  • This histological study was performed to investigate the effects of .Paljeungsan-gagambang on the epididymal fat cell size, number and the fat drop area of hepatic lobule of rats fed High Fat Diet. Paljeungsan-gagambang was orally administered to the rats of sample group 26mg per 200g of rat's weight everyday for 4 weeks, and the control group were just fed with equal amount of saline solution. Then they are the epididymal fat cell size, epididymal fat cell numberand fat drop area of hepatic lobule that were measured. The results were summarized as follows; 1. The size of the epididymal fat cell was decreased significantly as compared with the control group. 2. The number of the epididymal fat cells was decreased significantly as compared with the control group in distribution chart classified by cell size 3. The Area(%) of the Fat drops in the hepatic lobule was decreased significantly as compared with the control group. According to the above results, it is considered that Paljeungsan-gagambang have effects on the decrease of the epididymal fat cell size, number and fat drop area of hepatic lobule of rats fed high fat diet; Thus, the adminisrtation of Paljeungsan-gagambang is considered to have an improving effect on obesities and liver diseases caused by high fat diet.

  • Rat liver sinusoids were observed by scanning electron microscopy. The sinusoids were provided with fenestrations which were divided into three types; small, medium-sized and large. The small fenestrations were usually gathered into clusters. The medium-sized fenestrations were either individually organized or in the cluster of small fenestrations. The large fenestrations were usually accompanied by small or medium-sized fenestrations. The lobule was divided into three zones; peripheral, intermediate and central, according to the distribution pattern of the fenestrations. The sinusoid providing with small fenestrations (type I) were predominant in the peripheral zone. The sinusoid showing medium-sized fenestrations (type II) were frequently observed in the central zone. The sinusoid having large fenestrations (type III) were abundant in the intermediate zone. This evidence indicate that the sinusoidal fenestrations become larger toward the intermediate zone from the peripheral zone of the lobule and progressively smaller toward the central zone. However, the fenestrations observed in the central zone seem to be larger than those seen in the peripheral zone of the lobule.

  • Intrauterine growth retardation (IUGR) leads to the dysfunction in digestive system, as well as the alteration in the expression of some functional proteins. Heat shock protein 70 (Hsp70) could be induced by various stress factors, but whether Hsp70 expression is changed in neonatal IUGR infants has not been demonstrated. This study was conducted to explore the expression of Hsp70 in the liver by using the IUGR piglet model. Liver and plasma samples were obtained from IUGR and normal birth weight (NBW) piglets at birth. The neonatal IUGR piglets had significantly lower liver weight than their counterparts. The activities of aspartate aminotransferase and alanine aminotransferase in serum were enhanced significantly in IUGR indicating liver dysfunction. The activities of superoxide dismutase (p<0.01), glutathione peroxidase (p<0.01) and catalase (p>0.05) were lower and the level of malondialdehybe was higher (p<0.05) in IUGR liver compared with in NBW. According to the results of histological tests, fatty hepatic infiltrates and cytoplasmic vacuolization were present in the liver of IUGR piglets, but not in NBW liver. The expression of Hsp70 protein was significantly higher (p<0.05) in IUGR piglet liver than in NBW. Similar to where the hepatic injuries were observed, location of Hsp70 was mostly in the midzonal hepatic lobule indicating that oxidative stress might be responsible for the increased expression of Hsp70.

  • To treat fulminant hepatic failure (FHF) patients, various extracorporeal bioartificial liver (BAL) systems have been developed. Several requirements should be met for the development of BAL systems: hepatocytes should be cultured in a sufficiently high density; their metabolic functions should be of a sufficiently high level and duration; and the BAL systems module should permit scaling-up and aseptic handling. Several investigators have found that freshly isolated primary hepatocytes can be cultured into three dimensional, tightly packed, freely suspended, multicellular aggregates, or spheroids. These specialized cell structures exhibited enhanced liver specific functions and a prolonged differentiated state compared to cells maintained in a monolayer culture. Cells in spheroids appear to mimic the morphology and ultrastructure of the in vivo liver lobule. The ability of hepatocytes to organize into three-dimensional structures was hypothesized to contribute to their enhanced liver-specific activities. In this study, the ammonia removal rate and urea secretion rate of pig hepatocytes spheroids encapsulated in Ca-alginate bead were determined. A packed-bed bioreactor with encapsulated pig hepatocytes was devised as BAL support system. The efficacy of the system was evaluated in vitro.
