Effect of $Cr^{6+}$ Stress on Photosynthetic Pigments and Certain Physiological Processes in the Cyanobacterium Anacystis nidulans and Its Chromium Resistant Strain

  • A MNNG (N-methyl-N'-nitro-N-nitrosoguanidine) induced chromium resistant strain ($Cr^{r}18$) of unicellular cyanobacterium Anacystis nidulans has been isolated and characterized. The resistant strain could grow (although restricted to $50\%$ of control) in chromium concentration (180${\mu}M$) lethal to the wild-type. Sublethal ($160{\mu}M$) concentration of $Cr^{6+}$ significantly reduced (13-$37.5$) all the photosynthetic pigments of A. nidulans with maximum reduction in phycoerythrin followed by ChI $\alpha$. Pigments of A. nidulans were drastically decreased in lethal concentration of Cr^{6+} with maximum reduction in phycoerythrin ($75\%$) and allophycocyanin ($67.5\%$). Resistant strain $Cr^{r}18$ resisted toxic effects of sublethal and lethal concentrations of $Cr^{6+}$ on photosynthetic pigments as revealed by less decrease in pigments as compared to A. nidulans. Effect of $Cr^{6+}$ stress was also studied on nitrogen assimilation and phosphate uptake. Sublethal concentration of $Cr^{6+}$ drastically reduced ($71.5\%$) nitrate uptake by A. nidulans while a decrease of $29\%$ was observed in strain $Cr^{r}18$. Short (2 day) exposure of A. nidulans and its resistant strain $Cr^{r}18\;to\;Cr^{6+}$ did not affect nitrate reductase and glutamine synthetase (transferase), whereas longer (10 day) exposure to $Cr^{6+}$ lowered activities of both enzymes in A. nidulans but not significantly in the strain $Cr^{r}18$. Ammonium uptake by both strains was not affected by $Cr^{6+}$. Thus, $Cr^{6+}$ affected photosynthetic pigments, nitrogen assimilation, and phosphate uptake of A. nidulans, while strain $Cr^{r}18$ was able to resist toxic effects of the metal. Advantages of using strain $Cr^{r}18$ for bioremediation purposes have been evaluated by studying $Cr^{6+}$ removal from the solution. Resistant strain $Cr^{r}18$ was able to remove $33\%$ more $Cr^{6+}$ than A. nidulans and thus it can prove to be a good candidate for bioremediation of $Cr^{6+}$ from polluted waters.

수은 노출에 대한 기수산 물벼룩 Diaphnosoma celebensis의 항산화 반응 (Antioxidant Responses in Brackish Water Flea Diaphanosoma celebensis - Exposed to Mercury)

  • 수은은 다양한 산업활동에 사용되어 해양 환경 내에 지속적으로 유입되며 먹이사슬을 따라 축적되며 생물체 내로 유입 시 해양 생물의 성장, 발생, 생식, 대사 등에 악영향을 미칠 수 있다. 본 연구에서는 기수산 물벼룩 Diaphanosoma clelbensis에 대한 수은의 급성 독성과 산화적 스트레스 지표(총 glutathione 함량, GST, GR, GPx 활성)를 이용한 항산화 반응을 조사하였다. 24시간 수은을 노출시킨 결과 생존율이 농도 의존적으로 감소하는 양상을 보였으며, 24시간 LC50 값은 0.589 mg/l (95% C.I. 0.521~0.655 mg/l)로 나타났다. 수은(0.08과 0.4 mg/l)을 24시간 노출시킨 D. celebensis에서 총 glutathione 함량은 유의하게 감소하는 양상을 보였으며, GST, GR, GPx 활성은 유의하게 증가하는 양상을 보였다. 이러한 결과는 수은 노출에 의해 D. celebensis에서 산화적 스트레스가 유도되었음을 의미하며, 이들 산화적 스트레스 지표의 변화는 세포내에서 방어기전으로 작용하였음을 나타낸다. 본 연구는 D. celebensis에서 수은 독성의 분자메커니즘을 이해하는데 도움이 되며, 중금속 오염된 해양 생태계 모니터링과 해양 생물의 건강성 평가에서 이들 분자지표의 활용 가능성을 제시한다.

