단기간의 인진쑥 투여가 개의 혈액상에 미치는 영향 (The Effect of Artemisia capillaris Extract on Hematological Changes in Dogs)

  • This study was conducted to investigate the hematological changes of the dogs when administrated excessive amount of the Artemisia capillaris extract for a short period. In this experiment, clinically healthy dogs(n = 20, average weight = 7.68 kg) were divided into 2 groups : Artemisia capillaris rude juice extract group(CR) and concentrated extract group(CE). They were inserted stomach tube and administrated the extracts (5 ml/kg) for 8 days. The followings are the results of this experiment.: 1). The number of red blood cell(RBC) was significantly (p<0.05) decreased in CE group on day 4, 5, 6, whereas CJE group showed significantly (p<0.05) difference on day 5. 2). The packed cell volume(PCV) and hemoglobin concentration were significantly (p<0.05) decreased in both groups during experimental periods. 3). The met hemoglobin was significantly (p < 0.05) increased from 12 hrs after administration to day 8 in CJE group wheres, it was significantly (p<0.05) increased on day 6,7 in CE group.

Measurement of Hemoglobin Adducts in Female Mice Inhaled with 1,3-butadiene by Using GC/MS

  • 1,3-butadiene(DB) is a well-established rodent carcinogen, and a probable carcinogen to humans. This study was investigated the formation of hemoglobin adduct in ICR female mice inhaled with BD for 3 weeks (5 hr/day, 5 days/week). Body weights of mice were significantly low from onward day 4 or 9 of exposure comparing the control. Hemoglobin adducts formed by DB exposure were (N-2-hydroxy-3-butenyl) valine (HB Val adduct) and (N-2,3,4-trihydroxy-butyl)valine(THB Val adduct). The levels of HB Val adducts were 1.8, 3.7 and 6.2 pmol/mg globin for the 500 ppm BD inhalation group, and 5.7, 7.4 and 16.0 pmol/mg globin for the 1000 ppm BD inhalation group, when observed on the $1^{st},\;2^{nd},\;and\;3^{rd}$ week after inhalation exposure, respectively. The levels of THBVal adducts were 32.0, 42.0 and 55.0 pmol/mg globin for the 500 ppm DB inhalation group, and 67.8, 72.7 and 83.5 pmol/mg globin for the 1000 ppm BD inhalation group, when observed on the $1^{st},\;2^{nd},\;and\;3^{rd}$ week after inhalation exposure, respectively. Ratios of THBVal and HBVal adducts were higher at earlier exposure period and lower concentration. Ratios on the $1^{st},\;2^{nd},\;and\;3^{rd}$ week were 17.8, 11.4 and 8.87 for the 500 ppm BD inhalation group, and 11.9, 9.8 and 5.2 for the 1000 ppm BD inhalation group, respectively. In conclusion, THB Val and HB Val adducts were the important hemoglobin adducts in ICR female mice inhaled with BD, and the latter was more appropriate biomarker than the other for biological monitoring of BD exposure.

Effects of supplementation cysteine-coated Fe3O4 nanoparticles compared to FeSO4, on reproductive performance in male quail

  • Iron has a crucial role in growth as part of metalo-proteins like haemoglobin or myoglobin, enzymes; they are also involved in energetic reactions. Iron plays a vital role in fertility. At high doses, Iron has a harmful consequence on the reproductive system, which can be strongly reflected the final stage of spermatogenesis. Nutritional products are claiming to use nanotechnology and it is important to recognize the potential toxicity of nano-sized nutrients. Recently iron nanoparticles were proposed as a food additive for poultry. The objective of this study was to investigate the effects of L-cystein coated iron oxide nanoparticles on reproductive performance in male quails. The results of Fourier Transform Infrared Spectrometer, Alternating Gradient Force Magnetometer and Scaning Electron Microscopy showed that iron oxide nanoparticles was produced and have been coated with L-cycstein (Fe3O4-Cys NPs). A total of 100 one-week-old quail chicks were randomly placed to five groups of five replicates. Four quails (two male and two females) were raised in an individual cage for each replicate. The five experimental treatment diets consisted; negative control diet, with no Iron supplementation; positive control diet supplemented with 60 mg/kg of Fe3O4; treatment diets supplemented with 0.6, 6 and 60 mg/kg of L-cystein coated iron oxide nanoparticles. The hemoglobin, Red blood cell, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, gonadal somatic index, daily sperm production, total testicular sperm and sperm viability of the male quails that were fed with diet supplemented by 0.6 mg/kg of Fe3O4-Cys NPs were improved as compare with negative control. This study showed that not only the use of the Fe3O4-Cys nanoparticles had no side effects but also it can be used as a feed additive to improve the reproductive performance in male quails.

