Norovirus Quantification in Oysters Crassostrea gigas Collected from Tongyeoung, Korea (통영시 연안의 양식굴(Crassostrea gigas)에서 검출된 노로바이러스의 정량분석)

  • Norovirus (NoV) is a major cause of food poisoning outbreaks in Korea. Most NoV outbreaks originate from environmental contamination, but bivalves such as oysters are also important vectors. Oyster Crassostrea gigas contamination by NoV has been reported in Korea, but no quantitative analyses of NoV have been performed. We investigated the NoV concentration in 21 oyster samples from a Korean commercial oyster-growing area with confirmed fecal contamination from January to December 2012, using real-time reverse transcription-polymerase chain reaction. Additionally, we assessed the NoV concentration after heating to investigate the effects of heat treatment on NoV-infected oysters. In NoV-positive samples, the cycle threshold (Ct) values were 37.43-39.41 and 36.77-39.30, while viral concentrations were $8.97{\times}10^2-2.24{\times}10^2$ and $3.05{\times}10^2-7.47{\times}10^1$ copies/g for genogroups I and II, respectively. After heat treatment, NoV genogroup I decreased by 83.4%, 88.0%, 89.4% and 100% at $60^{\circ}C$, $68^{\circ}C$, $70^{\circ}C$, and $100^{\circ}C$, respectively, for 15 min, while genogroup II respectively decreased by 67.3%, 76.3%, 80.1%, and 89.8% under the same conditions.

Real Time Reverse Transcriptase-PCR to Detect Viable Enterobacteriaceae in Milk

  • This study was conducted to develop a real time reverse transcriptase-PCR (RT-PCR) method for the detection of viable Enterobacteriaceae in milk using primers based on the genes of ribosomal proteins S11 and S13 and to determine effects of heating and subsequent treatments on the threshold cycle (Ct) of the real time RT-PCR. Total RNA was isolated from 17 strains of bacteria including 11 strains of Enterobacteriaceae suspended in milk using a modified Tri reagent method. SYBR Green Master Mix was added to the RNA and the mixture was subjected to the real time RT-PCR. The Cts of eleven type strains of the Enterobacteriaceae in milk ($10^7$ cells) in the real time RT-PCR ranged from 21.5 to 24.6. However, the Cts of Pseudomonas fluorescens, Acinetobacter calcoaceticus, and three gram-positive bacteria were more than 40. The real time RT-PCR detected as low as $10^3$ cells in agarose gel electrophoresis. The Cts increased from 22.0 to 34.2 when milk samples contaminated with Escherichia coli ($10^7$ cells/mL) were heated at $65^{\circ}C$ for 30 min. In addition, subsequent incubation at $37^{\circ}C$ for 6 and 24 h increased the Cts further up to 36.2 and 37.2, respectively. Addition of RNase A to the bacterial suspension obtained from the heated milk and subsequent incubation at $37^{\circ}C$ for 1 h increased the Cts to more than 40. The results of this study suggests that pretreatment of bacterial cells heated in milk with RNase A before RNA extraction might enhance the ability to differentiate between viable and dead bacteria using real time RT-PCR.

Selective Separation of Hydrogen from Gas Mixture using LaNi5 (LaNi5를 이용한 혼합기체로부터 수소의 선택적 분리)

  • The selective separation of hydrogen from gas mixture containing hydrogen was experimentally studied using $LaNi_5$. The capacity and the rate of hydrogen separation, the purity of recovered hydrogen and the optimum condition of the regeneration of deactivated $LaNi_5$ were investigated. The separation rate and the recovery ratio of hydrogen were slowly decreased with the increase of the number of hydrogen absorption cycle. It was found that this result comes from the deactivation of $LaNi_5$ partly because of the blocking of hydrocarbon compounds in the $LaNi_5$ lattice and partly because of the poisoning of $LaNi_5$ surface by carbon monoxide contained in the gas mixture. The optimum condition for the regeneration of deactivated $LaNi_5$ was obtained by heating in a vacuum to about 637 K. The recovery ratio of hydrogen at the optimum condition was observed to be about 80%. The rates of hydrogen separation were measured in the ${\alpha}$-phase and two phase regions. The rate equations could be expressed as follows ; ${\alpha}$ - phase : $$-\frac{dP{_{H_2}}}{dt}=9.836{\times}10^{-3}(P{_{H_2}}_{-P_{eq}})$$ two phase region : $$-\frac{dP_{H{_2}}}{dt}=1.6909{\times}10^2\exp(-17560/RT)(P{_{H_2}}_{-P_{eq}})$$.

