Optimization of In Vivo Embryo Production and Pregnancy following Embryo Transfer in Hanwoo Cattle

  • Embryos formed in vivo were collected from 171 donors housed in Chung Cheong Buk-Do Institute of Livestock and Veterinary Research of the Chungbuk community during the years 2009~2012. We evaluated annual embryo collection, effect of follicle stimulating hormone (FSH), controlled internal drug release (CIDR) and prostaglandin (PG) administration to the donor for superovulation and controlling the estrus cycle, seasonal effects of embryo collection and compared the number of embryos recovered as per the collection days and pregnancy rate. In all, 1,243 embryos were collected from 118 donors with an average of $7.31{\pm}5.35$ embryos per donor, out of which 69.4% were transferable. Dosages of FSH required for inducing superovulation in various donors were compared. Average number of embryos collected from donors administered with 30 AU of FSH ($7.13{\pm}5.74$ per donor) was not significantly different from that of donors who were given an injection of 24 AU of FSH ($7.53{\pm}4.91$ per donor). However, the percentage of transferable embryos in the 30AU FSH-administered group (63.2 %, 449 of 711) was higher than that in the 24AU FSH-administered group (77.8%, 414 of 532). In the group of donors under a natural estrus cycle, the FSH dose administered did not influence the number of transferable embryos produced ($7.49{\pm}6.25$ per donor for 30 AU of FSH vs $7.49{\pm}4.92$ per donor for 24 AU of FSH). However, in donors administered with CIDR and PG for controlling the estrus cycle, the FSH dose affected the average number of transferable embryos collected ($4.25{\pm}2.87$ per donor for 30 AU of FSH vs $8.50{\pm}6.36$ per donor for 24 AU of FSH). We collected embryos from donors 6, 7 or 8 days after artificial insemination (AI). Results showed that the percentage of transferable embryos among those collected 8 days after AI was significantly higher than that among embryos collected 6 or 7 days after AI. Seasonal variations did not affect number of recovered embryos and pregnancy rates in natural estrus cycle and CIDR treatment groups (48.28% and 42.55%) but higher than pregnancy rate of frozen embryos (19.63%). These results indicated that administration of FSH beyond a threshold dose (at least 24 AU) has no beneficial effect on the production embryos and that collection of embryos 7~8 days after AI is optimal for embryo recovery. CIDR treatment induced superovulation in short term and had no influence on the natural estrus cycle. Finally, although good-quality embryos were transferred, freezing significantly reduced the pregnancy rates after transfer.

Prediction of genomic breeding values of carcass traits using whole genome SNP data in Hanwoo (Korean cattle) (한우에 있어서 유전체 육종가 추정)

  • Genomic breeding value (GEBV) has recently become available in the beef cattle industry. Genomic selection methods are exceptionally valuable for selecting traits, such as marbling, that are difficult to measure until later in life. One method to utilize information from sparse marker panels is the Bayesian model selection method with RJMCMC. The accuracy of prediction varies between a multiple SNP model with RJMCMC (0.47 to 0.73) and a least squares method (0.11 to 0.41) when using SNP information, while the accuracy of prediction increases in the multiple SNP (0.56 to 0.90) and least square methods (0.21 to 0.63) when including a polygenic effect. In the multiple SNP model with RJMCMC model selection method, the accuracy ($r^2$) of GEBV for marbling predicted based only on SNP effects was 0.47, while the $r^2$ of GEBV predicted by SNP plus polygenic effect was 0.56. The accuracies of GEBV predicted using only SNP information were 0.62, 0.68 and 0.73 for CWT, EMA and BF, respectively. However, when polygenic effects were included, the accuracies of GEBV were increased to 0.89, 0.90 and 0.89 for CWT, EMA and BF, respectively. Our data demonstrate that SNP information alone is missing genetic variation information that contributes to phenotypes for carcass traits, and that polygenic effects compensate genetic variation that whole genome SNP data do not explain. Overall, the multiple SNP model with the RJMCMC model selection method provides a better prediction of GEBV than does the least squares method (single marker regression).

