G009, a polysaccharide isolated from the mycelia of Ganoderma lucidum IYO09, showed a hepatoprotective activity against $CCl_4$ induced cytotoxicity in primary cu ltured rat hepatocytes. Incubation of $CCl_4$-intoxicated hepatocytes with G009 significantly reduced the levels of glutamic pyruvic transaminase and sorbitol dehydrogenase released from hepatocytes in the medium. G009 showed antioxidative effect by elevating the activities of glutathione reductase and superoxide dismutase, and the content of glutathione in $CCl_4$-intoxidcated primary cultured rat hepatocytes. Furthermore, G009 significantly elevated glutathione-S-transferase activity in $CCl_4$-intoxicated primary cultured rat hepatocytes. G009 also reduced the production of malondialdehyde, a byproduct of lipid peroxidation. From these results, it could be concluded that G009 exerted hepatoprotective activity against $CCl_4$-induced cytotoxicity through antioxidation.
Objectives : The aim of this study was to investigate the hepatoprotective effect of Soshiho-tang (SSH) in mouse primary liver cells against hydrogen peroxide ($H_2O_2$)-induced oxidative stress. We also elucidated the molecular mechanism of hepatoprotective effect by SSH. Methods : Cell viability, level of ALT, AST and LDH, intracellular ROS level, mRNA expression and activity of antioxidant enzymes were used to evaluate hepatoprotection of SSH against $H_2O_2$. Target gene expressions were analyzed by real-time PCR. Results : Pre-treatment with SSH for 1 hour prevented cytotoxicity against $H_2O_2$. $H_2O_2$-induced ROS level decreased under SSH pre-treatment. mRNA expression of GPx and SOD increased in SSH-treated cells. In addition, HSP72 and HSP40 gene expression were elevated under SSH-treatment. Conclusions : These results indicate that SSH protects mouse primary liver cells from $H_2O_2$-induced oxidative injury. This hepatoprotective activity of SSH is mediated by decreasing intracellular ROS and increasing antioxidant enzyme expression (GPx and SOD) and stress response protein (HSP72 and HSP40).
국내에서 자생하고 있는 75균주의 고등균류를 액체배양하여 얻은 다당류를 $CCl_4$에 의해 독성이 유발된 초대배양 간세포에 가하여 간세포 독성에대한 다당류의 보호효과를 glutamic pyruvate transaminase(GPT)활성을 측정하여 검색한 결과는 다음과 같다. 75균주의 고등균류 중 간세포 보호효과를 나타낸 것은 60균주로 Ganoderma lucidum 15균주 중 13균주, Lentinus edodes 7균주 중 5균주, Pleurotus ostereatus 1균주, Coriolus versicolor 5군주 중 4균주, Lyophyllum spp. 2균주, Grifori frondosa 7균주, Agaicus spp. 3균주, Schizophyllum commune 16균주 중 14균주, Cordyceps spp. 18균주 중 11균주등이었다. 상기 고등균류 중 GPT활성이 80%이하로 나타나, 비교적 간세포 보호효과가 우수한 것은 Ganoderma lucidum IY003 및 IY009, Lentinus edodes IY103, Lyophyllum sp. IY402, Agaicus sp. IY701 및 IY703, Schizophyllum commune IY804, IY810 및 IY818, Cordyceps sp. IY902등 10균주였으며, 이들의 GPT활성은 각각 80.0%, 75.5%, 78.2%, 75.2% 73.6%, 75.0%, 75.1%, 77.7%, 77.5% 및 78.4%로 나타났다.

