고강도 인터벌 트레이닝이 D-Gal/LPS로 유도된 마우스의 급성 간 부전에 미치는 효과 (Effect of High-Intensity Interval Training on Acute Liver Failure Induced by D-Galactosamine/Lipopolysaccharide in Balb/c Mice)

  • PURPOSE: This study investigated the protective role of high-intensity interval training against acute liver injury induced by D-galactosamine (D-Gal)/lipopolysaccharide (LPS). METHODS: A total of 30 male BALB/c mice aged 5-week were randomly assigned to high-intensity, interval training group (EX, n=10) or control group in cage (Non-EX, n=20) for 10 weeks. Peritoneal injection of D-Gal (700 mg/kg body weight) and LPS ($10{\mu}g/kg$ body weight) was applied to induce acute liver injury, and liver tissue was harvested 6 hours after the injection. Hematoxylin and Eosin (H&E) staining was used for liver histology. Real-time PCR was used to quantify expression of pro-inflammatory and anti-inflammatory genes in the liver. RESULTS: The liver histology showed that D-Gal/LPS treatment resulted in hepatic damage and increased number of neutrophils in conjunction with upregulation of hepatic IL-6 and $TNF-{\alpha}$ mRNAs and downregulation of hepatic $PPAR{\alpha}$ and SIRT1 mRNAs. On the other hand, the 10-week interval training resulted in a significant improvement in cardiorespiratory fitness assessed as run time to exhaustion on a treadmill. In addition, the interval training attenuated the D-Gal/LPS-induced liver damage and increased number of neutrophil in conjunction with downregulation of hepatic IL-6 and $TNF-{\alpha}$ mRNAs and upregulation of hepatic $PPAR{\alpha}$ and SIRT1 mRNAs. CONCLUSIONS: This study suggests that high-intensity interval training suppresses the D-Gal and LPS-induced acute liver damage and inflammatory responses.

Pharmacokinetics of Acebutolol and Its Main Metabolite, Diacetolol After Oral Administration of Acebutolol in Rabbits with Carbon Tetrachloride-Induced Hepatic Failure

  • Pharmacokinetic characteristics of Acebutolol and its main metabolite, diacetolol, following a single 10 mg/kg oral dose, were investigated in rabbits with carbon tetrachloride-induced hepatic failure. Plasma concentrations of acebutolol and diacetolol were determined by a high performance liquid chromatography assay. The area under the plasma concentration-time curves (AUC) and maximum plasma concentration ($C_{max}$) of acebutolol were significantly increased in moderate and severe carbon tetrachloride-induced hepatic failure rabbits. The ratio of the diacetolol to total acebutolol in plasma (i.e., metabolite percentage rate) was significantly decreased in moderate and severe carbon tetrachloride-induced hepatic failure rabbits. Volume of distribution ($V_{d}$) and total body clearance ($CL_{t}$) of acebutolol were significantly decreased in moderate and severe carbon tetrachloride-induced hepatic failure rabbits. Slope of terminal phase ($\beta$) of acebutolol was significantly decreased in hepatic failure rabbits. These findings suggest that the $V_{d},{\;}CL_{t}$ and $\beta$ of acebutolol were significantly decreased as a result of inhibition of the hepatic metabolism in moderate to severe hepatic failure rabbits. Therefore, dose adjustment may be necessary for acebutolol in hypertensive patients with hepatic damage.

Common Hepatic Artery Originating from Left Gastric Artery: A Rare Variant Encountered in Gastric Cancer Surgery

  • The hepatic artery can have numerous variations. However, a common hepatic artery originating from the left gastric artery and the entire hepatic blood supply furnished by the left gastric artery is an extremely rare anomaly. We encountered this anomaly in a patient with advanced gastric cancer. A surgeon should recognize this image appearance and identify the anomaly. Without knowledge of this anomaly and given the strategy for extensive lesser sac dissection generally employed during gastric cancer surgery, a surgeon could easily inadvertently divide this vessel, resulting in critical liver damage. We report a case of common hepatic artery originating from left gastric artery and review of the literatures.