Cigarette Smoking and Serum Bilirubin Subtypes in Healthy Korean Men: The Korea Medical Institute Study

  • Objectives: Cigarette smoking is a modifiable risk factor for cardiovascular disease. Bilirubin is a potent antioxidant and its concentration decreases in smokers. However, studies about the association between cigarette smoking and bilirubin are scarce and most are limited to total bilirubin. Additionally, bilirubin is highly related to hemoglobin. Therefore, this study evaluates the association between bilirubin subtypes and cigarette smoking in healthy Korean men independently of hemoglobin. Methods: This study included 48 040 Korean men aged 30 to 87 years who visited the Korea Medical Institute for routine health examinations from January to December, 2007. The association of smoking with total, direct, and indirect bilirubin was assessed by logistic regression analysis taking into consideration differences in subjects and smoking characteristics. Results: Current smokers had lower bilirubin concentrations than never-smokers and ex-smokers. Smoking amount and duration were inversely significantly associated with total, direct, and indirect bilirubin. In a multivariable adjusted model, compared to never-smokers, the odds ratios (ORs) and 95% confidence intervals (CIs) of current smokers with the highest number of pack-years were 1.7 (1.6 to 1.9) for total, 1.5 (1.4 to 1.6) for direct, and 1.7 (1.6 to 1.9) for indirect bilirubin. After further adjustment for hemoglobin, this association became stronger (OR [95% CI], 2.1 [1.9 to 2.2] for total; 1.9 [1.8 to 2.0] for direct; 2.0 [1.9 to 2.2] for indirect bilirubin). Conclusions: In this study, bilirubin subtypes are inversely associated with smoking status, smoking amount, and smoking duration in healthy Korean men independently of hemoglobin. Further studies are needed to investigate this association in healthy Korean women.

Electrochemical Behavior of Redox Proteins Immobilized on Nafion-Riboflavin Modified Gold Electrode

  • Electron transfer of a redox protein at a bare gold electrode is too slow to observe the redox peaks. A novel Nafion-riboflavin functional membrane was constructed during this study and electron transfer of cytochrome c, superoxide dismutase, and hemoglobin were carried out on the functional membrane-modified gold electrode with good stability and repeatability. The immobilized protein-modified electrodes showed quasireversible electrochemical redox behaviors with formal potentials of 0.150, 0.175, and 0.202 V versus Ag/AgCl for the cytochrome c, superoxide dismutase and hemoglobin, respectively. Whole experiment was carried out in the 50 mM MOPS buffer solution with pH 6.0 at 25 oC. For the immobilized protein, the cathodic transfer coefficients were 0.67, 0.68 and 0.67 and electron transfer-rate constants were evaluated to be 2.25, 2.23 and 2.5 s?1, respectively. Hydrogen peroxide concentration was measured by the peroxidase activity of hemoglobin and our experiment revealed that the enzyme was fully functional while immobilized on the Nafion-riboflavin membrane.

부항시술 후 나타난 색소반응에 따른 혈액성분 분석 (Analysis of Blood Components in Skin Color Reaction after Cupping Glass Therapy)

