Outward movement of hemoglobin and $K^+$ ion across rabbit erythrocyte membrane after heating, cooling and in acid medium was studied. One milliliter of rabbit blood was centrifuged and packed red cells were obtained. Packed red cells were resuspended by addition of 4 ml of 0.9% NaCl solution and were subjected to heating $(57^{\circ}C\;for\;5\;minutes)$ or cooling $(-4^{\circ}C{\sim}-8^{\circ}C\;of\;-10^{\circ}C{\sim}-11^{\circ}C\;for\;10\;minutes) $. For acid medium experiment packed ref cells were resuspended by addition of 4 ml of acid medium of PH 4.5 consisting of 0.01% glacial acetic acid in 0.85% NaCl solution and kept standing for 10 minutes. All red cell suspensions were centrifuged again and packed red cells were separated. This packed red cells were again suspended in 4 ml of NaCl solution of 0.8%, 0.7%, 0.6%, and 0.5% concentration respectively and kept standing for 20 minutes. The concentration of hemoglobin and $K^+$ in the supernatant of the above red cell suspensions were measured and the following results were obtained. 1. Outward movement of hemoglobin and $K^+$ was greatest in red cells subjected to heating. The movement paralled to the osmolal concentration gradient between extra- and intra-cellular phase of red cells. 2. In acid medium the outflux of hemoglobin and $K^+$ increased as compared to the control. 3. In red cells subjected to the cold of $-10^{\circ}C{\sim}-11^{\circ}C$ the outflux of hemoglobin and $K^+$ increased. Whereas in the environment of $-4^{\circ}C{\sim}-8^{\circ}C$ there was no change in the outflux of $K^+$. The he-moglobin outflux showed rather a decreased as compared to tile control.
Objectives: This study was performed in order to investigate the anti-hemolytic effect of Chunghyuljihwang-tang(CHJHT, Qingxuedihuang-tang) on phenylhydrazine-induced hemolytic anemia in Rats. Materials and Methods: Hemolytic anemia model of rats were induced by phenylhydrazine(PHZ, 40mg/kg/day, i.p.) for 3days. Male Sprague-Dawley rats were divided into three groups for their treatment. Normal group was injected normal saline for 3 days with distilled water(d.w.) orally, control group was injected PHZ for 3 days with d.w. and CHJHT group was injected PHZ for 3 days with administration of CHJHT for 10 days. RBC(Red blood cell), Hb(hemoglobin), mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, reticulocyte were determined on day 0, 4, 7, 10. Body weight was investigated on day 0, 2, 4, 6, 8, 10 and the weight of spleen was measured on day 10. Results: For CHJHT group, the levels of RBC and Hb were significantly higher than control group on day 4, and the concentration of hemoglobin on day 7 was increased more than control group. Moreover, reticulocyte was significantly decreased on day 10 more than control group. These results could be concluded that CHJHT suppressed the hemolytic anemia by PHZ and improved hematological values of anemia earlier.
This study was conducted to investigate changes in blood properties of high potassium (HK) phenotype Jindo dogs (15kg$\pm$2kg) after daily oral administration with water celery extracts (10 ml/kg) for 7 days. Blood samples were collected for three days in a row before administration of water celery extracts. After water celery extracts administration, blood samples were collected at 3h, 6h, 9h and then on daily basis until day 10 post administration (PA). At day 15, final sample was collected. Blood samples were analyzed on the basis of red blood cell (RBC), white blood cell (WBC), packed cell volume (PCV), hemoglobin (Hb) concentration, mean corpuscular volume(MCV), mean corpuscular hemoglobin concentration(MCHC), gluthathione concentration(GSH) and met-hemoglobin(Met-Hb) concentration. The significant changes (p<0.01, p<0.05) of RBCs were shown at 3 h to day 5, and days 7 and 9 after administration. PCV values were decreased form 3 h to day 10 after administration. Mean Hb concentration showed significant increase as 3 h to day 3, and day 6 to day 9 after administration. The significant changes (p<0.05) of WBCs were shown at 9 h and day 1 after administration. The increased numbers of MCV were detected at days 6 to 9 after administration (p<0.05, p<0.01). The significant changes of MCHC were shown at 9h and day 1 after administration. The significant increases (p<0.01, p<0.05) of GSH concentration were detected at days 1, 6 and 7 after administration. In Met-Hb concentration, the significant increases (p<0.05) occurred at only 9h and day 7 after administration, The significant increases (p<0.01, p<0.05) of reticulocyte were detected at days 2, 4, 5, 6 and 7. Data from blood samples collected at day 15 after administration showed that all of blood analysis results returned to normal level, compared to controls.
