Effect of Far-Infrared Irradiation and Heat Treatment on the Antioxidant Activity of Extracts from Peanut (Arachis hypogaea) Shell (땅콩껍질 추출물의 항산화능에 대한 원적외선과 열처리 효과)

  • The effects of far-infrared (FIR) irradiation and heat treatment on the antioxidant activity of extracts from peanut shells was evaluated. Peanut shells were placed in pyrex petri dishes (8.0cm diameter) and irradiated at $150^{\circ}C$ for 5, 10, 15, 20, 40 or 60min with a FIR heater. After FIR irradiation or simple heat treatment at same conditions, methanol extracts of peanut shells were prepared and total phenol contents (TPC), radical scavenging activities (RSA) and reducing powers of the extracts were determined. The antioxidant activities of the extracts increased as the time of heating or FIR-irradiation increased. When peanut shells were FIR­irradiated at $150^{\circ}C$ for 5 min, the values of TPC, RSA, and reducing power of the extracts increased from 40.17mg/mL to 42.30mg/mL, $67.7\%\;to\;76.3\%$, and 0.569 to 0.639, respectively, compared to the untreated controls. Simple heat treatment of peanut shell under the same conditions $(150^{\circ}C\;for\;5min)$ also increased the TPC, RSA, and reducing power of the extracts from 40.17mg/mL to 43.52mg/mL, 67.7\%\;to\;79.3\%$ and from 0.569 to 0.623, respectively. The results indicate that appropriate FIR-irradiation or heat treatment on peanut shells could increase the antioxidant activities of methanolic extracts.

  • Recently, the construction of onshore waste landfill sites has been studied due to the increase of waste and geosynthetics are widely utilized to enforce and protect waste landfill. Geosynthetics comprises the interface with soil and the seismic behavior and stability mostly depend on the dynamic shear behavior of the geosynthetic-soil interface. Therefore, the understanding of dynamic shear behavior and dynamic relative displacement of the interface is critical. The dynamic shear behavior of the interface is affected by surrounding conditions and loading and shows very complicated response, and, it is difficult to study theoretically. In this study, laboratory test to investigate dynamic relative displacement is performed under chemical condition. Dynamic interface apparatus is utilized and cyclic simple shear tests are conducted under short term (60 days of submerging period) and long term (840 days of submerging period) conditions. Consequently, relative displacement of the interface shows the largest values under acid condition, which means more severe damage of the interface.

Characteristics of Soft Soybean Curds prepared with the Ultra Fine Whole Soybean Flour and Proteinases (미세 전지 대두분말과 단백분해효소를 이용하여 제조한 연두부의 특성)

  • To prepare soybean curd utilizing effectively bioactive component of soybean, ultra fine whole soybean flour(UFWSF) was used as a soybean curd material for preparation of uncompressed whole soybean flour curd(UWSFC) in this study. The UWSFC was made with treatment by proteinase and coagulant, and proximate composition, color, textural properties and sensory evaluation of that were analyzed. Protease produced from Aspergillus sojae, bromelain and papain showed soybean curd coagulation ability and the pretense produced from Aspergillus sojae showed the highest soybean curd coagulation ability. When, after first heating and homogenizing, the proteinase was added to soybean milt textural properties of UWSFC were the best. Hunter's L and b values and textural properties (hardness, fracturability springiness, gumminess and chewiness) of UWSFC using proteinase and coagulant were higher than commercial soft soybean curd and adhesiveness and cohesiveness showed contrary tendency.

