GPS-X Based Modeling on the Process of Gang-byeon Sewage Treatment Plant and Design of Recycle Water Treatment Process (GPS-X 기반 모델링에 의한 강변사업소 처리효율 분석 및 반류수 처리 공정 설계)

  • The efficiencies of Gang-Byeon sewage treatment facilities, which are based on GPS-X modelling, were analysed and used to design recycle water treatment processes. The effluent of an aeration tank contained total kjeldahl nitrogen (TKN) of 1.8 mg/L with both C-1 and C-2 conditions, confirming that most ammonia nitrogen ($NH_3{^+}-N$) was converted to nitrate nitrogen ($NO_3{^-}-N$). The concentrations of $NH_3{^+}-N$ and $NO_3{^-}-N$ were found to be 222.5 and 227.2 mg/L, respectively, with C-1 conditions and 212.2 and 80.4 mg/L with C-2 conditions. Although C-2 conditions with higher organic matter yielded a slightly higher nitrogen removal efficiency, sufficient denitrification was not observed to meet the discharge standards. For the total nitrogen (T-N) removal efficiency, the final effluent concentrations of T-N were 293.8 mg/L with biochemical oxygen demand (BOD) of 2,500 mg/L, being about 1.5 times lower than that (445.3 mg/L) with BOD of 2,000 mg/L. Therefore, an external carbon source to increase the C/N ratio was required to get sufficient denitrification. During the winter period with temperature less than $10^{\circ}C$, the denitrification efficiency was dropped rapidly even with a high TKN concentration (1,500 mg/L). This indicates that unit reactors (anoxic/aerobic tanks) for winter need to be installed to increase the hydraulic retention time. Thus, to enhance nitrification and denitrification efficiencies, flexible operations with seasons are recommended for nitrification/anoxic/denitrification tanks.

Evaluation of Organic Matter and Trace Metal Contaminations of Intertidal Sediments from Coastal Islands in the Southern Region of Jeollanam Province (전남 남부 도서갯벌 퇴적물의 유기물 및 미량금속 오염 평가)

  • We measured the grain size, ignition loss (IL), chemical oxygen demand (COD), acid volatile sulfide (AVS) and trace metals (Al, Fe, Cu, Pb, Zn, Cd, Cr, Mn, Hg, and As) of intertidal surface sediment collected from 11 islands (62 stations) in the southern region of Jeollanam Province. The objective of this research was to evaluate the organic matter and trace metals contaminations of sediments from coastal island tidal flats. Surface sediment texture was characterized as follows: mud, sandy silt, muddy sand, and slightly gravelly sand facies. The finer sediments are mainly dominated in the northern part of each island. The concentrations of IL, COD, AVS and some trace metals (Al, Fe, Zn, Cr, Cu, and Hg) were higher in the northwestern part of Wan Island and the area between Gogeum and Sinji Islands, and were associated with relatively finer sediment, as compared to other locations. The concentrations of Mn, Pb, Cd, and As were higher in the northwestern and southeastern parts of Geoguem and Pyungil Islands, but were not correlated with mean grain size. Based on sediment quality guidelines (SQGs), the concentrations of trace metals were lower than the values of effect range low (ERL), used in United States, and threshold effects level (TEL), used in Korea, with exception of As. Similarly, the intertidal sediments were moderately contaminated with As, based on the the enrichment factor (EF) and the geoaccumulation index ($I_{geo}$). The high concentration of As in intertidal sediments from this study region may be due to the input of naturally or artificially contaminated submarine groundwater, contaminated waste from seaweed aquaculture operations and/or land-based seaweed processing facilities. Further studies are needed to identify the sources of As in this study region, and to determine the effects of As contamination on coastal ecosystem.

Experimental and Computational Studies on Particle Behavior in High Temperature Gas with the Various Temperatures of a Solid Wall (고체의 벽면온도에 따른 고온가스 내의 입자거동에 대한 실험 및 수치해석 연구)

  • The effect of a wall temperature on the soot deposition process from a diffusion flame to a solid wall was investigated in a microgravity environment to attain in-situ observations of the process. The fuel for the flames was an ethylene ($C_2H_4$). The surrounding oxygen concentration was 35% with surrounding air temperatures of $T_a=600K$. In the study, three different wall temperatures. $T_w$=300, 600, 800K, were selected as major test conditions. Laser extinction was adopted to determine the soot volume fraction distribution between the flame and burner wall. The experimental results showed that the maximum soot volume fractions at $T_w$=300, 800 K were $8.8{\times}10^{-6},\;9.2{\times}10^{-6}$, respectively. However, amount of soot deposition on wall surface was decreased because of lower temperature gradient near the wall with increasing wall temperature. A numerical simulation was also performed to understand the motion of soot particles in the flame and the characteristics of the soot deposition to the wall. The results from the numerical simulation successfully predicted the differences in the motion of soot particles by different wall temperature near the burner surface and are in good agreement with observed soot behavior that is, the 'soot line', in microgravity.

