Grapefruit seed extracts(GFSE) have some unknown compounds which exhibit the antibiotic activities aganist microorganisms including bacteria and fungi. We have examined the effects of GFSE on the growth of Enterobacter pyrinus which was isolated from necrotic lesions of pear trees. During the cultivation, the growth of the bacteria was strongly inhibited at the low concentration(0.01%, w/w) of GFSE. Hydrophobic fraction extracted from GFSE by mixed solvents (chloroform : methanol : water, 1 : 2 : 0.8, v/v/v) had components which inhibited the growth of bacteria. There was, however, no inhibitory effect of GFSE on the activities of several enzymes including hexokinase, glucose 6-phosphate dehydrogenase, malate dehydrogenase and succinate dehydrogenase. $O-nitrophenyl-{\beta}-D-galactopyranoside(ONPG)$, the artificial substrate of ${\beta}-galactosidase$ was hydrolyzed in the presence of GFSE, indicating that the membrane was pertubated by the GFSE. From the results it was suggested that the antibiotic activity of GFSE is due to the change of membrane permeability of cell. GFSE was fractionated by high performance liquid chromatography equipped with $C_{18}$ reverse phase column. Among active fractions, three peaks were identified as 1-chloro-2-methyl-benzene (o-toluene), N,N-dimethyl-benzenemethaneamine, 1-[2-(2-ethylethoxy)ethoxy]-4- (1,1,3,3-tetramethyl)-bezene, respectively, while the other three remained unidentified.

Quinacrine (QU), a phospholipase-A2 (PLA-2) inhibitor has been used clinically as a chemotherapeutic adjuvant. To understand the mechanisms leading to its chemotherapeutic effect, we have investigated QU-induced apoptotic signaling pathways in human cervical squamous carcinoma HeLa cells. In this study, we found that QU induced cytochrome c-dependent apoptotic signaling. The release of pro-apoptotic cytochrome c was QU concentration- and time-dependent, and preceded activation of caspase-9 and -3. Flow cytometric FACScan analysis using fluorescence intensities of $DiOC_6$/ demonstrated that QU-induced cytochrome c release was independent of mitochondrial permeability transition (MPT), since the concentrations of QU that induced cytochrome c release did not alter mitochondrial membrane potential (${\blacktriangle}{\Psi}_m$). Moreover, kinetic analysis of caspase activities showed that cytochrome c release led to the activation of caspase-9 and downstream death effector caspase-3, Caspase-3 inhibitor (Ac-DEVD-CHO) partially blocked QU-induced apoptosis, suggesting the importance of caspase-3 in this apoptotic signaling mechanism. Supplementation with arachidonic acid (AA) sustained caspase-3 activation induced by QU. Using inhibitors against cellular arachidonate metabolism of lipooxygenase (Nordihydroxyguaiaretic Acid, NDGA) and cyclooxygenase (5,8,11,14-Eicosatetraynoic Acid, ETYA) demonstrated that QU-induced apoptotic signaling may be dependent on its role as a PLA-2 inhibitor. Interestingly, NDCA attenuated QU-induced cytochrome c release, caspase activity as well as apoptotic cell death. The blockade of cytochrome c release by NDCA was much more effective than that attained with cyclosporin A (CsA), a MPT inhibitor. ETYA was not effective in blocking cytochrome c release, except under very high concentrations. Caspase inhibitor z-VAD blocked the release of cytochrome c suggesting that this signaling event is caspase dependent, and caspase-8 activation may be upstream of the mitochondrial events. In summary, we report that QU induced cytochrome c-dependent apoptotic signaling cascade, which may be dependent on its role as a PLA-2 inhibitor. This apoptotic mechanism induced by QU may contribute to its known chemotherapeutic effects.

A large amount of acidic waste solution is generated from the practical electrokinetic decontamination equipments for the remediation of soil contaminated with uranium. After filtration of uranium hydroxides formed by adding CaO into the waste solution, the filtrate was recycled in order to reduce the volume of waste solution. However, when the filtrate was used in an electrokinetic equipment, the low permeability of the filtrate from anode cell to cathode cell due to a high concentration of calcium made several problems such as the weakening of a fabric tamis, the corrosion of electric wire and the adhension of metallic oxides to the surface of cathode electrode. To solve these problems, sulfuric acid was added into the filtrate and calcium in the solution was removed as $CaSO_4$ precipitate. A decontamination test using a small electrokinetic equipment for 20 days indicated that Ca-removed waste solution decreased uranium concentration of the waste soil to 0.35 Bq/g, which is a similar to a decontamination result obtained by distilled water.