카타마란 알루미늄 보강판의 열영향부 효과를 고려한 최종강도 붕괴 해석 (Collapse Analysis of Ultimate Strength Considering the Heat Affected Zone of an Aluminum Stiffened Plate in a Catamaran)

  • 선박 및 해양구조물에서 사용하고 있는 고강도 알루미늄 합금들은 스틸과 비교해서 많은 이점을 가지고 있다. 최근 고강도 알루미늄 합금들은 육상 및 해양에 폭넓게 사용되고 있으며, 특히, 특수목적 선박의 선체 외판구조에 많이 이용되고 있고, 교량 구조물에 사용되는 상자 구조물, 그리고 고정식 해양플랫폼의 상부구조에서 소비율이 증가하고 있다. 알루미늄 재료는 스틸보다 1/3의 중량 구성비를 통하여, 구성 중량을 줄이게 하여 연비 절감을 가능하게 한다. 일반적인 강구조물의 응력-변형률 관계와 비교하면, 용접가공에 따라 발생하는 열영향부의 존재로 인하여 상당히 다르게 나타난다. 왜냐하면, 강구조물과 비교하면 열전도율이 높아서, 열영향부(heat affected zone, HAZ)가 남아 있어 구조 강도 저하를 가져온다. 본 논문에서는 MIG(Metal inert gas) 용접 때문에 발생하는 열영향부를 고려하고, 종방향 압축 하중에 대한 알루미늄 보강판의 좌굴 및 최종강도 특성을 분석하였다. MIG 용접에 따른 열영향부를 고려한 경우, 좌굴 및 최종강도 모두 감소하며, 열영향부의 범위가 15 mm부터 항복 이후 에너지 소실률이 크게 나타나며, 25 mm 이상부터는 그 차이가 크지 않다. 따라서, 알루미늄 합금재료를 적용한 보강판의 구조 거동을 파악하기 위해서는 열영향부 영향에 대한 검토 및 분석이 중요하다.

자생 수생식물 노랑꽃창포와 창포의 카드뮴 축적 및 내성 (Cadmium Accumulation and Tolerance of Iris pseudacorus and Acorus calamus as Aquatic Plants Native to Korea)

  • 국내 자생 수생식물인 노랑꽃창포와 창포를 수질 내 카드뮴(Cd) 제거용 식물재료로 이용하고자 Cd 축적과 내성을 조사하였다. Cd 농도에 따른 치사율을 근거로 Cd에 대한 반수치사량($LD_{50}$)을 계산한 결과, 노랑꽃창포는 $78.5{\mu}M$, 창포는 $47.6{\mu}M$로 나타났다. Cd 스트레스에 대해 노랑꽃창포에서는 POD와 SOD, 창포에서는 CAT와 SOD, 폴리페놀이 활성화되었다. 노랑꽃창포에서는 폴리페놀이 측정되지 않았다. 노랑꽃창포와 창포 모두 Cd 처리 농도가 증가하고 기간이 길어질수록 식물체 내 Cd 축적량이 증가하였고, 두 종 모두 지상부보다는 지하부 축적량이 현저하게 많았다. 지상부에 축적된 Cd 함량은 노랑꽃창포 $548.1mg{\cdot}kg^{-1}$(지하부 축적 대비 82.1%) 창포 $121.4mg{\cdot}kg^{-1}$(지하부 축적 대비 13.7%)로 두 종 모두 Cd 고축적식물로 확인되었으며, 특히 노랑꽃 창포가 Cd 축적능력과 내성이 우수하였다.

부자사심탕(附子瀉心湯)이 산화적 손상, 염증 및 골관절염 병태모델에 미치는 영향 (Effects of Bujasasim-tang Ethanol Extract on Oxidative Stress, Inflammation and Osteoarthritic Rat Model)

  • Objectives This study was performed to investigate the effects of Bujasasim-tang ethanol extract (BST) on oxidative stress, inflammation and osteoarthritic rat model. Methods To ensure safety of BST, heavy metal levels were measured and cytotoxicity test was done. In vitro, To evaluate antioxidative effects of BST, total phenolic contents, 1,1-diphenyl-2-picryl-hydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) scavenging activity, reactive oxygen species (ROS) levels were measured. Also, to evaluate anti-inflammatory effects of BST treated group, total nitric oxide (NO) and pro-inflammatory cytokines (IL-$1{\beta}$, IL-6, TNF-${\alpha}$) levels were measured in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. In vivo, We injected MIA $50{\mu}l$ (60 mg/ml) into knee joints of rats to induce osteoarthritis. Rats were divided into total 3 groups (normal, control, BST treated group, each n=7). Normal group was not treated at all without inducing osteoarthritis and taken normal diet. Control group was induced osteoarthritis by MIA and taken with 2 ml of distilled water once a day for 4 weeks. BST treated group was induced osteoarthritis by MIA and taken BST 2 ml (200 mg/kg/mouse) once a day for 4 weeks. We evaluated dynamic weight bearing with the Incapacitance Test Meter. At the end of experiment, the rats were sacrificed to observe the functions of liver and kidney, changes of WBC, neutrophil, lymphocyte, monocyte levels in blood, to evaluate the levels of pro-inflammatory cytokines, tissue inhibitor of metallopreteinases-1 (TIMP-1), matrix metalloproteinase-9 (MMP-9), prostaglandin $E_2$ ($PGE_2$), leukotriene $B_4$ ($LTB_4$) within serum. We observed change of articular structures by Hematoxylin & Eosin (H&E), safranin-O staining method and measured amount of cartilage by micro CT-arthrography. Statistical analysis was done by unpaired student's t-test with significance level at p<0.05 in SPSS 11.0 for windows. Results 1. Safety of the BST was identified. 2. AST, ALT, BUN, creatinine levels of BST treated group were within normal limit. In vitro, 1. DPPH and ABTS free radical scavenging activities of BST showed dose-dependent increase. 2. ROS production were significantly decreased. 3. Total nitric oxide (NO) and IL-$1{\beta}$ production were decreased. 4. IL-6 and TNF-${\alpha}$ production were significantly decreased. In vivo, 1. Weight bearing ability was significantly increased. 2. WBC, neutrophil, lymphocyte, monocyte levels in blood were decreased. 3. IL-$1{\beta}$ and TNF-${\alpha}$ levels in serum were significantly decreased. and the IL-6 level was decreased. 4. TIMP-1, MMP-9, $LTB_4$, $PGE_2$ levels in serum were significantly decreased. 5. Cartilage volume of BST treated group was significantly increased. Also changes of cartilage, synovial membrane, fibrous tissue were suppressed. Conclusions The results obtained in this study Bujasasim-tang have effects of antioxidative, anti-inflammatory, relieve pain and protection of cartilage. Therefore we expect that Bujasasim-tang is effective treatment for osteoarthritis.

