Characterization of Heterodera sojae Virulence Phenotypes in Korea

  • The white soybean cyst nematode Heterodera sojae, isolated from the roots of soybean in Korea, is widespread in most provinces of the country and has the potential to be as harmful to soybean as H. glycines. Determining the virulence phenotypes of H. sojae is essential to devising management strategies that use resistant cultivars. Consequently, virulence phenotypes of 15 H. sojae populations from Korea were determined on seven soybean lines and one susceptible check variety. Two different HS types were found to be present in Korea; the more common HS type 2.5.7, comprising 73.3% of the H. sojae populations and the less common HS type 0, constituting only 26.7% of the tested populations. Considering the high frequency of H. sojae adaptation to soybean indicator lines, the PI 88788 group may not be a possible source of resistance while PI 548402, PI 90763, PI 437654, and PI 89772 can be used as resistance sources for soybean breeding programs aimed at developing H. sojae-resistant soybean cultivars in Korea.

  • A new soybean cyst nematode, H. sojae was reported from Korea in 2016. This species is closely related to H. glycines which has the same host plant. Morphological observations of the cyst, female, male and second-stage juveniles indicated that this species is differed from H. glycines. Especially, cysts of H. sojae appeared more round, shining and darker than that of H. glycines. The distribution of this new cyst nematode in soybean field in Korea is unknown so far. In 2016, 270 soil samples were collected from soybean fields and examined the existence of H. glycines and H. sojae. Total of 111 samples contained cysts (41.1%). Among them 77% were H. glycines and 23% were H. sojae. H. sojae is future threatening in soybean production area.

강원과 충북지역 콩씨스트선충과 반짝이콩씨스트선충의 분포상 및 토양 특성 (Plant Parasitic Nematode Fauna of Heterodera glycines and H. sojae, and Soil Characteristics of Soybean Fields in Gangwon-do and Chungcheongbuk-do)

  • 콩 재배지에서 콩씨스트선충(Heterodera glycines)과 반짝이씨스트선충(Heterodera sojae)의 지리적 분포에 관한 조사를 강원도와 충청북도 지역에서 수행하였다. 강원지역 10개시군, 67개 포장에서는 콩씨스트선충이 13개소, 반짝이콩씨스트선충이 5개소에서 검출되었고, 두 선충 혼재하는 포장도 2곳이었다. 충북지역 9개 시군에서는 84개 조사 포장 중 27개소에서 콩씨스트선충이 검출되었고, 반짝이콩씨스트선충 4개소, 두선충 혼재 발생지 5곳이었다. 강원, 충북 지역에서 콩씨스트선충류 검출 여부에 따라 발생하는 다른 식물기생성선충 속의 다양성과 우점도 지수는 콩 재배지 사이에 유의한 차이를 나타내지 않았다. 콩씨스트선충과 토양 화학성 사이의 관계 분석에서는 결정계수가 낮게 나타났으며 콩씨스트선충의 밀도가 높은 토양에서는 pH와 가용성 인산 함량이 높은 것으로 나타났다. 씨스트선충이 검출된 토양의 토성은 사질토가 전체 샘플의 60%를 차지하였다. 점토와 미사의 함량이 높은 토양에서는 콩씨스트선충의 밀도가 낮은 경향을 보였고, 모래 함량이 높은 토양에서 밀도는 높게 나타났다.

Identification of Heterodera glycines (Tylenchida; Heteroderidae) Using qPCR

  • The soybean cyst nematode, Heterodera glycines, is a major plant-parasitic nematode that has caused important economic losses to Korea's soybean production. Four species of cyst nematodes, H. schachtii, H. glycines, H. trifolii, and H. sojae, all belong to schachtii group are coexist in field soil in Korea. The rapid identification of the nematode is crucial for preventing crop damage and in decision making for controlling this nematode. This study aimed to develop a species-specific primer set for quantitative PCR (qPCR) assay of H. glycines. The specific primer set (HGF1 and HGR1) for H. glycines was designed based on the cytochrome c oxidase subunit I (COI) sequence of mitochondrial DNA. After optimization, it is possible to identify the H. glycines using a qPCR assay with DNA extracted from a single cyst and single second-stage juvenile (J2). The specificity was confirmed by the absence of SYBR fluorescent signals of three other Heterodera species. A serial dilution of DNA extracted from a single cyst was obtained for the sensitivity test. The result showed that the standard curve of the test had a highly significant linearity between DNA concentration and Ct value (R2 = 0.996, slope = -3.49) and that the detection limit concentration of DNA of the primer set was 10 pg of DNA per reaction. Our findings suggested that H. glycines could be distinguished from H. sojae and other Heterodera species when a qPCR assay is used with a specific primer set.

