Genomic Sequence Variability of the Prion Gene (PRNP) in Korean Cattle

  • In this study, we have investigated sequence variants in the PRNP gene of 20 individuals belonging to the Korean cattle, and have analyzed and compared genetic features between varieties of other cattle breeds. Of the 73 sequence variants identified in Korean cattle, 27 were identified for the first time in this study, whereas 46 of these polymorphisms had previously been isolated. We discovered a 2.6 kb SNP hot spot region localized on the putative promoter region of the PRNP gene. Furthermore, the copy numbers of the octapeptide repeat (24 bp indel) which is detected on the coding sequence (CDS) of the PRNP exhibited a completely homozygous 6/6 genotype which is dominant in other cattle breeds. We also characterized a new 19 bp/10 bp allele located on the putative promoter region of the PRNP gene, which represented 0.71 in allele frequency. To the best of our knowledge, this report is the first to address polymorphisms of the PRNP gene structure in Korean cattle in which BSE has yet to be discovered. Therefore, our findings may prove useful with regard to our current understanding of allelic diversity in bovine species, and may also provide new insights into the genetic factors associated with susceptibility or resistance to BSE.

한우송아지의 전지골절에서 아크릴릭 프레임을 이용한 외부고정 (External Fixation with Acrylic Frame in Forelimb Fracture of Korean Native Two Calves)

  • Two day-old male and 1 month-old female Hanwoo calves presented to Chonbuk Animal Medical Center in Chonbuk National University. Two calves were stepped by mother cow and had right (2 day-old male) and left (1 month-old female) forelimb lameness. The male calf was diagnosed as right radius-ulnar fracture through physical and radiographic examination. The other female calf was diagnosed as left third and fourth metacarpal bone fracture by the same examinations as male calf. Acrylic frame external fixator was used to reduce the fractures. Arcrylic frame external fixator in two calves lasted for 5~6 weeks. After removing the acrylic frame external fixator, radiographic evaluation and physical examination were performed. Fractured radius-ulnar and metacarpal bones were unionized and the calves were able to stand and bear weight. In these two calves, application of acrylic frame external fixator in calf fracture was effective. Acrylic frame external fixator should be considered for the repair of radius-ulnar and metacarpal bone fracture in calf.

Antibacterial effect of ethylacetate fraction of Orostachys japonicus on Enterococcus faecalis causing Endophthalmitis

  • Endophthalmitis is a disease that causes ocular inflammation and has a catastrophic effect on eyesight. Recent studies show that Enterococcus faecalis is rapidly increasing causative bacterium of endophthalmitis. It is predicted that the increased endophthalmitis by E. faecalis is presumable due to the high resistance of E. faecalis to moxifloxacin (MFX), which is a common antibiotic used for eye drop. Because of the need for therapeutic agents to overcome this problem, this study sought to explore the feasibility of developing a combination therapy using Orostachys japonicus. The ethylacetate fraction of O. japonicus (OJA) used in this study. Antimicrobial activity was tested 13 E. faecalis strains including one E. faecalis standard strain, eight clinically isolated E. faecalis strains and four quinolone resistant E. faecalis strains using CLSI antibiotic susceptibility test method. Minimal Inhibitory Concentration (MIC) of OJA was confirmed to be $500{\mu}g/ml$ for all 13 strains. Then we tested for the synergistic effect of OJA to MFX using checkboard test method. The MIC of MFX was $0.25{\mu}g/ml$ for the standard strain and 8 for the clinical isolates, and $16{\sim}64{\mu}g/ml$ for the quinolone - resistant strains. When OJA was mixed with MFX, no synergistic effect was observed in all strains, but the antibacterial activity of OJA remained unchanged. Most ocular other strains can be removed by MFX except the MFX resistant E. faecalis, which can be removed by OJA in combination therapy. Therefore, OJA can be a potential candidate for the combined treatment endophthalmitis.

