A Plant Metabolomic Approach to Identify the Difference of the Seeds and Flowers Extracts of Carthamus tinctorius L.

  • Carthamus tinctorius L. (known as safflower) is a valuable oil plant whose importance is increasing rapidly in the world due to its high adaptation to arid regions. The seeds of this unique plant are especially used in edible oil, soap, paint, varnish and lacquer production. Its flowers are used in vegetable dye production and medicinal purposes beside its features as a coloring and flavoring in food. After the oil is removed, the remaining pulp and plant parts are used as animal feed, and dry straw residues are used as fuel. Beside all these features, its usage as a herbal medicinal plants for various diseases has gained importance on recent years. In this study, it was designed a plant metabolomic approach which transfers all the recent data processing strategies of untargeted metabolomics in clinical applications to the present study. Q-TOF LC/MS-based analysis of the extracts (70% ethanol, hexane, and chloroform) for both seed and flowers was performed using a C18 column (Agilent Zorbax 1.8 µM, 100 × 2.1 mm). Differences were observed in seed and fruit extracts and these differences were visualized using principal component analysis (PCA) plots. The total number and intersections of the peaks in the extracts were visualized using peak count comparison graph. Based on the experimental results, the number of the detected peaks for seeds was higher than the ones for the flowers for all solvent systems to extract the samples.

큰느타리버섯 수확후 관리기술 최근 연구 동향 (Recent research trends of post-harvest technology for king oyster mushroom (Pleurotus eryngii))

  • 큰느타리버섯은 에르고스테롤 및 글루칸과 같은 기능성 물질의 함량이 높아 항산화 활성 및 프리바이오틱 효과를 일으키는 버섯이다. 큰느타리버섯은 향이 강하며 단단해 기호도가 높지만 장기간 소요되는 선박수출 등에서는 선도유지 기간 확보가 필요하다. 버섯의 품질에 영향을 미치는 외부요인은 온도, 상대습도, 공기조성 및 통풍 상태가 있으며 이로 인해 버섯의 호흡량, 수분 함량, 경도, 색도, pH 및 이취 등에 영향을 준다. 큰느타리버섯의 주요 장해는 갈변, 이취, 부패가 있으며, 이를 막기 위해 상품성이 높은 품종을 개발하거나 저장성을 높이는 처리 방법이 고안되고 있다. 버섯의 품질을 유지하기 위한 방법으로는 건조, 감마선 조사, 키토산 처리와 같은 수확 후 처리 방법과 CA 저장과 MA 포장을 통한 저장 방법이 있다. 버섯의 건조는 자연건조, 열풍건조, 동결건조, 진공건조, 전자파진공건조 방법이 있으며 동결건조 방법을 사용한 버섯의 성분저하가 비교적 낮았다. 1 kGy 감마선 조사가 고선량에 비해 저장기간 연장에 효과적이었으며 오일, protocatechuic acid, 왁스 등의 친수성 colloid를 키토산에 첨가하여 코팅처리 하였을 때 경도유지, 호흡 및 갈변이 억제되었다. 큰느타리버섯의 CA 저장조건은 $10^{\circ}C$ 이하의 온도에서 5kPa $O_2$ + 10~15kPa $CO_2$의 공기 조성이 제시되었으며 MA 포장으로는 미세천공을 갖는 PP필름을 이용한 능동적 기체조성 방법이 저장 중 품질유지에 효과적이었다.