  • Purpose: The purpose of this study was to examine the relationship between pigmental reaction and blood components, such as white blood cell (WBC), lymphocyte, monocyte, red blood cell (RBC), hemoglobin, mean corpuscular index (mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC)), hematocrit, platelet and neutrophil segment, after Cupping Glass Therapy (CGT). Subjects: Twenty-five healthy adults participated in this study; Methods: Subjects had undergone CGT for 5 days and were divided into two groups (dark color (DCG) and light color (LCG)) depending the level of pigmental reaction. Blood was collected from each subject at the beginning of this study and recollected after 5 day s'CGT. Results: The percentage of lymphocytes was higher in LCG than in DCG after CGT (p<.01) and was significantly increased in LCD after CGT (p<.01), The number of RBC was more in DCG than in LCG both before (p<.01) and after CGT (p<.05). The amount of hemoglobin was more in DCG than in LCG both before (p<.01) and after CGT (p<.01) and was significantly increased in LCD after CGT (p<.01). Mean corpuscular index showed that both MCH (p<.05) and MCHC (p<.01) were higher in DCG than in LCG after CGT, but only MCHC before CGT (p<.01). It also showed either decreased MCV (p<.01) or increased MCHC in LCD after CGT (p<.01). Hematocrite was higher in DCG than in LCG both before (p<.01) and after CGT (p<.01). The percentage of neutrophil segments was higher in BCG than In LCG after CGT (p<.01) and was significantly decreased in LCD after CGT (p<.05). However, neither the number of WBC and platelets nor the percentage of monocytes was significantly different between DCG and LCG either before or after CGT. Conclusion: Pigmental reaction was significantly related to the changes of blood conponents after CGT. The results of this study suggest that CGT may have an effect on the components of blood cells.

임신성 고혈압 산모의 태아의 혈액상 (Hematologic Status of Newborn Infants of Mother with Pregancy-induced Hypertension)

  • 1993년 9월 1일부터 1994년 9월 30일 까지 영남대학교 의과대학 부속병원 산부인과에서 진단, 분만한 35례의 임신성 고혈압 산모와 37례의 정상 만삭임선 산모의 혈액소견과 그 선생아의 혈액소견 및 혈청 철 상태를 조사하여 다음과 같은 결과를 얻었다. 임신성 고혈압 임부에서 혈색소의 증가가 있었다. 임신성 고혈압 및 정산 만삭 임부의 혈색소 농도와 그 신생아의 혈청 철 상태사이에는 유의한 상관관계가 없었다. 임신성 고혈압 임산부의 신생아와 정상 만삭 임산부의 신생아 제대혈액내의 혈색소 농도 및 hematocrit치는 서로 유의한 차이가 없었다. 임신성 고혈압 임산부의 신생아는 정상 만삭 임산부의 신생아에 비해 제대혈청 철이 다소 증가하고 ferritin이 감소하는 양상을 보였으나 유의한 차이는 없었다. 임신성 고혈압 임산부의 선생아는 정상 만삭 임산부의 신생아에 비해 총 철 결합능 및 불포화철 결합능이 현저히 증가되어 있었다. 임신성 고혈압 임산부의 신생아는 잠재적인 철 결핍의 가능성이 있으며, 출생 후의 급속한 철분의 소모에 대비하여 철 결핍에 대한 주의깊은 관찰과 예방이 필요할 것으로 판단되었다.

Phenylhydrazine으로 유도한 용혈성 빈혈 흰쥐에 대한 녹용 추출물의 조혈 효과 (Hemopoietic Effect of Extracts from Four Parts of Deer Antler on Phenylhydrazine-Induced Hemolytic Anemia in Female Rats)

  • 본 연구는 부위별 녹용 추출물이 PHZ 투여로 용혈성 빈혈을 유도한 흰쥐의 조혈작용에 대한 효과를 검증하고자 하였다. 4주령의 암컷 Sprague-Dawley 흰쥐에 PHZ 10 mg/kg을 4일간 미정맥 주사하여 용혈성 빈혈을 유발한 후 각 부위별 녹용 추출물 200 mg/kg을 일주일간 경구투여 하였다. 용혈성 빈혈 유발군인 PHZ군은 hemoglobin, hematocrit치, 적혈구수가 대조군에 비해 유의적으로 감소하였고, 망상적 혈구수는 유의적으로 증가하였다. 반면, 녹용 추출물 군에서 는 hemoglobin, hematocrit치, 적혈구수가 PHZ 투여군보다 증가하였다. 특히 분골, 상대, 중대 추출물 투여 시 유의성있게 증가하였다. 상대 추출물 투여군에서는 망상적혈구수 증가가 유의적으로 억제되었다. PHZ 투여로 증가된 혈청 EPO 함량은 녹용 추출물 투여군에서 모두 정상적으로 회복 되었다. Hemoglobin 합성에 관여하는 $\delta$-ALAD 활성은 녹용 추출물 투여군에서 활성이 증가되는 결과를 보여주었다. 따라서 녹용 추출물은 PHZ으로 유도한 용혈성 빈혈에 대한 바람직한 조혈효과가 있는 기능성 물질로 사료된다.

