Molecular Cloning of Maltooligosyltrehalose Trehalohydrolase Gene from Nostoc flagelliforme and Trehalose-Related Response to Stresses

  • A genomic DNA fragment encoding a putative maltooligosyltrehalose trehalohydrolase (NfMTH) for trehalose biosynthesis was cloned by the degenerate primer- PCR from cyanobacterium Nostoc flagelliforme. The ORF of NfMTH is 1,848 bp in length and encodes 615 amino acid residues, constituting a 70 kDa protein. The deduced amino acid sequence of NfMTH contains 4 regions highly conserved for MTHs. By expression of NfMTH in E. coli, the function of this protein was demonstrated, where the recombinant protein catalyzed the hydrolysis of maltooligosyl trehalose to trehalose. The expressions of MTH and maltooligosyltrehalose synthase in the filaments of N. flagelliforme were upregulated significantly under dehydration stress, NaCl stress, and high temperature-drought stress. The accumulations of both trehalose and sucrose in the filaments of N. flagelliforme were also improved significantly under the above stresses. Furthermore, trehalose accumulated in smaller quantities than sucrose did when under NaCl stress, but accumulated in higher quantities than sucrose did when under temperature-drought stress, indicating that both trehalose and sucrose were involved in N. flagelliforme adapted to stresses and different strategies conducted in response to various stress conditions.

  • The petrochemical data of the porphyritic granites of Cretaceous age in the Yonghwa-Seolcheon area show the trend of subalkaline magma, calc-alkaline magma, I-type granitoid and magnetite series. This granite is the relevant igneous rock of gold-silver mineralization in this mining district Fluid inclusions have been studied in phenocryst quartz from the Cretaceous porphyritic granite. Three main types of fluid inclusion were found : liquid-rich inclusion(I type), gas-rich inclusion(II type) and solid-bearing inclusions(III-A, III-B). The solid-bearing inclusions(III-A,B) represent the earliest trapped fluids. They have salinities between 41.0 and 67.5 wt% equivalent to NaCl. These are high saline inclusions containing NaCl and KCl daughter crystals. Homogenization temperature inferred from the fluid inclusion study ranges from 650 to $75^{\circ}C$ Type I and II inclusions were observed within the same fracture. This cause for these differences in degree of filling is evidence of boiling. Salinities of type I and II inclusions range from 9.87 wt% to 15.29 wt%, from 8.40 wt% to 14.64 wt% NaCl equivalent, respectively.

A Study on the Foaming Properties of Small red bean Protein Isolates at Various Conditions (분리 팥 단백질의 기포 특성에 영향을 주는 제 요인에 관한 연구)

  • The purpose of this study was to determine the foaming properties of two small red bean protein isolates at various conditions. Data concerning the effects of pH, temperature, MaCl concentration, sugar concentration and protein concentration on the properties such as solubility, foam expansion, foam stability were presented. The results were summarized as follows : 1. The crude protein contents of two small red beans were 26.14% and 22.71%. The percentage of nonpolar amino acid group was the highest and that of sulfur containing amino acid group was the lowest. 2. Protein solubility showed the minimum at pH 4.5 which is isoelectric point of small red bean protein isolate adn heat treatment lowered solubility(P<0.05). At pH 4.5, solubility increased sighificantly as 0.4M NaCl was added. However, the effect of sugar concentration in the solubility was not significant. 3. Foam expansion of two small red bean protein isolates was high at pH 4.5 and heat treatment at 10$0^{\circ}C$ lowered foam expansion(P<0.05). While addition of NaCl, sugar did not affect the foma expansion, gradual increment of the protein isolates concentration up to 9% decreased the foma expansion slightly. 4. Foam stability was significantly high at pH 4.5 and heat treatment at 10$0^{\circ}C$ lowered foam stability. Addition of sugar caused slight decrease in foam stability. From 1% to 9% suspension, foma stability increased significantly as protein concentration increased(P<0.05)

  • Nanofiltration[NF45] and reverse osmosis membrane(HR98PP) separation treatment of dyestuff wastewater was carried out In order to separate relatively pure water from synthetic dyestuff wastewater, which consists of reactive dye, acid dye, basic dye, direct dye, and disperse dye. The experiments were performed by using the plate and frame membrane module. In the nanofiltration and reverse osmosis membrane separation, When the NaCl concentration was 0.1, 5.0, and 20.091, retention was 63.0, 46.0, 0.9%, respectively. When permeate flux was 125.0, 67.5, and 45.0 L/$m^2$ h, the osmotic pressure increased with Increasing the NaCl concentration. Permeate flux of two membranes Increased as temperature Increased due to segmental movement of polymer of the membrane and the rejection rate of dyestuff was decreased gradually. It was found that the rejection rate was about 95% in the nanofiltratlon, while the reverse osmosis membrane showed a high rejection rate of 99% under all temperature and pressures conditions.

