  • P. oleovorans의 유가식 배양에서 탄소원으로 octanoic acid, 질소원으로 $NH_4NO_3$를 이용한 혼합기질을 배양액의 pH 변화에 따라 공급하는 pH-stat 기질공급전략을 개발하였다. 공급기질의 탄소원/질소원 비 (C/N 비)를 변화시킴으로써 최종 균체농도, PHA 농도, PHA 함량 등을 변화시킬 수 있었으며, 최대 균체농도는 C/N 비가 10 (g octanoic acid/g $NH_4NO_3$)일 때 65 g/L, 최대 PHA 농도는 C/N 비가 20일 때 41 g/L, 최대 PHA 함량은 C/N 비가 20일 때 75%였으며 최대 PHA 생산성은 C/N 비가 10일 때 1.03 g/L/h였다.

Performance of Self-Manufactured Ion Selective Microelectrode (ISME) for Continuous Monitoring of Ammonia and Nitrate Ions

  • The ion selective microelectrodes (ISME) have been applied to observe the continuous profiles of NO3-N and NH4-N in bulk solutions or biofilms. In order to evaluate the performance and applicability of ion concentration measuring system, the characteristics, such as slope of calibration curve, detection limit and potentiometric selectivity coefficient were investigated. The slopes of calibration curve showed high degree of correspondence for each target ion concentrations. And the detection limits of nitrate and ammonia ion selective microelectrode were 10-4.7 M and 10-4.4 M, respectively. These ion selective microelectrodes were proved that their own performance could be maintained for 16 days after making. NO3-N and NH4-N selective microelectrodes were also adapted to detect the continuous ion profiles of cilia media packed MLE (Modified Ludzack-Ettinger) process. And the monitored nitrate and ammonia ion profiles with the ion selective microelectrode were stable and well corresponded to the results with conventional ion chromatograph. However, the electric potential was unstable until 8 hr because of the unknown noise. The tip shape and performance of the ion selective microelectrode was stably kept over 2 days continuous monitoring.

고농도 질산성 질소를 함유한 산세폐수의 생물학적 처리에 관한 연구 (A Study on the Biological Treatment of Acid Pickling Wastewater Containing a High Concentration of Nitrate Nitrogen)

  • The purpose of this study is the efficient biological treatment of highly concentrated nitrate nitrogen by calcium ion control present within the pickling wastewater. In laboratory scale's experiments research was performed using a sequencing batch reactor and the evaluation of denitrification reaction in accordance with the injection condition of calcium ions, the surface properties of microorganisms, and the evaluation of sludge precipitability were performed. Results of the study showed that the denitrification reaction was delayed when injecting more than 600 mg/L of the calcium ion within the denitrification process. In addition, we observed the absorption form of calcium ions absorbed on the surface of microorganisms following an increase in the calcium ion dose. It was found that as the calcium ion dose increased the sludge precipitability also increased continuously and it is judged that a smooth denitrification induction is possible when treating the nitrate nitrogen by the calcium ion control of pickling waste water and the shortening of precipitation time enables a liquid operation to increase the reaction time.

도시 대기 부유분진 중 음이온의 농도 (Concentrations of Anions in Airbone Particulate Matters in Metropolitan Area)

  • Total suspended particles were collected and size-fractionated by an Andersen high-volume air sampler at Chungang University located near the Han River, Seoul, Korea, during the period form March, 1986 to August, 1986 and from March, 1987 to February, 1988. The concentrations and the particle size distributions of anions such as chloride, nitrate and sulfate were determined by ion chromatography. The averages of concentrations were $125.43\;{\mu}g/m^3$ in 1986 and $189.19\;{\mu}g/m^3$ in 1987 for total suspended particles (TSP), $2.12\;{\mu}g/m^3$ in 1986 and $4.14\;{\mu}g/m^3$ in 1987 for chloride, $4.39\;{\mu}g/m^3$ in 1986 and $5.95\;{\mu}g/m^3$ in 1987 for nitrate and $11.98\;{\mu}g/m^3$ in 1986 and $19.29\;{\mu}g/m^3$ in 1987 for sulfate. Size distribution of TSP was found to be generally bimodal. Chloride exhibited a seasonal variation in the distribution; fine particles were predominant in the winter whereas the concentration of coarse ones was almost same through four seasons. Nitrate showed a distribution similar to that of chloride. Such variation was less significant for sulfate. For chloride and nitrate, the relationship between the monthly averaged distribution pattern and air temperature was analyzed in terms of fine fraction (FF). The FF decreased with increasing air temperature.

