Heat Shock Factor 1 Predicts Poor Prognosis of Gastric Cancer

  • Purpose: Heat shock factor 1 (HSF1) is a key regulator of the heat shock response and plays an important role in various cancers. However, the role of HSF1 in gastric cancer is still unknown. The present study evaluated the function of HSF1 and related mechanisms in gastric cancer. Materials and Methods: The expression levels of HSF1 in normal and gastric cancer tissues were compared using cDNA microarray data from the NCBI Gene Expression Omnibus (GEO) dataset. The proliferation of gastric cancer cells was analyzed using the WST assay. Transwell migration and invasion assays were used to evaluate the migration and invasion abilities of gastric cancer cells. Protein levels of HSF1 were analyzed using immunohistochemical staining of tissue microarrays from patients with gastric cancer. Results: HSF1 expression was significantly higher in gastric cancer tissue than in normal tissue. Knockdown of HSF1 reduced the proliferation, migration, and invasion of gastric cancer cells, while HSF1 overexpression promoted proliferation, migration, and invasion of gastric cancer cells. Furthermore, HSF1 promoted the proliferation of gastric cancer cells in vivo. In Kaplan-Meier analysis, high levels of HSF1 were associated with poor prognosis for patients with gastric cancer (p=0.028). Conclusion: HSF1 may be closely associated with the proliferation and motility of gastric cancer cells and poor prognosis of patients with gastric cancer. Accordingly, HSF1 could serve as a prognostic marker for gastric cancer.

Evidence of Interaction of Phage P22 Tailspike Protein with DnaJ During Translational Folding

  • Phage P22 tailspike is a thermostable homotrimeric protein, and temperature-sensitive folding (tsf) and global suppressor mutations affect its folding yields at elevated temperatures. We earlier suggested that the folding of the tailspike protein in Escherichia coli requires an unidentified molecular chaperone. Accordingly, in the present study, the interactions of purified DnaK, DnaJ, and GrpE heat-shock proteins with the tailspike protein were investigated during the translation and folding of the protein. The cotranslational addition of DnaJ to the tailspike protein resulted in the arrest of folding, when Dnak and GrpE were missing. However, the presence of DnaK, DnaJ, and GrpE had no effect on the folding yield of the tails pike protein, thus, providing evidence for the binding of the nascent tailspike protein with DnaJ protein, a member of DnaK chaperoning cycle.

T-and cross-reactive B-cell epitopes of Porphyromonas gingivalis and human heat shock protein 60 in atherosclerosis (동맥경화증에 있어서 Porphyromonas gingivalis와 인체 열충격단백의 T-세포 및 교차성 B-세포 epitope)

  • 본 연구의 목적은 인간의 동맥경화증에서 Porphyromonas gingivalis (P. gingivalis)와 인체 열충격단백 60의 T-세포 및 교차성 B-세포 epitope를 규명하고 수립된 T-세포주의 T-세포 주요조직적합체 양상을 파악하려는 데 있다. P. gingivalis 열충격단백-반응성 T 세포주와 환자의 혈청을 이용하여 P. gingivalis 열충격단백60 분자를 구성하는 104개의 중복성 합성 펩타이드의 T-세포 epitope과 B-세포 epitope을 규명하였다. 인체 열충격단백60에 대한 B-세포 epitope도 같은 방법으로 파악하였다. P. gingivalis, P. gingivalis 열충격단백60 또는 인체 열충격단백60에 대한 IgG 항체는 모든 동맥경화증 환자에서 상승하였다. P. gingivalis 열충격단백60의 3, 15, 24, 33, 45, 53, 64, 84, 88, 99번 펩타이드가 주요한 T-세포 epitope였고 이것들은 T-세포 및 B-세포 공동 epitope이기도 했다. 또한 인체 열충격단백60 교차반응 B-세포 epitope은 15, 29, 53, 56, 69, 74번 펩타이드로 판명되었다. 대부분 환자의 주요조직적합체는 $HLA-DRB1^{\ast}1504$$HLA-DZB1^{\ast}0603$으로 나타났다. 결론적으로 P. gingivalis 열충격단백60은 제 2급 주요조직적합제-제한적으로 분해되고 전달되었으며 이 단백질이 공통적인 T-세포 및 B-세포 epitope를 가지면서 동시에 인체 열충격단백60과 교차성 B-세포 epitope을 가지면서 동맥경화증의 면역조절기능에 관여한다고 볼 수 있다.

