현미 종류별 발아현미의 발아 전 후 생리활성물질 함량의 변화 (Pre- and Post-Germination Changes in Pharmaceutical Compounds of Germinated Brown Rice)

  • 발아현미에 함유된 기능성 물질의 발아 전후 함량 변화를 평가하기 위하여 일반미, 녹미, 적미, 흑미, 찹쌀현미 등 5종의 현미를 대상으로 발아 전 상태 및 발아현미 제조 후 상태에서의 tocopherol 및 tocotrienol류, squalene 및 phytosterol 류 등 기능성물질 함량 변화를 평가한 본 실험의 결과를 요약하면 다음과 같다. 1. 비타민 E 함량은 발아전보다 발아후의 현미에서 약 10% 수준의 함량 감소가 관찰되었으나 통계적 유의성은 인정되지 않았다. 2. Squalene 함량의 경우 발아찹쌀현미는 발아 과정 중 31% 증가하는 경향이었으나 다른 종류의 현미는 발아 전후 큰 변화가 없었다. 3. Phytosterol류 중 stigmasterol은 모든 종류의 발아현미에서 발아 전 대비 5-27%의 뚜렷한 증가가 관찰되었으며, campesterol, sitosterol 및 총 phytosterol 함량은 발아 전후 간 대차 없거나 다소 감소하는 경향이었다. 4. 이상의 결과를 종합할 때, 비록 현미 종류별 차이는 있었으나 대부분의 경우 발아 현미는 발아전과 비교할 때 stigmasterol은 뚜렷한 증가를 나타내며 본 실험 분석 대상이었던 tocopherol, tocotrienol, squalene, campesterol, sitosterol 등 기능성 물질은 발아 전후 대차 없는 것으로 판단되었다.

Antioxidant and Tyrosinase Inhibitory Activities from Seed Coat of Brown Soybean

  • Soybeans with brown, black, and yellow seed coats were compared to total phenolic contents and antioxidant activities including 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radicals. Also, 3 seed coats were examined for inhibitory activities on tyrosinase and lipoxygenase-1 on the basis of spectrophotometric and polarographic methods. Among seed coat extracts, 80% methanol extract of brown soybean seed coat showed the highest total phenolic contents ($68.9{\pm}3.29\;mg$ GAE/g) as well as exhibited potent scavenging effects on the DPPH ($IC_{50}=4.3\;{\mu}g/mL$) and ABTS ($IC_{50}=3.7\;{\mu}g/mL$) radicals. In a polarographic experiment, this extract was potentially inhibited the oxidation of L-tyrosine and L-3,4-dihydroxy-phenylalanin (L-DOPA) catalyzed by mushroom tyrosinase with $IC_{50}$ values of 12.4 and $63.7\;{\mu}g/mL$, respectively. It was also detected inhibition of the tyrosinase catalyzed oxidation of L-DOPA with an $IC_{50}$ value of 120.3 mg/mL in UV spectrophotometric experiment. In addition, this extract inhibited the linoleic acid peroxidation catalyzed by lipoxygenase-1 with an $IC_{50}$ value of $4.0\;{\mu}g/mL$. These results suggest that brown soybean may possess more beneficial effect on human health than black and yellow soybeans.

Effectiveness of Aromatherapy with Light Thai Massage for Cellular Immunity Improvement in Colorectal Cancer Patients Receiving Chemotherapy

  • Background: Patients with colorectal cancer are usually treated with chemotherapy, which reduces the number of blood cells, especially white blood cells, and consequently increases the risk of infections. Some research studies have reported that aromatherapy massage affects the immune system and improves immune function by, for example, increasing the numbers of natural killer cells and peripheral blood lymphocytes. However, there has been no report of any study which provided good evidence as to whether aromatherapy with Thai massage could improve the immune system in patients with colorectal cancer. The objectives of this study were to determine whether the use of aromatherapy with light Thai massage in patients with colorectal cancer, who have received chemotherapy, can result in improvement of the cellular immunity and reduce the severity of the common symptoms of side effects. Materials and Methods: Sixty-six patients with colorectal cancer in Phichit Hospital, Thailand, were enrolled in a single-blind, randomised-controlled trial. The intervention consisted of three massage sessions with ginger and coconut oil over a 1-week period. The control group received standard supportive care only. Assessments were conducted at pre-assessment and at the end of one week of massage or standard care. Changes from pre-assessment to the end of treatment were measured in terms of white blood cells, neutrophils, lymphocytes, CD4 and CD8 cells and the CD4/CD8 ratio and also the severity of self-rated symptom scores. Results: The main finding was that after adjusting for pre-assessment values the mean lymphocyte count at the post-assessment was significantly higher (P=0.04) in the treatment group than in the controls. The size of this difference suggested that aromatherapy with Thai massage could boost lymphocyte numbers by 11%. The secondary outcomes were that at the post assessment the symptom severity scores for fatigue, presenting symptom, pain and stress were significantly lower in the massage group than in the standard care controls. Conclusions: Aromatherapy with light Thai massage can be beneficial for the immune systems of cancer patients who are undergoing chemotherapy by increasing the number of lymphocytes and can help to reduce the severity of common symptoms.