  • A pure oxygen combustion technology is crucial in Carbon Capture and Storage (CCS) technology especially in capturing of $CO_2$, where CCS will reduce 9 $GtCO_2$ by 2050, which is 19% of the total $CO_2$ reduction amount. To make pure oxygen combustion feasible, a regenerative system is required to enhance the efficiency of pure oxygen combustion system. However, an existing air combustion technology is not directly applicable due to the absence of nitrogen that occupies the 78% of air. This study, therefore, investigates the heat and fluid flow in a regenerative system for pure oxygen combustion by using commercial CFD software, FLUENT. Our regenerative system is composed of aluminium packed spheres. The effect of the amount of packed spheres in regenerator and the effect of presence or absence of a bypass of exhaust gas are investigated. The more thermal mass in regenerator makes the steady-state time longer and temperature variation between heating and regenerating cycle smaller. In the case of absence of bypass, the regenerator saturates because of enthalpy imbalance between exhaust gas and oxygen. We find that 40% of exhaust gas is to be bypassed to prevent the saturation of regenerator.

A Study on Effect of Intake Mixture Temperature upon Fuel Economy and Exhaust Emissions in Diesel Engines with a Scrubber EGR System

  • The effects of intake mixture temperature on performance and exhaust emissions under four kinds of engine loads were experimentally investigated by using a four-cycle. four-cylinder. swirl chamber type. water-cooled diesel engine with scrubber EGR system operating at three kinds of engine speeds. The purpose of this study is to develop the scrubber exhaust gas Recirculation (EGR) control system for reducing $\textrm{NO}_{x}$ and soot emissions simultaneously in diesel engines. The EGR system is used to reduce $\textrm{NO}_{x}$ emissions. And a novel diesel soot-removal device of cylinder-type scrubber with five water injection nozzles is specially designed and manufactured to reduce soot contents in the recirculated exhaust gas to the intake system of the engine. The influences of cooled EGR and water injection. however. would be included within those of scrubber EGR system. In order to survey the effects of cooled EGR and moisture on $\textrm{NO}_{x}$ and soot emissions. the intake mixtures of fresh air and recirculated exhaust gas are heated up using a heater with five heating coils equipped in a steel drum. It is found that intake and exhaust oxygen concentrations are decreased, especially at higher loads. as EGR rate and intake mixture temperature are increased at the same conditions of engine speed and load. and that $\textrm{NO}_{x}$ emissions are decreased. while soot emissions are increased owing to the decrease in intake and exhaust oxygen concentrations and the increase in equivalence ratio. Thus ond can conclude that $\textrm{NO}_{x}$ and soot emissions are considerably influenced by the cooled EGR.

Thermodynamic Characteristics Associated with Localized Torrential Rainfall Events in the Middle West Region of Korean Peninsula (한반도 중서부 국지성 집중호우와 관련된 열역학적 특성)

