이상지질혈증 동물 모델을 이용한 솔잎 착즙액 첨가 설기떡의 지질개선 효과 (Effect of Sulgidduk containing pine needle juice on lipid metabolism in high fat-cholesterol diet induced dyslipidemic rats)

  • 본 연구는 솔잎착즙액이 고지방식이로 이상지질혈증을 유도한 흰쥐의 지질대사에 미치는 영향을 분석하기 위해 실험동물을 정상식이 (ND)군, 고지방식이군으로 나누어 각각의 식이를 4주간 급여한 후 고지방식이군을 설기떡 (SD)군 및 솔잎착즙액설기떡 (PSD)군으로 분류하여 6주간 사육하고, 혈장과 간 및 분변의 지질관련 지표를 분석하였다. PSD군은 설기떡 섭취에 의해 유의하게 높아진 체중증가량을 유의하게 낮추었으며 (p < 0.05), 설기떡 섭취에 의해 유의하게 높아진 혈장 총 콜레스테롤 및 LDL-콜레스테롤을 낮추는 경향을 나타내었다 (p > 0.05). 간 조직과 분변의 지질대사 관련 지표를 분석한 결과 PSD군은 설기떡 섭취에 의해 ND군보다 유의하게 높아진 간 조직의 중성지방 및 총 콜레스테롤과 간 조직의 SREBP2 유전자 발현을 유의하게 낮추는 효과를 나타내었다 (p < 0.05). 또한 PSD군은 SD군보다 분변으로 배출되는 중성지방 함량을 14.8% 높였으며 (p > 0.05), 혈장과 간의 지질과산화를 SD군보다 각각 7.7%, 20.2% 낮추는 효과를 나타내었다 (p > 0.05). 혈장과 적혈구의 항산화활성 관련 지표를 분석한 결과 PSD군은 설기떡 섭취에 의해 낮아진 혈장 ORAC을 높이는 경향을 나타내었으며, 설기떡 섭취에 의해 유의하게 낮아진 간조직의 Mn-SOD 및 CAT 효소활성을 SD군보다 각각 21.5%, 19.6% 유의하게 높이는 효과를 나타내었다. 이상의 결과에서, 고지방식이로 이상지질혈증을 유도한 흰쥐에서 솔잎착즙액설기떡의 섭취는 체중증가억제, 혈장 콜레스테롤 감소, 간 내 콜레스테롤 생합성 억제, 간 조직의 지질수준 감소, 분변으로의 지질배출 증가, 간 조직의 지질과산화 감소, 혈장과 간 조직의 항산화 활성 증가 효능을 나타내는 것으로 확인되었다. 이에 본 연구에서 개발된 솔잎착즙액을 첨가한 설기떡은 지질 감소 효능을 가진 기능성 떡으로써 활용될 수 있을 것으로 기대된다.

Green perilla leaf extract ameliorates long-term oxidative stress induced by a high-fat diet in aging mice

