Isolation and Characterization of Antioxidant Components in Epimedium koreanum NAKAI extract (음양곽 추출물 중의 항산화성분의 분리 및 성질)

  • The antioxidant activities of Epimedium koreanum N. methanol extract on the oxidation of fats and oils were studied by measuring peroxide values during storage at $75^{\circ}C$. The methanol extract showed high antioxidant activity on the oxidation of lard and corn oil, and the antioxidant effectiveness increased as the concentrations of the extract in lard increased. After heating the extract at $180^{\circ}C$ for 120 min, the heat-treated extract exhibited above 66% of the original antioxidant effectiveness on the oxidation of lard during storage at $75^{\circ}C$. Epimedium koreanum N. extract, however, accelerated the oxidation of soybean oil during storage under irradiation of ultraviolet light. The main antioxidant components in Epimedium koreanum N. extract seemed to be tocopherols and flavonoids. The contents of ${\alpha}$, ${\beta}$, ${\gamma}$ and ${\delta}-tocopherol$ in the extract were 21.61, 1.48, 5.50 and 3.75 mg%, respectively.

Effect of Ethanol Extract of Quercus mongolica Leaf as Natural Food Preservative (신갈나무 잎 에탄올 추출물의 식품보존제 효과)

  • This study was investigated to determine the antimicrobial effect of the ethanol extract of Quercus mongolica leaf on microbial growth. The ethanol extract at the concentration of $250\;\mu\textrm{g}/mL\;and\;500\;\mu\textrm{g}/mL$ inhibited the growth of gram positive and gram negative food-borne disease bacteria for 40 hours in tryptic soy broth, respectively. Antimicrobial activity of the ethanol extract from Quercus mongolica leaf was not affected by pH and heat treatment. The comparision between ethanol extract and commercially available preservatives on antimicrobial activity in food system was conducted. When the 0.1% ethanol extract of Quercus mongolica leaf was added to pine needle drink and carrot juice, antimicrobial activity was similar to those of containing 0.05% benzoic acid and 0.5% grapefruit seed extract. Also addition of 2~3% ethanol extract to the soybean paste inhibited the microbial growth up to 7 week, comparable to the inhibition of 2% ethanol. Thus, this results indicate that the ethanol extract of Quercus mongolica leaf may be useful as natural antimicrobial agents.

Inhibitory Effects of Water Extracy of Prunellae Spica on the Production of Pro-inflammatory Mediator in LPS-activated Raw 264.7 Cells (하고초물추출물이 LPS로 활성화된 Raw 264.7 cell에서의 염증매개물질 억제효과)

  • Prunellae Spica is the spike or whole plant of Prunella vulgaris Linne, which has been used for clearing heat from the liver, brightening the eyes and treating headache in traditional oriental medicines. This study was conducted to evaluate the inhibitory effects of the aqueous extract of Prunellae Spica (PSE; PS extract) on the production of NO and PGE2 in LPS-activated Raw 264.7 cells. Cell viability was determined by MTT assay, and all three doses of PS extract (0.03, 0.1 and 0.3 mg/ml) had no significant cytotoxicity during the entire experimental period. The cells were treated with 1 ${\mu}g/ml$ of LPS 1 h before adding PS extract, and increased NO and PGE2 production were detected in LPS-activated cells compared to control. However, these increases were dose-dependently attenuated by treatment with PS extract. The inhibition of NO by PS extract was due to the suppression of iNOS expression via inhibition of $NF{\kappa}B$ nuclear translocation and proteolytic degradation of $I{\kappa}B{\alpha}$. The decreased level of PGE2 was derived from inhibition of COX-2 activity, but expression of COX-2 protein was not affected by PS extract. Moreover, PS extract reduced the elevated production of IL-${\beta}$ and IL-6 by LPS. These results demonstrate that PS extract has inhibitory effects on the production of NO and PGE2 as a consequence of the reduction of proinflammatory cytokines, especially IL-${\beta}$ and IL-6 in LPS-activated Raw 264.7 cells.