Natural ground is a composite consisted of the three phases of water, air and soil paircies. Among the three components, water as a material is weU understood but soil particles are not in foundation engineering. Especially, weathered granite soil generally shows a large volumetric expansion when they freeze. And, the stability and durability of the soil have shown decreased with repetitive freezing and thawing processes. These unique charcteristics may cause various construction and management problems if the soil is used as a construction material and foundation layers. This project was initiated to investigate the soil's physical and engineering characteristics resulting from freezing and freezing-thawing processes. Research results may be used as a basic data in solving various problems related to the soil's unique characteristics. The following conclusions were obtained: The degree of decomposition of weathered granite soil in Kangwon-do was very different between the West and East sides of the divide of the Dae-Kwan Ryung. Soil particles distributed wide from very coarse to fine particles. Consistency could be predicted with a function of P200 as LL=0.8 P200+20. Permeability ranged from 10-2 to 10-4cm/sec, moisture content from 15 to 20% and maximum dry density from 1.55 to 1.73 g /cmΥ$^3$ By compaction, soil particles easily crushed, D50 of soil particles decreased and specific surface significantly increased. Shear characteristics varied wide depending on the disturbance of soil. Strain characteristics influenced the soil's dynamic behviour. Elastic failure mode was observed if strain was less than 1O-4/s and plastic failure mode was observed if strain was more than 10-2/s. The elastic wave velocity in the soil rapidly increased if dry density became larger than 1.5 g /cm$^3$ and these values were Vp=250, Vg= 150, respectively. Frost heave ratio was the highest around 0 $^{\circ}C$ and the maximum frost heave pressure was observed when deformation ratio was less than 10% which was the stability state of soil freezing. The state had no relation with frost depth. Over freezing process was observed when drainage or suction freezing process was undergone. Drainage freezing process was observed if freezing velocity was high under confined pressure and suction frost process was occurred if the velocity was low under the same confined process.
Crack widths play an important role in the serviceability limit state. When crack widths are controlled sufficiently, the reinforcement corrosion can be reduced using only existing concrete cover thickness due to low permeability in the region of finely distributed hair-cracks. Thus, the knowledge about the tensile crack opening is essential in designing more durable concrete structures. Therefore, numerous researches related to the topic have been performed. Nevertheless accurate measurement of a crack width is not a simple task due to several reasons such as unknown potential crack formation location and crack opening damaging strain gages. In order to overcome these difficulties and measure precise crack widths, a displacement measurement system was developed using digital image correlation. Accuracy calibration tests gave an average measurement error of 0.069 pixels and a standard deviation of 0.050 pixels. Direct tensile test was performed using ultra high performance concrete specimens. Crack widths at both notched and unnotched locations were measured and compared with clip-in gages at various loading steps to obtain crack opening profile. Tensile deformation characteristics of concrete were well visualized using displacement vectors and full-field displacement contour maps. The proposed technique made it possible to measure crack widths at arbitrary locations, which is difficult with conventional gages such as clip-in gages or displacement transducers.
A stochastic numerical analysis for predicting the groundswater fluctuations in hillside slopes is performed in this paper to account for the uncertainties associated with the rainfall and site characteristics. The effect of spatial variabilities of aquifer parameters and the effect of temporal variability of recharge on the groundwater fluctuations are studied in depth. The Kriging is used to account for the spatial tariabilities of aquifer parameters. This technique prolevides the best linear unbiased estimator of a parameter and its minimum variance from a litsitem number of measured data. A stochastic one-dimensional numerical model is delreloped b) combining the groundwater flow model, the Kriging, and the first-order second-moment analysis. In addition, a two dimensional detelministic groundwater model is developed to study the change of ground water surfas in the transverse direction as well as in the downslope direction. It is revealed that the undulations of the impervious bedrock in addition to the permeability and the specific yield have an important influence on the fluctuations of the groundwater surface. It is also found that th'e groundwater changes significantly in the transverse direction as well as in the downslope direction. The results obtained in this analysis may be used for evaluation of landslide risks due to high porewater pressure.

