Cryopreservation of Mouse Embryos by Vitrification (Mouse 배의 Glass화 보존)

  • Eight-cell mouse embryos equilibrated with vitrification solution(VS3), consisting of glycerol and polyethlene glycol, were plunged directly into liquid nitrogen. The embryos were cryopreserved by vitrification without intra- and extracellular ice formation. After the vitrified embryos were warmed at 4$^{\circ}C$, sucrose dilution procedures were examined and the survival embryos were transferred to recipients after 48h incubation in vitro. The results were obtained as follows. 1. Mouse embryos equilibrated with VS3 were diluted with 1.5m sucrose-HB 1 solution, 0.5M sucrose-HB1 solution for 5min at room temperature, respectively. The proportion of vitrified-warmed embryos developed to blastocyst(85.7%) was as high as that of the embryos diluted with 1.04M sucrose-HB1 solution at 4$^{\circ}C$(85.5%). 2. Normal live youngs were obtained in 53.9%(55/102) of the vitrified-warmed embryos after tranfer to pseudopregnant recipients.

  • It is believed that age controls horizontal-branch (HB) morphology of Globular Clusters besides metallicity (first parameter). However, other second parameter candidates, such as He abundance and alpha elements enhancement, could also affect HB morphology. These effects may influence to the age measurements from the main-sequence turnoff (MSTO) as well. We will measure the variations of age caused by each second parameter candidate at HB and MSTO respectively, using the well known second parameter pair M3 and Ml3. (omitted)

  • Let X be a Banach space and Z a closed subspace of a Banach space Y. Denote by L(X, Y) the space of all bounded linear operators from X to Y and by K(X, Y) its subspace of compact linear operators. Using Hahn-Banach extension operators corresponding to ideal projections, we prove that if either $X^{**}$ or $Y^{*}$ has the Radon-Nikodym property and K(X, Y) is an M-ideal (resp. an HB-subspace) in L(X, Y), then K(X, Z) is also an M-ideal (resp. HB-subspace) in L(X, Z). If L(X, Y) has property SU instead of being an M-ideal in L(X, Y) in the above, then K(X, Z) also has property SU in L(X, Z). If X is a Banach space such that $X^{*}$ has the metric compact approximation property with adjoint operators, then M-ideal (resp. HB-subspace) property of K(X, Y) in L(X, Y) is inherited to K(X, Z) in L(X, Z).

  • Let be $X_1$,...,$X_p$, $p\geq2$ independent random variables where each $X_i$ has a gamma distribution with $\textit{k}_i$ and $\theta_i$ The problem is to simultaneously estimate $\textit{p}$ gamma parameters $\theta_i$ and $\theta_i{^-1}$ under entropy loss where the parameters are believed priori. Hierarch ical Bayes(HB) and empirical Bayes(EB) estimators are investigated. And a preference of HB estimator over EB estimator is shown using Gibbs sampler(Gelfand and Smith 1990). Finally computer simulation is studied to compute the risk percentage improvements of the HB estimator and the estimator of Dey Ghosh and Srinivasan(1987) compared to UMVUE estimator of $\theta^{-1}$.

  • We investigated the effects of annealing on the rearrangement of H-bonding structure and its influence on the thermal and rheological properties of $2^{nd}\;and\;4^{th}$ pseudo-generation aliphatic hyperbranched (HB) polyesters based on 2,2-bis(methylol)propionic acid. During annealing of amorphous HB polyesters, the structure becomes more ordered as a consequence of multiple H-bonds formation between linear sequences. Structure ordering is more pronounced for the lower pseudo-generation HB polyester with low molar mass, low degree of branching and incompletely reacted core hydroxyl groups which greatly increases the possibility for multiple H-bond interactions.

  • The water celery, Oenanthe javanica DC., is widely distributed in Korea and the other South East Asian countries. From ancient times, the water celery has widely been used for food or medicine. In herbal medicine, the water celery has a medical actions which are defervescence, diuresis, hypotensor, appetizer. Recently, studies on the water celery are briskly being conducted. For example, studies on the morphologic property and classifications of origin of the water celery, studies on the chemical constituents of the Water celery, studies on the bio activities of the water celery which contains flavonoids. However, there is not studies of hematological changes in dogs administrated the water celery until now. Consicontly, this study was conducted to observe the hematological changes in dogs administrated excessive amount of the water celery concentrated extract for a long period. In this studies, all dogs showed significantly decrease of the RBC, PCV, Hb and MCHC, and increase of the WBC, MCV, GSH and reticulocytes, but did not significantly change in the Met-Hb.