Korean Red Ginseng (Panax ginseng Meyer) with enriched Rg3 ameliorates chronic intermittent heat stress-induced testicular damage in rats via multifunctional approach

  • Background: Panax ginseng Meyer, known as Korean Red Ginseng (KRG), is one of the important age-old traditional herbs used in boosting libido and improving male fertility. In this study, the effects of Rg3-enriched KRG extract (KGC04P) on heat stress-induced testicular damage in experimental rats was evaluated. Methods: Male rats (Sprague-Dawley) were divided into four groups (n = 10): normal control (NC), heat-stressed control (HC), heat-stressed plus KGC04P-100 mg/kg (HK100), and heat-stressed plus KGC04P-200 mg/kg (HK200) groups. Starting 1 week prior to heat stress, animals were administered orally with KGC04P (100 and 200 mg/kg) mixed with a regular pellet diet and continued for 25 weeks. Heat stress was induced to HC, HK100, and HK200 groups by intermittently exposing the animals to high temperatures ($32{\pm}1^{\circ}C$, 2 h/day). After 6 months, animals were euthanized under general anesthesia with carbon dioxide and evaluated for various parameters in serum and testicular tissue by using Western blotting, biochemical kits, and reverse transcription-polymerase chain reaction. Results: Significant (p < 0.05) alterations in several parameters, such as body/organ weight, sperm kinematics, and lipid metabolism marker levels, in the serum and testis of rats were observed. Further, the expression of testicular antioxidant enzymes, inflammatory cytokines, sex hormonal receptors, and spermatogenesis-related genes were also affected significantly (p < 0.05) in the heat-stressed group. However, KGC04P prevented the heat stress-induced changes in rats significantly (p < 0.05) at both concentrations. Conclusion: KGC04P attenuated heat stress-induced testicular damage by a multifunctional approach and can be developed as an excellent therapeutic agent for hyperthermia-mediated male infertility.

Inhibitory Effects of Sargassum thunbergii Ethanol Extract against α-amylase (지충이 에탄올 추출물의 α-amylase 저해활성)

  • This study was performed to investigate the inhibitory activity of Sargassum thunbergii (ST) against ${\alpha}$-amylase and elucidate the availability of ST extract as a functional food agent. To test the inhibitory activity of ST against ${\alpha}$-amylase, porcine pancreatic ${\alpha}$-amylase and potato starch were used as substrates. It was revealed that ST crude ethanol extracts have high ${\alpha}$-amylase inhibitory activity. Subsequently, ST crude ethanol extract was separated into five partition layers by solvent extraction: n-hexane, chloroform, ethyl acetate, butanol, and water. Chloroform and n-hexane fractions showed higher inhibitory activities than did acarbose (positive control). To confirm the changes in enzyme inhibitory activity by physical treatments, ST crude ethanol extract was subjected to heat, pH, and ${\gamma}$-irradiation treatments. In all heat treatments with the exception of one ($121^{\circ}C$, 15 min), the inhibitory activity was increased compared with the untreated group. With regard to pH stability, ST extract showed no significant changes at pH 4.6, but somewhat decreased inhibitory activity was revealed at pH 2, 8, and 10. On the other hand, ST ethanol extract was stable under ${\gamma}$-irradiation under all conditions (3.20 kGy). In summary, ST ethanol extract can be used in the food industry as a natural ${\alpha}$-amylase inhibitor.

Production of Brewer's Yeast Extract by Enzymatic Method (효소 분해법에 의한 맥주효모 추출물의 제조)

  • Cell lytic enzyme, 5'-phosphodiesterase, and AMP-deaminase were used to produce yeast extract as a natural seasoning from beer yeast cells. Prior to the addition of cell lytic enzyme, heat treatment was performed to increase the cell wall degradation` the optimum condition of the cell lytic enzyme was 50C at pH 7.0. The production yields by the enzymatic method and conventional autolysis method were 42% and 35%, respectively. The total quantity of 5'-nucleotides, GMP and IMP, produced by enzymatic method was increased by 45% than that by the conventional method. Futhermore, the operation time of enzymatic method was only 6.5 hrs, significantly reduced from 24 hrs of the conventional method.

