Fractionation of Plasma Protein on the Several Fresh Water Fishes by Disc Electrophoresis (Disc 전기영동법에 의한 수종 담수어 혈장 단백질의 분획)

  • The plasma proteins of fresh water fishes have been fractionated by disc electrophoresis in acrylamide gels utilized as an electrophoretic supporting medium. The species of fishes examined in this experiment were Anguilla japonica, Misgurnus mizolepis, Parasilurus asotus, Siniperca scherzeri, Pelteobagrus fulvidra, Carassius carassius, Cyprinus carpio, and Hemibarbus labeo, obtained in the Han River. Disc electrophoresis was performed as described by Ornstein and Davis. Gels and buffer solution were prepared by the method developed by W.J.Kim. The separation gels were 7% acrylamide gel. The fractionation of plasma proteins showed 13 bands in Anguilla japonica, 10 in Misgurnus mizolepis, 15 in Parasilurus asotus, 12 in Siniperca scherzeri, 11 in Pelteobagrus fulvidra, 13 in Carassius carassius, 9 in Cyprinus carpio, and 13 in Hemibarbus labeo. The patterns of plasma protein on the each species of fishes were different in the number of bands, ratio of contents, relative mobilities, and forms of fractionation.

Age and Growth of Barbel Steed Hemibarbus labeo in Goe-san Lake in Korea

  • Age and growth of Hemibarbus labeo caught from Goe-san Lake in South Korea from March to November, 2011, were studied. A total of 201 specimens was collected, ranging from 110 to 580 mm in total length (TL). Males and females made up 47.9% and 52.1% of the sample, respectively. Marginal increment analyses showed that vertebral increments, each composed of one opaque and one hyaline zone, were deposited annually. Opaque edges were prevalent from June to July. The relationship between TL and vertebral radius was linear, with equations of R = 0.008TL - 0.208 (male) and R = 0.009TL - 0.272 (female). Regression equations between TL and total weight (TW) were $TW=9{\times}10^{-6}TL^{2.987}$ (male), $TW=8{\times}10^{-6}TL^{3.014}$ (female), and $TW=9{\times}10^{-6}TL^{2.988}$ (combined sexes), according to the von Bertalanffy growth equation. Back-calculated TL was expressed using the von Bertalanffy equation as follows: $L_t=438.25(1-e^{-0.175(t+0.164)})$ for males, $L_t=483.36(1-e^{-0.147(t+0.115)})$ for females, and $L_t=464.86(1-e^{-0.162(t+0.176)})$ for the sexes combined. The growth performances were 4.526, 4.536, and 4.544, respectively.

Development of Eggs, Larvae and Juveniles of the Hemibarbus labeo from Wicheon Stream, Nakdonggang-River (낙동강 수계 위천에 서식하는 누치 Hemibarbus labeo의 난발생 및 자치어 형태발달)

  • This study investigated the early life history of Hemibarbus labeo from in Wicheon, stream Nakdonggang-River, and compared their characteristics with closely related species. In April 2021, egg formation and development of autonomous fish were observed in fertilized eggs collected at four spawning sites. The size of the fertilized egg was 1.93~2.39 (average 2.22±0.15, n=30) mm. The water temperature was 22.2~24.1°C, and the hatching time took 109~115 hours. Newly after hatching, the total length of the yolk-sac larvae was 7.50~8.80 (average 7.99±0.46) mm, and the mouth and anus did not develop and had difficulty in being yolk. 6 days after hatching, the preflexion larvae were fed with a total length of 9.49~10.2 (9.78±0.23, n=30) mm. 10 days after hatching, the flexion larvae was 9.97~11.9 (10.7±0.72, n=30) mm in total length, and the tail of the vertebrae began to bend. 20 days after hatching, the postflexion larvae was 12.6~15.2 (13.9±0.77, n=30) mm in total length, and the tailbone was completely bent to 45°. 29 days after hatching, the total length of the juvenile was 16.9~19.8 (18.1±0.91, n=30) mm, and the number of fins reached an integer with 10 dorsal fins, 9 anal fins, and 7 ventral fins. The distribution of melanophore, such as the head, the center of the body, and the upper part of the fin, was different from that of the allied species during the postflexion larvae period, so the morphological characteristics could be distinguished.

