초고압 시스템을 이용한 생물 산업의 적용 (Application of Biological industry using High Hydrostatic Pressure (HHP) system)

  • 본고에서는 초고압 시스템 (High hydrostatic pressure (HHP))의 특성, 종류, 응용을 예로 들어 설명하였다. 초고압 기술은 식품의 살균, 식품 보존기간연장, 유효성분의 추출 등에 선택적으로 적합하게 적용하여 생물산업공정에 효율적으로 응용 할 수 있다. 초고압 시스템을 효율적으로 적용하기 위한 최적조건을 선정하는 것은 많은 조업 변수에 대한 고려를 해야 하는 힘든 작업이다. 따라서 하드웨어에 대한 기초 실험과 예비 생산을 통하여 공정에 대한 모사를 검증하고 생산 효율을 증가시킬 수 있다. 또한 초고압 시스템의 중요성이 증가함에 따라 고성능 및 사용이 더욱 편리한 시스템이 나타날 것이며, 많은 영역의 바이오산업에서 필수적으로 이용될 것이다.

초고압 처리에 의한 감귤의 추출률 및 특성변화 (Quality Characteristics of Citrus Fruit by Cyclic Low Pressure Drying and High Hydrostatic Pressure Extraction)

  • 본 연구에서는 순환형 감압건조 및 초고압 추출공정을 이용하여 전통적인 기존 추출공정과 비교함으로써 복합 추출공정에 의한 감귤의 항산화 활성 증진을 확인하고자 연구를 수행하였다. 건조공정을 거친 후 초고압 처리 추출물의 수율이 20.41~28.19%로 높은 추출 수율을 나타내어 건조전 열수추출공정(17.21%)과 비교하여 약 1.6배의 높은 추출수율을 나타내었다. 순환형 감압건조와 초고압 공정을 병행하였을 시 총 폴리페놀과 플라보노이드 함량이 순환형 감압건조공정을 거치지 않은 것보다 다소 증가되는 것으로 보아 활성성분의 용출이 증진된 것으로 보인다. DPPH radical 소거 활성은 15분 초고압 처리한 추출물이 48.21%로 높은 활성을 나타내었다. 전처리 공정에 따른 감귤 시료의 주사전자현미경(SEM)을 통해 순환형 감압건조 후 초고압 추출이 감귤 내부 조직까지 영향을 주어 세포벽이 깨어지면서 조직 및 구조가 변화한 것으로 판단되며, 이를 통해 수율 및 활성 성분의 용출 증가가 이루어 진 것으로 사료된다. 따라서, 감귤의 건조 및 초고압 추출공정의 최적화를 통한 활성물질의 추출 극대화를 통해 추출수율을 향상시킬 것으로 판단된다.

Ginsenoside composition of Panax ginseng flower extracts obtained using different high hydrostatic pressure extraction conditions

  • Ginsenosides are active constituents of ginseng (Panax ginseng) that have possible anti-aging, physiological and pharmacological activities, such as anti-cancer and anti-inflammatory effects. Although the ginseng root is generally used more often than the aerial parts for medicinal purposes, the flowers also contain numerous ginsenosides, including Rb2, Rc, Rd, Re and Rg1. Therefore, an extract from the flowers of the P. ginseng could have the pharmacological efficacy of bioactive compounds including ginsenosides. The high hydrostatic pressure extraction (HHPE) is a method that is used for the efficient extraction of bioactive compounds from plant materials. In this study, we compared the yield of ginsenosides from ginseng flowers under different conditions of extraction pressure and time of HHPE. The results indicate that the total yield of the ginsenosides improved as the pressure increased from 0.1 to 80 MPa and treatment duration increased to 24 hours. In addition, the ginsenoside extracts from HHPE at 80 MPa, which possessed a higher total ginsenoside concentration, decreased the viability of the primary human epidermal keratinocytes (HEKs) significantly than the ginsenoside extracts from HHPE at 0.1 MPa. Collectively, we found that the method of HHPE that was performed for 24 hours at 80 MPa showed the highest yield of ginsenosides from the flowers of P. ginseng. In addition, our study provides a foundation for the efficient extraction of ginsenosides, which had a potent bioactivity, from flowers of P. ginseng through HHPE.

Extract of high hydrostatic pressure-treated danshen (Salvia miltiorrhiza) ameliorates atherosclerosis via autophagy induction

  • Danshen (Salvia miltiorrhiza) is a traditional medicinal plant widely used in Asian countries for its pharmacological activities (e.g., amelioration of cardiovascular diseases). In this study, we investigated the anti-atherosclerotic activity of raw danshen root extract prepared using high hydrostatic pressure (HHP) at 550 MPa for 5 min and hot water extraction. This method was useful for elimination of bacteria from cultured danshen plants and for better extraction yield of active principles. The HHP-treated danshen extract (HDE) inhibited proliferation of human umbilical vein endothelial cells (HUVECs) and induced autophagy that was assessed by LC3 conversion and p62 degradation. HDE suppressed foam cell formation in oxLDL-induced RAW264.7 macrophages; lysosomal activity simultaneously increased, measured by acridine orange staining. HDE also reduced atherosclerotic plaque development in vivo in apolipoprotein E knock-out (ApoE-/-) mice fed a high cholesterol diet. Taken together, these results indicated that HDE exhibited anti-atherosclerotic activity via autophagy induction.

Effects of High-Hydrostatic Pressure on Ginsenoside Concentrations in Korean Red Ginseng

  • The effects of high-hydrostatic pressure (HHP) on the ginsenoside concentration in Korean red ginseng were investigated. HHP-pretreated Korean red ginseng samples were compared to samples produced by a conventional method. Six-year-old Korean fresh ginseng (Panax ginseng C.A. Meyer) samples were vacuum-packaged in polyethylene film and treated at room temperature for 1 min with HHP (200-600 MPa) and steamed at $98^{\circ}C$ for 3 hr. Major ginsenosides of red ginseng were analyzed by HPLC. HHP-pretreated red ginseng showed a 45% higher level of total major ginsenosides than conventionally prepared red ginseng. The levels of 4 protopanaxadiol-type ginsenosides increased 34-43% and the levels of 5 protopanaxatriol-type ginsenosides increased 45-66%. Scanning electron microscopy and electrical conductivity spectrum analysis showed that HHP pretreatment damaged ginseng plant cells and increased extraction efficiencies of ginsenosides from red ginseng products.

한약재 가공 기술의 특허 동향 연구 - 비가열 가공 기술을 중심으로 - (Analysis on Patent Trends in Nonthermal Processing Technologies for Medicinal Herbs)

  • The purpose of this study was to analyze the patent application trend in the processing technology for medicinal herbs. Recently, in processing technology for medicinal herbs, experimental researches have frequently been published through papers in journals. However, the research results about the patent area were fewer than the others. We tried to analyze the patent application trend in nonthermal processing technologies for medicinal herbs by country as Korea, Japan, U.S.A. and Europe. The detailed technologies consisted of pulsed electric field, oscillatory magnetic field, intense pulsed light, ultrasonification, high hydrostatic pressure, microwave, radiation, Ohmic heating, and supercritical extraction. As a result we found that patents of nonthermal processing technologies has been growing steadily in quantity from 1980s and growing quickly since 2000s. The number of patent in Korea is larger than others as making up 70% in that whole. The number of patent in ultrasonification field was larger than others in portfolio analysis. Patent application trend in nonthermal processing technologies for ingestion occupies high share compared to other usage applications. In conclusion, patent trends of nonthermal processing technologies for medicinal herbs belong to the period in the development.