On-site Water Nitrate Monitoring System based on Automatic Sampling and Direct Measurement with Ion-Selective Electrodes

  • Purpose: In-situ monitoring of water quality is fundamental to most environmental applications. The high cost and long delays of conventional laboratory methods used to determine water quality, including on-site sampling and chemical analysis, have limited their use in efficiently managing water sources while preventing environmental pollution. The objective of this study was to develop an on-site water monitoring system consisting mainly of an Arduino board and a sensor array of multiple ion selective electrodes (ISEs) to measure the concentration of $NO_3$ ions. Methods: The developed system includes a combination of three ISEs, double-junction reference electrode, solution container, sampling system consisting of three pumps and solenoid valves, signal processing circuit, and an Arduino board for data acquisition and system control. Prior to each sample measurement, a two-point normalization method was applied for a sensitivity adjustment followed by an offset adjustment to minimize the potential drift that could occur during continuous measurement and standardize the response of multiple electrodes. To investigate its utility in on-site nitrate monitoring, the prototype was tested in a facility where drinking water was collected from a water supply source. Results: Differences in the electric potentials of the $NO_3$ ISEs between 10 and $100mg{\cdot}L^{-1}$ $NO_3$ concentration levels were nearly constant with negative sensitivities of 58 to 62 mV during the period of sample measurement, which is representative of a stable electrode response. The $NO_3$ concentrations determined by the ISEs were almost comparable to those obtained with standard instruments within 15% relative errors. Conclusions: The use of the developed on-site nitrate monitoring system based on automatic sampling and two-point normalization was feasible for detecting abrupt changes in nitrate concentration at various water supply sites, showing a maximum difference of $4.2mg{\cdot}L^{-1}$ from an actual concentration of $14mg{\cdot}L^{-1}$.

통계적 기법을 이용한 광주지역 민방위비상급수용 지하수 수질 오염도 분석 및 시각화 연구 (Visualization and contamination analysis for groundwater quality of CDEWSF in Gwangju area using statistical method)

  • 본 연구에서는 광주지역 민방위비상급수시설 101 개소를 대상으로 2006년부터 2016년까지 11년 동안 측정한 지하수 수질자료를 통계적으로 분석하였으며, 각 시설별로 먹는물 수질기준 초과횟수와 평균농도 분석 및 경향성 분석을 실시하고, 그 분석결과를 이용하여 4 개의 오염등급으로 구분하였다. 또한 이를 바탕으로 토지이용 현황별로 각 항목의 지하수 오염도를 평가하였다. 통계적으로 유의한 민방위 비상급수 15 개 항목에 대한 수질기준초과횟수, 평균농도분석, 경향성분석의 세가지 오염지시인자를 분석하여 합산한 결과 Turbidity (51.5 %) > Color (32.7 %) > Nitrate nitrogen (28.7 %) > Hardness (25.7 %) 순으로 오염 지시율이 상대적으로 높게 평가되었다. 오염등급 분석결과 안전(0)부터 오염심각(3)까지 오염등급이 다양하게 분포한 Turbidity, Color, Nitrate nitrogen의 3 개 항목을 제외하면 12 개 항목의 수질 오염등급은 전체의 25 %는 '오염가능한 등급(1)'으로, 75%는 '안전한 등급(0)'으로 평가되었다. QGIS를 이용하여 오염 지시율이 높게 평가된 4 개 항목(Turbidity, Color, Nitrate nitrogen, Hardness)은 오염지도에 작성하여 오염등급을 시각화하였다.