Applicability of Next-Generation Sequencing for Analysis of Stomach Contents in Fish (Next-Generation Sequencing을 활용한 어류 위 내용물 분석의 적용 가능성)

  • The predator-prey interaction in freshwater ecosystems is a crucial area in the ecological study field and one of example to find such interaction is to investigate stomach contents. However, traditional method through visual inspection often induce misidentification, as it depends critically on intactness of physically visible data. In this study, we utilized Next-Generations Sequencing (NGS) technology to test the applicability stomach content analysis and overcome such limitation. NGS was applied to analyze the stomach contents of the Hemibarbus labeo, Tachysurus fulvidraco, and Plecoglossus altivelis collected in the lower part of Nakdong River. As a result, T. fulvidraco had a higher number of Animalia operational taxonomic units (OTUs) intake rate than H. labeo. At the same time, P. altivelis had higher number of Plantae OTUs intake rate than T. fulvidraco and higher Protozoa OTUs intake rate than H. labeo respectively. Therefore, NGS technology application enable to overcome traditional method's limitation and discover hidden interspecific interaction which can further be used in appropriate habitat assessment.

Fish Fauna and Weight-Length Relationships for 9 Fish Species in Andong Reservoir (안동호의 어류상과 9개 어종의 길이-무게 상관관계에 관한 연구)

  • The fish fauna and weight-length relationship in Andong reservoir were investigated from May 2005 to March 2006. During surveyed period, 2,540 individuals 20 species belonging to 7 families were collected. Dominant species were Hemibarbus labeo(19.6%) and sub-dominant species were Carassius cuvieri (19.2%). The korea endemic species were Hemiculter eigenmanni(7.1%), Squalidus chankaensis tsuchigae (0.1 %), Corroperca herzi (0.1 %) and introduced fishes were Carassius cuvieri (19.21 %) Lepomis macrochirus (4.2%), Micropterus salmoides (0.2%). Dominace, species diversity, species richness, species even-ness were 0.39, 3.16, 2.42, 0.73 respectively. The values of the exponent b in the weight-length relation-ship $W=aL^{b}$ ranged from 2.9162(H. labeo, $R^{2}$=0.9567) to 3.2509(Zacco pla쇼pus, $R^{2}$=0.9928) and the median value was 3.1079, whereas 50% of the values ranged between about 3.05 to 3.20. The Condition factor(K) showed good state of health in the surveyed species (excepts H. labeo and Cyprinus carpio).

Ichthyofauna and Fish Community Structure in Lake Chungju, Korea (충주호의 어류상과 어류군집)

  • The ichthyofauna and fish community structure were investigated at three areas in Lake Chungju from April to October 2004. During the surveyed period,32 two species belonging to 12 families were collected. Dominant species in number was hemibarbus labeo (relative abundance : 23.2%) and subdominant was Erythroculter erythropterus (12.1%). Siniperca scherzeri (9.2%), and Pseudogobio esocinus (8.6%) were also numerous. There were nine Korean endemic species belonging to six families including Acheilognathus yamatsutae. The number of fish species collected was 28 in Danyang, 17 in Jecheon, and 15 in Chungju. Highest dominant index calculated was 0.670 in Chungju, and highest diversity index calculated was 2.442 in the Danyang area. We need closer observation of population fluctuation of some species, including H. labeo, the dominant species by biomass in Lake Chungju.

Analysis of Vitellogenin Gene Expression by RT-PCR in Hemibarbus labeo (Cyprinidae) for the Analysis of Estrogenic Activity in Aquatic Environment (수환경 내 Estrogen 에스트로젠 활성 검출을 위한 누치 난황전구단백질 유전자 발현의 RT-PCR시험법)