High-Pass (ETC ; Electronic Toll Collection) system is one of the basic elements, which adopts a wireless communication method using 5.8 GHz and can realize a part of ITS (Intelligent Traffic System). On the other hand, communication errors occur frequently in Hi-Pass system due to signal erros, multi-path reflection, and/or system-to-system interferences. To solve these problems, an EM (Electro-Magnetic) wave absorber can be used. To solve these Problems, we fabricated some samples in the different composition ratios of Carbon, Sendust, and CPE, and it was confirmed that the optimum composition ratio of Carbon : Sendust : CPE is 10 : 40 : 50 wt.%. The complex relative permittivity and complex relative permeability were derived by using the measured data. In addition, the optimum design parameters for the absorber were determined by simulation. Then the absorption abilities were calculated by changing the thickness of the EM wave absorbers. As a result, the optimum thickness of the developed EM wave absorber was 2.85 mm with absorption ability over 22.4 dB at 5.8 GHz. Futhermore, the EM wave absorber was fabricated based on the simulated and designed values. The measured values agreed well with the simulated ones. Therefore, it was clearly shown that the developed EM wave absorber in this paper is to be applied in actual situations.

The effects of annealing in rotating magnetic field after deposition on electromagnetic properties of $Co_{82}Zr_6Mo_{12}$ thin (200~1200 $\AA$) films prepared by RF-magnetron sputtering were investigated in terms of microstructure and surface morphology. The coercivity decreases, but $4{\pi}M_5$ does not change with increasing the film thickness. The coercivity of the films was decreased below 300 $^{\circ}C$ due to stress relief and decreasing the surface roughness, while increased at 400 $^{\circ}C$ due to partial grain growth. And then, $4{\rho}M_5$ was almost independent of annealing temperatures below 200 $^{\circ}C$, but increased from 7.4 kG to 8.0 kG at 300 $^{\circ}C$ and at 400 $^{\circ}C$, which was caused by precipitation and growth of fine Co particles in the films. The electrical resistivity of films was decreased with increasing annealing temperatures and the magnetoresistance was a negative value of nearly 0 $\mu$$\Omega$cm. After annealing at 300 $^{\circ}C$, maximum effective permeability was 1200 to the hard axis of the thin films according to high frequency change. Considering the practical application of biasing layers of the films for magnetoresistive heads, optimal annealing conditions was obtained after one hour annealing at 300 $^{\circ}C$ in 400 Oe rotating magnetic field.
Objective: The use of nanoparticle products is expected to present a potential harmful effect on consumers. Also, the lack of information regarding inhaled nanoparticles may pose a serious problem. In this study, we addressed this issue by studying pulmonary toxicity after nasal instillation of Al-NPs in SD rats. Methods: The animals were exposed to Al-NPs at 1 mg/kg body weight (low dose), 20 mg/kg body weight (medium dose) and 40 mg/kg body weight (high dose). To determine pulmonary toxicity, bronchoalveolar lavage (ts.AnBAL) fluid analysis and histopathological examination were conducted in rats. In addition, cell viability was investigated at 24 hours after the treatment with Al-NPs. Results: BAL fluid analysis showed that total cells (TC) count and total protein (TP) concentrations increased significantly in all treatment groups, approximately two to three times. Also, lactate dehydrogenase (LDH) and cytokines such as TNF-alpha and IL-6 dose-dependently increased following nasal instillation of Al-NPs. However, polymorphonuclear leukocytes (PMNs) levels showed no significant changes in a dose dependant manner in BAL fluid. In the cytotoxicity analysis, the treatment of Al-NPs significantly and dose-dependently induced cell viability loss (20 to 30%) and damage of cell membrane (5 to 10%) in rat normal lung epithelial cells (L2). Conclusions: Our results suggest that inhaled Al-NPs in the lungs may be removed quickly by alveolar macrophages with minimal inflammatory reaction, but Al-NPs have the potential to affect lung permeability. Therefore, extensive toxicity evaluations of Al-NPs are required prior to their practical application as consumer products.
