The Positive Rates of Anti-HBs and Titers of Antibody after Hepatitis B Vaccination (B형 간염백신 접종 후 항체형성률과 역가)

  • The purpose of this study was to survey the anti-HBs positivity and titers of antibody in current students who received nationwide vaccination against Hepatitis B virus which targeted infants in 1995. The subjects were 262 students in Gyeongnam province from April 2014 to October 2014. The positive rate of anti-HBs was 55.3% (145 people) and the negative rate of anti-HBs was 44.7% (117 people). Positivity was shown to be higher in women than men. However, there was no statistically significant difference. Of the HBV-vaccinated subjects, 117 (44.7%) had anti-HBs titer <10, which is judged to be negative, 126 (47.8%) had anti-HBs titer 10-499.9 mIU/mL, which is judged to be positive, and 22 (7.3%) had anti-HBs titer more than 500 mIU/mL. The rate of anti-HBs with lower titer (10-99.9 mIU/mL) was 62% in the positive group. Classifying the antibody titer according to age, the rate of anti-HBs positivity in titer with less than 100 mIU/mL was indicated to be 78.3% in cases of 19-20 year old and 46.7% in 21-22 year old, 52.3% in 23-24 year old. A case of the lower titer with 10-99.9 mIU/mL, showed significant difference according to age. As a result of research, the antibody titers is decreased depending on the passage of time. Hence, the checking of anti-HBs titer is needed after Hepatitis B vaccination and many healthy adults will need periodic boosters of hepatitis B vaccine to maintain production of antibody to hepatitis B surface antigen.

A Study on Serum Zinc Concentration in some Methamphetamine Abusers (Methamphetamine 남용자에 있어서 혈청 아연농도에 관한 연구)

  • The author investigated the zinc concentrations in sera of 110 methamphetaimine abusers (male 87, female 23) and of 41 healthy official workers(male 29, female 12) using inductively coupled plasma emission spectrometer. The serum zinc concentrations were analysed according to hepatitis C virus antibody(Anti HCV), hepatitis B surface antigen(HBsAg) and alanine aminotransferase(ALT) value in both groups. The analytical results were obtained as follows 1. The mean zinc concentration of MAP abuser group($298.5{\pm]246.9{\mu}g/100ml$) was 2.5 times higher than that of control group($118.8{\pm}25.4{\mu}g/100ml$). 2. The distributions of zinc concentration in both groups were skewed pattern. 3. The zinc concentrations were higher in Anti HCV-positive group and abnormal ALT value group than Anti HCV-negative group and normal ALT value group. But HBs-Ag positive group had lower zinc concentration than HBs-Ag negative group. 4. The number of anti HCV-positive subjects was increased and the zinc concentrations were decreasd in proportion to the abusing duration of MAP.

Seroprevalence of antibody to the hepatitis C virus in methamphetamine abusers (Methamphetamin 남용입원환자들의 C형간염 항체 양성률에 관한 조사)

  • This study was designed to estimate the prevalence of hepatitis C virus(HCV) infection in drug abusers. The subjects were 141 inpatients who had been admitted to a general hospital with the symptoms and signs of methamphetamine intoxication. Seroprevalence of antibody to the hepatitis C virus(anti-HCV) was 60.3%(85/141) and it was higher in the group with increased frequency and duration of drug abuse, but such a relationship was not found in the seroprevalence of hepatitis B surface antigen(HBsAg). These findings suggested the possibility of high prevalence of HCV infection in methamphetamine abusers, and the importance of repetitive percutaneous injection in the transmission of HCV infection.

A study of the current(2003-2005) prevalence of anti-HBs and immunologic memory of hepatitis B vaccine in children from the central area of Korea (최근(2003-2005) 우리나라 중부지역 소아에서 B형 간염 항체 보유율과 백신의 면역학적 기억에 대한 연구)

  • Purpose : This study was conducted to assess the current(2003-2005) prevalence of anti-HBs and immunologic memory for Hepatitis B vaccine in children from the central area of Korea. Methods : Subjects were chosen from children and adolescents who received tests for hepatitis B surface antigen(HBsAg) and anti-HBs at Dankook University Hospital from March 2003 to May 2005. Among these, antibodies to hepatitis B core antigen(IgG anti-HBc) were checked. A single booster vaccination was performed on children whose anti-HBs titers were under 10 mIU/mL. One month after booster vaccination we rechecked the anti-HBs titer. Results : A total of 3,277 subjects were tested for HBsAg/anti-HBs, and 1,913(58.4 percent) of them were positive for anti-HBs. Of these, 29 subjects(0.9 percent) were positive for HBsAg. Positive results for anti-HBs by age were 78.6 percent for 6-12 months of age, 62.7 percent for 1-3 years of age, 51.9 percent for 4-6 years of age, 49.5 percent for 7-12 years of age, 63.4 percent for 13-15 years of age and 72.2 percent for 16-18 years of age. The 80 subjects who were tested negative for HBsAg/anti-HBs received a single booster vaccine, 71 subjects were tested positive for antibodies. IgG anti-HBc titer was checked for 169 of the subjects, 5 subjects were positive. Conclusion : In our study, a significant anamnestic response was observed in 88.8 percent of children. This is believed to be a result of the relatively long immunologic memory effect of the hepatitis B vaccination in children from the central area of Korea.