Changes in Free Sugars, Organic Acids and Fatty Acid Composition of Kanjang Prepared with Different Cooking Conditions of Whole Black Bean (원료콩의 증자조건을 달리한 검정콩 간장의 유리당, 유기산 및 지방산의 변화)

  • pH values in all samples were in the range of 5.04 to 5.32. pH of kanjang prepared with normal pressure(NPK) was higher than that of kanjang prepared with high pressure(HPK) and kanjang prepared with steam(SPK). Titratable acidity(TA) in all samples were found to be 4.26~7.72 mL. TA in HPK(5.99~6.96 mL) was lower than that in SPK(4.26~5.61 mL) and NPK(7.32 mL). Reducing sugar(RS) contents in all samples were in the range of 25.18 to 194.81 mg%. RS contents of HPK and SPK were a similar pattern and were higher than that of NPK. Major free sugars(FS) were glucose, fructose and xylose. In total FS, SPK was higher than that of NPK and HPK. Major organic acids(OA) were succinic acid, oxalic acid, tartaric acid, malic acid and citric acid. In total OA contents, SPK(40.38~76.96 mg%) was higher than that of NPK(55.85 mg%)and HPK(27.78~34.21mg%). OA contents in HPK and SPK were decreased slightly according to the heating time of whole black bean. The highest percentage of fatty acid (FA) in all samples was palmitic acid(33.28~43.51 % as area % of GC chromatogram), followed in a decreasing order by stearic acid(14.15~19.16%) and myristic acid(5.93~19.14%). The composition ratios of three FA to total FA ranged from 55.97 to 72.85&. In all samples, there was myristic acid that was not contained in raw soybean.

  • The purified $\beta$-galactosidase from L. sporogenes was most active at pH 7.0 and 6$0^{\circ}C$ with O-nitrophenyl-$\beta$-D-galactopyranoside (ONPG) in 0.05 M phosphate buffer. It was stable over a pH range from 5.0 to 9.0 and lost less than 10% of its activity after heating for 30 minutes at 6$0^{\circ}C$ and pH 7.0. All the mineral ions examined in this work showed no significant activating effect, whereas L-cysteine exerted a great stimnlatory effect on the enzyme activity at the concentration of 10 mM. The Km values were 1.2 mM for ONPG and 33.3 mM for lactose. Approximately 85% of lactose in cow's milk, in 10% skim milk and in 5% lactose solution was hydrolyzed after 4 hours incubation at 6$0^{\circ}C$ with 2 units of the purified $\beta$-galactosidase per $m\ell$ of the substrate solutions. The $\beta$-galactosidase from L. sporogenes, therefore, is considered to be suitable for hydrolysis of lactose in milk and other dairy products.

Effect of storage condition on the quality of the wine and Yakju made by Lycium chinense Miller (구기자술의 저장에 따른 성분변화)

  • Fruit of Lycium chinense Miller was known to Korean as traditional medicine that has effective components for strengtherning function of human body, especially liver. To study characteristics of the wine and Yakju made by fruit Lycium chinense Miller, general components such as organic acid, methanol, ester, fusel oil and $SO_2$ were analyzed after storing them for 10 and 20 days at $20^{\circ}C\;and\;30^{\circ}C$, respectively. Organic acid contents of the wine were mainly lactic, malic, tartaric and acetic acid. Ester and fusel oil contents of the wine were $4.0{\sim}5.0\;mg%$ and $12{\sim}30\;mg%$, respectively, and the values were smaller than those of the Yakju. Methanol content of the wine was $25{\sim}27\;mg%$. Storage temperature in terms of the quality of the wine and Yakju was better in $20^{\circ}C\;than\;30^{\circ}C$. Treatment of the wine and Yakju either by adding 100 ppm of $SO_2$ or heating at $60^{\circ}C$ for 30 min was better in quality than control for storage.

Optimization of Crack-Free Polytypoidally Joined Dissimilar Ceramics of Functionally Graded Material (FGM) Using 3-Dimensional Modeling (폴리타이포이드 경사 방식으로 접합 된 이종 세라믹간의 적층 수의 최적화 및 잔류응력 해석에 대한 연구)