  • BACKGROUND/OBJECTIVES: Oxidative stress is caused by an imbalance between harmful free radicals and antioxidants. Long-term oxidative stress can lead to an "exhausted" status of antioxidant defense system triggering development of metabolic syndrome and chronic inflammation. Green perilla (Perilla frutescens) is commonly used in Asian cuisines and traditional medicine in southeast Asia. Green perilla possesses numerous beneficial effects including anti-inflammatory and antioxidant functions. To investigate the potentials of green perilla leaf extract (PE) on oxidative stress, we induced oxidative stress by high-fat diet (HFD) in aging mice. MATERIALS/METHODS: C57BL/6J male mice were fed HFD continuously for 53 weeks. Then, mice were divided into three groups for 12 weeks: a normal diet fed reference group (NDcon), high-fat diet fed group (HDcon), and high-fat diet PE treated group (HDPE, 400 mg/kg of body weight). Biochemical analyses of serum and liver tissues were performed to assess metabolic and inflammatory damage and oxidative status. Hepatic gene expression of oxidative stress and inflammation related enzymes were evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). RESULTS: PE improved hepatopathology. PE also improved the lipid profiles and antioxidant enzymes, including hepatic glutathione peroxidase (GPx) and superoxide dismutase (SOD) and catalase (CAT) in serum and liver. Hepatic gene expressions of antioxidant and anti-inflammatory related enzymes, such as SOD-1, CAT, interleukin 4 (IL-4) and nuclear factor erythroid 2-related factor (Nrf2) were significantly enhanced by PE. PE also reduced the levels of hydrogen peroxide (H2O2) and malondialdehyde (MDA) in the serum and liver; moreover, PE suppressed hepatic gene expression involved in pro-inflammatory response; Cyclooxygenase-2 (COX-2), nitric oxide synthase (NOS), interleukin 1 beta (IL-1β), and interleukin 6 (IL-6). CONCLUSIONS: This research opens opportunities for further investigations of PE as a functional food and possible anti-aging agent due to its attenuative effects against oxidative stress, resulting from HFD and aging in the future.

Hypolipidemic Properties of Fermented Capsicum and Its Product

  • This study was conducted to investigate the effects of fermented capsicum and a capsicum product on lipid metabolism. Fermented capsicum was prepared from red pepper puree tov three months. After 9() days of fermentation, capsaicin and dihydrocapsaicin concentrations were reduced from 24.7 and 14.7 g/mL to 15.5 and 6.45 g/mL, respectively. The capsicum product was prepared from the fermented capsicum mixed with prune extract, green tea extract, neroli extract and oligo-saccharide. Thirty-two male Sprague-Dawley rats were as- signed to four dietary groups (control, high-fat control (BE-control), high-fat-fermented capsicum (HF-S-1), high- fat-capsicum product (HF-S-2)). Plasma and hepatic lipid profiles were examined after three weeks of experimental diet. Food intakes were significantly lower in the HF-S-1 and HF-S-2 groups compared to the control group (p<0.05). The weight of perirenal fat pads was lowest in animals on the control diet (low-fat) and highest in high-fat control diet. The addition of fermented capsicum to high fat diets, HF-S-1 and HE-S-2 groups, resulted in significantly lower fat pad weights compared with the HF-control group. Both fermented capsicum (HF-S-1) and the capsicum product (HF-S-2) groups had lower plasma TG levels, atherogenic-index, and liver TG levels than the BE-control group (p <0.05). Liver TC levels were significantly lower in the HF-S-2 group than the HF-control group. The results demonstrate a hypolipidemic effect of fermented capsicum and the fermented cap-sicum product.

Effects of Dietary Coconut Oil as a Medium-chain Fatty Acid Source on Performance, Carcass Composition and Serum Lipids in Male Broilers

  • This study was conducted to investigate the effects of dietary coconut oil as a medium-chain fatty acid (MCFA) source on performance, carcass composition and serum lipids in male broilers. A total of 540, one-day-old, male Arbor Acres broilers were randomly allotted to 1 of 5 treatments with each treatment being applied to 6 replicates of 18 chicks. The basal diet (i.e., R0) was based on corn and soybean meal and was supplemented with 1.5% soybean oil during the starter phase (d 0 to 21) and 3.0% soybean oil during the grower phase (d 22 to 42). Four experimental diets were formulated by replacing 25%, 50%, 75%, or 100% of the soybean oil with coconut oil (i.e., R25, R50, R75, and R100). Soybean oil and coconut oil were used as sources of long-chain fatty acid and MCFA, respectively. The feeding trial showed that dietary coconut oil had no effect on weight gain, feed intake or feed conversion. On d 42, serum levels of total cholesterol, low-density lipoprotein cholesterol, and low-density lipoprotein/high-density lipoprotein cholesterol were linearly decreased as the coconut oil level increased (p<0.01). Lipoprotein lipase, hepatic lipase, and total lipase activities were linearly increased as the coconut oil level increased (p<0.01). Abdominal fat weight/eviscerated weight (p = 0.05), intermuscular fat width (p<0.01) and subcutaneous fat thickness (p<0.01) showed a significant quadratic relationship, with the lowest value at R75. These results indicated that replacement of 75% of the soybean oil in diets with coconut oil is the optimum level to reduce fat deposition and favorably affect lipid profiles without impairing performance in broilers.

