Hibiscus hamabo, called "Hwang-geun", growing about 3m in height is a deciduous shrub or subtree of Malvaceae. Because the number of these species is very limited in the world, the Ministry of Environment has designated H. hamabo as a preserved plant. The Korea Forest Service also protects it strictly by law since H. hamabo is an out-of-the-way plant and possibly may be exterminated soon in Korea. Investigation for distribution and ecological characteristics of the habitat for H. hamabo was carried out on Soan Island. Two wild H. hamabo were found at the forest edge (equation omitted) along the sea coast located in the southern part of Soan Island and this was the first report in the Korean academic world. These two wild H. hamabos were growing in a naturally inhibited area. The diameters at the base were 12cm and 15cm. The Tree heights were 150cm and 210cm and the number of branches of each wild H. hamabo was 4 and 7. However, the present condition of these plants was not good. Environmental conditions of the naturally inhibited area of H. hamabo were very mild because it is located at the edge of the forest and is always sunny during the daytime since the slope of the inhibited area is facing South. The ground drained very well since the soil was made of gravels and sand. Because the percent of vegetation of the subtree layer where H. hamabo was growing was 40%, the cover degree and sociability of flex crenata trees and Eurya japonica were found to be high. In the naturally inhibited area of H. hamabo, a dominant value of Rubus parvifolius in the lower part of the herb layer was very high and many plants in Compositae, such as Artemisia princeps var. orientalis and Erigeron annuus, were also present. A dominant value of liana, such as Vitis thunbergii var. sinuata, Rosa multiflora, Clematis terniflora and Hedera rhombea, and Gramineae plants that rhizomes were well developed and aggressively propagated, such as Miscanthus sinensis var. purpurascens, Phragmites communis, Spodiopogon cotulifer and Oplismenus undulatifolius which were surveyed as high, too. These results imply that H. hamabo might be exterminated soon through a natural selection if the proper management of the naturally inhibited area of H. hamabo is not conducted continually.
Lilium cernum Komarvo. is an important endangered plant belonging to the family Liliaceae. A method was developed for the rapid micropropagation of L. cernum through plant regeneration from bulb scales explant-derived calli. The bulb scales segments were cultured on Murashige and Skoog (MS) medium supplemented with 0, 0.5, 1.0, $3.0mg{\cdot}L^{-1}$ kinetin and 0, 0.1, 0.5, $1.0mg{\cdot}L^{-1}$ NAA or 2,4-D for callus induction. In media with $0.5{\sim}3.0mg{\cdot}L^{-1}$ kinetin and $0.1{\sim}1.0mg{\cdot}L^{-1}$ NAA and 2,4-D, 95~100% of explants produced callus. They were then transferred to MS medium supplemented with various concentrations of NAA (0, 0.01, 0.05 and $1.0mg{\cdot}L^{-1}$) in combination with BA (0, 1.0 and $2.0mg{\cdot}L^{-1}$) for bulbet formation. Bulbet induction (78%), weight (468 mg) and size (15.5 mm) were obtained the highest on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA. In vitro frequency of plant regeneration was not significantly treated in strength of MS and sucrose concentration. Chlorophyll contents in 1/2MS with $50g{\cdot}L^{-1}$ sucrose treatments were higher than those in control and another treatment. This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.
Cultivated wild ginseng is a widely used dietary supplement and medicinal herb. The aim of this study was to optimize the ginseng using high performance liquid chromatography (HPLC)- charged aerosol detection (CAD) for ginsenoside analysis. CAD measures the physical property of an analyte and responds to almost all non-volatile species, independent of their nature, spectral properties, or particle size. It has become widely employed in pharmaceutical analysis. The cultivated wild ginseng extracts were analyzed for compositions of ginsenosides Rb1, Rd, Rg1, Rf, Re, and Rh1. The optimal analysis condition was set up from an experiment using a gradient. Ten grams of cultivated wild ginseng were extracted with 95% EtOH 100 ml for 24 hr at 80℃. The contents of the 6six major ginsenosides in the cultivated wild ginseng extract were Rb1 (5.48±0.12 mg/g), Rd (5.33±0.14 mg/g), Rg1 (12.80± 0.05 mg/g), Rf (19.08±0.68 mg/g), Re (19.87±0.05 mg/g), and Rh1 (16.47±0.16 mg/g), respectively. HPLC showed that the protopanaxatriol group (Rg1, Rf, Re, Rh1) had more content than the protopanaxadiol group (Rb1, Rd) in cultivated wild ginseng extract. In summary, the ginsenosides were identified with HPLC-CAD analysis, and their presence and quantity imply the importance of quality control, as well as the pharmacological activity of the ginseng root.