Effect of Fermentation Temperature on Quality of Doenjang (숙성온도가 된장의 품질에 미치는 영향)

  • There were four types of Doenjang fermentation as following conditions for investigation ; 1) low temperature fermentation at $13^{\circ}C$ for 180 days, 2) low temperature at $13^{\circ}C$ for 7 days to room temperature at $30^{\circ}C$ for 10 days, to low temperature at $13^{\circ}C$ for 163 days, and for 173 days, 3) low temperature at $13^{\circ}C$ 7 days to room temperature at $30^{\circ}C$, 4) room temperature at $30^{\circ}C$ for 180 days. There were no changes of moisture, NaCl and total nitrogen content during fermentation period of four types conditions, but pH and amino type nitrogen decreased in room temperature at $30^{\circ}C$ for 180 days. It required 3 times more fermentation period until same quantity of the amino type nitrogen. The low temperature fermentation sample was lower than room temperature fermentation sample in pH and amino type nitrogen. The yeast decreased in low temperature fermentation sample taken 15 to 30 days longer than room temperature sample. The yeast is increased up to 30 days, and decreased little by little. After 60 days, it remained a few without effectiveness on the Doenjang quality. The low temperature fermentation sample showed brighter than room temperature fermentation sample. Different fermentation condition affected Doenjnag quality, especially, low temperature fermentation sample showed bright color in Doenjnag. So low temperature fermentation must be expected as good method for getting high quality Doenjnag.

  • An air-conditioning system based on the chemical heat storage principle was considered. $H_2O$ was chosen as the reaction gas and the working fluid as well. Na$_2$S, CaCl$_2$, MnCl$_2$, BaCl$_2$, MgCl$_2$, Fe$_2$(SO$_4$)$_3$ and MnSO$_4$ were tested as the solid reactants by using Cahn pressure balance. Na$_2$S was superior to other salts in respect of high capability of absorption of water gas, 5 moles of $H_2O$ per unit mole of Na$_2$S, and adequate temperature of adsorption, $65^{\circ}C$ at 7torr, and of desorption, 13$0^{\circ}C$ at 76torr. Clausius-Clapeyron diagram of Na$_2$S was obtained via adsorption experiments at several vapor pressures of water gas. To enhance heat and mass transfer characteristics, usually below 1W/m K, of the reactor bed of general adsorption systems, expanded graphite block was adapted as the support of Na$_2$S salt. Expanded graphite blocks had thermal conductivity values of 20~80W/mK with respect to 100~400kg/㎥ of block bulk density. Permeability values of expanded graphite blocks were 10$^{-13}$ ~ 10$^{-14}$ $m^2$ with respect to 100~300kg/㎥ of block bulk density showing highly decreasing values of permeability, below 10$^{-l4}$$m^2$, in the range of above 150kg/㎥ of block bulk density.y.

  • We have investigated the properties of poly (vinyl alcohol)-film obtained by casting from the solution PVA/electrolytes (NaCl, KCl, MgCl$_2$, CaCl$_2$)/water systems at room temperature. The removed electrolytes films have got high crystallinity as well as high draw ratio. The removed electrolytes films had layer-structure while a film without electrolytes didn't have it. A relation between draw ratio and thickness or number of layers, were recognized.