  • A low-cost, simple strip reader system using a linear movement mechanism of CD-ROM deck has been developed to characterize a lateral flow membrane-based immunochromatographic assay. The test strip reader was assembled by a CD-ROM deck and home-made optical head especially designed for immunoassays. The optical head for detecting reflected light from the test strip surface consists of green light-emitting diode, large area silicon photodiode, and anodized aluminum mounting block providing a slit structure for cutting light from the LED. The stepping motor of the deck was operated in the full step mode, whose distance of each reading point is about 0.15mm. The performance of the strip reader was tested by analysis of HBV(hepatitis B virus) antigen test kit. This strip reader can be useful for inexpensive, disposable, and membrane-based assays that provide visual evidence of the presence of an analyte in a liquid sample.

Effect of Sodium Hydrosulfite Solution on Agrobacterium-Mediated Chinese Cabbage Transformation and Transient Expression

  • We investigated chemical-wounding effect on Agrobacterium-mediated Chinese cabbage transformation via vacuum infiltration. Pre-germinated or germinating Chinese cabbage seeds were infiltrated with Agrobacterium tumefaciens LBA4404 cells carrying either GUS gene (pBI121) or hepatitis B virus surface antigen DNA (pBIHBsAg). Prior to agroinfiltration process, the seeds were soaked in sodium hydrosulfite (SHS) solution or just in sterile water as a control. Comparative transformation efficiency was determined by both of histochemistry and ELISA. We could demonstrate that SHS solution treatment especially to 1-day or 2-days old germinating seeds efficiently improved transformation process, and therefore, transient expression level. This strongly indicated that Agrobacterium infection could be facilitated indeed by SHS-causing wounds on Chinese cabbage seeds.

  • To improve the growth of mouse hybridoma 2c3.1 secreting anti-Hepatitis B surface antigen monoclonal antibody (anti-HBsAg MAb), we had constructed an enriched medium and observed the effects of fetal bovine serum and serum-free supplements including human serum albumin, 'insulin and transferrin', and monoethanolamine. For further enhancement of growth, the concentrations of two major energy sources, glucose and glutamine, were strengthened with various ratios in the enriched medium. Maximum cell growth and monoclonal antibody production obtained in various ratios of glucose/glutamine with an inoculation concentration of 2$\times$105 cells/ml were 0.73$\times$106-4.62$\times$106 cells/ml and 65.1-422.6 $\mu\textrm{g}$/ml, respectively. Glutamine was round to be a major energy source and a limiting nutrient in comparison to glucose for 2c3.1 cell cultivation in enriched media with low serum.

Development of Transient Gene Expression System using Seedlings

  • An efficient transient expression system has been developed and characterized for the production of foreign genes in seedlings. The seedlings can be easily produced from commercial seeds used for vegetable sprouts. In principal, a chemical abrasive was employed to generate wounds in seedlings prior to vacuum-infiltration with Agrobacterium tumefaciens bearing the target gene. This optimized chemical wounding-assisted agro-infiltration process resulted in up to 15-fold increase in $\beta$-glucuronidase (GUS) enzyme activity. This procedure has been used efficiently to express hepatitis B surface antigen (HBsAg) protein in a transient mode. Therefore, seedlings with proper wounds can be suggested as a convenient tool for the production of useful recombinant proteins.

Positive Rate of HBsAg in School Children in Incheon Area (인천 지역 초, 중, 고등학생의 B형 간염 바이러스 표면항원 양성률에 대한 조사 연구)

  • Purpose : The incidence of hepatitis B virus infection has gradually decreased since 1983 when hepatitis B vaccine was firstly produced. This study was performed to evaluate the efficacy of hepatitis B immunization. Methods : The elementary, middle and high school children in Incheon area were enrolled in this study in 1997 and 1998. Hepatitis B virus surface antigen(HBsAg) was measured using reversed passive hemagglutination(RPHA). Results : The results were as follows The positive rates of HBsAg in elementary, middle and high school children were 0.7%(337/46,861), 2.5%(381/15,026) and 3.1%(681/21,938) respectively in 1997 and 0.6%(257/41,946), 2.7%(379/13,652) and 2.4%(628/25,277) respectively in 1998. The positive rates of HBsAg in children under 19 years of age in 1985, 1990 and 1995 were 6.1, 5.2 and 3.5% respectively. Conclusion : The positive rates of HBsAg in elementary school children in 1997 and 1998 has decreased compared with those of the previous studies. The majority of elementary school children were given hepatitis B vaccination. These results suggest that hepatitis B vaccines used in Korea were effective for prevention of hepatitis B infection in school children.

Effect of Various Additives on the Production of Recombinant HBsAg during Methanol Induction in Pichia pastoris (Pichia pastoris에서 메탄올 유도시 첨가물이 재조합 HBsAg 생산에 미치는 영향)

  • Methanol induction conditions with various additives for the enhanced production of recombinant hepatitis B surface antigen(HBsAg) were investigated in Pichia pastoris, which can utilize methanol as a carbon source and produce recombinant proteins under the control of strong, tightly-regulated alcohol oxidase(AOX) promoter. The presence of non-methanol carbon sources such as glycerol and glucose fully repressed the expression of AOX promoter. Various additives were tested to improve the production of recombinant protein and it was found that sorbitol could be a good carbon source during methanol induction period. An optimized concentration of amino acid mixture enhanced the production of HBsAg significantly. Pluronic F-68, a non-ionic surfactant, also improved the production of HBsAg without inhibiting cell growth. Addition of oleic acid at 0.01%(v/v) during the induction period showed positive effect on the production of HBsAg. Finally, 1.2%(v/v) of trace salts enhanced the production of HBsAg 1.9 times compared to that of control culture.