Gene Expression Profiling of Liver and Mammary Tissues of Lactating Dairy Cows

  • Gene expression profiling is a useful tool for identifying critical genes and pathways in metabolism. The objective of this study was to determine the major differences in the expression of genes associated with metabolism and metabolic regulation in liver and mammary tissues of lactating cows. We used the Michigan State University bovine metabolism (BMET) microarray; previously, we have designed a bovine metabolism-focused microarray containing known genes of metabolic interest using publicly available genomic internet database resources. This is a high-density array of 70mer oligonucleotides representing 2,349 bovine genes. The expression of 922 genes was different at p<0.05, and 398 genes (17%) were differentially expressed by two-fold or more with 222 higher in liver and 176 higher in mammary tissue. Gene ontology categories with a high percentage of genes more highly expressed in liver than mammary tissues included carbohydrate metabolism (glycolysis, glucoenogenesis, propanoate metabolism, butanoate metabolism, electron carrier and donor activity), lipid metabolism (fatty acid oxidation, chylomicron/lipid transport, bile acid metabolism, cholesterol metabolism, steroid metabolism, ketone body formation), and amino acid/nitrogen metabolism (amino acid biosynthetic process, amino acid catabolic process, urea cycle, and glutathione metabolic process). Categories with more genes highly expressed in mammary than liver tissue included amino acid and sugar transporters and MAPK, Wnt, and JAK-STAT signaling pathways. Real-time PCR analysis showed consistent results with those of microarray analysis for all 12 genes tested. In conclusion, microarray analyses clearly identified differential gene expression profiles between hepatic and mammary tissues that are consistent with the differences in metabolism of these two tissues. This study enables understanding of the molecular basis of metabolic adaptation of the liver and mammary gland during lactation in bovine species.

The effects of $Angelica$ $keiskei$ $Koidz$ on the expression of antioxidant enzymes related to lipid profiles in rats fed a high fat diet

  • This study was performed to examine the feeding effects of $Angelica$ $keiskei$ $Koidz$ (AK) and its processed products on serum, liver, and body fat content and the expression of antioxidant genes in rats fed a high fat diet. AK and its processed products were added at 3-5% to a high fat diet and fed to adult rats for 6 weeks. In experiment 1 (EXP 1), the rats were fed with one of six diets including a control diet (normal fat), high fat diet (HF), and HF + AK additives groups (four groups). In experiment 2 (EXP 2), the rats were separated into three groups of HF, HF + AK whole leaves, and HF + fermented juice (FS) + squeeze (SA). Body weight was not different among the groups in either experiment. The liver weight was lower in the FS and SA groups compared to that in the other groups (P<0.05). Serum luteolin was higher in the AK and processed products groups compared to that in the HF group (P<0.05). Gene expression of the antioxidative enzymes catalase and glutathione-s-reductase in the liver was higher in the AK processed products group than that in the other groups (P<0.05). The results suggest that the intake of AK and its processed products increased the expression of antioxidant enzymes in animals fed a high fat diet, reduced hepatic cholesterol content, and increased the effective absorption of luteolin.