  • Thermodynamic conditions related with localized torrential rainfall in the middle west region of Korean peninsula are examined using radar rain rate and radiosonde observational data. Localized torrential rainfall events in this study are defined by three criteria base on 1) any one of Automated Synoptic Observing System (ASOS) hourly rainfall exceeds $30mmhr^{-1}$ around Osan, 2) the rain (> $1mmhr^{-1}$) area estimated from radar reflectivity is less than $20,000km^2$, and 3) the rain (> $10mmhr^{-1}$) cell is detected clearly and duration is short than 24 hr. As a result, 13 cases were selected during the summer season of 10 years (2004-13). It was found that the duration, the maximum rain area, and the maximum volumetric rain rate of convective cells (> $30mmhr^{-1}$) are less than 9hr, smaller than $1,000km^2$, and $15,000{\sim}60,000m^3s^{-1}$ in these cases. And a majority of cases shows the following thermodynamic characteristics: 1) Convective Available Potential Energy (CAPE) > $800Jkg^{-1}$, 2) Convective Inhibition (CIN) < $40Jkg^{-1}$, 3) Total Precipitable Water (TPW) ${\approx}$ 55 mm, and 4) Storm Relative Helicity (SRH) < $120m^2s^{-2}$. These cases mostly occurred in the afternoon. These thermodynamic conditions indicated that these cases were caused by strong atmospheric instability, lifting to overcome CIN, and sufficient moisture. The localized torrential rainfall occurred with deep moisture convection result from the instability caused by convective heating.

  • The purpose of this study is to assess economic effects for an advanced technology-based HVAC system. The study has been performed using LCC technique for the economic analysis. Data for LCC analysis are collected from estimation and interview of estimators and maintenance experts of buildings. Based on the LCC analysis, the economic effect of a new HVAC system has been predicted as follows : for the investment during 15 years of study period, (1) $21.6\%$ of LCC saving is predicted, (2) return rate for the investment is 4.8 times in case of new construction, and 14.4 times in case of building remodelling.

  • The main function of conventional insulation materials is only to block the heat transfer and reduce heat loss from the building. On the other hand, thermal storage materials can work as an energy saver by absorbing or emitting heat within a specific temperature range. Thermal storage materials for building can maintain a constant temperature by effectively regulating the cycle of indoor temperature. As a result, we can enhance the performance of a cooling and heating system efficiently. In this study, phase change materials (PCMs) were added as thermal storage materials into gypsum boards which are extensively used for building material and we found out the thermal environmental characteristics. In addition, we checked out some problems when applying the thermal storage materials to buildings. Finally, This study set out to examine the degree of environmental-friendly characteristics of thermal storage building materials by analyzing the amount of TVOC and HCHO contents with the possibility of pollutants emission.

  • The improvement of energy conservation mandates decrease consumption of fossil fuels and minimize negative impacts on the environment, which originates from large cooling and heating demand. The absorption heat pump technology has a large potential for energy saving in this respect. Adsorption heat pump is a means to upgrade waste heat without addition of extra thermal energy. The increase of absorbed amount is of great importance for absorption heat pump cycle. In this study, in order to improve the performance of absorber, the absorbers of two different types have been investigated using methanol-glycerine as a working fluid. The former was tangential feed of liquid phase without spiral tube in the absorber and the latter was with spiral tube in the absorber. The latter was found to be more effective in enhancing the mass and heat transfer to increase the absorption performance.

Study on the Effects of Flows on the Acceleration of the Grooving Corrosion in the ERW Pipe (ERW강관에서 홈부식의 가속화에 미치는 유동의 영향에 관한 연구)

  • The grooving corrosion is caused mainly by the different microstructures between the matrix and weld which is formed during the rapid heating and cooling cycle in welding. By this localized corrosion reaction of pipes, it evolves economic problems such as the early damage of industrial facilities and pipe lines of apartment, and water pollution. So lots of researches were carried out already about grooving corrosion mechanism of ERW carbon steel pipe but there is seldom study for water hammer happened by fluid phenomenon and corrosion rate by flow velocity. In this study, the analysis based on hydrodynamic and fracture mechanics was carried out. ANSYS, FLUENT and STAR-CD were used for confirmation of flow phenomenon and stress on the pipe. As the results, fatigue failure is able to be happened by water hammer and grooving corrosion rate is increased cause by turbulent. Grooving corrosion is happened on the pipe, then friction loss of fluid is occurred from corroded part. Erosion can be happened enough in corroded region of microscopic size that wear "V" form. Also pipe is able to be damaged by water hammer effects because of corroded region is general acting as a notch effects. Corrosion depth was more than half of total thickness, it can be damaged from water hammer pressure.