군집분석을 이용한 전시장비의 취약성 그룹 재분류 (Reclassification of the vulnerability group of wartime equipment)

  • 현재 한국군의 전시장비소요산정은 미군의 모델인 ELCON을 기반으로 하고 있다. 하지만 전시장비소요산정을 위해 ELCON에서 적용하는 22개의 취약성그룹에 대한 구체적인 분류기준은 알려져 있지 않다. 따라서 이번 연구에서는 2014년 국방전시기본품목 505종에 대해 데이터를 수집하고, 군집분석 방법을 통해 ELCON에서 사용되는 취약성그룹과 다른 새로운 취약성그룹을 제시하였다. 이번 연구를 통해 한국의 상황에 맞는 전시장비들의 취약성그룹을 분류하는 방법을 제시하였을 뿐만 아니라 새로 추가될 장비 역시 해당 장비의 특성을 통해 분류 할 수 있는 기준을 제시함으로 전시 정확한 장비소요를 예측할 수 있도록 하였다.

유전자 행렬 맵핑을 활용한 우수 유전자형 조합 선별 (Detection of major genotypes combination by genotype matrix mapping)

  • 인간의 질병 및 가축의 특성치에 관한 유전자 규명은 매우 중요한 과제이다. 유전자원 보존과 유전능력향상을 위한 기술 개발 역시 매우 중요한 관심사로써 이와 관련된 많은 연구들을 진행해왔다. 통계모형의 상호작용 효과를 분석하기 위해 로지스틱 회귀분석과 같은 전통적인 방법과 비모수적인 방법들이 개발되었지만 이들은 상호작용에 영향을 준 양적형질위치들의 하위 집단의 유전자형에 대해서 고려하지 않은 문제점이 있다. 따라서 많은 양적형질위치들을 한 번에 비교하여 특성치에 영향을 주는 양적형질위치의 상호작용과 그 하위집단인 유전자형을 규명하는 방법으로 유전자형 행렬 맵핑이 개발되었다. 본 연구에서는 EST_based SNP 연관지도에 의해 선정된 17개의 후보 단일염기다형성을 대상으로 유전자 행렬 맵핑을 활용하여 한우의 주요 경제형질인 일당증체량, 도체중, 등심단면적, 근내지방도에 영향을 주는 우수 유전자형 조합을 선별한다. 그리고 선별된 조합에 대해 유전자 행렬 맵핑 방법에서 적용되지 않은 순열검정을 도입함으로써 우수유전자형 조합에 대한 통계적인 유의성을 확인한다.

Combined Effect of Irradiation and Ageing Condition on Physicochemical and Microbial Quality of Hanwoo Eye of Round

  • The combined effects of electron-beam irradiation and ageing of beef were examined. The irradiated samples at dose of 0 or 2 kGy were kept and analyzed for the microbial growth, shear values, meat color, and nucleotide-related flavor compounds at different ageing temperatures (2, 10, or 25℃) for 8 d. The irradiation effect on inactivation of foodborne pathogens was also investigated. The population of Listeria monocytogenes and E. coli O157:H7 inoculated in beef samples decreased in proportion to the irradiation dose, showing D10 values of 0.66 and 0.65 kGy respectively. The irradiated beef eye of round had lower number of total aerobic bacteria (TAB) than nonirradiated one during the storage, but the TAB increased with higher ageing temperature (p<0.05). Especially, TAB increased sharply in non-irradiated samples aged at 25℃ after 4 d (p<0.05). With increasing ageing temperature and ageing time, shear force values decreased (p<0.05). The color a* values of the irradiated beef were lower than those of the non-irradiated throughout the ageing period (p<0.05). As ageing time and temperature increased, the amounts of inosine monophosphate decreased and the hypoxanthine increased (p<0.05). Relatively high ageing temperature could be used at irradiated beef eye of round to shorten the ageing time.

Potential functional roles of follistatin on bovine somatic cell nuclear transfer embryos

  • To demonstrate that follistatin treatment enhances the efficiency of nuclear transfer (SCNT), cell allocation and preimplantational development were determined in bovine SCNT embryos in the present study. Treatment of activated SCNT embryos with 10 ng/ml follistatin significantly increased the proportion of blastocyst development compared to untreated SCNT embryos. In addition, an increase in trophectoderm (TE) cell numbers and relatively higher proportion of TE cells to total cells were observed, but the number of inner cell mass (ICM) cell and total cell numbers were not changed (P < 0.05). No significant effect of other doses of follistatin was observed for the above endpoints. However, treatment with 1 and 10 ng/ml follistatin reduced the proportion of nuclear transfer blastocysts with an ICM ratio of > 60% relative to untreated nuclear transfer blastocysts at Day 7. No significant effect of follistatin treatment on proportions of nuclear transfer blastocysts with ICM ratio of 20-40% or 40-60% was observed. Taken together, these results suggested that follistatin can be used to increase developmental competence of SCNT embryos in terms of cell allocation, particularly TE cells, during preimplantation stages, subsequently enhancing placentation and birth of live offspring.