충남지역 두류작물 재배지 식물기생선충 감염현황 (Incidence of Plant-parasitic Nematodes from Legume Fields in Chungnam Province)

  • To survey the incidence of plant-parasitic nematodes, 187 soil samples from leguminous plant fields in Chungnam province, Korea were assessed from June to August 2018. As the result, four nematode genera - the cyst nematode (Heterodera spp.), root-knot nematode (Meloidogyne spp.), root-lesion nematode (Pratylenchus spp.), spiral nematode (Helicotylenchus spp.) - were detected. Among them, the incidence of cyst nematode was the highest in Taean (73%) and lowest in Hongseong (10%). The cyst nematode was detected from the adzuki and kidney bean fields as well as soybean fields. The incidence of cyst nematode in upland fields was higher than in paddy fields. In addition, the extracted cyst nematodes consisted of two species, which was identified as Heterodera glycines and H. sojae. The incidence of H. glycines (22%) was higher than H. sojae (10%), and a few of the fields (5%) was mixed with two cyst nematodes. This result suggested that we should consider that the cyst nematode is the major plantparasitic nematode when controlling the nematodes in soybean fields in Chungnam province.

PCR-RFLP를 이용한 국내 분포 씨스트선충 4종의 동정 (Identification of Four Cyst Nematodes using PCR-RFLP in Korea)

  • To identify four cyst nematodes (Heterodera schachtii, H. trifolii, H. glycines, H. sojae) that are economically important plant-parasitic nematodes in Korea, restriction fragment length polymorphism (RFLP) by 8 endonucleases (PstI, VspI, AlwI, RsaI, MvaI, EcoRI, Eco72I, Hinf I) was performed based on sequence difference of mitochondrial DNA cytochrome c oxidase subunit I (COI) gene. As a result, species-specific DNA band patterns by RsaI endonuclease were observed in H. schachtii. The specific patterns was in H. trifolii by 3 endonucleases (VspI, AlwI, Hinf I), and was in H. glycines by Hinf I. While, H. sojae was not digested by 4 endonuclease (VspI, AlwI, RsaI, Hinf I). This study showed that four cyst nematodes could be distinguished using RFLP by 4 endonucleases (RsaI, VspI, AlwI, Hinf I) based on the sequence difference of COI gene.

Differential Morphological, Structural and Biological Characteristics of Cysts in Heterodera Species in Korea

  • Morphological (cyst shape, color, and sizes [length (L), maximum width (W), volume and "a" (L/W)]), structural (vulvar cone slope angle [VCSA], surface wrinkle [VCSW], cyst wall thickness, composition, and texture) and biological characteristics (fecundity, hatching, and emergence [number of second-stage juveniles (J2) from a cyst]) in preceding Heterodera glycines (Hg), currently-recorded H. sojae (Hs) and H. trifolii (Ht) were examined by microscopy. Cysts were lemon-shaped, indicating the genus is Heterodera except for Hs that formed frequently globular cysts with significantly flatter VCSA (102.2°) with smooth VCSW than Hg (50.6°) and Ht (82.0°), but not genus Globodera because of the presence of vulvar cone in Hs. Ht was significantly larger in all morphological characteristics than Hg and Hs, suggesting Ht may be diagnosed differentially by cyst sizes and also host plant preferences. Hs showed smaller "a" value with more globular shape and stronger structures with more thickened and strengthened collagen-like texture of cyst wall than Hg and Ht. This suggests Hs may be diagnosed differently by structural characteristics from the others, especially Hg with similar cyst sizes. There were no significant differences in emergence (inoculum potential) among cyst nematodes due to the offset of fecundity and hatching rate; however, the inoculum potential of Hs may be not so persistent as Hg and Ht in fields because of its lower fecundity and higher hatching rate (causing rapid inoculum loss) than the others. These characteristics of cysts provide information useful for simple and differential diagnoses and reliable management of cyst nematodes.