  • In an effort to develop the biomarker for monitoring the contamination of xenoestrogen in the freshwater environment of Korea, reverse transcription-polymerasechain reaction (RT-PCR) analysis of vitellogenin (VTG) gene expression was optimized in Hearisarsus Iaseo, Based on the homology of the VTG cDNA sequences between the common carp and zebra fish, a set of PCR primers for VTG mRNA amplification for H; labo was designed. VTG mRNA level in livers from female and male fishes was analyzed by RT-PCR following single injection of 17 beta estradiol($E_2$ 10 mg $kg^{-1}$ B.W.). As an internal control, beta actin mRNA was amplified. One us of total liver RNA was subjected to RT-PCR. In female the amount of PCR productof VfC gradually increased in the range from 16 to 34 cycles of amplification. On the contrary, in control male, PCR product first detected at 32 cycles of amplification and linearly increased up to 40 cycles of amplification. In $E_2$ injected male liver, the VTC mRNA level was similar to that in the female. Taken together, this result suggests that liver of male H. labo expresses minute amount of VTG mRNA which are2-l6 equivalent of female and that induction of VTG mRNA occurs in male liver after estrogen treatment. In conclusion, the optimized protocol for RT-PCR analysis of VTG mRNA expression in liver of male H. labo will provide the environmental monitoring method for the xenoestrogen contamination in the rivers in Korea.

The Ichthyofauna and Fish Community in the Lower Course of the Imjin River (임진강 하류역의 어류상과 어류군집)

  • The Ichthyofauna and fish community were studied in the lower course of Imjin River from April 2003 to October 2004. During the survey period 80 species belonging to 23 families were collected. Twenty-five species (31.1%) of them were Korean endemic species. Exotic fishes were Carassius cuvieri, Lepomis macrochirus and Micropterus salmoides, and they complied 1.3% of total catch. The species of Hemibarbus mylodon was identified as natural monument, and acheilognathus signifer, Pseudopungtungia tenuicorpa, Gobiobotia macrocephala and Gobiobotia brevibarba were endangered species. Migration fish were Coilia ectens, Anguilla japonica, Plecoglossus altivelis and Takifugu obscurus. Dominant species were Zacco platypus (St. 1), Microphysogobio yaluensis (St. 2), Hemibarbus labeo (St. 3, 4) and coilia ectens (St. 5). The lower course of Imjin River was more abundance of species, high diversity and richness, and lower dominance index than those of the lower course of Han River.

Monitoring of Pathogens and Characteristics of Fish Community in the Taewha River (태화강의 어류군집에 대한 병원체 모니터링)

  • The pathogens and community structure of the fishes in Taehwa river were investigated from March 2007 to January 2009. During the study period, 3,504 individuals belonging to 35 species, 17 families and 9 orders were collected. The numerically dominant and subdominat species were Opsarichthys uncirostris (relative abundance 39.7%) and Hemibarbus labeo (relative abundance 30.9%). There were five Korean endemic species (20.8%) including Squalidus chankaensis tsuchigae, Zacco koreanus, Cobitis hankugensis, Coreoperca herzi, Odontobutis platycephala. The large fishes like Hemibarbus labeo or Opsarichthys uncirostris were gathered around the Samho bridge, sampling site 2 according to a season. The reaction to which two kinds of fish pathogenic virus is all negative and no fish pathogenic bacteria was isolated from 220 individuals. The fish pathogenic parasite not present variously with 7 species. Especially, Trichodina sp. was detected monthly and the infective density was high. But it is cosidered that temporary overcrowding of fish is not influenced mass mortality causing diseases in the specific site of river.

Ichthyofauna and Structure of the Fish Community in Hapcheon Lake on the Hwang River (합천호의 어류상과 어류 군집구조)

  • The ichthyofauna and structure of the fish community of Hapcheon Lake on the Hwang River were surveyed at six localities from May 2004 to Feb. 2005. There were 36 species belonging to 31 genera and 12 families in the Hapcheon Lake system. Of these, 24 species (66.7%) were cyprinids; cobitids and centropomids were next with 2 species (5.6%) each. The dominant species in the Hapcheon Lake system was Hemiculter eigenmanni (relative abundance 28.4%), the subdominant species Hemibarbus labeo (24.1%). Ten species (28.3%) of the 36 were Korean endemic species. Among the endemic species, H. eigenmanni and Squalidus chankaensis tsuchigae were abundant at each locality except site 6, but the eight other species were rare. In biomass, H. labeo comprised about 36.5% of the total weight, H. eigenmanni 22.3%, Cyprinus carpio 7.5%, Hypomesus nipponensis 6.9%, Culter brevicauda 3.7% and Lepomis macrochirus 3.6%. Average dominance, diversity, and evenness index of the fish community collected from six localities in the Hapcheon Lake system were 0.21, 0.85, and 0.67, respectively.