파킨슨씨병 유발물질인 황산철로 손상된 배양 신경아교세포에 대한 물매화 추출물의 항산화 효과 (Antioxidative Effects of Parnassia palustris L. Extract on Ferrous Sulfate-Induced Cellular Injury of Cultured C6 Glioma Cells)

  • 본 연구는 배양 C6 glioma 세포를 재료로 파킨슨병의 유발 물질인 황산철(FeSO4)의 신경독성과 이에 대한 물매화(Parnassia palustria L., PP) 추출물의 영향을 조사하였다. 이를 위하여 세포생존율을 비롯한 슈퍼뮤타제(SOD)-유사능 및 과산소음이온 라디칼(SAR)-소거능과 같은 항산화 효과를 분석하였다. 본 연구에서 FeSO4는 처리농도에 비례하여 세포생존율을 유의하게 감소시켰으며, 이 과정에서 XTT50값이 63.3 μM로 나타나 Borenfreund and Puerner의 독성판정기준에 따라 고독성 (highly-toxic)인 것으로 나타났다. 또한, 항산화제의 일종인 quercetin은 FeSO4의 독성으로 손상된 세포생존율을 유의하게 증가시켰다. 한편, PP 추출물은 FeSO4만을 처리한 것에 비하여 세포생존율을 유의하게 증가시켰으며, 동시에 SOD-유사 능과 SAR-소거능과 같은 항산화능을 나타냈다. 이상의 결과로부터, FeSO4의 세포독성에 산화적 손상이 관여하고 있으며, PP추출물은 항산화 효과에 의하여 FeSO4에 의한 세포독성을 효과적으로 방어하였다. 결론적으로, 물매화 추출물과 같은 천연물은 FeSO4와 같이 산화적 손상과 관련된 질환을 유발시키는 중금속화합물에 의한 독성을 개선 내지는 치료하는데 유용한 소재라고 생각된다.

자동차 루프랙의 형상에 따른 구조 해석을 통한 융합 연구 (A Convergence Study through the Structural Analysis due to the Shape of Automotive Roof Rack)

  • 최근 취미로 다양한 레포츠를 즐기는 인구가 증가하였다. 그에 따라 차량 지붕위에 다양한 물건들을 적재한 차량들을 거리에서 쉽게 볼 수가 있다. 차량 지붕위에 적재를 할 수 있게 하는 장치는 랙 이라는 장치이며 차량마다 각기 다른 형상을 가지고 있다. 다양한 종류들이 있지만 무거운 짐을 적재하기 위해 강도 및 내구성을 가져야 한다. 본 연구에서는 루프 랙의 지지대 방식과 고정대의 형상에 따른 구조 해석을 하였다. 세 가지의 모델들 중, Model C가 가장 좋은 내구성을 가지고 있음을 보였다. 따라서 어떤 형상을 가진 루프랙이 가장 안정성이 있는 것을 본 연구 결과로서 알 수 있다. 본 결과를 토대로 얻은 자동차 루프랙의 형상에 따른 구조 해석에 통한 융합 연구에 대한 설계데이터를 활용함으로서 실생활에서의 자동차 부품에 융합하여 그 미감을 보일 수 있다.