  • Crack-free joining of $Si_3N_4\;and\;Al_2O_3$ using 15 layers has been achieved by a unique approach introducing Sialon polytypoids as a functionally graded materials (FGMs) bonding layer. In the past, hot press sintering of multilayered FGMs with 20 layers of thickness $500{\mu}m$ each has been fabricated successfully. In this study, the number of layers for FGM was reduced to 15 layers from 20 layers for optimization. For fabrication, model was hot pressed at 38 MPa while heating up to $1700^{\circ}$, and it was cooled at $2^{\circ}$/min to minimize residual stress during sintering. Initially, FGM with 15 layers had cracks near 90 wt.% 12H / 10 wt.% $Al_2O_3$ and 90 wt.% 12H/10 wt.% $Si_3N_4$ layers. To solve this problem, FEM (finite element method) program based on the maximum tensile stress theory was applied to design optimized FGM layers of crack free joint. The sample is 3-dimensional cylindrical shape where this has been transformed to 2-dimensional axisymmetric mode. Based on the simulation, crack-free FGM sample was obtained by designing axial, hoop and radial stresses less than tensile strength values across all the layers of FGM. Therefore, we were able to predict and prevent the damage by calculating its thermal stress using its elastic modulus and coefficient of thermal expansion. Such analyses are especially useful for FGM samples where the residual stresses are very difficult to measure experimentally.

Studies on the Use of Repeat Breeders as Donors for Embryo Transfer in Dairy Cow (젖소 수정란 이식에 있어서 공란우로서 저수태우의 이용에 관한 연구)

  • This study was carried out to examine the effects of seasonality, age, abnormalities of reproductive tract and the frequency of superovulatory treatments on the superovulatory responses and rate of pregnancy in repeat breeder for embryo transfer. Thirty eight eggs were obtained from 6 donors by flushing the uterine horns and 33 embryos were transfered to 33 recipients. (29 holstein heifers, 3 holstein cows and 1 Korean native heifer). From this study, the following results could be obtained: 1. It seemed that spring (Feb.-Apr.) was the better season than any others to produce good embryos. Rate of pregnancy decreased significantly after August because of low heating rate. 2. Rate of pregnancy in the cows with normal reproductive tract showed much better superovulatory responses (80%) than the cows with abnormal reproductive tract (33%), as expected. 3. 8 years old cows produced higher rater of pregnancy than 4 years old cow (79.3 vs 50), and also in superovulatory responses, 8 years old cows showed slightly better results than 4 years old cow. 4. The number of ovulated eggs and rate of pregnancy in the repeated superovulatory treatment more than once were 41 eggs and 85.7%, and those of the donors treated the first superovulatory treatment were 32 eggs and 68.4%, respectively. 5. These results are showing the possibility that utilizing the repeat breeders as donors in embryo transfer could be able to elevate their economic values.

  • Micro climatic conditions within the livestock facility are affected by various factors such as ventilation, cooling, heating, insulation and latent and sensible heat generation from animals. In this study, numerical BES method was used to simulate energy flow inside the poultry house. Based on the BES method and THI concept, degree of thermal stress of poultry was evaluated according to the locations in South Korea. Comparison of THI values within the poultry house was also carried out according to the stocking densities to reflect recent animal-welfare issue. Significant decrease in thermal stress of poultry was observed when the stocking density of $30kg/m^2$ was applied in the change of the seasons(p<0.05) however, there was no statistically significant difference in summer season(p>0.05). It meant that installation of proper cooling system is urgently needed. For Iksan city of Jeollabuk-do province, total 252 hours of profit for thermal stress was found according to decrease in the stocking density.

  • In this research, we describe the methodology and the quantification about GHG reduction effects, expected by optimization of operation mode according to establishing additional heat storage system of Bundang Combined Cycle Power Plant. As an intermediate form of General Combined Cycle Power Plant and Heat supply only district heating plant, Bundang Combined Cycle Power Plant(and Ilsan, Anyang, Bucheon) is possible to satisfy demand for the electrical load and thermal load capacity at the same time through changes to the operation mode itself. Therefore, through the operating transition of high-efficiency mode that the condenser cooling water is recovered and supplied to district heat and cooling, establishing additional heat storage system have flexible supply ability at the power and heat market. In this research, We calculated using the operating performance for the last three years(2008~2010) and efficiency of each mode-specific values. As a result, GHG reduction effects were calculated as $97.95kg_{-}CO_2/Gcal$ per heat energy 1 Gcal supplied at the heat storage system and we expected emmision reduction effect about $13,500Ton_{-}CO_2/yr$.
