Effects of TongBiEum(TBE) on Hyperlipidemia Induced by a Sucrose-rich Diet

  • Objectives : This study aimed to elucidate the effects of TBE on hyperlipidemia. Methods : We studied the effects of TBE on hyperlipidemia through gene expressions related with lipid metabolism and serum triglyceride as well as total and HDL-cholesterol levels, and perceived histological changes. Results : The present studies demonstrate that TBE can reduce the rise in plasma cholesterol and TG levels induced by a high-cholesterol diet and also reverse pre-established hypercholesterolemia and hypertriglycemia. In the TBE group total cholesterol levels decreased, TG levels decreased, but HDL-cholesterol levels also decreased. In the analysis of absolute and relative liver weight, TBE inhibited the weight gain induced by a high-cholesterol diet. In the histological observations, lipid droplet and apoptotic change in the TBE treated group were less compared with the control group. In the serum biochemical analysis, a difference of serum AST and ALT changes among groups was not shown, but TG and total cholesterol levels were less and HDL level decreased compared with the control group. In the gene expression related with TG and cholesterol metabolism, DGAT decreased slightly but ACAT decreased more as compared with control and Lipidil groups. Conclusion : From this study, we can infer that TBE possesses a hypolipidemic effect by inhibiting the intestinal absorption and storage of exogenous and endogenous cholesterol.

모시잎이 고지방-고콜레스테롤 식이 흰쥐의 지질대사 개선 및 항비만 효과에 미치는 영향 (Effects of Ramie Leaves on Improvement of Lipid Metabolism and Antiobesity Effect in Rats Fed a High Fat/High Cholesterol Diet)

  • 본 연구는 모시잎의 혈청, 간 및 지방조직의 체내 지질대사 개선 및 항비만 효과를 살펴보기 위하여 in vivo에서 5주령 된 흰쥐 수컷 24마리를 1주일간 적응시킨 후 정상 식이군(N), 고지방-고콜레스테롤 식이군(HFC), 고지방-고콜레스테롤 식이와 5% 모시잎 분말 첨가군(HFC-RL) 및 고지방-고콜레스테롤 식이와 10% 모시잎 분말 첨가군(HFC-RH)으로 나누어 4주간 실시하였다. 고지방-고콜레스테롤 식이로 인하여 체중이 유의적으로 증가하였으나, 모시잎 분말의 첨가 수준이 증가할수록 체중이 유의하게 감소하였다. 식이섭취량은 대조군(HFC)이 가장 낮았으며, 모시잎 분말 급여로 감소하는 경향이었다. 식이효율은 10% 모시잎 분말첨가로 유의하게 감소하여 정상 식이군(N)의 식이효율 수준과 유사하였다. 장간막지방조직과 부고환지방조직의 무게는 10% 모시잎 분말 첨가로 유의하게 감소하여 정상 식이군(N)보다 오히려 더 감소하는 경향을 보였다. 고지방-고콜레스테롤 식이로 증가되어진 혈청 중 AST, ALT, ALP 및 LDH 활성은 모시잎 분말 급여로 저하되었다. 혈청 중 중성지방 함량과 총콜레스테롤 함량도 고지방-고콜레스테롤 식이 급여로 증가되었으나, 모시잎 분말 첨가에 따라 농도 의존적으로 감소하였다. 고지방-고콜레스테롤 식이로 감소되어진 혈청 중 HDL-콜레스테롤 함량은 모시잎 분말 급여로 모두 유의하게 증가하였으며, 증가되어진 혈청 중 LDL-콜레스테롤 함량은 모두 유의하게 감소하였다. 동맥경화지수와 심혈관위험지수는 5%와 10%의 모시잎 분말 급여로 모두 유의하게 감소하였으며, 특히 10% 모시잎 분말을 급여하였을 경우 감소효과가 커 정상 식이군(N)과 유사한 경향이었다. 간조직 중 중성지방과 총콜레스테롤 함량은 모시잎 분말 급여로 모두 대조군(HFC)에 비하여 유의하게 감소하였다. 장간막지방조직과 부고환 지방조직의 중성지방 및 총콜레스테롤 함량도 모시잎 분말 급여로 유의하게 감소하였다. 지방조직의 HR-LPL과 TE-LPL 활성은 모시잎 분말의 첨가 급여로 농도 의존적으로 유의하게 감소하였다. 이상의 실험 결과 고지방-고콜레스테롤 식이와 함께 급여한 모시잎 분말은 체중, 간 및 지방조직의 무게 감소와 더불어 혈청 및 지방조직의 지질대사 개선에 긍정적인 영향을 미치는 것으로 나타났다. 이러한 결과는 모시잎 분말에 다량 함유된 식이섬유소와 클로로필 등의 생리활성 물질이 지질대사를 개선시키고 지방축적을 억제하여 고지혈증의 예방 및 치료효과와 비만을 억제하는 효과를 나타내 대사성 합병률과 심혈관계 질환의 발생을 감소시키는데 효과가 있을 것으로 생각되어진다. 그 외에 어떠한 생리활성 성분이 어떻게 작용을 하는지에 대해서는 추후 더 체계적인 연구가 필요할 것으로 사료된다.