세포 내 지방생성과 Glut-4 의존성 포도당 운반에 미치는 발효복합한약 물추출물(F-MAPC)의 영향 (Water Extract of Fermented New Korean Medicinal Mixture (F-MAPC) Controls Intracellula Adipogenesis and Glut-4 dependent Glucose Uptake in 3T3-L1 Adipocytes and L6 Myoblasts)

  • Objectives : The aim of this study was to investigate the effects water extract of fermented new korean medicinal mixture, combinations of Mori Folium, Adenophorae Radix, Phllostachyos Folium and Citri Pericarpium (F-MAPC), on adipocyte differentiation, adipogenesis and glucose uptake using undiffernentiated 3T3-L1 adipocytes and L6 myoblasts. Methods : Each herb and those mixture were respectively fermented and then extracted with water. We carried on MTT assay for check-up on cell toxicity, Oil Red O staining for determination of cell differentiation and intracelluar adipogenesis. Western blot analysis for measurement of pAMPK and pACC, $C/EBP{\alpha}$, $PPAR{\gamma}$ and Glut-4 protein expressions were performed. Results : F-MAPC showed significant inhibitory activity on adipocyte differentiation in 3T3-L1 preadipocytes without affecting cell toxicity as assessed by measuring fat accumulation, and this effect was 2 fold higher in 0.2 mg/ml F-MAPC than that of the same dose of each fermented herbal extract alone. In addition, these effects were associated with modulation of adipogenic transcription factors, such as $C/EBP{\alpha}$, $PPAR{\gamma}$, as well as stimulated phosphorylations of AMPK and ACC. Translocation of Glut-4 was significantly increased by 10.2% in L6 cells treated with 0.2 mg/ml F-MAPC compared with that of control. Conclusions : These results demonstrate that F-MAPC may be an ideal candidate for therapy of obesity and diabetes by disturbing the differentiation into adipocytes, as well as the inducement of intramuscular glucose uptake from blood.

Acute and Repeated 28-Day Oral Dose Toxicity Studies of Thymus vulgaris L. Essential Oil in Rats

  • Thymus vulgaris L. is widely used as an ingredient in cooking and in herbal medicine. However, there is little information about its toxicity. The present study was performed to evaluate the acute and repeated 28-day oral dose toxicity of thyme essential oil in rats. For the acute toxicity test, two groups of three rats were used. The rats received a single dose of essential oil: 300 or 2,000 mg/kg of body weight (bw). The rats were observed individually during the first four hours, and then daily until day 14. For the toxicity test with repeated doses, four groups of 10 rats were used. Doses of 100, 250, and 500 mg/kg/day were tested for 28 days. At the end of the experiment, blood was collected and the animals were sacrificed. Histopathological examination showed that in the lungs of rats given the 2,000 mg/kg bw dose, polymorph nuclear infiltrates, hemosiderin macrophages, and interstitial space thickening were present. In the repeated dose study, all rats survived the 28-day treatment period and apparently showed no signs of toxicity. The hematological and biochemical parameters were not altered. The histopathological study of the organs showed severe changes in the lung, with the dose of 500 mg/kg/day; in the other organs, no alterations were observed or the changes were slight. The body weight was only altered in male rats given the 500 mg/kg dose. The relative weight of the organs did not show any significant changes. Our studies revealed that the essential oil of Thymus vulgaris has moderate oral toxicity according to the results of the acute test, whereas the results of the 28-day oral toxicity test suggest that the no-observed-adverse effect level (NOAEL) is greater than 250 mg/kg/day.

과체중 및 비만 환자의 체중감량을 위한 식사량 조절 및 복합 한약 단기 치료(5일 레스큐 프로그램)의 효과와 안전성: 진료기반 연구 (Effect and Safety of Calorie Restriction and Complex Herbal Medicine Short-term Treatment ('Oil-rescue' Program) for Weight Reduction of Obesity or Overweight Patients: A Practice Based Research)