Ecophysiology of Seed Germination in Chinese Milk Vetch (Astragalus sinicus L.)

  • Germination and emergence habits of Chinese milk vetch (CMV) were examined to obtain the basic information for the effect of environmental conditions and cultural practices on the seedling density. Seed germination tests with different water potentials, temperatures, and soil and water depths showed the environmental effects on the characters related to seed germination in CMV. Imbibition under different temperatures reflects that initial velocity was rapid at higher temperature, however, the times to full imbibition were not different between 15 and $25^{\circ}$. The optimal germination temperature for CMV germination was ranged from 15 to 20 and the germination was highly affected by water potential of media at relatively high temperature above $20^{\circ}$. When the seeds were sown in flooded condition the germination was not proportionally affected by water depth. In addition, there was no correlation between water depth and oxygen concentration. The germination of seeds flooded by 2cm water depth were poorly germinated compared to other depths. Results indicated that the germination of submerged seeds was more highly influenced by flooding depth than the temperature, it was also affected more strongly at 10 than $20^{\circ}$. Emergence of CMV depending on the thickness of covered soil was poor when the soil layer was greater than 5cm. In the experiment with seeds collected between 22 days after flowering (OAF) and 52 DAF, the highest germination ability of CMV seeds was observed at 39 DAF and germinability was decreased subsequently as seeds became mature. The lower germinability may be due to the enhanced seed dormancy.

Nitrogen Removal using Autotrophic Microorganism in Membrane-Attached Biofilm Reactor (MABR) (Membrane-Attached Biofilm Reactor(MABR)에서의 독립영양 미생물을 이용한 질소 제거)

  • The purpose of this study is to investigate the performance of nitrogen removal using autotrophic microorganism in the Membrane-Attached Biofilm Reactor (MABR). The treatment system consists of an aerobic MABR (R1) for nitrification and an anaerobic MABR (R2) for hydrogenotrophic denitrification. Oxygen and hydrogen were supplied through the lumen of hollow-fiber membranes as electron acceptor and donor, respectively. In phase Ι, simultaneous organic carbon removal and nitrification were carried out successfully in R1. In phase II, to develop the biofilm on the hollow-fiber membrane surface and to acclimate the microbial community to autotrophic condition, R1 and R2 were operated independently. The MABRs, R1 and R2 were connected in series continuously in phase III and operated at HRT of 8 hr or 4 hr with $NH_4{^+}-N$ concentration of influent, from 150 to 200 mgN/L. The total nitrogen removal efficiency reached the maximum value of 99% at the volumetric nitrogen loading rate of $1.20kgN/m^3{\cdot}d$ in the combined MABR system with R1 and R2. The results in this study demonstrated that the combined MABR system could operate effectively for the removal of nitrogen in wastewater not containing organic materials and can be used stably as a high rate nitrogen removal technology.

Performance Evaluation of Microorganisms Immobilized Reactive Capping Materials on Elution Blocking of Organic, Nitrogen, and Phosphorus Compounds (미생물이 고정화된 반응성 피복재의 유기물, 질소 및 인 용출 차단성능 평가)

  • This study aims to evaluate the effect of capping materials on blocking pollutant elution from contaminated sediment to water body. Experiments were carried out under conditions in which the elution rate was intensified artificially using compost with high concentration of organic compound and nutrient salts instead of sediments. Activated carbon (AC), modified activated carbon (MAC), P. putida immobilized activated carbon (PBAC) and effective microorganisms immobilized activated carbon (EBAC) were used as capping materials. Zeolite (ZT) and two kinds of commercially available microorganisms immobilized zeolite products (ZC, ZN) were used for comparison experiment. The elution rate of organic compound, nitrogen and phosphorus were compared with that of control experiment. The experiments were conducted for 56 days. Concentrations of chemical oxygen demand, total nitrogen, and total phosphorus were measured to use the comparison of release rate of organic compound, nitrogen and phosphorus. From the experimental results, AC based materials showed better performance to block the elution of organic compound and nitrogen than ZT based materials. Although ZT based materials were more effective than AC and PBAC to block phosphorus, MAC and EBAC showed the best performance of phosphorus elution blocking among the all candidate materials. In conclusion, EBAC is considered as the most effective capping materials, because organic compound, nitrogen and phosphorus will be degraded continuously by EM in the long term.

Effects of Submerged Aerator on the Growth of Algae in Daechung Reservoir (대청호에 설치된 수중폭기시설이 조류 발생에 미치는 영향)

  • To evaluate the effects of submerged aerators installed at Chudong and Muneui areas in Daechung Reservoir on improvement of water quality and reduction of algal bloom, the water quality was monitored at the effected and control areas at the time of operation. The water temperature and concentrations of dissolved oxygen, total phosphorus, and total nitrogen in depth at the effected and control areas were not different each other, indicating the submerged aerators at these areas are not effective for circulation of water body and reduction of nutrients. In warmer season, the concentrations of total phosphorus in deep water, which was probably released from contaminated sediment or inflowed from watershed, was high. To decide the operation of aerators in this season, the concentration of total phosphorus in water should be considered because the dispersed phosphorus by operation of aerators can enhance the algal growth.