Hepatoprotective effect of kasni against aflatoxin B1 induced hepatic damage in mice

  • The efficacy of alcoholic extract of Kasni (Cichorium intybus L.) to control hepatic damage induced by aflatoxin $B_1$ was explored in Swiss albino mice. Aflatoxin $B_1$ was administered orally to the mice with a daily dose of $66.6{\mu}g/kg$ body weight till three months. A signifi-cant increased in thio barbituric acid reactive substances (TBARS) levels with concomitant reduction in enzymatic (glutathione-s-transferase, glutathione peroxidase, superoxide dismutase, and catalase) and nonenzymatic (reduced glutathione) antioxidants were shown in aflatoxin treated group of mice. However, there was a significant reduction in increased TBARS levels and elevation in enzymatic. and non enzymatic antioxidant levels in group of mice which received alcoholic extract of kasni (300 mg/kg bw / day) along with aflatoxin. Histopathological analysis of liver samples also confirmed the hepatoprotective effect of kasni extract. These results suggest that kasni shows hepatoprotective effect against aflatoxin $B_1$ induced hepatic damage in mice.

족삼리 전침자극이 Carbon Tetrachloride 장기투여로 유발된 흰쥐의 만성 간손상에 미치는 영향 (The Effect of Electro-acupuncture at ST36 on Carbon Tetrachloride-induced Chronic Hepatic Damage in Rats)

  • Objectives : The purpose of this study is to investigate the effect of electro-acupuncture(EA) at ST36 on $CCl_4{^-}$ induced chronic hepatic damage in rats. Methods : The authors performed several experimental items, including measurements of liver index, hematological analysis for WBC, lymphocytes, neutrophils, monocytes, hemoglobin, and biochemical assays for ALT, AST, ALP in serum, and histological analysis of liver tissue. Results : Liver index(liver weight/body weight), the levels of WBC, Neutrophils and Monocytes in blood, the levels of ALT, AST and ALP in serum which were increased by $CCl_4{^-}$ intoxication were reduced significantly by EA at ST36. The levels of lymphocyte and hemoglobin in blood which were decreased by $CCl_4{^-}$ intoxication were significantly increased by EA at ST36. Conclusions : EA at ST36 has a hepatoprotective effect on chronic hepatic damage in $CCl_4{^-}$ intoxicated rats.

Antioxidant Activities of Isoflavones from the Rhizomes of Belamcanda chinensis on Carbon Tetrachloride-Induced Hepatic Injury in Rats

  • The present study was carried out to clarify whether tectorigenin and tectoridin isolated from the rhizomes of Belamcanda chinensis (Iridaceae) inhibit hepatic damage induced by carbon tetrachloride ($CCl_4$)-intoxication in rats by the experimental methods in vitro and in vivo. Tectorigenin and tectoridin exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4$-intoxication in rats, as well as in a lipid peroxidation causing a significant decrease in malondialdehyde (MDA) production by thiobarbituric acid (TBA)-reactant assay. Both compounds also showed strong increase in the antioxidant enzymes such as hepatic cytosolic superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-px) activities in $CCl_4$-intoxicated rats. These results suggested that tectorigenin and tectoridin isolated from the rhizomes of B. chinensis possess not only the antioxidative, but also the hepatoprotective activities in $CCl_4$ -intoxicated rats.

  • This study was undertaken to investigate the inhibition effects of Auricularia auricula-judae methanol extract in edible mushroom on lipid peroxidation and liver damage in benzo(a)pyrene(B(a)P)-treated mice. The activities of serum aminotransferase, cytochrome P-450, superoxide dismutase, catalase, glutathione peroxidase and the hepatic content of lipid peroxide after B(a)P-treatment was markedly increased than control but those levels were significantly decreased by the treatment of Auricularia auricula-judae methanol extract. Glutathione S-transferase activity and the hepatic glutathione content were decreased by B(a)P-treatment than control, but those were also inhibited by the treament of Auricularia auricula-judae methanol extract. These results suggest that Auricularia auricula-judae methanol extract have a protective effect on liver damage by B(a)P.