  • Recently, it was reported that enamel matrix derivative may be beneficial in periodontal regeneration procedures in expectation of promoting new bone and cementum formation. The aim of present study was to evaluate the effect of enamel matrix derivative($Emdogain^?$)and Caso4 sulfate paste in 1-wall intrabony defects in beagle dogs. Surgically created 1-wall intrabony defects were randomly assigned to receive root debridement alone or $Emdogain^{(R)}$ or $Emdogain^{(R)}$ and Caso4. Clinical defect size was 4 X 4mm. The control group was treated with root debridement alone,and Experimental group I was treated with enamel matrix derivative application, and Experimental group II was treated with enamel matrix derivative and Caso4 sulfate paste application,. The healing processes were histologically and histometrically observed after 8 weeks and the results were as follows: 1. The length of junctional epithelium was $0.41{\pm}0.01mm$ in the control group, $0.42{\pm}0.08mm$in the experimental group I and $0.50{\pm}0.13mm$in the experimental group II. 2. The connective tissue adhesion was $0.28{\pm}0.02mm$ in the control group, $0.13{\pm}0.08mm$ in the experimental group I and $0.19{\pm}0.02mm$ in the experimental group II. 3. The new cementum formation was $3.80{\pm}0.06mm$ in the control group, $4.12{\pm}0.43mm$ in the experimental group I and $4.34{\pm}0.71mm$ in the experimental group II. 4. The new bone formation was $1.43{\pm}0.03mm$ in the control group, $1.53{\pm}0.47mm$ in the experimental group I and $2.25{\pm}1.35mm$ in the experimental group II. Although there was limitation to present study, the use of enamel matrix derivative in the treatment of periodontal 1-wall intrabony defect enhanced new cementum and bone formation. Caso4 sulfate paste will be the candidate for carriers to deliver enamel matrix derivative, and so enhance the regenerative potency of enamel matrix derivative.

Branched-chain Amino Acids are Beneficial to Maintain Growth Performance and Intestinal Immune-related Function in Weaned Piglets Fed Protein Restricted Diet

  • As a novel approach for disease control and prevention, nutritional modulation of the intestinal health has been proved. However, It is still unknown whether branched-chain amino acid (BCAA) is needed to maintain intestinal immune-related function. The objective of this study was to determine whether BCAA supplementation in protein restricted diet affects growth performance, intestinal barrier function and modulates post-weaning gut disorders. One hundred and eight weaned piglets ($7.96{\pm}0.26kg$) were randomly fed one of the three diets including a control diet (21% crude protein [CP], CON), a protein restricted diet (17% CP, PR) and a BCAA diet (BCAA supplementation in the PR diet) for 14 d. The growth performance, plasma amino acid concentrations, small intestinal morphology and intestinal immunoglobulins were tested. First, average daily gain (ADG) (p<0.05) and average daily feed intake (ADFI) (p<0.05) of weaned pigs in PR group were lower, while gain:feed ratio was lower than the CON group (p<0.05). Compared with PR group, BCAA group improved ADG (p<0.05), ADFI (p<0.05) and feed:gain ratio (p<0.05) of piglets. The growth performance data between CON and BCAA groups was not different (p>0.05). The PR and BCAA treatments had a higher (p<0.05) plasma concentration of methionine and threonine than the CON treatment. The level of some essential and functional amino acids (such as arginine, phenylalanine, histidine, glutamine etc.) in plasma of the PR group was lower (p<0.05) than that of the CON group. Compared with CON group, BCAA supplementation significantly increased BCAA concentrations (p<0.01) and decreased urea concentration (p<0.01) in pig plasma indicating that the efficiency of dietary nitrogen utilization was increased. Compared with CON group, the small intestine of piglets fed PR diet showed villous atrophy, increasing of intra-epithelial lymphocytes (IELs) number (p<0.05) and declining of the immunoglobulin concentration, including jejunal immunoglobulin A (IgA) (p = 0.04), secreted IgA (sIgA) (p = 0.03) and immunoglobulin M (p = 0.08), and ileal IgA (p = 0.01) and immunoglobulin G (p = 0.08). The BCAA supplementation increased villous height in the duodenum (p<0.01), reversed the trend of an increasing IELs number. Notably, BCAA supplementation increased levels of jejunal and ileal immunoglobulin mentioned above. In conclusion, BCAA supplementation to protein restricted diet improved intestinal immune defense function by protecting villous morphology and by increasing levels of intestinal immunoglobulins in weaned piglets. Our finding has the important implication that BCAA may be used to reduce the negative effects of a protein restricted diet on growth performance and intestinal immunity in weaned piglets.

