The study was performed to investigate the effects of isoflavone and/or grape seed oil supplementation on serum and liver lipid profiles and bone strength in ovariectomized female rats. Female Spraque-Dawley rats were assigned into 4 groups of ovariectomized (O), isoflavone (0.085 mg/100 g b.w/day) in ovariectomized rats, grape seed oil in ovariectomized rats and isoflavone and/or grape seed oil in ovariectomized rats. After 8 weeks, biochemical profiles of serum, liver and bone were analyzed. Total food intakes, body weight gains and FER (food efficiency ratio) were not statistically significantly different among groups. Total cholesterol, triglyceride and LDL-cholesterol levels in serum were decreased by fed of isoflavone and/or grape seed oils. However, crude lipid and total cholesterol contents in liver were not affect of isoflavone and/or grape seed oil. The hepatic glutathione contents were increased by isoflavone and/or grape seed oil fed. The hepatic glutathione-S-transferase activity in isoflavone and/or grape seed oil supplemented groups were higher than that O group. Bone (scapular and femur bone) dry weight, femur of max weight and bending strength were no significant difference among groups. Our finding suggest that isoflavone/grape seed oils might have potential role for serum lipid profiles improvement and bone strength in vivo.
To evaluate the effect of defatted sesame flour(DSF) on the oxidative stress of ethanol feeding in rats, Wistar male rats were divided into 4 groups of control, ethanol, DSF and DSF ethanol. Each group was sacrificed after feeding for 4 weeks and was examined by measuring the formation of 2 thiobarbituric acid reactive substance(TBARS), total cholesterol(TC) in serum, redox glutathione S transferase(GST) enzyme activity and the contents of glutathione(GSH) in the liver. The formation of TBARS in the liver after ethanol feeding was significantly increased comparing to the control, but the levels were significantly decreased by the DSF as compared to the ethanol feeding group(p<0.05). When compared to fed control diet, we found that serum TC levels were significantly lower in the DSF fed group than control group (p<0.05). The activity of hepatic GST was significantly increased by DSF as compared to the control and was decreased by ethanol feeding. On the other hand, the hepatic contents of GSH were unaffected by DSF feeding. Our findings suggest that feeding DSF may inhibit ethanol induced oxidative stress may be due to the stimulation of antioxidative activity by sesaminol glucosides in DSF.
This paper examines the effects of dietary fats on the fatty acid composition and market enzyme activites during liver damage in 2-acetylaminofluorene treated rats. Weaning Sprague-Dawley male rats were fed the diet of beef tallow(BT source of sturated fatty acid) corn oil(CO source of n-6 fatty acid) and perilla oil(PO source of n-3 fatty acid) at the level of 15% fat. Ten days after feeding 2-acetylaminofluorene(2-AAF) was injected intraperitoneally twice every week at the level of 50mg/kg body weight for 7 weeks. Liver microsomal and cytosolic fractions were collected to determine the microsomal fatty acid composition lipid peroxide(malondialdehyde MDA) contents glucose 6-phosphatase(G6 Pase) activity cytochrome(Cyt) P-450 contents and cytosolic glutathione S-transferase(G6 Pase) activity cytochrome(Cyt) P-450 contents and cytosolic glutathione S-transferase(GST) activity. The fatty acid composition in microsomal fraction was reflected by different dietary fats. By 2-AAF treatment linoleic acids were increased regardless of the diet MDA contents were higher in CO group than it was in BT group. However 2-AAF treatment decreased MDA contents in all dietary groups. G6Pase activity of BT group was higher than those of the other gropus. CO group had the highest Cyt P-450 contents and 2-AAF treatment lowered Cyt P-450 contents only in CO gropu GST activites were higher in CO than in BT group whereas the enzyme activites were increased by 20AAF treatment in all dietary groups. These results suggest that dietary fats and 2-AAF treatment in all dietary groups,. These results suggest that dietary fats and 2-AAF treatment affect microsomal fatty acid composition The enzyme activities concerned with liver damage were influenced differently by dietary fats and 2-AFF treatment Although PO diet contains much more polyunsaturated fatty acids than CO diet PO diet doesn't cause more oxidant stress compared with CO diet in these data.