질산태 질소 정량을 위한 환원 증류법에서 Devarda's Alloy의 입자크기 및 함량이 미치는 영향 (Particle Size Effects of Devarda's Alloy on the Recovery of Nirate N Determined by the Steam Distillation Method)

  • 본 연구는 환원증류법을 사용하여 축사, 매립지, 시설재배지 등 높은 수준의 질산태 질소로 오염된 토양의 질산태질소 함량을 정확히 측정하는데 있어서, Devarda's alloy 의 입자 크기가 질산태 질소의 회수율에 미치는 효과와 Devarda's alloy의 처리량과 환원된 질산태 질소의 양과의 상관관계, 고농도의 질산태 질소를 함유한 토양 추출액의 정확한 분석을 위한 적절한 접근방법을 파악하고자 수행되었다. 본 연구에서 시험된 각 Devarda's alloy는 입자 크기의 분포가 서로 달랐으며, 이는 다소 높은 질산태 질소 조건인 1 mg과 2 mg $NO_3$-N에서 서로 다른 질산태 질소 회수율로 반영 되었다. 한편, 고농도의 질산태 질소 조건에서는 모든 Devarda's alloy들이 용액 중 질산태 질소의 함량이 증가할수록 급격히 질산태 질소의 회수율이 감소하는 경향을 보였으나, 시험된 모든 Devarda's alloy들은 예상과 달리, 단위 질량 당 환원된 질산태 질소의 양이 용액중 질산태 질소의 양에 비례하여 감소하는 경향을 보였다. 이상의 연구결과들은 높은 수준의 질산태 질소로 오염된 토양 시료를 안정적으로 분석하기 위해서는 Devarda's alloy의 입자 크기 분포를 감안한 충분한 처리, 그리고 두 수준 이상의 Devarda's alloy를 처리한 후 회수된 질산태 질소량 변화를 살피는 것이 필요하다는 것을 제시한다. 아울러, 본 연구에서 발견된 질산태 질소량의 화학적 비당량성은 앞으로의 연구를 통해 보다 자세히 조사되어야 할 것으로 사료된다.

채소(菜蔬) 및 과실중(果實中) 질산염(窒酸鹽)과 아질산염(亞窒酸鹽)의 축적(蓄積)에 관(關)한 연구(硏究) (Studies on the Accumulation of Nitrite and Niarite in Vegetables and Fruits)

  • Vegetables and fruits purchased from several markets in Seoul from July to October in 1977 were analyzed to know the level of nitrate-and nitrite-nitrogen accumulation in relation to a public health. Radishes and chinese cabbages utilized mainly as pickled vegetables in Korea resulted in the highest concentration of nitrate-nitrogen. Some of the levels for radishes and chinese cabbages were notably high and exceeded a recommended upper limit of 300 ppm $No_{3}-N$, and thus these levels would render these samples unsafe for use. The levels in some of vegetables other than radish and chinese cabbage, e.g., spinach, lettuce, green onion, cabbage were relatively high and considered to be unsafe for use in feeding infants, where as those of green pepper, bean sprouts and parsely were very low and safe. And also the levells in fruits were very low and safe. Nitrite-nitrogen contents in all tested vegetables and fruits ranged to trace and appeared not to be accumulated in fresh vegetables and fruits. Stems and roots of radishes and chinese cabbages accumulated approximately 2 fold more nitrate-nitrogen than leaves in 5 samples of each vegetable tested.

Biological nitrogen removal of ammonium-rich industrial wastewater by suspended bacterial growth

  • Industrial wastewater with high ammonium concentration was treated in batch biological systems which was a modified Ludzack- Ettinger process. Up to 78% conversion of $NH_4\;^+-N$ to $NO_x\;^--N$ was achieved in batch culture condition. Under anoxic condition with methanol as the carbon source, the denitrifiers decreased $NO_x\;^--N$ concentration from 608 mg/L to 5.6 mg/L in 22 d. As well as anoxic denitrification of $NO_x\;^-$ to $N_2$, dissimilatory nitrate reduction to ammonium also occurred under the condition as respiratory denitrification.