  • Study the gene about economical characteristic of human disease or domestic animal is a matter of grave interest, preserve and elevation of gene of Korea cattle is key subject. Studies have been done on the gene of Korea cattle using EST based SNP map, but it is based on statistical model, therefore there are difference between real position and statistical position. These problems are solved using both EST_based SNP map and Gene on sequence by Lee et al. (2009b). We have used multifactor dimensionality reduction(MDR) method to study interaction effect of statistical model in general. But MDR method cannot be applied in all cases. It can be applied to the only case-control data. So, method is suggested E-MDR method using CART algorithm. Also we identified interaction effects of single nucleotide polymorphisms(SNPs) responsible for average daily gain(ADG) and marbling score(MS) using E-MDR method.

Characteristics of the Absorption of Cherry Tomato by the Application of Chelated Calcium and Germanium (킬레이트화 칼슘 및 게르마늄의 방울토마토 시용에 따른 흡수 특성)

  • Bloods from slaughtered Korean native cattle (Hanwoo : Bos taurus coreanae) can be used in agriculture with various beneficial effects on the environment. This study was performed to find out the effect of the application of calcium and germanium, which are chelated with peptides made of cattle bloods on the absorption of them by cherry tomato (Lycopersicon esculentum Mill.). Peptide was purified by enzymatic hydrolysis and ultrafiltration from serum protein, which is composed of Asp, Glu, Lue etc. Chelated Ca and Ge were produced by reacting the peptide with $CaCl_2$ and Ge-$132^{(R)}$. Then, they were applied to cherry tomato during cultivation period at the rate of $300mL{\times}3times$ (10 days interval) for each tomato tree. Application of chelated-Ca increased the Ca contents in leaves and fruits of cherry tomato showing as 19.9% and 23.4% in newerly prepared chelated-Ca-200 ($200mg\;L^{-1}$), 8.1% and 6.8% in commecrial Calciolid Ca-300 ($300mg\;L^{-1}$) compared to 3152.6 and $63.2mg\;kg^{-1}$ in control, respectively. Application of chelated-Ge showed the increase in the germanium contents in both leaves and fruits of cherry tomato by over 6 times than those of control and over 4 times than Ge $132^{(R)}$-20 application. As a result of this study, it seemed that the application of chelated-Ca and chelated-Ge would be more effective than the existing commertial one in the absorption of calcium and germanium by cherry tomato.

Effect of Embryo Factors on Pregnancy and Abortion Rate after Transfer of In Vitro Produced Korean Native Cattle Embryos (한우 체외수정란이 이식된 수란우의 임신과 유산에 영향을 미치는 수정란측 요인)

  • In this study, we investigated the effects of number, grade, stage and day of embryos on pregnancy and abortion rates of Holstein heifer recipients after transfer of in-vitro produced Korean Native Cattle (Hanwoo) embryos. When one, two or three blastocysts were transferred to recipients, pregnancy (32.0, 44.0 and $32.0\%$) and abortion (12.5, 13.6 and $6.3\%$) rates did not differ among treatments. The pregnancy and abortion rates of different grades of blastocysts were $40.8\~44.2\%\;and\;9.4\~16.3\%$, respectively. However, they were not significantly different. According to the stage of blastocyst, the pregnancy rates were similar between $39.1\%\;and\;45.3\%$. However, the abortion rate in hatched blastocyst stage $(20.8\%)$ was significantly higher than that in late blastocyst stage $(9.0\%)$. The pregnancy rates in day $7\cdot7$ and day $7\cdot8$ blastocyst (47.1 and $53.9\%$, respectively) was significantly higher than that in the day $9\cdot9\;and\;8\cdot9$ blastocyst (29.8 and $27.7\%$, respectively). The abortion rate was the highest in the day $9\cdot9$ blastocyst $(30.8\%)$ among treatments, and it was significantly different compared to that in the day $7\cdot7$ blastocyst $(10.1\%)$. Therefore, this study suggested that the pregnancy rate was affected by only day of embryos, while the abortion rate was affected by stage and day of embryos.