Prospect of new variety breeding of Italian ryegrass in South Korea

  • The objective of this study was to know prospect and present state of new variety breeding of Forage and Grassland in South Korea. The industry of forage in Korea is going up to expend through utilization of good quality forage, Italian ryegrass, forage corn and oat rather than rice straw. Especially, since 2007, Italian ryegrass(IRG) had been very important major winter forage crop in South Korea and developed 13 varieties including very early maturity variety (three varieties), early maturity variety (three varieties), medium maturity variety (one variety) and late maturity variety (six varieties). But the disadvantage of Italian ryegrass was weak winter hardness and drought but has good advantage of forage nutritive value, high-yielding and high sugar content, and like livestock as like hanwoo, dairy cattle, goat so on. The Ko-variety (Korea developed variety) of Italian ryegrass has high cold-tolerant and adaptability more than any other country developed variety, and expend to cultivation area from southern area (below Daejeon) to middle-northern area (upper Han river). Although the cultivation area of Italian ryegrass of South Korea was 21,700 ha in 2007, right now, that of Italian ryegrass is about 123,600ha due to expend cultivation area and know famer to good forage crop and have a various maturity varieties (very early, early, medium, late) according to local situation (before-crop harvesting stage or double cropping system). The seed market of Italian ryegrass in South Korea becoming extended to around 100 million Korean won and seed export get nearer to foreign country. We are going to develop of new variety for stress tolerant and high yield and quality forage variety, good adaptability to the Korean environmental conditions including reclaimed area, make self-sufficiency system for forage seed (Italian ryegrass), export our seed to foreign countries.

한우 비육우 전업농가 시설투자한계 분석 (Analysis of Investment in Equipment Capital for Fattening Cattle Farms)

  • The objective of this study was to estimate the investment limit in equipment capital by Capital Recovery Method. The data used was collected from 23 Hanwoo(Korean Native Cattle) farms in 1994. The necessary time for herd size expansion was average 5 years from opening(9.7 heads) to 50 heads, 4 years from 50 to 100heads, 3 years from 100 to 150 heads, and 2 years from 150 to 200 heads, respectively. It took 14 years from opening to 200 heads of herd size. The debts for fattening cattle farms were 15.2million won for the size of 50 heads, 37.1 million won for the size of 100 heads and 89.0 million won for the size 200 heads, respectively. Average rates of debt interest were 5.1 % 7.2 and 10.8, correspendingly. As the hear size increased, debt interest rates also increased due to government funds limit. The investment limits in equipment capital perfarm household were 77.7 million won for 50 heads, 135.1 for 100 heads, and 294.3 for 200 heads for 5,500 won per kg liveweight, 70.7 million won for 50 heads, 122.6 for 100 heads and 269.3 for 200 heads for 5,000 won per kg liveweight, 63.6 million won for 50 heads 110.0 for 100 heads and 244.2 for 200 heads for 4,500 won per kg liveweight.

체외에서 생산된 하우 배반포의 발달에 미치는 삼투압의 효과 (Effect of Hyper tonic Solution on In Vitro Development of Hanwoo Blastocysts Produced In Vitro)

  • This study was conducted to enhance the efficiency of embryo transfer and embryos produced in vitro according to select good quality blastocyst among in vivo or in vitro produced embryos. After middle blastocys(MB) and late blastocyst(LB) stages embryos that developed in vitro for 7∼8 days were treated hypertonic solution(380 mOsm or 600 mOsm) and the rate of development to hatched blastocyst(HB) was examined. The results obtained were summarized as follows: The proportion of embryos developed to HB was higher than that of control regardless of hypertonic solution treatment. However, it was related to culture time in hypertonic solution The propotrion of embryos developed to HB of morphologically recovered embryos in 380 nOsm solution ws higher than that of morphologically shrinked embryos. However, treatment time was not significantly different the rates of HB development. Especially, the proportion of embryos developed to HB of morphologically recovered embryos of the 30-min-treated group was significantly higher than that of control (p<0.05). The results suggest that hypertonic solution treatment should enhance the efficiency for criterion of transferable blastocyst quality.