산란계에 있어 델타-아미노레블린산의 급여가 생산성 및 계란 품질에 미치는 영향 (Effects of Dietary Delta-Aminolevulinic Acid on Egg Production and Egg Quality in Laying Hens)

  • 본 연구는 델타-이미노레블린산(ALA)의 급여가 산란계의 생산성 및 계란 품질에 미치는 영향에 대해 알아보기 위하여 시험을 실시하였다. 사양 시험은 30주령 ISA Brown 144수를 공시하였으며 28일간 실시하였다. 시험 설계는 1) CON (기초 사료), 2) 0.05(기초 사료 + ALA 0.05%) 3) 0.1(기초 사료 + ALA 0.1%) 및 4) 0.2(기초 사료 + ALA 0.2%)로 4개 처리를 하여 처리당 4반복, 반복당 12수씩 완전 임의 배치하였다. 전체 사양 시험 기간 동안, 델타-아미노레블린산의 첨가 수준이 증가함에 따라 산란율의 감소가 나타났으나, 0.2% 첨가 수준에서 다시 증가하는 경향을 나타내었다(quadratic effect, P=0.01). 난중은 델타-아미노레블린산의 첨가 수준이 증가함에 따라 무게가 증가하였다(linear effect, P=0.01; quadratic effect, P=0.01). 난황색은 전체 사양 시험 기간 동안, 0.1% 첨가 수준까지 난황색이 높아지다가 0.2% 첨가 수준에서 낮아지는 경향을 나타내었다(quadratic effect, P=0.03). 델타-아미노레블린산의 첨가 수준이 증가함에 따라 혈액 내 hemoglobin 농도가 유의적으로 증가하였다(linear effect, P= 0.01). 결론적으로, 산란계에 델타-아미노레블린산의 급여는 난중 및 난황색 그리고 혈액 내 hemoglobin 농도를 증가시키는 것으로 사료된다.

고농도 Heme-iron의 생산을 위한 Hemoglobin의 가수분해 및 분리 조건 (Conditions of Hemoglobin Hydrolysis and Separation for the Production of Enriched Heme-iron)

  • Hemoglobin(Hgb)의 효소 가수분해액으로부터 고농도 heme-iron의 분리에 영향을 주는 기질의 농도, 가수분해도 (DH, degree of hydrolysis) 및 분리 pH의 효과 등을 조사하였으며, pilot scale에서 고농도 heme-iron을 생산하였다. 최적 반응조건(pH 10.0, $50^{\circ}C$, 5 hr)에서의 Esperase에 의한 Hgb의 DH는 $24{\sim}25%$였다. Hgb의 농도는 heme-iron의 분리에 영향을 미쳐서 Hgb의 농도가 2.5%, 5.0%, 10.0%로 증가할수록 heme-iron/peptide(heme-iron의 분리도)는 각각 47.4%, 35.6%, 21.8%로 감소하였으며, 이에 따라 농축율도 감소하였다. Hgb의 가수분해액으로부터 고농도 heme-iron의 분리에 효율적인 pH 영역은 DH에 따라 달라졌으며, DH가 6.3%의 경우는 pH $5.0{\sim}6$에서 heme-iron/peptide의 비가 8.92%, DH가 14.5%인 경우는 pH $4.0{\sim}6.0$에서 $12.1{\sim}19.5%$였으며, DH가 24.3%인 경우는 pH $3.0{\sim}5.0$에서 $34.7{\sim}36.4%$로 가수분해도가 높을수록 고농도 heme-iron의 분리시 최적 PH 범위가 산성 영역으로 이동되었으며, 이 영역의 pH에서 peptide의 용해도 증가로 분리도가 증가되었다. 가수분해액으로부터 분리된 heme-iron의 분자량은 약 1 kDa정도로 hematin과 유사한 분리패턴을 보였다. Pilot scale로 제조한 고농도 heme-iron 제품에서의 heme-iron 함량은 27.1%, heme-iron/peptide 비는 38.7%, 생산수율은 9.3%이었다.