대두 추출물이 지방식이를 급여한 흰쥐의 혈청 지질 성분에 미치는 영향 (Effects of Soybean Extract on Serum Lipid Contents in Fed Fat Diet Rats)

  • 각 지방 식이를 달리 처리하여 물과 대두 추출물을 섭취한 흰쥐의 혈청 지질 함량에 미치는 영향을 조사하기 위해 체중 $100{\pm}10g$의 Sprague-Dawley계 수컷 흰쥐 98마리를 콜레스테롤 비투여군, 표준지방식이, 동물성 지방식이, 식물성 지방식이, 고지방식이에 각각 1% 콜레스테롤을 첨가하여 물과 대두 추출물을 달리 섭취하여 처리한 실험군을 14개군으로 분류하여 4주 동안 사육하였다. 체중 증가량은 각 지방식이를 달리 하여 물만 섭취한 군과 대두 추출물 처리를 달리하여 섭취한 군들 사이에서 대두 추출물 군이 약간 증가하였으나 유의적인 큰 차이는 없었다. 혈청 중 중성지질과 콜레스테롤 함량에서 물만 섭취한 군에 비하여 대두추출물군에서 낮은 함량을 나타냈다. HDL-, LDL-cholesterol은 고지방식이와 동물성 지방식이에서 대두콩 추출물의 투여후 혈중 지질 개선 효과가 뚜렷하게 관찰되었고, 특히 식물성 지방 식이와 대두 추출물을 같이 투여한 군은 상승 작용을 가져와 혈중 지질 증가 억제 효과가 컸다.

The effect of seamustard on blood lipid profiles and glucose level of rats fed diet with different energy composition

  • Recently, Korean people are consuming seaweeds almost 3.5 times more now than three decades ago. It is well known that seaweeds contain lots of soluble dietary fiber in addition to micronutrients such as ${\beta}$-carotene, iodine and some bioactive components. Seaweeds are considered to be effective for preventing chronic diseases including obesity, diabetes mellitus, atherosclerosis, cancer or constipation. This study was conducted to investigate the effect of seamustard intake on body weight gain, blood glucose level and lipid profiles in rats fed diets with different energy nutrient composition. Male Sprague-Dawley rats (average initial weight 103.7g) were divided into groups for two experiments as follows; Control, M2.5 & M5 groups (Exp. I) and M5, M10, HCM5, HCM10, HFM5 & HFM10 groups (Exp. II). The rats were fed diet and water ad libitum for 4 weeks. In general, there was no significant difference in blood glucose and triglyceride concentration among groups. In Exp. I, serum LDL-cholesterol level of rats fed diet with 5% seamustard powder (M5) was significantly lower than that of control group, while HDL-cholesterol level, TC/LDL ratio and weight of adrenal gland were higher. In Exp. II, food intake, body weight gain and EER of high fat diet with 10% seamustard group (HFM10) were the lowest among groups. Except gastrocnemius muscle, all organ weights of HFM10 group were the lowest. Fecal cholesterol excretion and serum LDL-cholesterol concentration of HFM10 group were the highest, while serum HDL-cholesterol level was the lowest among groups. Interestingly, HDL-cholesterol concentration was the highest in HCM5 group among groups. From these results, it was suggested that seamustard intake might be more effective for body weight control, but not for improving blood lipid profiles in high fat diet than in high carbohydrate diet.