간의 $^{18}F-Fluorodeoxyglucose$ 섭취 양상과 지방간의 관계 (Correlation of Hepatic $^{18}F-Fluorodeoxyglucose$ Uptake with Fatty Liver)

  • 목적: 다른 기관들에 비해 간에서는 상대적으로 FDG 섭취 양상이 매우 불규칙하며, 악성 병변이 없는 간에서 비정상적으로 FDG 섭취가 증가하기도 한다. 하지만, 이제까지 이러한 다양한 간의 FDG 섭취를 간의 다른 양성 질환과 연관시킨 연구가 없어, 저자들은 이 연구에서 간에서의 FDG 섭취 양상과 지방간, 간 기능 검사, 지질 프로필을 포함하는 다양한 임상 요인과의 관계를 밝히고자 하였다. 대상 및 방법: 건강검진 목적으로 $^{18}F-FDG$ PET/CT를 시행한 188명 (남/여: 120/68, 평균나이: $50{\pm}9$세)을 대상으로 분석하였다. 간의 FDG 섭취에 영향을 줄 수 있는 당뇨 및 내당증 장애 환자, 심한 간질환이나 오랜 약물 복용의 기왕력이 있는 환자는 연구대상에서 제외하였다. PET/CT의 횡단면에서 간의 5번 분절에 해당하는 부위에 관심영역을 설정하고 이 부위의 평균 SUV를 구하여, 지방간이 있는 군과 없는 군 사이의 차이를 분석하였다. 또한 혈중 간 기능(ALT, AST), 알코올성 간질환(${\gamma}-GT$), 총 콜레스테롤, 중성지방 수치와 간의 FDC 섭취와의 관계를 알아보았다. 지방간 환자들의 체질량지수, 음주력을 조사하여 이 인자들이 간의 FDG 섭취에 미치는 영향도 분석하였다 결과 지방간이 있는 군의 평균 SUV가 지방간이 없는 군보다 통계적으로 유의하게 높았다(p<0.05). 혈중 총 콜레스테롤과 중성지방 수치는 간의 FDG 섭취와 유의한 상관관계를 보인 반면, 다른 요인들은 상관관계를 보이지 않았다. 또한, 지방간 환자에서 음주나 체질량 지수의 위험군과 비위험군 사이에 간의 FDG 섭취 정도는 차이는 보이지 않았다. 다변량 분석 결과, 지방간의 유무와 혈중 중성지방 수치가 간의 평균 SUV에 영향을 주는 유의한 독립 인자였다. 결론: 결론적으로 간의 FDG 섭취 정도는 지방간의 유무에 영향을 받으며, 중성지방 수치와 유의한 상관관계를 보였다.

Skate cartilage extracts containing chondroitin sulfate ameliorates hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet-fed LDL receptor knockout mice in comparison with shark chondroitin sulfate

  • BACKGROUND/OBJECTIVES: In this study, we investigated the beneficial effects of skate cartilage extracts containing chondroitin sulfate (SCS) on hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet (HCD)-fed mice in comparison with the effects of shark cartilage-derived chondroitin sulfate (CS). MATERIALS/METHODS: Low-density lipoprotein receptor knockout (LDLR-KO) mice were fed HCD with an oral administration of CS (50 and 100 mg/kg BW/day), SCS (100 and 200 mg/kg BW/day), or water, respectively, for ten weeks. RESULTS: The administration of CS or SCS reduced the levels of serum triglyceride (TG), total cholesterol (TC), and LDL cholesterol and elevated the levels of high-density lipoprotein cholesterol, compared with those of the control group (P < 0.05). Furthermore, CS or SCS significantly attenuated inflammation by reducing the serum levels of interleukin (IL)-1β and hepatic protein expression levels of nuclear factor kappa B, inducible nitric oxide synthase, cyclooxygenase-2, and IL-1beta (P < 0.05). In particular, the serum level of tumor necrosis factor-alpha was reduced only in the 100 mg/kg BW/day of SCS-fed group, whereas the IL-6 level was reduced in the 100 and 200 mg/kg BW/day of SCS-fed groups (P < 0.05). In addition, lipid peroxidation and nitric oxide production were attenuated in the livers of the CS and SCS groups mediated by the upregulation of hepatic proteins of antioxidant enzymes, such as superoxide dismutase, catalase, and glutathione peroxidase (P < 0.05). CONCLUSIONS: These results suggest that the biological effects of SCS, similar to those of CS, are attributed to improved lipid profiles as well as suppressed inflammation and oxidative stress induced by the intake of HCD.