식물화합물 다량 함유 제주조릿대 잎 추출물의 제조와 특성 (Preparation and Characterization of Phytochemical-Rich Extract from Sasa quelpaertensis Leaf)

  • 조릿대 속 식물의 잎은 한의학적으로 염증, 해열, 이뇨 관련 질환 치료에 사용되어 왔다. 제주조릿대는 제주도 한라산 일대에서만 분포되어있는 토착식물이다. 본 연구에서는 제주조릿대 잎 추출물의 산업적 활용 효율을 높이기 위해 추출방법에 따른 식물화합물의 함량과 그 생리 활성을 비교함으로써 기능성이 강화된 식물화합물 다량함유 추출물(phytochemical-rich extract; PRE)의 제조방법을 개발하였다. PRE는 열수추출물과 알코올추출물에 비하여 높은 폴리페놀 함량을 나타내었고, 조릿대 주요성분인 p-coumaric acid (44.10 mg/g)와 tricin (5.35 mg/g)의 함량은 열추추출물(p-coumaric 23.39 mg/g, tricin 0.18 mg/g)과 알코올 추출물(p-coumaric 10.8 mg/g, tricin 0.38 mg/g)에 비해 매우 높았다. RPE는 다른 추출물에 비해 총 폴리페놀 함량과 항산화 활성[1,1-diphenyl-2-picrylhydrazy(DPPH) 소거활성, 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) 소거활성, nitric oxide(NO) 소거활성, xanthine oxidase 저해활성]이 가장 높았다. 그리고 RPE는 LPS로 자극시킨 RAW 264.7 세포에서 NO 생성($IC_{50}=59.1{\mu}g/ml$)과 혈액 암(HL-60) 세포의 성장을 효과적으로 억제하였다. 본 연구결과는 PRE가 식물화합물을 다량 함유하고 있어 항산화 및 항염소재로서 활용가치가 있음을 제시해 준다.

한국 된장에서 Equol의 검출 및 미생물 동정 (Bacterial Identification and Detection of Equol in Korean Soybean Paste)

  • 에쿠올은 인간의 건강에 유익한 효과를 나타낸다. 발효된 콩 식품들은 에쿠올을 함유하고 있으며, 많은 미생물들이 에쿠올 생산과정에 참여하는 것으로 밝혀졌다. 본 연구에서는 한국의 전통 발효 식품인 된장에 대해 조사하였다. 먼저 서로 다른 제조자로부터 수집 된 37개의 된장 샘플들을 대상으로 에쿠올의 농도를 측정하기 위해 LC-MS/MS를 시행하였다. 측정 결과 3개의 된장 샘플에서 에쿠올이 검출되었고, 507 ng/100 g의 농도가 가장 높게 나타났다. 에쿠올을 함유한 된장에서 15개의 미생물 종들이 16S rRNA gene sequence analysis와 2개의 MALDI-TOF MS분석법에 의해 분리, 동정되었으며 Bacillus spp, Paenibacillus spp, Tetragenococcus spp, Stapylococcus spp, and Clostridium species들이 가장 우세한 미생물들이었다. 이 연구결과로 한국의 전통 발효식품인 된장에서도 에쿠올이 검출되었음을 확인하였다.