${\alpha}$-Lipoic acid reduced weight gain and improved the lipid profile in rats fed with high fat diet

  • The purpose of this study was to investigate the effects of ${\alpha}$-lipoic acid on body weight and lipid profiles in Sprague-Dawley rats fed a high fat diet (HFD). After 4 weeks of feeding, rats on the HFD were divided into three groups by randomized block design; the first group received the high-fat-diet (n = 10), and the second group received the HFD administered with 0.25% ${\alpha}$-lipoic acid (0.25LA), and the third group received the high-fat diet with 0.5% ${\alpha}$-lipoic acid (0.5LA). The high fat diet with ${\alpha}$-lipoic acid supplemented groups had significantly inhibited body weight gain, compared to that in the HFD group (P < 0.05). Organ weights of rats were also significantly reduced in liver, kidney, spleen, and visible fat tissues in rats supplemented with ${\alpha}$-lipoic acid (P < 0.05). Significant differences in plasma lipid profiles, such as total lipids, total cholesterol, triglycerides, low-density lipoprotein, and high-density lipoprotein, were observed between the HFD and 0.5LA groups. The atherogenic index and the plasma high density lipoprotein-cholesterol/total cholesterol ratio improved significantly with ${\alpha}$-lipoic acid supplementation in a dose-dependent manner (P < 0.05). Total hepatic cholesterol and total lipid concentration decreased significantly in high fat fed rats supplemented with ${\alpha}$-lipoic acid in a dose-dependent manner (P < 0.05), whereas liver triglyceride content was not affected. In conclusion, ${\alpha}$-lipoic acid supplementation had a positive effect on weight gain and plasma and liver lipid profiles in rats.

Cholesterol-induced inflammation and macrophage accumulation in adipose tissue is reduced by a low carbohydrate diet in guinea pigs

  • BACKGROUND/OBJECTIVES: The main objective of this study was to evaluate the effects of a high cholesterol (HC) dietary challenge on cholesterol tissue accumulation, inflammation, adipocyte differentiation, and macrophage infiltration in guinea pigs. A second objective was to assess whether macronutrient manipulation would reverse these metabolic alterations. MATERIALS/METHODS: Male Hartley guinea pigs (10/group) were assigned to either low cholesterol (LC) (0.04g/100g) or high cholesterol (HC) (0.25g/100g) diets for six weeks. For the second experiment, 20 guinea pigs were fed the HC diet for six weeks and then assigned to either a low carbohydrate (CHO) diet (L-CHO) (10% energy from CHO) or a high CHO diet (H-CHO) (54% CHO) for an additional six weeks. RESULTS: Higher concentrations of total (P < 0.005) and free (P < 0.05) cholesterol were observed in both adipose tissue and aortas of guinea pigs fed the HC compared to those in the LC group. In addition, higher concentrations of pro-inflammatory cytokines in the adipose tissue (P < 0.005) and lower concentrations of anti-inflammatory interleukin (IL)-10 were observed in the HC group (P < 0.05) compared to the LC group. Of particular interest, adipocytes in the HC group were smaller in size (P < 0.05) and showed increased macrophage infiltration compared to the LC group. When compared to the H-CHO group, lower concentrations of cholesterol in both adipose and aortas as well as lower concentrations of inflammatory cytokines in adipose tissue were observed in the L-CHO group (P < 0.05). In addition, guinea pigs fed the L-CHO exhibited larger adipose cells and lower macrophage infiltration compared to the H-CHO group. CONCLUSIONS: The results of this study strongly suggest that HC induces metabolic dysregulation associated with inflammation in adipose tissue and that L-CHO is more effective than H-CHO in attenuating these detrimental effects.

식이내 Ca 수준과 지방 종류를 달리하였을 때 흰쥐의 체내 지방대사에 미치는 영향 (Effects of Dietary Ca Levels and Kinds of Lipids on the Lipid Metabolism in the Rats)

  • Effects of a change in dietary Ca content and an alteration in dietary lipid type on lipid metabolism have been investigated in male Sprague-Dawley rats. The results obtained were summarized as follows ; 1) There was no comparable changes in food consumption and body weight gain among all 9 groups. 2) Serum total lipids and cholesterol were lowest when high Ca-corn all diet was fed, and highest when low Ca-butter diet was fed. 3) The contents of total lipids and cholesterol per g liver and total liver showed no consistent tendency with the dietary treatment. 4) Addition of Ca to the diet caused the significant increase in the dry fecal matter and fecal Ca. And fecal total lipids and fecal cholesterol were much greater if the high-Ca diet was fed than if control and low-Ca diet were fed. It is concluded that Ca and polyunsaturated acids have tendency of hypocholesterolemic effect.