Antimicrobial Activity and Characteristics of Amblytropis pauciflora Kitagawa Extract (털새동부 추출물의 항균효과 및 특성)

  • Methanol extract of roots of Amblytropis pauciflora Kitagawa showed the antimicrobial activity to three test strains. Antimicrobial spectra of various extracts of Amblytropis pauciflora Kitagawa were tested against 24 strains of bacteria and fungi. The crude methanol extract inhibited the growth of 12 strains of bacteria and Asp. fumigatus with the exception of yeasts. The properties of the antimicrobial substance were very stable under heat(at 12$0^{\circ}C$), acid(pH 3.0) and alkali(pH 11.0) treatment. Only the root harvested in spring showed the antimicrobial activity. Among the components extracted by butanol, ginseng saponin Rg1 and various saponin-like materials were detected by TLC analysis using a plate of silica gel 60F254. The antimicrobial compound was purified by methanol extraction, activated charcoal column chromatography, Sep-pak(C18) pretreatment and reverse phase HPLC. The purified compound was detected at 13.520 min as a single peak(about 98% purity) through the HPLC analysis.

  • Staibilization effect of Panax ginseng C. A. Meyer on .betha.-D-Galactosidase inactivation was proved by kinetic studies of thermal inactivation of the enzyme. The water extract Panax ginseng C. A. Meyer showed stabilization activity at minimal concentration of 10ppm. The methanolic extract was purified to obtain ginseng saponins, and two groups of the ginsenosides, i. e. protopanaxadiol and protopanaxatriol were isolated. They also showed a protective effect against the thermal and chemical inactivation of the enzyme; p-chloromercuribenzoic acid and hydroxylamine known as protein modifier greatly inactivated the enzyme but inactivation was significantly balocked by the ginseng component MG$^{2+}$, known as a cofactor, stabilized the enzyme and the poor stabilization effect by it was potentiated by ginseng components.s.

Assessment of Stability and Safety of Maskne Cosmetic

  • Wearing a mask is still advised since COVID-19 continues to spread. However, masks may also irritate the skin and cause mask acne, often known as "maskne", which is a type of acne mechanica caused by friction between the skin and clothing. Therefore, there is a need to develop an effective maskne cosmetic. In this study, we made the maskne cosmetics containing humulus lupulus extract and copper tripeptide-1 and investigated its stability and safety. To measure stability, a centrifugation test and heat-cool cycling were done, and changes in viscosity and pH were measured for 8 weeks. The Cumulative Irritation Test (CIT, WKIRB-202111-HR-096) was performed and positive reactions were determined by the ICDRG criteria. The results indicated that the samples were stable after centrifugation, temperature cycling, viscosity, and pH tests. In addition, cosmetic safety test results revealed that maskne cosmetics containing humulus lupulus extract and copper tripeptide-1 did not cause any skin responses. These findings indicate that prepared maskne cosmetics' stability and safety were comparable to those of currently available commercial cosmetics.

  • Public demand for the heat pump system as a next generation energy equipment is increasing for its eco-friendly and cost-effective advantage. Many researches have been concentrated on how to calculate and develop its own efficiency, while the possible effect of the heat pump operation on the whole subsurface temperature distribution is relatively less considered, During the current study, subsurface temperature disturbance caused by seasonal surface temperature cycle in Busan area and general W-tube heat pump operation is simulated in 3-dimensional heterogeneous medium. It shows that subsurface deeper than 10m from the surface remains nearly unchanged throughout the 4 seasons and groundwater convect ion in highly permeable layer near the surface acts like a main path of heat plume from heat pump system, This implies the significance of detail descript ion in shallow sedimentary layer or highly permeable layer which plays an important role on the regional flow advection and heat transfer. Also, the effect of groundwater convection increases when the arrangement of the 2 injection pipes and 2 extract ion well is maintained parallel to groundwater flow. Therefore, more careful and detail investigation is required before installation and operation of heat pump system that it may not cause any possible change of microbial ecosystem in the shallow subsurface environment or 'contamination of temperature' for groundwater use as well as the loss of efficiency of the equipment itself. This can also help to design the optimized grouting system for heat pump.