해조성분 강화 기능성소금에 대한 연구 (Studies on Functional Salt Fortified with Seaweed Components)

  • 해조성분 강화 기능성소금 3종(Hizikia mineral salt, fucoidan salt, laver salt)을 제조하여 일반적 품질특성 및 무기성분 조성, 용해도, 관능특성 및 ACE 저해활성과 bile acid 결합력을 알아보았다. 수분함량은 Hizikia mineral salt의 경우 대조시료와 유의한 차이가 없었으나 fucoidan salt와 laver salt는 상대적으로 높은 수분함량을 나타내었다. 염도는 해조성분 강화 기능성소금 3종 모두 대조시료 보다는 약간 낮은 값을 나타내었고 pH는 Hizikia mineral salt의 경우 11.34로 다소 높은 값을, fucoidan salt와 laver salt은 6.88과 6.53으로 중성에 가까운 값을 나타내었으며, ORP는 -229 mV에서 38 mV로 월등히 낮은 값을 나타내었다. 기능성 소금의 L, a, b 값은 모두 정제염 보다는 약간 어두운색을 나타내었다. 무기물 함량은 Hizikia mineral salt에서 혈압 저하에 효과가 있는 무기성분인 $K^{+}$ 함량이 다량 함유되어 있었으며 용해도는 Hizikia mineral salt가 다소 떨어지는 경향을 나타내었으나 큰 차이는 나타나지 않았다. 관능특성을 알아본 결과 $Na^{+}$ 이외의 미네랄 성분을 다량 함유하고 있는 Hizikia mineral salt를 제외한 나머지 fucoidan salt, laver salt에서는 색을 제외한 모든 항목에서 높은 점수를 받아 조미기능성 염으로서도 사용가치가 기대되었다. 또한, laver salt는 54.8% 수준의 ACE 저해 활성값을 나타내었으며, fucoidan salt는 54.1-80.7% 수준의 높은 bile acid 결합특성을 각각 나타냄으로서 콜레스테롤 대사 및 혈압조절 기능성을 갖고 있는 것으로 사료되었다.

리보오스 보충이 최대강도 운동 후 혈중 피로물질 생성에 미치는 영향 (The Effects of D-Ribose Supplementation on the Production of Blood Fatigue Factors after Maximal Intensity Exercise)

  • 본 연구에서는 두 가지 투여그룹을 비교하여 ergogenic aid로서 D-ribose의 효과를 운동영양학적으로 규명하는데 그 목적이 있었고, 이를 위하여 2,000 m 조정운동 전, 운동종료 직후, 그리고 운동 종료 후 회복기 30분의 변화를 연구하였으며, 다음과 같은 결론을 얻었다. 첫째, 투여그룹에 따른 2,000 m 조정운동 수행력의 차이는 없었으며 둘째, 투여그룹에 따른 젖산과 암모니아 수치의 차이도 나타나지 않았다. 셋째, 회복 30분에서 무기인산은 PS에 비해서 RS투여그룹의 수치가 더 낮게 나타났으며, 마지막으로 회복 30분에서 hypoxanthine의 축적은 PS투여그룹에 비해 RS투여그룹의 수치가 더 낮게 나타났다. 따라서, 7-8분 정도의 고강도 운동 시 운동기록의 단축에 D-ribose 구강투여의 효과는 없었지만, 휴식 기 피로회복에 긍정적인 효과를 줄 수 있을 것이라 사료된다.

Lifestyle modifications in an adolescent dormi­tory: a clinical trial

  • Purpose: Childhood obesity is an increasing public health issue worldwide. We examined dietary patterns among adolescents in a dormitory school, identified obese adolescents and tried to intervene to improve food habits and physical activity. Methods: We conducted an experimental prospective longitudinal study based on 36 obese (body mass index $[BMI]{\geq}95th$ percentile) adolescents (aged 12-18 years) compared with controls (healthy children: normal age-appropriate BMI ($BMI{\leq}85th$ percentile). Six months' intervention included lifestyle-modification counseling (once a week by a clinical dietician), and an exercise regimen twice a week, 60 minutes each time, instructed by a professional pediatric trainer). Both groups underwent baseline measurements at the beginning of the study and 6 months later (arterial stiffness, blood pressure, pulse, weight and height, hemoglobin, creatinine, liver enzymes, highly sensitive C-reactive protein and complete lipid profile). Results: Twenty-one participants completed the study. Low compliance from participants, school staff and parents was observed (participation in planned meetings; 71%-83%). BMI significantly decreased from $32.46{\pm}3.93kg/m^2$ to $30.32{\pm}3.4kg/m^2$ (P=0.002) in the study group. Arterial stiffness was not significantly different between the 2 groups and did not change significantly after 6 months' intervention (P=0.494). No significant changes in CRP and lipid profile were observed after the intervention. Conclusion: Making lifestyle modifications among adolescents in a dormitory school is a complex task. Active intervention indeed ameliorates BMI parameters. However, in order to maximize the beneficial effects, a multidisciplinary well-trained team is needed, with emphasis on integrating parents and the school environment.
