The phototoxicity inhibitory activity of 15 natural products having antiinflammatory effect was screened by three in vitro methods: yeast growth inhibition test with Candida albicans, RBC photohemolysis and MTT assay. We induced phototoxic reaction by irradiating UVA (365 nm) on chlorpromazine (CPZ) that has been widely documented as phototoxic agent in clinical and experimental studies and then observed the effects of the natural products after treating them with CPZ. In yeast growth inhibition test, X. stramonium showed the inhibitory effect on the UVA phototoxicity and E. officinalis, Yeast, P. suffruticosa showed phototoxicity inhibitory effect in that their % hemolysis compared with control were 36.14${\pm}$ 2.69, 42.82${\pm}$1.35, 36.41${\pm}$0.48 on UVA. In MTT assay, all tested natural products increased cell viability compared with the control.
These biologic test procedures are designed to test the suitability of P.V.C. made in Korea intended for parenteral preparation, which were based on the U.S. Pharmacopeia XIX "Biologic Test-Plastic Container", Official from July 1, 1975. Healthy adult human blood and rabbits weighing 2\ulcorner.2Kg were used for test materials. Sample P.V.C. were sampled from the medical equipments made in Korea randomly and Control P.V.C. were sampled from the standardized Cobe and Polystan P.V.C. tubes. P.V.C. extract was prepared from a homogeneous P.V.C. samples by incubating 60 square centimeters of the sample per 20 millimeters of sterile pyrogen-free saline at 70\ulcorner for 72 hours or autoclaving at 120\ulcorner for 1 hour. The Implantation Test was designed to evaluate the reaction of living tissue to the plastic by the method of the implantation of the Sample itself into animal tissue. The Systemic Injection Test, the Intracutaneous Test, and the remainders were designed to determine the biological response of animals to plastics by the single-dose injection of specific extracts prepared from a Sample. The results are as follows; 1.Implantation Test - No significant difference for reactions was noted between the Sample treated animal and the Control after 72 hours of implantation. 2.Systemic Toxicity Injection Test - No sign of toxicity and/or death immediately after injection and at 4, 24, 48 hours respectfully after injection. 3.Intracutaneous Test - None of the animals treated with the Sample showed a significantly greater reaction than the observed in the animals treated with Blank. 4.Pyrogen Assay-Only one animal treated with the Sample showed the maximal rise of rectal temperature about 0.2\ulcorner after 3 hours of injection, but remainders showed no change. 5.Hemolytic Index - The positive Control tube of distilled water exhibited complete hemolysis while the negative Control tube and P.V.C. extract were negative demonstrating no hemolysis. 6.Cell Morphology of Erythrocytes and Leukocytes on Stored, Heparinized Human Blood -- There was no significant difference in the morphology of either the Control or Sample extract. 7.Clotting Mechanism of Human Blood in vitro - After allowing to the P.V.C. extract at room temperature for 5 Hours and at 10\ulcorner for 24 hours, there was no appreciable difference in Prothrombin Time under these conditions. 8.Clotting Mechanism of Rabbit in vivo - At the termination of 5 days after intraperitoneal injection of the P.V.C. extract, no significant changes in Clotting Time were observed. According to the above results, it could be concluded that the P.V.C. made in Korea was acceptable for parenteral preparation, especially treated with physiologic saline and/or human blood.man blood.
Korean rockfish, Sebastes schlegelii, is a representative bony fish that belongs to the family Scorpaenidae and the order Scorpaeniformes. It has high ecological and economic value and is widely cultivated in many East Asian countries, including South Korea, Japan and China. One of streptococci, Streptococcus iniae, is Gram-positive cocci with a negative reaction for catalase and oxidase. The Korean rockfish shows clinical signs when infected with S. iniae, such as body darkening, bleeding, enlarged kidneys, blurred eyes, abdominal distension, etc., ultimately leading to death. The Korean rockfish causes significant economic losses every year in South Korea due to streptococcosis. In this study, we identified bacteria from the fish using polymerase chain reaction and conducted analyses of hemolytic activity and biochemical tests using API 20 STREP and API ZYM systems. Results of confirming the hemolytic activity (n=4) observed in alpha-type hemolysis (25%), beta-type hemol- ysis (50%), and gamma-type hemolysis (25%) of isolates. The biochemical test results exhibited sig- nificant variation among S. iniae. Additionally, we performed intraperitoneal injection with S. iniae in the fish and analyzed the phylogenetic tree using housekeeping genes of S. iniae, including cpsD, arcC, glnA, groEL, gyrB, mutS, pheT, prkC, rpoB, and tkt, via multilocus sequence typing (MLST). The lethal dose (LD50) showed strong pathogenicity, such as 3.34 × 10 colony-forming unit (CFU)/ml for 23FBStr0601 strain and 7.16 × 10 CFU/ml for 23FBStr0602 strain. 23FBStr0603 strain showed relatively low pathogenicity at 1.73 × 105 CFU/ml. The strains 23FBStr0601 and 23FBStr0602, which showed strong pathogenicity, clustered into one monophyletic group. The 23FBStr0603 strain showed weak pathogenicity and formed a monophyletic group with KCTC 3657.
Aqua-acupuncture therapy is a new acupuncture-therapy that cures diseases by needling with direct medication of oriental or western medicine at acupuncture points which are related to a disease, pressure pain points or positive reaction points according to meridian theory, followed by controlling one's body function and relieving the pathogenic state. Recently, the method is widely applied clinically owing to its handiness and usefulness, but it is pointed that it has some problems. Therefore, the present studies have been carried out to introduce history of the method, materia-medica which is ordinarily used, acupuncture points to be selected, handling method, mechanism of action, range of application and some points to be careful. Moreover, through the experiences that we have made by applying the method clinically, we suggest that the following study should be carried out : First, clinical application of needling with direct medication should be made by using materia-medica which is already proved safe. Second, it is essential to study the side-effects accompanying fever, pains, mass and hemolysis after the method is applied. Third, the study on the mechanism of action of the method should be accomplished continuously.