Effect of Scoria, Quartz Porphyry and Oak Charcoal Powder Feeding on the Meat Quantity and Quality of Fattening Cross Bred Hanwoo Bulls (점토광물 급여가 한우교잡우의 산육능력 및 육질에 미치는 영향)

  • This study was conducted to investigate the effects of Scoria, Quartz porphyry and Oak charcoal powder feeding by 1% level of concentrate on the fattening cross bred bulls(5/8 Korean Native Cattle, 2/8 Charolais, 1/8 Brahman germ plasma) Feeding trial was conducted with 4 treatment groups which were T1(Full feeding of concentrate and roughage for 12 months), T2(T1 and Scoria addition), T3(T1 and Quartz porphyry addition) and T4(Oak charcoal addition) for 360 days, consisting of 32 heads whose initially weights were about 299.7kg. The results obtained are summarized as follows; 1. During the over-all period, average daily gains by T3, T4, T2 and T1 were 1.024, 0.987, 0.977kg and 0.964, respectively, without a significant difference by treatments. 2. Concentrate intakes per body weight and TDN intakes required per unit of kilogram gain were lower in addition agent groups than in control, and in all the treatments feed efficiency was higher at early stage for 360 fattening day than at end stage. 3. Carcass weight, dressing percent, back-fat thickness and loin eye muscle area by treatments were 330.9 to 348.4kg(average 340.3kg), 55.2~56.2(average 55.8%), 0.41~0.55(average 0.46cm)and 76.0~80.9$\textrm{cm}^2$(average 77.6$\textrm{cm}^2$), respectively, and were improved in addition agent groups than in control. 4. Meat quality grade is higher in addition agent groups than in control, and heads of 1st grade by treatments was 3, 2, 1 and none in T4, T2, T3 and T1, respectively. 5. Incidence heads of diseases by treatment were not apparently different, but diseases of digestive system was decreased in addition agent groups than in control. 6. The nitrate nitrogen content of fattening bull dung which collected in rectum were 0.082~0.089% (average 0.084%), and the content in addition agent groups was smaller than in controll. 7. Income was higher in order of T4, T3, T2, and T1 and incomes of treatment groups were grater than that of control group by 21.4 to 33.5 percent. According to the above results it may be concluded that fattening bulls may be required to feed no more than 1% of Scoria, Quartz porphyry and Oak charcoal powder based on the concentrate feeding level during the fattening period(12 to 24 month of age) to produce high quality meat and increase income.

Genetic Correlation of Carcass and Meat Production Traits with Hormones and Metabolic Components in Hawoo (가축의 혈청 호르몬 및 대사물질 농도와 도체 및 산육형질에 대한 유전상관에 관한 연구)

  • This study was aimed to investigate genetic relationships, variables, and correlations between economic traits and metabolic materials in serum components according to bleeding periods and breeding locations for the castrated and not castrated Hanwoo cattle at National Livestock Research Institute. Analysis of variance for serum hormones and metabolic materials showed significant differences by breeding locations except for testosterone and globulin. Statistical differences for serum components were detected by birth year except for cortisol, total protein, globulin and creatinine, and by castration except for total protein and BUN. All the serum components were tended to have sire effects except for testosterone resulting in some degree of additive gene actions. Breeding locations showed statistical significances for carcass weight and back fat thickness, but not in carcass rate, KPH, live weight and transportation weight loss. Effects of breeding locations and castration were significant for all weight measurement periods except for 9 month and 6 month, respectively. A significant sire effect was observed in all weight measurements. Least squared means for concentration of serum components by breeding year, season and castration were not significant. High concentration of cortisol, creatinine and triglyceride and low concentration of IGF-1 and glucose were detected in castrated cattle. Concentration of testosterone with castrated cattle was $5.2\%$ corresponding to non castrated cattle. Estimation of heritabilities of serum components using a sire model with restricted maximum likelihood were ranged 0.07 to 0.58. High heritabilities were estimated for total protein, albumin, globulin, cortisol, creatinine and BUN were 0.53, 0.54, 0.42, 0.45, 0.58 and 0.54, respectively. Low heritabilities were estimated fur calcium, testosterone and IGF-1 for 0.07, 0.15 and 0.12, respectively. Heritabilities for carcass weight, back fat thickness, meat yield index, KPH, and IMF were estimated as 0.39, 0.45, 0.30 0.13, and 0.93. Heritabilities of weights on 18, 12, 9, 6, and 24 month were estimated as 0.78, 0.76, 0.62, 0.58 and 0.58. Estimated heritabilities for average daily gain on 6${\~}$2, 12${\~}$18, and 18${\~}$24 month were 0.80, 0.75 and 0.19, respectively.