The Development of Acupuncture- Moxibustion Device for 4 points around GV20(Baihui) (백회와 사신총 혈위에 적합한 침구 소자의 개발)

  • Objectives This paper focused on developing the acupuncture-moxibustion device for head. Disadvantages of the existing devices for head moxibustion are its hume and inconvenience in operating because of the hair. Methods The proposed acupuncture-moxibustion device for GV20(Baihui) is 10mm long as needle body and 6mm long as needle root whose feature are 1~2 turns spiral coil. The spiral coil contains the moxa in order not to drop the moxa while operating Results The direct temperature on the burning moxa were measured by TK-305 infrared thermometer. But the direct temperature on the needle of acupuncture-moxibustion device were not able to measure by TK-305 infrared thermometer. By definition equation, the indirect temperatures on the needle of acupuncture-moxibustion device were able to be calculated. Conclusion A very safe, small and cheap acupuncture-moxibustion device for GV20(Baihui) of head is proposed. It will be useful to generate the heat shock protein and to operate in preventing and curing the Alzheimer's disease.

High sensitivity of embryonic stem cells to proteasome inhibitors correlates with low expression of heat shock protein and decrease of pluripotent cell marker expression

  • The ubiquitin-proteasome system is a major proteolytic system for nonlysosomal degradation of cellular proteins. Here, we investigated the response of mouse embryonic stem (ES) cells under proteotoxic stress. Proteasome inhibitors induced expression of heat shock protein 70 (HSP70) in a concentration- and time-dependent manner, and also induced apoptosis of ES cells. Importantly, more apoptotic cells were observed in ES cells compared with other somatic cells. To understand this phenomenon, we further investigated the expression of HSP70 and pluripotent cell markers. HSP70 expression was more significantly increased in somatic cells than in ES cells, and expression levels of pluripotent cell markers such as Oct4 and Nanog were decreased in ES cells. These results suggest that higher sensitivity of ES cells to proteotoxic stress may be related with lower capacity of HSP70 expression and decreased pluripotent cell marker expression, which is essential for the survival of ES cells.

Differentiation and upregulation of heat shock protein 70 induced by a subset of histone deacetylase inhibitors in mouse and human embryonic stem cells

  • Inhibiting histone deacetylase (HDAC) activity modulates the epigenetic status of cells, resulting in an alteration of gene expression and cellular function. Here, we investigated the effects of HDAC inhibitors on mouse embryonic stem (ES) cells. The HDAC inhibitors trichostatin A, suberoylanilide hydroxamic acid, sodium butyrate, and valproic acid induced early differentiation of mouse ES cells and triggered induction of heat-shock protein (HSP)70. In contrast, class III HDAC inhibitors failed to induce differentiation or HSP70 expression. Transcriptional upregulation of HSP70 was confirmed by mRNA expression analysis, an inhibitor study, and chromatin immunoprecipitation. HSP70 induction was dependent on the SAPK/JNK, p38, and PI3K/Akt pathways. Differentiation and induction of HSP70 by a subset of HDAC inhibitors was also examined in human ES cells, which suggests that the phenomenon generally occurs in ES cells. A better understanding of the effects of HDAC inhibitors may give more insight into their application in stem cell biology.

  • The hydrolysis of sphingomyelin (SM), known as the SM pathway, is induced by the activation of sphingomyelinase (SMase) to generate the second messenger ceramide, which plays a key role in cellular responses such as apoptosis, differentiation, senescence, and inflammation. Fluctuation of Temperature Induces Pathogenicity of Streptococcus iniae and Changes of Immunology Related Genes of Korean Rockfish, Sebastes schlegeli

  • This study was designed to examine the immune response in Korean rockfish during water temperature fluctuation and to elucidate the factors contributing to streptococcal pathogenesis in cultured Korean rockfish, S. schlegeli. We investigated cumulative mortality against Streptococcus iniae (FP5228 strain) infection in the exposed Korean rockfish (39.7±5.8 g) to environmentally relevant temperature (Control, 23℃; High temperature, 28℃ and 23℃ and 28℃ with 12 hours interval exchange, 23↔28℃) for 48 hours. Also, the expression of the mRNA related to the immune response genes (heat shock protein 70, interleukin1β, lysozyme g-type and thioredoxin-like 1) were measured in spleen and head kidney by real-time PCR analysis in the exposed fish to thermal stress. In this study, the combined stress with bacterial challenge in fishes exposed to thermal stress lowered the survival rate than that of control (23℃). The cumulative mortality in the group of control, 28℃ and 23↔28℃ was 24%, 24% and 40% (P<0.05), respectively. Also, thermal stress modulated the mRNA level of immune related genes; heat shock protein 70, interleukin-1β, lysozyme g-type and thioredoxin-like 1 in Korean rockfish. The present study indicates that a high and sudden water temperature change affect immune responses and reduce the disease resistance in Korean rockfish.