  • Objectives: This study is a practice based research conducted to investigate the effectiveness and safety of a 5-day short-term diet program ('Oil-rescue' program) designed to reduce the adverse events and initial dropout rate in obesity treatment. Methods: We retrospectively analyzed 4 Korean medicine clinic patient data who completed 'Oil-rescue' program which is consisting of Gambi-hwan, Bium-hwan, Butgiban-hwan and Jayoon Kyungokgo. The weight change before and after participating 'Oil-rescue' program was primary outcome of our study. Changes in body mass index, body fat mass, skeletal muscle mass, body water content, lean body mass, protein, and minerals were also measured. Results: A total of 35 patients who satisfied the eligible criteria were finally included. The body weight decreased from 69.45±11.86 kg to 67.43±11.58 kg, a total of 2.02±1.03 kg (P<0.001). Body fat mass decreased from 25.77±7.45 kg to 24.98±7.26 kg, a total of 0.78±1.21 kg (P<0.001). Body mass index decreased from 26.39±3.64 kg/㎡ to 25.64±3.49 kg/㎡, a total of 0.75±0.41 kg/㎡. (P<0.001). A total of 15 patients had side effects such as diarrhea, abdominal pain, and insomnia, but all were mild. 16 out of 35 people switched to long-term obesity treatment programs. Conclusions: Through this retrospective practice based research, it was found that the 'Oil-rescue' program effectively reduced body weight, body fat, and body mass index, and the other obesity related parameters. It was a relatively safe and effective short-term obesity treatment program.

Pharmacological findings of an Indian traditional Ayurvedic herbalformulation Amritaristo

  • In this present study Indian traditional Ayurvedic herbal formulation Amritaristo has been studied to assess the general pharmacological effect on mice. The drug showed no significant activity on the neuropharmacological test models experimented. The increased pentobarbital sleeping time was considered related with hepatic metabolism of pentobarbital. The formulation exhibited a non-significant reduction of gastrointestinal motility, and devoid of any acute diuretic activity. The tested drug revealed antidiarrhoeal activity on castor oil-induced model, whereas on magnesium sulphate-induced model no effect was observed.

변비(便秘)의 식료방법(食療方法)에 관한 연구(硏究) (A study of dietetic on the constipation)

  • Objectives : This research examined about dietary therapy on the constipation based on the literary findings, for clinical treatment and prevention of the constipation. Methods : This Research examined on histories, compositions, applications, and effects of the constipation on about 100 cases of dietary therapy of the constipation from recent Chinese literatures Results : 1. Various vegetables, animals and mineral materials including herbs, grains, vegetables, fruits, food and drink were used for the dietary therapy. 2. Methods of the preparation for use as therapeutics were decoction, pulvis, gruel, medicinal wine, cake, tea, paste and gelatin and etc. 3. Frequently used materials were sesame oil, honey, ear mushroom, shiitake mushroom, kelp, walnut seed, hemp seed, groundnut, glutinous rice, apricot stone, yellow bean peel, radish, potato, spinage, and spring onion. Conclusion : Though dietary therapy for the constipation is not based on clinical or experimental data, but through experience. It is mostly based on Yin-Yang and five elements, visceral manifestation, channels and their collateral channels and chinese herbal medicine theories. If we use them properly according to oriental medicine method, it will be effective on treating and preventing the constipation.

  • Introduction: Liver fibrosis is defined unbalance of collagen metabolism, especially a stimulation of collagen synthesis and inhibition of collagen degradation, and antifibrotic effect is delaying or inhibition of new collagen synthesis and deposition in liver tissue. In this study, we investigated the antifibrotic and antioxidative effect of Solanum lycopersicum (SL) in liver fibrosis induced rats.   • As the part of our continuing study for antihyperlipidemic agents from the herbal medicinal resources. we examined the possibility of the ethylacetate fraction of Alpinia officinarum water extract in vitro and in vivo. We isolated some compounds from the ethylacetate fraction of Alpinia officinarum and measured their antihyperlipidemic activities. The active components isolated by silicagel column lmproved serum TG. HDL and LDL level in corn oil feeding and triton WR-1339 induced hyperlipidemic mice. The most active compound was 3-Methylethergalangin. Consequently. these biologically active gerbal functional foods could be used for preparing the healthy food which might diabetas. hyperlipidemia and other disease.