Phellinus gilvus (PG) is a widely used mushroom for health promotion. We studied the hepatoprotective effect of the polysaccharide aqueous extract of PG (PGP) against carbon tetrachloride ($CCl_4$)-induced liver injury in rats. Sprague Dawley rats were divided into 5 groups: Normal control, $CCl_4$ control, PGP 50, 100, and 200 mg/kg + $CCl_4$. The levels of serum biochemical parameters, liver lipid peroxide and antioxidant enzymes, and histological appearances were evaluated. The $CCl_4$-induced increments of alanine aminotransferase, asparate aminotransferase, and alkaline phosphatase levels in serum were significantly decreased by PGP-pretreatments. The PGP dose-dependently decreased hepatic malondialdehyde formations in $CCl_4$-treatment groups. Hepatic antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase) were elevated by PGP in $CCl_4$-treatment groups. Histopathological evaluation of liver showed that the loss of hepatocytes, fatty changes, swelling and extensive necrosis of hepatocytes in centrilobular regions of the $CCl_4$-treated rats were ameliorated by PGP pretreatment. The PGP has hepatoprotective and antioxidative effects in $CCl_4$-induced liver injury of rat.
Lactiplantibacillus plantarum DSR330 (DSR330) has been examined for its antimicrobials production and probiotics. In this study, the hepatoprotective effects of DSR330 were examined against nonalcoholic fatty liver disease (NAFLD) in a high-fat diet (HFD)-fed C57BL/6 mouse model. To induce the development of fatty liver, a HFD was administered for five weeks, and then silymarin (positive control) or DSR330 (108 or 109 CFU/day) was administered along with the HFD for seven weeks. DSR330 significantly decreased body weight and altered serum and hepatic lipid profiles, including a reduction in triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels compared to those in the HFD group. DSR330 significantly alleviated HFD-related hepatic injury by inducing morphological changes and reducing the levels of biomarkers, including AST, ALT, and ALP. Additionally, DSR330 alleviated the expression of SREBP-1c, ACC1, FAS, ACO, PPARα, and CPT-1 in liver cells. Insulin and leptin levels were decreased by DSR330 compared to those observed in the HFD group. However, adiponectin levels were increased, similar to those observed in the ND group. These results demonstrate that L. plantarum DSR330 inhibited HFD-induced hepatic steatosis in mice with NAFLD by modulating various signaling pathways. Hence, the use of probiotics can lead to hepatoprotective effects.

Herbal medicinal mushroom Cordyceps militaris has been traditionally used as tonic medicine for metabolic syndrome. Cordycepin, main extract of C. militaris, has been reported with immunomodulatory, anticancer, and hepatoprotective effects. This study was conducted to evaluate the potential hepatoprotective effect of cordycepin-enriched Cordyceps militaris, against high fat diet (HFD)-induced hepatic steatosis (HS) in male obese (ob/ob) mice. HFD was provided to ob/ob mice ad libitum (except negative control). Cordycepin-enriched C. militaris extract powder (CM) was orally administered once daily at dose levels of 0, 125, 250, and 500 mg·kg-1 for 4 weeks. During the study, body weight gain was statistically increased in all HFD fed groups compared to negative control, but body weight gain in CM 500 mg·kg-1 treated group shows a low tendency compared to HS model group. In organ weights, absolute and relative weights (to body weight) in liver and perirenal adipose tissue were increased in all HFD treated groups except CM 500 mg·kg-1 treated group compared to the negative control. In clinical chemistry, serum glucose and total cholesterol levels in CM 250 and/or 500 mg·kg-1 treated groups were lower than HS model group. In microscopical examination, hepatocyte vacuolation with macrovesicles in HS model group was increased compared to negative control, but this finding was decreased in CM 500 mg·kg-1 treated group compared to HS model group. In this study, CM exhibited hepatoprotective effects against hepatic steatosis at mg·kg-1 in ob/ob mice.
Inhibitory effect of water and ethyl acetate extract of 60 kinds of herbal medicines was investigated on ${\beta}-glucuronidase$. Among water extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Termaliae Fructus, Amomi Tsa-ko Fructus and Arecae Semen were also potent inhibitors. Among ethyl acetate extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Nelumbinis Semen. Ephedrae Radix and Termaliae Fructus were also potent inhibitors. The extract of Galla Rhois had potent hepatoprotective effect on $CCl_4-induced$ hepatotoxicity of rats. These results suggest that the ${\beta}-glucuronidase$ seems to be closely related to the liver injury, which could be prevented by the inhibitor of ${\beta}-glucuronidase$.