The antiosidant activity of carnosine and related compounds such as anserine, homo-carnosine, histidine, and $\beta$-alanine which are found in most mammalian tissues, was investigated using hypochlorite (HOCl)-induced oxidant systems. Carnosine and related compounds were protective against HOCl-induced ascorbic acid oxidation, as determined by UV absorbance at 265nm. L-histidine was the most effective among them. The inhibitory effect of these compounds was strongly associated with a decrease in HOCl. It was also found that carnosine and related compounds significantly protected against the HOCl-mediated erythrocyte damage, as determined by hemoglobin release and gemolysis (p<0.05). Carnosine and anserine also inhibited of $\alpha$-antiprotease($\alpha$-AP) by HOCl, thereby inactivating porcine elastase. The inhibitory effect of carnosine on inactivation of $\alpha$-AP by HOCl depended on the concentration of carnosine and on the time preincubated with HOCl. Homocarnosine, histidine, and $\beta$-alanine did not inhibit the reaction. These results indicate that carnosine and related compounds can neutralize or scavenge HOCl. Thus, these compounds may play an important role in protecting against HOCl-mediated damage of biomolecules in vivo.
Thirteen strains of Clostridium perfringens were isolated from the pigs with hemorrhagic enteritis. The characteristics of the outbreaks, clinical signs and lesions were examined. The biochemical properties, type of toxin and susceptibility to antimicrobial agents of the isolates were investigated. The results could be summarized as follows ; 1. Almost of the pigs affected with hemorrhagic enteritis, 17 cases examined from 1989 to 1992, were piglets less than 7 day old. 2. The average mortality rate of piglet less than 7 day old affected with hemorragic and necrotic enteritis was 48.5%. 3. The clinical signs of pigs with hemorrhagic enteritis were depression, hemorrhagic diarrhea, anemia and dehydration. Necropsy of the infected pigs showed typical hemorrhage of upper intestine and necrosis of mucosal membrane. 4. The characteristic biochemical properties of the isolates were 2-band hemolysis, positive reaction of reverse CAMP test and formation of LV precipitate in egg yolk medium. 5. The toxin type of the 13 isolates, investigated by mouse inoculation test, was all type C strains of Clostridium perfringens. 6. In susceptibility test to antimicrobial agents, 13 isolates of Clostridium perfringens were highly sensitive to ampicillin, enrofloxacin(Baytril), cephalothin, penicillin and trimethoprim-sulfamethoxazole.
As a part of investigation for listeriosis, we attmepted iolationof Listeria spp. from commerical frozen and refrigerated foods at the supermarket level. Seven strains of Listeria spp. were isolate from 6 samples (7.15) among 84 samples of frozen foods, and ten strains of Listeria spp. were also isolated from 8 samples (7.6%)_ among 105 samples of refrigerated foods at the supermarkets in Pusan area. From a total of 189 commerical foods, the number of isolated Listeria spp. and ratio were 6 strains(3.25) of L. grayi, one strain (0.5%) of L. welshimeri, 6 strains (3.2%) of L. innocua and 4 strains (2.15) of L monocytogenes. Listeria spp. isolates except L. monocytogenes did not show $\beta$-hemolysis on blood agar and positive reaction in CAMP test with Staphyloccccus aureus. In the antibiotic susceptibility, most isolates of Listeria spp. were susceptible to 12 antibiotics such as ampicillin , cephalotin, penicillin, amikacin, gentamicin, erythromycin, kanamycin, vancomycin, tobramycin, carbenicillin , tetracycline and trimethoprim /sulfamethoxiazole. Four strains of L. monocytogenes were susceptible to ntibiotics used in this study except nitrofurantio. The serotype of 3 strains and one strain of L. monocytogenes were classified into Listeria O serotype 1 and 4, respectively.
S. parauberis는 S. uberis type Ⅱ로 분류되었으나 Williams and Collins (1990)에 의해서 S. parauberis라는 종명이 제시되었다.
This study was carried out to investigate the necrotic enteritis of chicken in Korea. Epidemiological, pathological and microbiological examination were done on 12 naturally occurred cases of necrotic enteritis of chicken. And the susceptibility of isolated bacteria to antimicrobial agents was also examined. The results obtained were as follows; 1. The Clostridium perfringens infection, necrotic enteritis of chicken, occurred in the chickens mainly raised in floor pens. Necrotic enteritis of chicken was occurred coincidently with coccidiosis and Gumboro disease frequently. And several cases were recurred at 2-3 weeks after recovery. 2. Clinical signs of the infected chickens were depression, decreased appetite, reluctance to move, diarrhea, ruffled feathers and acute death within several hours. 3. The characteristic biochemical properties of isolates were 2-band hemolysis, no motility, positive reaction of reverse CAMP test and the formation of LV precipitate in egg yolk medium. 4. Gross lesions of the infected chickens were distention of intestine with gas, thickened mucosa and formation of thick pseudomembrane in intestine. Livers were friable with yellowish brown color and, in some case, showed demarcated necrotic foci. 5. Histopathological findings of the infected chickens were severe necrosis of the intestinal mucosa and attachment of numerous large bacilli to the mucosal surface of necrotic villi. In liver, necrosis of liver tissue and numerous large bacilli in the necrotic foci were also observed. 6. In susceptibility test to antimicrobial agents, 12 isolates of Clostridium perfringens were highly sensitive to ampicillin, baytril, cephalothin and penicillin.