차전초 잎의 물 추출물이 고지방식이로 비만 유도된 C57BL/6 생쥐의 체지방 감소에 미치는 영향 (Effect of Plantaginis asiaticae Folium water extract on body fat loss in high fat-induced obese C57BL/6 mice)

  • Objective : Plantago asiatica L. has been used for many years in Korea, China, and Japan for the treatment of many diseases such as anti-inflammatory, anti-asthma and diuretic. But the anti-obesity effect of Plantaginis asiaticae Folium has not been studied yet. Thus, this study aimed to demonstrate the anti-obesity efficacy of Plantaginis asiaticae Folium water extract (PAF) in high fat diet-induced obesity animal model. Method : Male, five-weeks old C57BL/6 mice were divided into 5 groups; ND (normal diet + 0.9% saline), HFD (high-fat diet + 0.9% saline), PC (high-fat diet + Garcinia cambogia 500 mg/kg), PAF 100, 300 (high-fat diet + PAF 100, 300 mg/kg. Treatments were performed daily for 6 weeks per os. We evaluated the changes of body weight, white adipose tissues weight, hepatic and fecal lipids level, pancreatic lipase activity and lipid profiles in plasma. Result : Body weights, adipose tissue weights and FER were significantly lower in mice fed the high-fat diet with PAF treatment than in mice fed the high-fat diet alone. In plasma, TG, TC, LDL-C, ATS and ALT levels significantly decreased in PAF groups compared with HFD group. Also, hepatic triglyceride (TG) levels decreased and fecal TG increased in PAF groups compared to HFD group. In addition, the activity of pancreatic lipase was decreased in the PAF groups compared to that of HFD group. Conclusion: These results suggest PAF extract is effective in preventing and improvement obesity, so it might be a potential and safe material for anti-obesity agent of weight control or functional supplements.

고지방식이 마우스 비만모델에서 인제청금단에 의한 체중감량과 혈중 지질대사의 개선효과 (Injecheonggeumdan reduces body weight gain and blood lipid profiles in high fat diet-fed mice)

  • Objectives : We investigated the effects of Ingecheonggeumdan(ICD) on body weight and examined whether blood lipid levels and visceral fat are inhibited by it in high fat diet-fed obese male mice. Methods : 8 weeks old, high fat diet-fed obese male mice were divided into 6 groups: C57BL/6N normal, control, ICD-1(150mg/kg), ICD-2(300mg/kg), ICD-3(600mg/kg) and orlistat(10mg/kg). After mice were treated with ICD and orlistat for 8 weeks, we measured body weight gain, food intake, feeding efficiency ratio, fat weight, plasma leptin and lipid levels. We also performed histological analysis for liver and fat on the mice. Results : Compared with controls, ICD and orlistat-treated mice had lower body weight gain and adipose tissue weight, the magnitudes of which were prominent in ICD-3. Compared with controls, ICD and orlistat-treated mice had lower blood leptin levels, the magnitude of which was prominent in ICD-3. Compared with controls, ICD and orlistat-treated mice had higher blood HDL-cholesterol and lower blood plasma LDL-cholesterol, free fatty acid and triglyceride levels, the magnitudes of which were prominent in ICD-3. Blood plasma AST and ALT concentrations were not changed by ICD and orlistat, indicating ICD and orlistat do not show any toxic effects. Consistent with their effects on body weight gain, the size of adipocytes and hepatic lipid accumulation were significantly decreased by ICD and orlistat. Conclusions : These results demonstrate that ICD and orlistat effectively reduce body weight gain, blood plasma LDL-cholesterol, free fatty acid and triglyceride levels and improves abdominal fat, the magnitudes of which were prominent in ICD-3.
