아질산 노출이 흰다리새우, Litopenaeus vannamei의 생존율 및 독성 생리에 미치는 영향 (Effects of nitrite exposure on survival and physiology of white leg shrimp, Litopenaeus vannamei)

  • Juveniles of the white leg shrimp Litopenaeus vannamei (Weight 0.18±0.08 g) were exposed to nitrite-N at 0, 25, 50, 100, 200 and 400 mg/L for 72 hours, and the lethal concentration, heamolymph and genes regulation were evaluated. The lethal concentration 50 (LC50) of L. vannamei exposed to nitrite-N was 141.2 mg/L at 25℃ and 33 psu. In Total protein, total cholesterol, and BUN in heamolymph temporarily increased after the start of the experiment and then stabilized, but glucose, an indicator of stress, decreased over time in the entire experimental group, and creatines, an indicator of tissue damage, decreased with nitrite concentration until the first 12 hours. The genes of immune-related showed that masquerade-like serine proteinase(Mas) increased at 50 and 400 ppm for 24 hours, and then gradually decreased depending on concentration. In the case of prophenoloxidase, it was highest at 400 ppm for 40 hours, and other genes(Ras-related nuclear protein, Masquerade-like serine proteinase, proPO-activating enzyme) showed a response for 48 hours and then gradually decreased. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of L. vannamei.

Expression of Human Stem Cell Factor with Recombinant Baculovirus in BmN Cell Line and Silkworm

  • A recombinant transfer vector pBacSCF was constructed by inserting huamn stem cell factor (hSCF) cDNA into plasmid pBacPAK8. BmN cells were co-transfected with modified Bombyx mori, nuclear polyhedrosis virus (BmBacPAK) DNA and the recmbinant transfer vector to construct a recombinant baculovirus containing hSCE gene. DNA dot blotting and RNA dot blotting demonstrated that the hSCE gene was contained in the recombinant virus and transcribed. The recombinant baculovirus was infectious to BmN cells and to silkworm. SDS-PAGE analysis showed a specific band of expressed product in the extract of infected cells and in the heamolymph of infected larvae. Bioactivity of the recombinant hSCE was determined with W-1 cell line and MTT colorimetric method in synergy with interlukin-3 (IL-3). These results revealed that the hSCF gene was over-expressed in cultured cells and lavae of silkworm.

  • 누에의 성충특이체액단백질(adult specific protein : ASP)의 생리 생화학적 특성 및 성충화 발육에 따른 기능을 구명하기 위한 연구의 일환으로 ASP의 분리·정제 및 분자적 특성을 조사하였다. ASP는 Sephadex G-100 gel 여과 및 CM52 ion exchange chromatography에 의해 분리·정제되었으며, 정제된 단백질의 순도는 native-PAGE 및 면역전기영동에 의해 확인되었다. ASP는 분자량 19.5kDa 및 17.5kDa으로 추정되는 2종류의 subunits으로 구성되어 있고 또한 당 및 지질을 포함하지 않는 단순단백질임이 밝혀졌다.

Biology of a predatory bug Eocanthecona furcellata Wolff (Hemiptera : Pentatomidae) on Vapourer tussock moth larvae: a major pest of tasar silkworm food plants

  • Tropical Tasar culture is forest and agro-based activity that covers agricultural and industrial activity. Tasar silk is produced by the silkworm, Antheraea mylitta which is polyphagous in nature. A large number of pests are reported to attack both the host plant as well as silkworm. As the rearing is conducted outdoor silkworms are exposed to attack of various pests during their life cycle. Eocanthecona furcellata (Hemiptera: Pentatomidae) is the major predator of tasar silkworm during young age rearing. Both nymphs and adults of the stink bug suck the heamolymph of silkworm leading to death. The present work was undertaken to study the life cycle of E. furcellata on larvae of vapourer tussock moth which is a major pest of tasar host plants. The incubation period, nymphal duration and total developmental period from egg to adult was $8.0{\pm}0.45d$, $16.0{\pm}3.24$ and $22.0{\pm}4.20d$, respectively. Pre-oviposition, oviposition and longevity of male and female stink bug was $3.20{\pm}0.38$, $17.60{\pm}1.86$, $42.40{\pm}1.94$, $37.00{\pm}3.18d$, respectively. Analysis of the data revealed no significant difference in life cycle of the predator when provided silkworm and vapourer tussock moth larvae as hosts. Which clearly indicates that E. furcellata can multiply on vapourer tussock moth larvae during non rearing periods. Adoption of recommended pest control measures for defoliators of tasar host plants during non-rearing periods will help in reducing the incidence of host plant pests and also in reducing the incidence of stink bug during rearing season. During the study it was also observed that larvae semilooper, boll headed caterpillar and nymphs of leaf hopper serve as alternate/secondary hosts to E. furcellata during non rearing seasons.