This study was done to investigate the protective effect of Alnus japonica Steude on hepatotoxicity in carbon let-rachloride $(CCl_4)$ intoxicated rats. Alnus japonica Steud was extracted with methanol and fractionated with hexane, chloroform, ethylacetate, butanol and water. Rats were treated with those orally once a day for 6 days. The activities of aminotransferase and ${\gamma}-glutamyltranspeptidase$ and contents of cholesterol, TG and hepatic lipid peroxide in butanol fraction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats but the content of glutathione was significantly increased compared to the only $CCl_4$ treated rats. Also activities of hepatic superoside dismutase, catalase, glutathione perpxidase in butanol fraction pretreated rats were signigicantly decreased compared to the only $CCl_4$ treated rats. These result indicated that butanol fraction of Alnus japonica Steude showed hepatoprotective effect in carbon tetrachloride intoxicated rats.
Objectives : Polygoni Multiflori Ramulus has been widely used as a traditional medicinal herb for the treatment of insomnia, limb pain and itch. The extract of Polygoni Multiflori Ramulus (PMRE) is known to have a modulatory effect of many inflammatory responses. This study was performed to investigate the hepatoprotective effect of PMRE against arachidonic acid (AA) + iron-induced oxidative stress on HepG2 cell and carbon tetrachloride ($CCl_4$)-induced liver injury on mice. Methods : The effects of PMRE on cell viability was assessed by MTT assay. And flow cytometric analysis was performed to estimate the effects on mitochondrial function. To investigate its underlying mechanism, apoptosis-related proteins were analysed by using immunoblot analysis. In addition, ICR mouse were administrated (po) with the PMRE (30, 100 mg/kg) for 3 days and then, injected (ip) with $CCl_4$ (0.5 ml/kg body weight) to induce acute liver damage. The level of pro-caspase-3 was measured. Results : Treatment of PMRE increased relative cell viability, prevented a cleavage of poly (ADP ribose) polymerase and pro-caspase-3, and also reduced mitochondrial membrane permeability against AA + iron-induced oxidative stress. In addition, PMRE treatment decreased liver injury induced by $CCl_4$, as evidenced by increases in pro-caspase-3 level. Conclusions : These results demonstrate that PMRE has an ability to anti-oxidant and hepatoprotective effect against AA + iron-induced oxidative stress and $CCl_4$-induced liver injury.
다슬기와 마늘 혼합건조분말(3 g/kg b.w. in saline, 7 : 3 ratio, p.o.)의 전처리가 $CCl_4$(1 g/hg b.w in corn oil)의 경구 투여로 유도된 간손상 실험동물모델에 미치는 간보호 효능을 확인하기 위하여 혈청 중 간기능 지표효소 활성도 및 광학현미경적 관찰을 실시하여 다음과 같은 결과를 얻었다. 간기능 지표효소인 혈청 ALT, AST 및 SDH 활성도는 다슬기와 마늘의 전처리로 $CCl_4$단독 투여군에 비해 통계적으로 유의성 있게 감소하였으나 혈청 ALP 활성도는 $CCl_4$ 투여로 증가된 활성을 다슬기와 마늘의 전처리로 감소시키지 못했다. 광학현미경적 관찰에서 $CCl_4$단독 투여군의 경우 인접한 괴사부위와 bridge를 형성한 central necrosis 영역에서 염증 세포의 침윤과 울혈현상이 나타났으나, 다슬기와 마늘의 전처리군에서는 중심영역에서 소핵을 갖는 부푼 형태의 초기 가역적 손상을 입은 간세포가 관찰되었을 뿐 $CCl_4$ 단독 투여군과 같은 염증 세포의 침윤이나 괴사는 관찰되지 않았다. 이상의 실험 결과를 종합해 볼 때, 다슬기와 마늘의 혼합 건조 분말이 간세포의 기능을 유지시킴으로써 사염화탄소 투여에 의해 유발된 간손상을 억제하여 간조직을 보호할 수 있을 것으로 사료된다.