This research was conducted to investigate morphological and genetic variation of Allium victorialis val. platyphyllum which growed wild in Mt. Hambaek, Mt. Odae, and Ullungdo. The tree layer of Allium victorialis var. platyphyllum community in Mt. Hambaek and Mt. Odae was dominated by Quercus mongolica. The tree layer of Ullungdo generally consist of Fagus crenata var. multinervis, Acer triflorum, Sorbus commixta, and Tilia insularis. In the herb layer, Rumohra standishii, Trillium tschonoskii, and Lilium hansonii are common at Allium victorialis var. platyphyllum community in Ullungdo. The vegetation in Ullungdo was widely different from those in Mt. Hambaek and Mt. Odae by species composition. The result of Principal Component Analysis(PCA) and Canonical Discriminent Analysis of by the 8 characters showed that Allium victorialis var. platyphyllum could be classified into 2 groups: (one ; Mt. Hambaek and Mt. Odae, the other ; Ullungdo). In PCA, the major factors in the first principal component group was angle of leaf apex. Variation of band by isozyme GOT(glautamate oxaloaccetate transaminase) is similar between Mt. Hambaek and Mt. Odae. However, Ullungdo differed from Mt. Hambaek and Mt. Odae in variation of bands.
'Hwaweon 6' was developed from a cross between 'Hwaseongbyeo' and a wild species, Oryza minuta L. (Acc. No. 101154) based on marker-assisted selection and backcrossing. The recurrent parent 'Hwaseongbyeo' is a high grain quality cultivar with medium maturity. Hwaweon 6 is nearly isogenic to Hwaseongbyeo except a small O. minuta introgressed segment on chromosome 7 harboring genes related with spikelets per panicle. The preliminary and replicated yield trial was conducted at Chungnam National University and Chungcheongnamdo Agricultural Research & Extension (CARES), Yesan in 2009 and 2010. The local adaptability test was carried out by the National Seed Management Office (NSMO) in 2011 and 2012. This cultivar was registered to NSMO with a cultivar designated as Hwaweon 6. This cultivar averaged 80cm in culm length and has a medium growth duration. Milled rice of Hwaweon 6 is translucent and the grain quality traits are comparable to those of the recurrent parent. The average yield potential of Hwaweon 6 in grain was about 6.57 MT/ha at the ordinary fertilizer level for two years about 3.0% higher than that of Hwaseongbyeo due to an introgression of the quantitative trait locus (QTL) for spikelets per panicle (qSPP7) from O. minuta. This variety is comparable to Hwaseongbyeo in low temperature germinability and cold tolerance. The qSPP7 QTL would be useful in enhancing yield potential in rice breeding program.
Doellingeria scabra Thunb. (syn. Aster scaber Thunb.), a perennial herb in the family Asteraceae, is frequently found in the wild mountain regions of Korea. This aim of this work was to measure the characteristics of patchiness of D. scabra in a local population on Mt. Maebong in Taeback-ci, Gangwon-do. The spatial distribution pattern of this species was estimated by analyzing ecological data by methods including the index of dispersion, Lloyd's mean crowding, and Morisita's index. The mean population density of the D. scabra population was 2.94. The D. scabra individuals were uniformly or randomly distributed in small-scale plots and were aggregately distributed in two large-scale plots ($16{\times}32m^2$ and $32{\times}32m^2$). The mean crowding ($M^*$) was 0.916. The mean patchiness index (PAI) was 0.796. Morisita's coefficient tended to decrease the density of the population as the plot size increased. The expected value of Eberhardt's index ($I_E$) in the local population was 2.623. Moran's I of D. scabra significantly differed from the expected value in 6 of 8 cases (75.0%). The first five classes were positive, with four showing statistical significance, indicating similarity among individuals in the first four distance classes (I-IV, 8 m), The results presented here could provide a theoretical basis for the conservation of D. scabra (Korean: chamchwi) and for the rehabilitation and sustainable management of forest ecosystems on Mt. Maebong, as well as on other mountains.
Objectives: The seeds from Arctium lappa have been considered for its various pharmacological properties, which include anti-carcinogenic, anti-inflammatory, anti-diabetic, and anti-viral activities. Methods: In the present study, we investigated the anti-inflammatory effect of the ethanol extract from the seeds of Arctium lappa L (EAL) on cytokine-induced vascular inflammation in human umbilical vein endothelial cells (HUVEC). Results: Pretreatment with EAL significantly decreased tumor necrosis factor alpha ($TNF-{\alpha}$)-induced cell adhesion molecules expression such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and endothelial-selectin (E-selectin) in a dose-dependent manner. Cell adhesion assay showed that pretreatment with EAL suppressed HUVEC-monocyte adhesion by $TNF-{\alpha}$ over $1{\mu}g/ml$ concentration. We investigated the involvement of nuclear transcription factor kappa-B ($NF-{\kappa}B$) in $TNF-{\alpha}$-induced vascular inflammation. $NF-{\kappa}B$ p65 nuclear expression was induced by $TNF-{\alpha}$, however, pretreatment with EAL was attenuated that nuclear translocation. In cytoplasm, EAL was also attenuated $TNF-{\alpha}$-induced decrease of inhibitor of ${\kappa}B-{\alpha}$ ($I{\kappa}B-{\alpha}$) expression. Moreover, EAL significantly decreased $TNF-{\alpha}$-induced production of intracellular reactive oxygen species (ROS). Conclusions: Taken together, our findings suggest that seeds of Arctium lappa L could be a therapeutic herb for prevention of cardiovascular diseases throughout the inhibition of vascular endothelial inflammation.