A Role of Unsaturated Fatty Acid in Animal Reproductive Cells and Biology

  • As a one of unsaturated fatty acid, polyunsaturated fatty acids (PUFAs) have multiple actions: as precursor of prostaglandins (PGs), steroid hormone synthesis and energy production in animal reproduction. PUFAs, which include omega-3 (n-3) and omega-6 (n-6), are derived from the diet and changed by diet, species, breed and season. The plasma membrane of spermatozoa in mammals contain various PUFAs. These composition of PUFAs regulate the membrane fluidity and cause lipid peroxidation via generation of reactive oxygen species (ROS). Induced lipid peroxidation by ROS decreased viability and motility of spermatozoa, and it is reduced by addition of antioxidant and low concentration of PUFAs. Because oocytes of animal have a high lipid components, process of oocyte maturation and embryo development are influenced by PUFAs. In in vitro study, oocyte maturation, embryo development, intracellular cAMP and MAPK activity were increased by treatment of n-3 ${\alpha}$-linolenic acid (ALA) during maturation, whereas n-6 linoleic acid (LA) negatively influenced. Also, inhibition of fatty acid metabolism in oocyte influenced blastocyst formation of cattle. PGs are synthesized from PUFAs and various PUFAs influence PGs via regulation of PG-endoperoxide synthase (PTGS). Steroid hormone synthesis from cholesterol is regulated by expression of steroid acute regulator (StAR) protein and mRNA. Exogenous n-3 and n-6 PUFAs altered sex hormone in animal through stimulate or inhibit StAR activity. Because PUFAs altered PG and steroid hormone synthesis, follicular development was influenced by PUFAs. This effect of unsaturated fatty acid could provide information for improvement of reproductive ability in animals.

Antioxidative Effect of Commercial Lecithin on the Oxidative Stability of Fish Oil (어유의 산화안정성에 미치는 레시틴의 산화방지 작용)

  • The antioxidative effects of three kinds of commercial lecithin in fish oil(EPA 25%, DHA 10%) were investigated through active oxygen method (AOM, hrs. at $97.8^{\circ}C$), Oven test, polymer test by gel chromatography and coloring test. Although there were difference of antioxidative effect among commercial lecithins, antioxidative effects of the lecithins added to the fish oil increased with increasing the concentration of lecithin. Lecithin III(acetone insoluble content 65%) had the greatest antioxidative effect and the addition of 1%, 5 and 10% enhanced the oxidative stability to 310%, 620% and 840%, respectively. The results also showed that the polymerization in presence of 10% lecithin III did not occur up to 10 hours at the AOM condition, and the degree of color(Gardner number) increased as storage time went by and was accerated at high temperature.

Neuroprotective Effect of the Water-insoluble fraction of Root Barks of Dictamnus dasycarpus 70% Ethanolic Extract on Glutamate-Induced Oxidative Damage in Mouse Hippocampal HT22 Cells (백선피 70% 에탄올 추출물의 비수용성 분획물의 뇌세포 보호 효과)

  • Oxidative stress or accumulation of reactive oxygen species (ROS) leads neuronal cellular death and dysfunction, and it contributes to neuronal degenerative disease such as Alzheimer's disease, Parkinson's disease and stroke. Glutamate is one of the major excitatory neurotransmitter in the central nervous system (CNS). Glutamate contributes to fast synaptic transmission, neuronal plasticity, outgrowth and survival, behavior, learning and memory. In spite of these physiological functions, high concentration of glutamate causes neuronal cell damage, acute insults and chronic neuronal neurodegenerative diseases. Heme oxygenase-1 (HO-1) enzyme plays an important role of cellular antioxidant system against oxidant injury. NNMBS020, the water-insoluble fraction of the 70% EtOH extract of root barks of Dictamnus dasycarpus, showed dominant neuroprotective effects on glutamate-induced neurotoxicity in mouse hippocampal HT22 cells by induced the expression of HO-1 and increased HO activity. In mouse hippocampal HT22 cells, NNMBS020 makes the nuclear accumulation of Nrf2 and stimulates extracellular signal-regulated kinase (ERK) pathway. The ERK MAPK pathway inhibitor significantly reduced NNMBS020-induced HO-1 expression, whereas the JNK and p38 inhibitors did not. In conclusion, the water-insoluble fraction of the 70% EtOH extract of root barks of D. dasycarpus (NNMBS020) significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2 and ERK pathway in mouse hippocampal HT22 cells.