고콜레스테롤 식이에 있어 키토산 올리고당이 체내 콜레스테롤농도 및 항산화효소 활성에 미치는 영향 (Effect of Chitosan Oligosaccharides on Cholesterol Level and Antioxidant Enzyme Activities in Hypercholesterolemic Rat)

  • 본 연구는 고콜레스테롤을 급여한 흰쥐 에 키토산 올리고당의 첨가 수준을 달리한 식이가 흰쥐의 지방대사에 미치는 영향을 알아보고자 수행하였다. 체중이 174.7$\pm$6.3 g인 Sprague-Dawley종 수컷 흰쥐에게 고콜레스테롤혈증을 유발시키기 위하여 콜레스테롤을 식 이 무게의 0.5%를 급여하고 키토산 올리고당을 식이 무게의 1.0%와 2.0%로 하여 총 3군으로 나누어 4주간 사육하였다. 식이섭취량과 체중증가량은 식이에 의한 영향이 없었다. 혈청 중 총 콜레스테롤, 중성지방은 대조군에 비해 2.0% 키 토산 올리고당을 첨가한 군에서 유의적으로 그 농도가 크게 감소하였으며 HDL-콜레스테롤/총콜레스테롤 비는 키토산 올리고당 첨가에 의해 높아지는 경향을 보였다. 간 조직 중 TBARS 함량은 키토산 올리고당을 첨가하였을 경우 유의적으로 낮게 나타났으며 SOD와 catalase의 활성은 키토산 올리고당을 첨가한 군이 대조군에 비해 유의적으로 낮았다. 이상의 결과를 볼 때 키토산 올리고당은 체내 지방대사에 있어 총콜레스테롤과 LDL-콜레스테롤, 중성지방 및 TBARS 함량을 낮추고 HDL-콜레스테롤/총 콜레스테롤 비는 높이는 효과를 나타냈으며, 또한 생체내 고콜레스테롤에 의한 산화적 스트레스로부터 간을 보호하는 것을 알 수 있었다.

알코올 투여와 아연과 구리의 섭취비율 증가가 흰쥐의 혈청지질함량에 미치는 영향 (The Effect of the Ethanol Feeding and Increase in the Raion of Zine to Copper Ingested on Serum Lipid Control in the Rats)

  • This study was undertaken to investigate the effect of increase in the ratio of zinc to copper and alcohol consumption on serum lipid content in Sprague-Dawley male rats during five weeks. Sixty rats were divided into six groups, CZ < zinc 30ppm diet >, CZA < zinc 30 ppm diet plus alcohol 5% solution >, HZ < zinc 300ppm diet >, HZA < zinc 300ppm diet plus alcohol 5% solution >, SHZ < zinc 600ppm diet >, SHZA < zinc 600 ppm diet plus alcohol 5% solution >. High ratio of zinc to copper groups < HZ, SHZ > were significantly decreased in high-density lipoprotein (p<0.01) and increased in total cholesterol, triglyceride, total lipid in serum(p<0.001, p<0.001, p<0.01). Triglyceride levels of serum in alcohol groups < CZA, HZA, SHZA > were significantly higher (p<0.001) than non-alcohol group < CCZ, HZ, SHZ >. Copper levels of serum were reduced in group high ratio of zinc to copper. These results present that high ratio of zinc to copper increase cholesterol(p<0.001) and alcohol consumption increase triglyceride(p<0.001) in serum. This close association to lipid metabolism in rats draw attention to the potential significance of zinc and alcohol nutriture in relation to cardiovascular disease.

천남성이 고지방식이로 유발된 비만 흰쥐의 당지질대사 및 조직화학적 변화에 미치는 효과 (Effects of Rhizoma Arisaematis on Lipid and Glucose Metabolism and Histochemical Changes of Rats Fed High Fat Diet)

  • Objectives : This study was performed to investigate the effects of Rhizoma Arisaematis water extract on lipid and glucose metabolism and histochemical change of obese rats. Methods and materials : 10 rats were divided into normal, control and RA (Rhizoma Arisaematis) groups. We fed the control group a high-fat diet and administered normal saline for 8 weeks. We fed the experimental group of rats a high-fat diet and administered an extract of Rhizoma Arisaematis for 8 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. The groups were examined for effects on blood serum lipids, blood sugar, blood insulin concentration and epididymal fat cells. Results : 1. Serum total cholesterol, triglyceride, HDL-cholesterol and glucose of the RA group decreased compared with those of the control group. These decreased rates were significant(P<0.05). 2. Serum LDL-cholesterol, total lipid, free fatty acid and the average size of epididymal fat cells of the RA group decreased compared with those of the control group. These decreased rates were significant(p<0.01). Conclusions : These results suggest that RA may be used to prevent or cure the obesity iniduced by a high-fat diet.