Aster yomena (Kitam.) Honda is an edible vegetable and perennial herb belonging to the Asteraceae family, and has been used for a long time for the prevention and treatment of various diseases. Although leaf extracts of A. yomena are known to have antioxidant and anti-inflammatory effects, accurate efficacy assessments are still inadequate. In this study, we investigated whether the antioxidant efficacy of ethanol extract of A. yomena leaf (EEAY) is correlated with the anti-inflammatory effect in RAW 264.7 macrophages. The results showed that EEAY significantly inhibited the hydrogen peroxide ($H_2O_2$)-induced growth inhibition in RAW 264.7 cells, which was associated with increased expression of nuclear factor erythroid 2-related factor-2 (Nrf2) and heme oxygenase-1 (HO-1). EEAY pretreatment also effectively prevented $H_2O_2$-induced reactive oxygen species generation and apoptosis through inhibition of caspase-3 activation and poly (ADP-ribose) polymerase degradation. Additionally, EEAY significantly increased the expression and production of interleukin-10, a representative anti-inflammatory cytokine, which was associated with increased expression of toll-like receptor 4 and myeloid differentiation factor 88 at transcriptional and translational levels. Furthermore, the increased production of nitric oxide (NO) by lipopolysaccharide was markedly abolished under the condition of EEAY pretreatment, and the inhibitory effect of NO production by EEAY was further increased by hemin, an HO-1 inducer. Overall, our results suggest that EEAY is able to activate the Nrf2/HO-1 signaling pathway to protect RAW 264.7 macrophages from oxidative and inflammatory stress.

This study investigated the effect of traditional medicinal plants on the quality characteristics of Samgyetang breast meat and broth. The traditional medicinal plants used in this study were roots of Panax ginseng, Codonopsis lanceolata, Platycodon grandiflorum, Salvia miltiorrhiza, Adenophora triphylla, and Allium hookeri. There was no significant difference (P>0.05) in the moisture, protein, ash content, water holding capacity, and cooking loss of Samgyetang meat with the addition of traditional medicinal plants. The color values (lightness, redness, and yellowness) of Samgyetang meat and broth were significantly different (P<0.05); the 2-thiobarbituric acid reactive substances and shear force of Samgyetang meat were significantly different (P<0.05); and the pH, turbidity, and viscosity of Samgyetang broth were also significantly different (P<0.05) with the addition of traditional medicinal plants. In the overall acceptability of Samgyetang meat, the addition of S. miltiorrhiza showed the highest score compared to that of other treatments (P<0.05). Thus, the addition of traditional medicinal plants in the production of Samgyetang were found to affect the lipid rancidity beneficially with plant species when compared to Samgyetang with Panax ginseng. In conclusion, with the development of Samgyetang using S. miltiorrhiza, it will be possible to develop products with superior quality characteristics in antioxidant, shear force, and overall acceptability.

Background: This research aims to study the effect of climate change on the phenology, growth, and physiological traits of Silene capitata Kom., a Korean endangered species II. This study increased $CO_2$ concentration in a closed glass greenhouse, with the daily mean temperature and $CO_2$ concentration respectively being $4.61^{\circ}C$ and 93.63 ppm higher than the outside temperature (ambient conditions, control). The seeds of S. capitata were sown in control and treatment environments in March 2013 while seedlings were transplanted into individual pots in May 2013. To research phenological changes, the first day of the flowering and ripening of the plants transplanted in 2013 and first day of leafing in 2014 were observed. The growth and physiological responses of mature leaves were also studied in 2013. Results: There was no difference in the first day of flowering, but the first day of ripening was earlier in the treatment group than the control group. There was no difference in the number of rosette leaves between the two groups, but leaf area was wider in the treatment group than the control group. Transpiration rate and stomatal conductance were higher in the treatment group than the control group, chlorophyll content decreased, and photosynthetic rate and water use efficiency were the same for both groups. As a result of simple regression analysis among the transpiration rate, stomatal conductance, photosynthetic rate, and water use efficiency, stomatal conductance increased when transpiration rate increased. Stomatal conductance increased with photosynthetic rate in the control unlike in the treatment group. The photosynthetic rate and water use efficiency increased with transpiration rate in the control group unlike in the treatment group. Furthermore, water use efficiency increased as photosynthetic rate increased in both groups. Conclusion: Due to high $CO_2$ concentration, the photosynthetic rate was no longer controlled by the stomata, which appeared to suppress the excessive production of photosynthetic products by reducing chlorophyll content. It is believed that the phenological responses of S. capitata under climate change conditions will advance and that stable growth will be difficult in regions lacking moisture due to the high transpiration rate.