  • The Ssangjeon tungsten deposit is located within the Yeongnam Massif. Within the area a number of hydrothermal quartz veins were formed by narrow open-space filling of parallel and subparallel fractures in the metasedimentary rocks as Wonnam formation, Buncheon granite gneiss, amphibolite and/or pegmatite. Mineral paragenesis can be divided into two stages (stage I, ore-bearing quartz vein; stage II, barren quartz vein) by major tectonic fracturing. Stage I, at which the precipitation of major ore minerals occurred, is further divided into three substages (early, middle and late) with paragenetic time based on minor fractures and discernible mineral assemblages: early, marked by deposition of arsenopyrite with pyrite; middle, characterized by introduction of wolframite and scheelite with Ti-Fe-bearing oxides and base-metal sulfides; late, marked by Bi-sulfides. Fluid inclusion data show that stage I ore mineralization was deposited between initial high temperatures (≥370℃) and later lower temperatures (≈170℃) from H2O-CO2-NaCl fluids with salinities between 18.5 to 0.2 equiv. wt. % NaCl of Ssangjeon hydrothermal system. The relationship between salinity and homogenization temperature indicates a complex history of boiling, fluid unmixing (CO2 effervescence), cooling and dilution via influx of cooler, more dilute meteoric waters over the temperature range ≥370℃ to ≈170℃. Changes in stage I vein mineralogy reflect decreasing temperature and fugacity of sulfur by evolution of the Ssangjeon hydrothermal system with increasing paragenetic time.

Adsorption Characteristics of Biochar from Wood Waste by KOH, NaOH, ZnCl2 Chemical Activation (폐목재를 이용한 KOH, NaOH, ZnCl2 화학적 활성화로 생성된 바이오차의 흡착특성에 관한 연구)

  • There is a lot of interest in methods for pollutants using adsorption, and recent research is being conducted to show that biochar can be used to remove organic and inorganic pollutants. In particular, wood waste as waste biomass requires a biomass recycling method, and a method to increase the adsorption capacity of biochar produced using wood waste is needed. Biochar is created by Hydrothermal carbonization (HTC) using, which uses low temperature and high pressure, has low energy consumption and does not require moisture removal pretreatment, and biochar is created through chemical activation using KOH, NaOH, and ZnCl2 chemicals. The adsorption characteristics of biochar were determined by analyzing iodine adsorptivity, specific surface area, pore diameter, pore volume, pore distribution, and SEM according to the activation. The results of analyzing the selecting biochar by activating the biochar produced at HTC 300℃, 4 hr by KOH, NaOH, and ZnCl2 chemicals, the specific surface area was 774~1.387 m2/g, showing a high specific surface area similar to activated carbon, and it was confirmed that micropores with an average pore diameter in the range of 21~24 Å were formed. As a result of SEM observation, the surface was uniform with a certain shape depending on activation. It was confirmed that one pore was developed and the number of pores increased.

Effect of Electrolytic Material Feeding on Blood and Carcass Traits of Broiler under Intense Heat Condition in Summer (폭염 시 육계 출하 전 전해질 급여가 닭고기 도체 및 혈액에 미치는 영향)

  • This study was conducted to investigate the effect of feeding the electrolytic materials on blood and carcass traits of broiler during transportation exposed under intense heat condition in summer. The broilers were selected on the day when the outside temperature was about $32^{\circ}C$ to provide heat stressed environment. Broilers reared for 33 d were selected and fed with the electrolytic materials ($NaHCO_3$, NaCl, KCl) for 2 days. Treatments were as follows; feeding the underground water for control, $NaHCO_3$ (1.0%) + NaCl (0.5%) for treament 1, KCl (0.5%) + NaCl (0.5%) for treatment 2, KCl (1.0%) + NaCl (0.5%) treatment 3, KCl (0.5%) + $NaHCO_3$ (1.0%) + NaCl (0.5%) for treatment 4 and KCl (1.0%) + $NaHCO_3$ (1.0%) + NaCl (0.5%) for treament 5. pH of chicken meat increased for treatments group of electrolytic material, especially, that of treatment 3 was highest when compared to the other treatments. The frequency rate (%) of $1^+$ quality grade were 33.3, 60.0 and 83.3% at control, treatment 3, 4 and treatment 5, respectively. Occurrence rates of PSE were 50% for control and 13.3% for treatment 5. Corticosterone increased at the post-harvest period compared to the pre-harvest period of broiler and have small disparity between pre-and post-harvest only except treatment 3 when compared to control. $pCO_2$ partial pressure of blood at the pre-harvest period was low in all treatments by heat stress, the disparity value of control was high for control, and those of treatment 4 and 5 were low compared to other treatments.