Characterization of the Nanog 5'-flanking Region in Bovine

  • Bovine embryonic stem cells have potential for use in research, such as transgenic cattle generation and the study of developmental gene regulation. The Nanog may play a critical role in maintenance of the undifferentiated state of embryonic stem cells in the bovine, as in murine and human. Nevertheless, efforts to study the bovine Nanog for pluripotency-maintaining factors have been insufficient. In this study, in order to understand the mechanisms of transcriptional regulation of the bovine Nanog, the 5'-flanking region of the Nanog was isolated from ear cells of Hanwoo. Results of transient transfection using a luciferase reporter gene under the control of serially deleted 5'-flanking sequences revealed that the -134 to -19 region contained the positive regulatory sequences for the transcription of the bovine Nanog. Results from mutagenesis studies demonstrated that the Sp1-binding site that is located in the proximal promoter region plays an important role in transcriptional activity of the bovine Nanog promoter. The electrophoretic mobility shift assay with the Sp1 specific antibody confirmed the specific binding of Sp1 transcription factor to this site. In addition, significant inhibition of Nanog promoter activity by the Sp1 mutant was observed in murine embryonic stem cells. Furthermore, chromatin-immunoprecipitation assay with the Sp1 specific antibody confirmed the specific binding of Sp1 transcription factor to this site. These results suggest that Sp1 is an essential regulatory factor for bovine Nanog transcriptional activity.

Detection of major genotypes combination by genotype matrix mapping (유전자 행렬 맵핑을 활용한 우수 유전자형 조합 선별)

  • It is important to identify the interaction of genes about human disease and characteristic value. Many studies as like logistic analysis, have associated being pursued, but, previous methods did not consider the sub-group of the genotypes. So, QTL interactions and the GMM (genotype matrix mapping) have been developed. In this study, we detect the superior genotype combination to have an impact on economic traits of Korean cattle based on the study over GMM method. Thus, we identified interaction effects of single nucleotide polymorphisms (SNPs) responsible for average daily gain(ADG), marbling score (MS), carcass cold weight (CWT), longissimus muscle dorsiarea (LMA) using GMM method. In addition, we examine significance of the major genotype combination selected by implementing permutation test of the F-measure which was not obtained by Sachiko et al.

Prospect of new variety breeding of Italian ryegrass in South Korea

  • The objective of this study was to know prospect and present state of new variety breeding of Forage and Grassland in South Korea. The industry of forage in Korea is going up to expend through utilization of good quality forage, Italian ryegrass, forage corn and oat rather than rice straw. Especially, since 2007, Italian ryegrass(IRG) had been very important major winter forage crop in South Korea and developed 13 varieties including very early maturity variety (three varieties), early maturity variety (three varieties), medium maturity variety (one variety) and late maturity variety (six varieties). But the disadvantage of Italian ryegrass was weak winter hardness and drought but has good advantage of forage nutritive value, high-yielding and high sugar content, and like livestock as like hanwoo, dairy cattle, goat so on. The Ko-variety (Korea developed variety) of Italian ryegrass has high cold-tolerant and adaptability more than any other country developed variety, and expend to cultivation area from southern area (below Daejeon) to middle-northern area (upper Han river). Although the cultivation area of Italian ryegrass of South Korea was 21,700 ha in 2007, right now, that of Italian ryegrass is about 123,600ha due to expend cultivation area and know famer to good forage crop and have a various maturity varieties (very early, early, medium, late) according to local situation (before-crop harvesting stage or double cropping system). The seed market of Italian ryegrass in South Korea becoming extended to around 100 million Korean won and seed export get nearer to foreign country. We are going to develop of new variety for stress tolerant and high yield and quality forage variety, good adaptability to the Korean environmental conditions including reclaimed area, make self-sufficiency system for forage seed (Italian ryegrass), export our seed to foreign countries.
