level decision-support model is presented. With an adequate model implementation it is possible to efficiently develop and implement different strategies of productivity measurement and productivity increase, especially in the fields of internal transport productivity, entrance/exit truck gates operations and wagon manipulations. According to our observation a significant increase might be achieved in all three fields.

Fiscal Convergence and Total Factor Productivity: Firm-Level Evidence from Pakistan

  • This study examines the impact of corporate tax, a fiscal measure along with trade liberalization and research and development on total factor productivity for a panel of 153 industrial firms listed on the Pakistan Stock Exchange over the period 1997-2017. For empirical analysis, we employ the system generalized method of moment estimator. In the first step, we estimate industrial production function and the results reveal that raw material, industrial labour force and energy play vital role in enhancing industrial production. Whereas, industrial capital exerts negative impact on industrial output. We also measured total factor productivity using the production elasticities. In the second step, we examine the impact of corporate tax, trade liberalization and research and development on total factor productivity. The results indicate that higher level of corporate tax exerts negative impact on total factor productivity. The findings reveal that higher corporate taxation discourages industrial firms to undertake research and development thereby exerting adverse impact on total factor productivity of firms. The impact of trade liberalization proxied by average tariff is positive while customs duty and sales tax negatively impact firm-level total factor productivity. These findings provide useful insights for managers, investors and policy makers in Pakistan.

The Impact of Weakened Global Value Chain on the Productivity of Sectors and Firms in Chungnam Area (글로벌 가치사슬 약화가 충남지역 산업 및 기업 생산성에 미치는 영향)

  • This paper analyzes the impact of the recent change in global value chain on the productivity of manufacturing sectors and firms in Chungnam area using ADB MRIO. Forward participation significantly increases sector productivity in Chungnam area and backward participation affects the sector productivity after 2015. In the analysis for firm productivity in Chungnam area, GVC participation does not enhance the productivity of firms in Chungnam area. However, firms in the top 25% of productivity in 2007 improved their productivity with higher GVC participation and the productivity of firms in the top 25% of asset in 2007 was positively affected by forward pariticipation and negatively affected by backward participation. The productivity of firms that were in international market in 2007 was significantly worsened by GVC participation.

  • This paper aims to find out the R&D personnel management strategies which enhance the productivity of researchers. The R&D personnel management practices are examined focusing on career development and compensation scheme. As proxies for higher motivation and/or productivity, job satisfaction and propensity to leave of researchers are analyzed to evaluate the management practices. Using the survey data from researchers in the Korean automobile industry. this paper suggests the management strategies to raise job satisfaction and thus lower propensity to leave of R&D personnel.

  • The paradigm of the global logistics network has been rapidly changing from traditional concept of productivity in cost terms, toward the new characteristics of innovative productivity in terms of economies of scale, economies of scope, and economies of network. As the ship or airplane becomes larger and speedier, the less logistics hub required. The more diverse logistics parties integrate each other horizontally or vertically, the higher synergy effects they get. The more systematic concurrent engineering available for global supply chain management such as Korean manufacturers in China, the higher productivity they get. The paper analyzes these three paradigms of logistics into the application for the Korean manufacturers in China and concludes that the Korean government, the companies in China should be more focused on the governance of the logistics and the intermediary such as the 4th party logistics(4PL) is definitely developed its role and functions based on three paradigms above mentioned.

Enhancement of cis,cis-Muconate Productivity by Overexpression of Catechol 1,2-Dioxygenase in Pseudomonas putida BCM114

  • For enhancement of cis,cis-muconate productivity from benzoate, catechol 1,2-dioxygenase (C12O) which catalyzes the rate-limiting step (catechol conversion to cis,cis-muconate) was cloned and expressed in recombinant Pseudomonas putida BCM114. At higher benzoate concentrations (more than 15 mM), cis,cis-muconate productivity gradually decreased and unconverted catechol was accumulated up to 10 mM in the cae of wild-type P. putida BM014, whereas cis,cis-muconate productivity continuously increased and catechol was completely transformed to cis,cis-muconate for P. putida BCM114. Specific C12O activity of P. putida BCM114 was about three times higher than that of P. putida BM014, and productivity was enhanced more than two times.

  • The effectiveness of informatization can vary depending on the level of operation of the information system as well as investment and installation. This study investigates and analyzes the information system operating environment and quality level of small and medium-sized manufacturing companies in Korea, and investigates how it affects the performance indicators of companies. The influence of the environmental factors operating the information system on the productivity performance and financial performance through the quality level of the information system was analyzed through the structural model. As a result of the analysis, it can be said that the higher the level of information system operating environment factors, the higher the quality level, the higher the quality level, the higher the productivity performance, and the higher the productivity performance, the higher the financial performance. It is judged that the effect of informatization depends on the operating environment and quality level after the installation of the information system.

Relationship between the GVC participation and the productivity in the Chinese Manufacturing Industries (중국 제조업에서 GVC 참여 정도와 위치가 생산성에 미친 영향 실증 분석)

  • This paper investigates the effects of China's participation in global value chains (GVC) on the productivities focusing on the manufacturing industries. In this study, several indicators of participation in global value chains were used. These include GVC participation, forward GVC participation, backward GVC participation and GVC position index. In particular, we used the data obtained from 18 manufacturing industries in China during 15 years from 2000 to 2014. The main results of the analysis are as follows. First, the higher the degree of total participation in GVC, the higher the productivity. This means that with the increase in exports and imports of intermediate goods, productivity has increased through technology spillover effects or competition effects, and so on. Second, the backward participation does not increase the productivities significantly while forward participation leads higher productivity. Third, the productivity improvement effects of GVC participation was larger in the high-tech industries than in the low-tech industries. These results show that GVC participation was helpful for the economic growth of China and the efforts for moving toward upstream production stage in GVC is necessary for the improvement of international competitiveness in Chinese manufacturing industry.

Productivity Analysis by Region and Species of Fish Cage-culture (해상가두리 어류양식업의 지역별 어종별 생산성 분석)

  • This study is aimed to figure out the productivity and variability of cage-aquaculture changes. According to the analysis, the productivity of major fish species has been increasing, except mullet. Although the regional productivity has decreased in the last two years, it has been on the rise considering as a whole. Gyeongsangnam-do showed the highest level of productivity by region. Productivity by species was also higher than other regions in the cases of rock fish, mullet and sea bream followed by productivity of Chungcheongnam-do. The production of marine cage-culture in Jeollanam-do is the second largest in Korea in value/weight while its productivity is lower than that of Chungcheongnam-do. When it comes to comparison by region, Gyeongsangnam-do shows the lowest productivity variation. And Jeollanam-do shows the second-lowest variation in productivity that is only about half of that of Chungcheongnam-do province. Thus, it is found that Jeollanam-do region has an advantage in management stability while its productivity is low. On the other hand, productivity by species was also analyzed. Gyeongsangnam-do has the highest productivity by species for rock fish, mullet and sea bream whereas rock bream productivity is the highest in Jeollanam-do. Therefore, it probably needs to reflect these results when choosing regional-focused incubation fish species.
