Peony Stem Rots by Neopestalotiopsis clavispora and Sclerotinia sclerotiorum, and Antifungal Propineb and Bacillus siamensis H30-3 against the Two Fungal Species

  • In July 2022, stem rot symptom was found in a peony plant grown in a pot under a greenhouse at Jinju, Gyeongnam Province, South Korea. Two fungal species were isolated from the infected peony stems and cultured on 1/2-strength potato dextrose agar for identification. The morphological characteristics of the fungal isolates were examined, and nucleotide sequences of the internal transcribed spacer region, β-tubulin and translation elongation factor 1-α were analysed. The pathogenicity of the two isolates was confirmed in detached peony leaves, according to Koch's postulates. To our knowledge, this is the report of Neopestalotiopsis clavispora and Sclerotinia sclerotiorum as the causal agents of peony stem rots. Antifungal activity of chemical fungicide propineb and rhizobacterium Bacillus siamensis H30-3 was shown against the two plant pathogenic fungi N. clavispora and S. sclerotiorum.Unidentified diffusible and volatile compounds from B. siamensis H30-3 could suppress in vitro mycelial growths of N. clavispora JJ 8-2-1 and S. sclerotiorum JJ 8-2-2.

Effects of Intercrops on Growth and Yield of Paeonia Lactiflora PALLAS. (간작물(間作物)이 작약(芍藥)의 생육(生育) 및 수양(收量)에 미치는 영향(影響))

  • This study was carried out to select proper intercrops in peony cultivation because peo­ny is damaged easily by disease and cannot make proper income with it's single crop system, so it needs rational intercropping system to raise it's productivity and income. The results of this study are as follows. The kinds of soil nematodes were Meloidogyne sp, Pratyienchus sp, Aphelenchoides sp, Ditylenchus sp, Xiphinema sp, and damaging dominant nematode was Meloidogyne sp. The density of Meloidogyne sp wa suppressed remarkably in the combination of peony+sesame, and peony + job's tears. This study shows that sesame and job's tear were most resistant crops to the IvIeloi­dogyne sp. The quanity of herbaceous peony was the best in the combination of peony+red pepper. The results of income analysis per lOa for three years shows next conclusion. Peony+red pepper combination and peony+ sesame combination were most effective when it's compared with single cropping of peony (1,490 thousand won). But red pepper was damaged easily by soil nematode, therefore, sesame was the most stable and high yield crop as a intercrop in the cultivation of herbaceous peony.

Effect of Seed Size and Mucilaginous Substance on Seed Germination of Peony(Paeonia lactiflora Pall.) (작약(芍藥) 종자(種子)의 형태적(形態的) 특성(特性)과 점액물(粘液物)이 발아(發芽)에 미치는 영향(影響))

  • The experiments were conducted to determine the influence of seed size and mucilaginous substance on seed germination of herbaceous peony collected from farmers' fields in Korea. No significant difference was recognized between the size of seed and the rale of germination. The maximum percentage(68%) of seed germination was obtained from the seeds with thin brown colour treated at $20^{\circ}C$. The amount of mucilaginous substances exuded from peony seeds was the highest in the seeds with black brown colour. Germination rate of peony seeds was negatively correlated with the amount of mucilaginous substance exuded from the seeds. High frequency(93%) of seed germination was obtained from the seeds which treated in the mixture(1:1) of sands and activated charcoal.

Effect of Embryo Morphology on Plant Development in Anther Cultures of Paeonia lactiflora Pall. (작약(Paeonia Lactiflora Pall.)의 약배양에서 형성된 배의 형태가 유식물 발달에 미치는 영향)

  • The embryos formed from anther culture of peony exhibited divergent morphologies. The frequency of normal embryos with two cotyledons was about two times higher in the embryos formed through direct embryogenesis than those formed from callus. About 69% of the embryos with two cotyledons converted into normal plants, but convention rate of the abnormal embryos with one and three or four cotyledons was only 4 to 9%. The embryos of hem and bowling pin shape did not undergo development into normal plants.

  • The experiments were carried out to determine the optimum conditions for the direct embrogenesis from the cotyledon derived zygotic embryo culture of Paeonia lactiflora Pall. Different peony tissues derived from zygotic embryos were cultured on MS medium with and without 2,4-D. Somatic embryos were formed from the cotyledons cultured on the medium without 2,4-D. The somatic embryogenesis from cotyledons was promoted in the growth regulator-free MS medium containing 1.65~3.3 g/L $NH_4NO_3$ and 30~40 g/L sucrose. The maximum frequency (80.0%) of somatic embryo formation was obtained from the cotyledons excised from zygotic embryos that cultured on MS medium containing 3.3 g/L $NH_4NO_3$. Epicotyl and roots were elongated from a somatic embryo by adding 0.3 mg/L GA$_3$ in the medium or the cold treatment at 4$^{\circ}C$ more than three weeks at 4$^{\circ}C$.

Anti-inflammatory and Anti-Oxidant Effects of Oxypaeoniflorin, Paeoniflorin and Paeonia lactiflora cv. 'Red Charm' Flower Petal Extracts in Macrophage Cells

  • The root extracts of Paeonia lactiflora cv. 'Red Charm' has been studied by many groups, however, little attention has been paid to its flower petal. Paeonia is the genus in the Paeoniaceae family. 'Red Charm' Paeonia is a soft-stemmed herbaceous peony hybrid of P. officinalis and P. lactiflora. We previously showed the flower petal extract of Red Charm might have anti-oxidant and anti-inflammatory activities, however, it was not clear which components might be involved in this activity. Bioinformatics analysis previously indicated these extracts have potential anti-oxidant materials. One of them is identified as paeoniflorin, which is major component in root extract of Red Charm. In this study, we compared paeoniflorin and oxypaeoniflorin using DPPH assays to measure its anti-oxidant activities. Oxypaeoniflorin showed higher levels of radical scavenging activity, similar to ascorbic acid control, whereas paeoniflorin did not. Furthermore, nitric oxide assay showed they have similar anti-inflammatory effects. Taken together, these results suggest oxypaeoniflorin may play a more important role in the anti-oxidant activity of the flower petal and root extracts of Red Charm, compared to paeoniflorin. Further studies may be able to provide a platform to develop potential dual effects therapeutics for oxidant-mediated and inflammation-mediated disease in the near future.

Effect of Plant Growth Regulators on Somatic Embryogenesis from Cotyledon of Herbaceous Peony (Paeonia lactiflora Pall.) (芍藥(Paeonia lactiflora Pall.)의 子葉組織 培養시 식물생장조절제가 體細胞胚發생에 미치는 影響)

  • This experiments were carried out to determine the optimum concentrations of plant growth regulators for the direct embryogenesis from the cotyledon culture of Paeonia lactiflora Pall. Zygotic embryos rescued from true seeds were developed abnormally in the medium containing ABA. But somatic embryogenesis from cotyledons of abnormal seedlings was induced more effectively. Also the somatic embryogenesis from cotyledons was promoted in the medium containing ABA. The frequency of embryogenesis was maximum(59.9%) from the cotyledons cultured on MS medium containing 0.5 mg/L ABA on which the frequency of somatic embryos with two cotyledons was 22.6%.