This study was conducted to investigate the antioxidative effects of Paeonia lactiflora (PL) seeds on antioxidative defense system and lipid peroxidation of liver in rats fed high-cholesterol diet. Sprague-Dawley male rats weighing 100 $\pm$10g were randomly assigned into five experimental groups fed 0.5% cholesterol ; HC group which was not supplemented PL seeds extract, 0.1% methanol extract diet group (MP1 group), 0.2% methanol extract diet group (MP2 group), 0.05% ether-souble fraction diet group (EP1 group) and 0.1 % ether-souble fraction diet group (EP2 group). Experimental diets were fed ad libitum to the rats for 3 weeks. The activity of hepatic superoxide dismutase (SOD) was not significantly different among all the high cholesterol diet groups. The hepatic glutathione peroxidase (GSHpx) activity in MP2 group was increased to 27% compared to HC group. The activity of hepatic catalase (CAT) was not significantly different among the all high cholesterol diet groups. The hepatic glutathione S-transferase (GST) activity in the EP1 and EP2 groups were increased to 12% and 13%, respectively, as compared to HC group. The levels of hepatic TBARS in the MP1, MP2, EP1 and EP2 groups were reduced by 18%, 21%, 20% and 23%, respectively, as compared with HC group. The contents of lipofuscin in liver was not significantly different among all the experimental groups. The results indicated that PL seeds extract may be reduced oxidative damage by activating antioxidative defense system of hepatic in rats fed high-cholesterol diets. 본 연구에서는 액체식이로 알코올을 열량의 35%로 6주간 섭취시킨 흰주의 간조직내 지질과산화물과 glutathione 이용효소계의 활성도 및 cytochrome P-450에 미치는 영향을 살펴보고 아울러 간암의 발암원으로 알려진 2-AAF를 투여하여 이들의 상호효과를 조사한 결과 다음과 같은 결과를 얻었다. 1. 체중, 간무게, 그리고 체중에 대한 간무게의 알코올에 대한 효과는 유의적인 차이를 보여 체중은 알코올에 대한 효과는 유의적인 차이를 보여 체중은 알코올 섭취군이 유의적으로 감소하였고 간무게 및 체중에 대한 간무게는 알코올군의 유의적으로 증가하였다. 2. Microsome의 지질과산화물 함량 및 cytosol의 glutathione peroxidase, glutathione reductase 활성도는 알코올과 2-AAF 투여시 모두 유의적인 차이를 보이지 않았으나, cytosol의 glutathione S-transferase 활성도는 알코올과 2-AAF에 의해서 모두 유의적으로 증가하였고 알코올 섭추와 함께
투여시 GST 활성도가 가장 많이 증가하였다. 3. Microsome의 cytochrome P-450 및 cytochrome b5 함량에 대한 알코올 효과는 cytochrome P-450 함량을 증가시키는 경향이 있고 cytochrome b5는 유의적인 증가를 보여 주었으며 2-AAF 투여 역시 cytochrome P-450의 유의적인 증가를 유도하였다. 따라서 알코올 섭취와 2-AAF 함께 투여 시 cytochrome P-450의 함량이 대조군의 약 2.2배 정도 증가하였으며 cytochrome b5 함량이 1.7배로 높이 증가하였다. 이는 2-AAF가 cytochrome P-450을 유도하여 자신의 대사를 촉진시키며 알코올의 섭취 또한 2-AAF의 hydroxylation을 증가시킬 수 있는 것으로 생각된다. 이상의 결과에서 과량의 알코올을 만성적으로 섭취시 간조직내의 microsome의 MFO system에 영향을 미쳐서 발암물질의 생체 활성화를 촉진시킬 수 있고 또한 GST의 활성도를 증가시키므로 어느 정도 발암과정에 영향을 미치는 것으로 생각된다.
To study on antioxidant effects in the liver of 40-week-old mouse, the sample were orally pretreated 5mg/kg/day for 5 days with red ginseng saponin components(total saponin, protopanaxadiol saponin, protopanaxatriol saponin, ginsenoside-Rd, ginsenoside-Re, compound-K) for 5 days. The ability of saponin to protect the mouse liver from oxidative damage was examined by determining the activity of superoxide dismutase(SOD), glutathione peroxidase(GPx) and the contents of glutathione, the level of malondialdehyde, The only protopanaxadiol among the ginseng saponin fractions was significantly increased the hepatic SOD activity(p<0.01). The red ginseng saponin induced a slight increase of GPx activity, especially ginsenoside Rd, compound K and protopanaxatriol treatments significantly increased its activity. The content of glutathione was significantly increased by total saponin, protopanaxadiol and ginsenoside Rd(p<0.01), but the oxidized glutathione level was lowered in all the red ginseng saponin. Finally, the level of malondialdehyde was significantly decreased by ginsenoside Rd and protopanaxadiol. In conclusion, protopanaxadiol and ginsenoside Rd among the saponin fraction were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation. This comprehensive antioxidant effects of red ginseng saponin seems to be by a certain action of saponin other than a direct antioxidant action.

Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly assigned into five experimental groups fed 0.5% cholesterol ; HC group which was not supplemented FVL extract, 0.05% methanol extract diet group (MSI group), 0.1% methanol extract diet group (MS2 group), 0.025% ethylacetate-souble fraction diet group (ES1 group) and 0.05% ethylacetate-souble fraction diet group (ES2 group). Experimental diets were fed ad libitum to the rats for 3 weeks. The hepatic xanthine oxidase (XOD) activity in the MS2 group was decreased to 20% as compared to HC group. The activities of hepatic superoxide dismutase (SOD) and catalase (CAT) were not significantly different among all the high cholesterol diet groups. The hepatic glutathione peroxidase (GSHpx) activity in MS2, ES2 groups were significantly increased as compared to HC group. The hepatic glutathione S-transferase (GST) activity in the MS2 group was increased to 20% as compared to HC group. The levels of hepatic TBARS in the MS1, MS2, ES1 and ES2 groups were reduced by 13%, 21%, 13% and 21%, respectively, as compared with HC group. The contents of lipofuscin in liver tissue was not significantly different among all the experimental groups. The results indicate that FVL extract may reduce oxidative damage by activating antioxidative defense system of liver in rats fed high-cholesterol diets.
식이성 단백질 함량이 toluene에 의한 간 손상에 어떠한 영 향을 미치는지를 검토할 목적으로 흰쥐를 저 단백식이 (casein 7%, LP) 및 고단백식이 (casein 20%, SP)로 성장시킨 후 체중 100g당 toluene과 olive oil의 동량흔액 0.2ml를 1일 1회 4번 투여한 다음 24시간 후 처치하였다. 체중 100g당 간무게는 SP군이 대조군에 비하여 약 28%, LP군은 약 17%의 유의한 증가를 보였으며 혈청 xanthine oxidase(XO)활성은 SP군에서는 약 35%의 유의한 증가를 나타내었으나 LP군에서도 증가되었으나 유의성은 없었다. 한편 toluene투여로 인한 간 glycogen, glutathione(GSH) 함량 감소율에 있어서는 SP군이 LP군 보다 높게 나타났으며 간세포의 대조군에 대한 cytosol, mitochondria 단백함량 감소율 역시 SP군이 LP군 보다 증가되는 경향을 보였다. 그리고 병리조직검사 소견에서는 SP군에서 swelling 정도가 LP군에 비하여 심화되어 나타남을 알 수 있었다.
This study was to investigate the effects of Gibangganbang on the liver injury induced dimethylnitrosamine, CCl4, ethanol and partial hepatectomy. Hydroxyproline, hepatic fibrosis, hepatic inflammatory cell infiltration, fatty value, lipidoperoxide levels, glutathione levels, mitotic index, contents of protein in the serum and liver tissues were measured and observed. The results obtained were as follows. 1. The increasing level of hydroxyproline volume induced by dimethylnitrosamine in mice was decreased by the oral administration of Gibangganbang. 2. The degree of hitological fibrosis and hepatic inflammatory cell infiltration induced by $CCl_4$ was decreased by the oral administration of Gibangganbang. 3. The degree of fatty value and the increasing level of lipidoperoxide in liver tissues was decreased by the oral administration of Gibangganbang. 4. The level of glutathione in liver tissues was increasing by the oral administration of Gibangganbang. 5. The increasing level of microsomal lipidoperoxide in vitro assay was decreasing by the oral administration of Gibangganbang. 6. The increasing level of the mitotic index, weight of liver, contents of protein, RNA and DNA synthesis of the liver tissues after partial hepatectomy was activated by the oral administration of Gibangganbang. These results suggest that Gibangganbang not only inhibits liver cirrhotic change and ethanol-induced fatty change but also activates antioxidant enzymes and regeneration ability ocf liver.
This study was conducted to investigate the synergic effect of dietary selenium and methionine levels on hepatic lipid metabolism in ethanol treated rats. Sprague-Dawley male rats were fed diets containing three levels of methionine(0,3 and 9g/kg diet) with or without selenium(0.45mg/kg diet). Ethanol was administered with 25%(v/v) ethanol orally at the same time once a day in ethanol group and isocalori sucrose was administered to the control group. The rate were sacrificed after 5 and 10 weeks of feeding period. Glutathione content was decreased by ethanol treatment and significantly increased in proportion to level of dietary methionine and was higher in selenium deficiency group than that of selenium admin-istration group. Lipid peroxide content was significantly increased in deficiency of both methionine and selenium(LMet-Se+EtOH) group. Total lipid triglyceride and cholesteol contents in liver were increas-ed and phospholipid content was decreased in ethanol treated group and ethanol treatment accelerated those increment and decrement in methionine deficiency(LMt) group and excessive methionine admin-istration(HMet)group.