Hepatoprotective activity of Indigofera aspalathoides extract against CCl4-induced liver damage

  • The plant Indigofera aspalathoides are used by a large number of tribes in India for the treatment of various hepatic disorder. The methanol extract of Indigofera aspalathoides (MEIA) was evaluated for its effect on carbontetrachloride $(CCl_{4})$ induced liver damage. Biochemical parameters such as serum glutamine oxaloacetate trasaminase (SGOT), serum glutamine pyruvate transaminase (SGPT), serum alkaline phosphatase (ALP), total serum protein (TP), thiobarbituric acid reactive substances (TBRS) and glutathione content of the liver were estimated to assess liver function and metabolism. Biochemical observations suggest that methanol extract of Indigofera aspalathoides (MEIA) significantly restored the liver function and metabolism towards normal condition in $CCl_{4}$-induced hepatic damage.

흰쥐에 있어서 Bromobenzene전처치가 간조직 중 Glutathione 및 Glutathione S-transferase활성에 미치는 영향 (Effect of Bromobenzene Pretreatment on the Hepatic Glutathione Content and Glutathione S-transferase Activity in Bromobenzene Treated Rats)

  • To evaluate the effect of bromobenzene pretreatment on the bromobenzene metabolism, the animal group was induced the stage of slight liver damage with 7 times bromobenzene injection every two days (400 mg/kg body wt. i.p.). In the present experimental animal model, the single dose of bromobenzene(400 mg/kg body wt. i.p.) was injected to the bromobenzene-pretreated rats and the hepatic aniline hydroxylase(AH) activity, glutathione(GSH) content and glutathione S-transferase (GST) activity were determined at the intervals of 2, 4, 8, 24 hours throughout 24 hr. The activities of hepatic AH and GST were generally higher in bromobenzene-pretreated rats than those in normal group throughout the whole course of experiment. Furthermore, the decreasing rate of hepatic GSH content was also higher in bromobenzene pretreated rats than in normal rats. Moreover, the value of V$_{max}$ in hepatic GST was higher in bromobenzene pretreated rats than that in the normal rats. In conclusion, these results indicate that the pretreatment of bromobenzene may rather enhance the bromobenzene metabolism.

흰쥐에서 사염화탄소 투여로 유발된 간 손상에 대한 인진쑥 추출물의 치료효과 (The Therapeutic Effect of Artemisia Capillaris Extract on Hepatic Damage Induced by Carbon Tetrachloride in Rats)

  • This experiment was conducted to find out the therapeutic effect of Artemisia capillaris extracts on hepatic damage in rats induced by carbon tetrachloride ($CCl_{4}$). In this experiment, 96 Sprague-Dawley rats were used as experimental groups, which were divided into 4 groups; control group(A), $CCl_{4}$-treated group(B), $CCl_{4}$+Artemisia extract-treated group(C) and $CCl_{4}$+silymarin-treated group(D). The B, C, D group were administrated single dose of $CCl_{4}$(2.5 ml/kg) to induce acute hepatic injury. C group was administrated with Artemisia capillaris extract(200 mg/kg/day) and D group treated with silymarin(50 mg/kg/day) for 7 days. Hematological, ultrasonographical, histological examinations and examination of antioxidant activity were also performed in all groups. AST and ALT activities of C group were significantly decreased compared with B group. The activities of AST and ALT in C and D groups returned to the normal range more rapidly than those of B group. In ultrasonographic examination, the echogenicity of liver in C group was significantly decreased compared with B group. Also C and D group had tended to recover faster than B group on liver histogram. Histologically, the percentage of degenerative regions and degenerative cell numbers in peri-central vein hepatic parenchyma of C and D group were significantly decreased compared with B group. In examination of lipid peroxidation, malondialdehyde of hepatic tissue in C group was decreased as compared with B group. In examination of antioxidant enzyme activity in liver, glutathione peroxidase and catalase activities were significantly increased compared with B group. As results of this study, it is thought that A. capillaris extract has therapeutic effects on hepatic injury induced by carbon tetrachloride, and has the similar therapeutic effects as silymarin in rats.