Effect of Scratching Method Complex Therapy on the Patients with Peripheral Facial Paralysis (말초성 안면신경마비에 대한 괄법(括法) 복합치료 효과)

  • Objectives : This study was done for reporting the effect of scratching method on patients with peripheral facial palsy. Methods : We investigated 34 cases of patients with peripheral facial paralysis, and devided patients into two groups : We treated one group by complex oriental medical treatment with scratching method therapy, and did the other group by complex oriental medical treatment without scratching method therapy. gross grading system of House-Brackmann(HB score) and Yanagihara's unweighed grading system(Y score) at baseline and final were used for evaluating the effect of the treatment. Results : 1. In scratching method group and non-scratching method group, compared with baseline, at final, HB score was significantly decreased and Y score was significantly increased. 2. At final, scratching method group showed significant decrease on HB score and significant increase on Y score compared with non-scratching method group. Conclusions : scratching method showed significant improvement in peripheral facial palsy.

Animal Cell Culture and the Production of Monoclonal Antibody(MAb) Using Biopolymer Membrane (생물고분자 막 형성을 이용한 동물세포 배양 및 단클론항체 생산)

  • Biopolymer membrane was prepared using two oppositely charged natural biopolymers. The biopolymer membrane was used for the encapsulation of two hybridoma cell lines(ATCC CRL-1606, ATCC HB-8852) to produce monoclonal antibodies. In order to reduce the down stream steps, the pre size of the membrane was controlled to retain the monoclonal antibodies in the capsules based on the diffusion experiments with standard proteins. T-flask culture showed cell densities of 8$\times$107 cells/mL and 3$\times$107 cells/mL, and MAb concentrations of 506$\mu$g/mL and 109$\mu$g/mL for encapsulated ATCC CRL-1606 and HB-8852, respectively. Two liter perfusion cultures with encapsulated ATCC HB-8852 were performed to enhance the MAb production. The MAb production of the encapsulated hybridoma increased considerably comparing to the culture using silicon tubing for oxygen transfer.

Isolation of Hepatic Drug Metabolism Inhibitors from the Rhizomes of Curcuma zedoaria

  • The methanolic extract of the Rhizome of Curcuma zedoaria exhibited a significant prolongation of hexobarbital (HB)-induced hypnosis. Through liquid chromatography of an ether soluble fraction. monitoring by bioassay, three sequiterpenes, germacrone (A), curzerenone (B) and germacrone epoxide (C) were isolated as active consituents. A single treatment (100-200 mg/kg, i.p.) of each compound showed not only a significant prolongation of HB-induced sleeping time but also a significant inhibition of aminopyrine N-demethylase activity in mice, and further exhibited a typical type I binding spectra with oxidized rat hepatic cytochrome P-450 induced by phenobarbital. All of the compounds provoked a sleep episode at a subhypnotic dose of HB, implying that they possess CNS depressant properties.

The Effect of Horizontal Branch Stars on the Age-Dating of Simple Stellar Populations

  • Based on Yonsei Evolutionary Population Synthesis (YEPS), we have investigated the effect of horizontal branch stars (HBs) on the age-dating of simple stellar populations (SSPs). A detailed comparison of YEPS SSP with H_beta of M31 globular clusters (GCs) reveals (1) that the age dating without HB prescription gives ~5 Gyr younger ages for metal-poor M31 GCs, and (2) the age dating with HB prescription does not need any age gap between metal-poor and metal-rich GCs. This result is parallel to the well-known discrepancy in ages derived from integrated Balmer strengths and isochrone fittings of Milky Way GCs (MWGCs). Without a synthetic blue HB model, we cannot explain strong Balmer indices of metal-poor and old MWGCs. Our results suggest that the SSP model with well calibrated HBs should be used for the age-dating of SSPs to avoid a serious underestimation of ages due to the strong Balmer indices.