Benefit of the Drinking Water Supply System in Office Building by Rainwater Harvesting: A Demo Project in Hanoi, Vietnam

  • Vietnam is a developing country with the rate around 5%-6% per year, especially in urban areas. Rapidly developed urban areas lead to stress for infrastructure and the water supply is also stressed. In Hanoi city, total water capacity from the manufactories is around one million cubic meters per day and almost the entire main water source is groundwater but it is not enough to supply all of Hanoi's people, especially in the summer. A demo project is implemented in Hanoi University of Civil Engineering (HUCE) to produce drinking water by using the rainwater and membrane system and supply for people. In this project, rainwater is collected on the rooftop of the lecture building with an area of around $500m^2$ and $100m^3$ volumetric rainwater tanks. Afterwards, the rainwater is treated by the micro-membrane system and supplied to the tap water. Total cost for construction, technology and operation in the first year is around USD 48,558. In the long-term (15 yr) if HUCE invests in the same system, with $20m^3$ volumetric storage tank, it can provide drinking water for 500 staffs in every year. The cost of investment and operation for this system is lower than 30% compared to buying bottled water with the price USD 1.8/bottle. The drinking water parameters after treatment are pH, 7.3-7.75; turbidity, 0.6-0.8 NUT; total dissolved solids, 60-89 mg/L; coliform, 0; heavy metal similar with water quality in the bottle water in Vietnam.

Panax ginseng Extract as Protectant in Mercuric Chloride Induced Alterations in Protein Biochemistry in the Serum of Albino Rats

  • Adverse changes in individual's biochemistry under heavy metal stress are directly linked with its metabolic activity and health status. The present investigation highlights the differences in protecting role of Panax ginseng extract against mercuric chloride induced alterations in serum proteins. The assessment was based on dividing fifty albino rats into two sets, one for acute and the other for sub-acute study. All the sets had five groups with five albino rats in each i.e. control group, mercuric chloride treated group, Panax ginseng extract treated group, mercuric chloride followed by Panax ginseng extract treated group and Panax ginseng extract followed by mercuric chloride treated group. Mercuric chloride was given orally 0.926 mg/kg body weight for acute set and 0.044 mg/kg body weight for sub-acute set after LD50 (9.26 mg/kg body weight) determination by probitt analysis. 10 mg/kg body weight Panax ginseng extract was given in both acute and sub-acute sets after incorporating safety trials. The control group received tween-20 and distilled water only. The result exhibited significantly reduction (P<0.01) in serum protein, albumin and globulin following mercuric chloride intoxication whereas significant (P<0.01) enhancement in other groups with Panax ginseng extract as an ingredient confirming its protective role. All serum samples were also electrophoresed in 10% SDS with standard marker using discontinuous buffering system. Gradual disappearance of alpha-2 and beta-1 globulin bands from electrophoretic pattern was observed, while a single sharp band was observed between beta-2 and gamma globulin in serum protein pattern of acutely mercuric chloride treated rats. However, this band could not be visualized in sub-acute studies. Panax ginseng extract exhibits a better protection after acute intoxication.

Comparative and Interactive Biochemical Effects of Sub-Lethal Concentrations of Cadmium and Lead on Some Tissues of the African Catfish (Clarias gariepinus)

  • Cadmium is a strong toxic heavy metal which presents in paints and liquid wastes and causes oxidative stress in fish. On the other hand, lead is widely used for different purposes, e.g. lead pipes, it targets vital organs such as liver and kidney causing biochemical alterations. The present study evaluates the effects of 60 days exposure to Cd and Pb either single or combined together in African catfish. Sixty-four fishes were divided into 3 groups and exposed to $CdCl_2$ (7.02 mg/L) or $PbCl_2$ (69.3 mg/L) or a combination of them along with control group. Activities of acid phosphatase (ACP), lactate dehydrogenase (LDH) and glucose-6-phosphate dehydrogenase (G-6-PDH) were estimated. Moreover, gill, liver and kidney were assayed for activities of superoxide dismutase (SOD), catalase (CAT) and levels of glutathione (GSH) and malondialdehyde (MDA). Individual exposure showed that both Cd and Pb significantly decreased LDH activity and SOD activity in the kidney. Pb significantly increased G-6-PDH activity and decreased GSH level in the gill. CAT activity in liver and kidney elevated significantly on Cd exposure while lead caused a significant depletion in the liver and significant elevation in the kidney. Both Cd and Pb significantly increased MDA levels in liver and kidney while Pb increased its level in gills. The combined exposure resulted in normalization of LDH, G-6-PDH activity, and CAT activity in liver and kidney as well as GSH level in both tissues and MDA in gill and kidney. The combination increased SOD activity and MDA level in liver and decreased SOD activity in kidney and GSH level in gills. In conclusion, the antioxidant system of African catfish was adversely affected by prolonged exposure to Cd and Pb. The combined exposure caused less damage than individual exposure and returned most parameters to those of controls.