This study was designed to assess the iron nutritional status and growth development of children provided with and without the national school -lunch program(NLSP). The subjects consisted of 590 elementary school children (313 boys, 277 girls) in the 2nd, 4th and 6th grades provided with (n=390) and without (n=200) NSLP. anthropometric measurements were taken for body weight, height, triceps skinfold thickness and body fat percentage. Nutrient intakes were assessed by a modified 24 -hour recall method. Fastinig blood samples were obtained and analyzed for hemoglobin concentration, hematocrit, serum iron and total iron binding capacity. The results obtained are summarized as follows. No significant differences between children provided with and without NSLP were found in height and body weight, but triceps skinfold thickness and body fat(%) were significantly higher in children without NSLP than in those with NSLP. There was no significant difference in hemoglobin concentration and hematocrit between children provided with and without NSLP. However, serum iron concentration and transferrin saturation were higher in the children provided with NSLP(81.9$\mu\textrm{g}$/dl and 22.8%) than in the children without NSLP(73.1$\mu\textrm{g}$/dl and 20.9%). When comparing iron and vitamin C intakes , iron intake was significantly higher in children provided with school-lunch, but vitamin C intake was significantly higher in children provided without school-lunch. Percentages of iron -deficient anemia in underweight, normal and obese children when judged by total iron binding capacity were 14.9%, 12.5% and 25.8% respectively.
Common carp (Cyprinus carpio) (Weight 23.05 ± 6.98 g, Length 12.65 ± 1.38 cm) were exposed to waterborne nitrite at 0, 50, 100, 200, 400 and 800 mg NO2-/L for 96 hours. The lethal concentration 50 (LC50) of C. carpio exposed to waterborne nitrite was 398.6 mg NO2-/L. Hemoglobin, hematocrit and RBC count were significantly decreased by waterborne nitrite exposure. The MCV (mean mean corpuscular volume) (µl), MCH (mean corpuscular hemoglobin) (pg) and MCHC (mean corpuscular hemoglobin concentration) (%) were significantly increased. The inorganic component, plasma calcium, was significantly increased, and the organic components such as plasma cholesterol and total protein were significantly increased showing a similar tendency with calcium. In enzymatic components, the AST and ALT were also significantly increased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of C. carpio.
The effect of sodium n-dodecyl sulphate (SDS) on hemoglobin autoxidation was studied in the presence of a 100mM phosphate buffer (pH 7.0) by different methods. These included spectorphotometry, fluorescence technique, cyclic voltametry, differential scanning calorimetry, and densitometry. Spectroscopic studies showed that SDS concentrations up to 1 mM increased deoxy-, decreases oxy-, and had no significant effect on the met- conformation of hemoglobin. Therefore, a SDS concentration up to 1 mM increased the deoxy form of hemoglobin as the folded, compact state and decreases the oxy conformation. The turbidity measurements and differential scanning calorimetry techniques indicated a more stable conformation for hemoglobin in the presence of SDS up to 1mM. Electrochemical studies also confirmed a more difficult oxidation under these conditions. The induction of the deoxy form in the presence of SDS was confirmed by densitometry techniques. The compact structure of deoxyhemoglobin blocks the formation of met-conformation in low SDS concentrations.
This study has examined the removal of mixed soil of protein and fat by protease. Cotton and PET fabrics were soiled by spotting of hemoglobin and triolein, respectively. The soiling order and soil concentration were changed in this procedure. The soiled fabrics were aged at 130℃ for 20 minutes. Protease was added in the alcohol ethoxylate(AE) detergent solution. The removal effciency was evaluated by analysis of protein and/or fat on the fabrics before and after washing, respectively. The detergency of PET fabrics was higher than that of cotton fabrics. The removal efficiency of hemoglobin was improved by protease from cotton and PET fabrics. Especially the removal efficiency of hemoglobin was remarkably improved from cotton fabrics. With the increase of hemoglobin and triolein (1:1) mixed soil, the removal of mixed soil was increased in proportion to mixed soil content up to a certain point. but it began to decrease above the point from cotton fabrics, while it was generally increased from PET fabrics. The detergency of total mixed soil from cotton fabrics was higher in case of soiling order with triolein after hemoglobin than in case of soiling order with triolein before hemoglobin. But the soiling order was not greatly effected in the detergency of total mixed soil from PET fabrics.

To investigate the effects of strenuous physical exercise on commonly used hematological markers in subjects the intensive long running. Blood samples were obtained from nineteen participants in a 622 km ultra-marathon race before, 300 km and immediately after completion of the 622 km ultra-marathon. Samples were analyzed for total white cell count (WBC) and differential, total red cell count (RBC), hemoglobin, hematocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), red cell distribution width (RDW), platelets, mean platelets volume (MPV), platelets distribution width (PDW). Significant increases were found in WBC, neutrophil and platelets at 622 km compared to the pre-race. RBC, hemoglobin and hematocrit decreased statistically significantly the race at 300 km and 622 km compared to pre-race. A wide range of hematological perturbations occur during 622 km ultra-marathon running but it was physiological changes within a reference range. The 622 km ultra-marathon is less likely to cause clinically significant hematologic changes in athletes.
