Characteristic of bacterial flora from the uterus in HanWoo cattle (정상 한우 및 번식장애 한우에 대한 자궁 내 세균 분석)

  • Uterine sterilization is important for improving fertility in cattle. This study compared bacterial flora in the uterus between healthy and repeat breeder cows (RBCs). The uterine flushing of six heifers, 13 healthy HanWoo cows and eight RBCs (HanWoo) were sampled, and 15 frozen semen samples were selected. Overall, 35 bacteria were identified from in HanWoo uterine flushing and semen. The bacterial genera identified from HanWoo uterine flushing were Alloiococcus, Bacillus, Enterobacter, Enterococcus, Erysipelothrix, Gardnerella, Granulicatella, Kocuria, Pantoea, Pasteurella, Rothia, Serratia, Sphingomonas, Staphylococcus, Stenotrophomonas and Streptococcus. The bacterial genera identified from HanWoo semen were Bacillus, Escherichia, Kocuria, Oligella, Pseudomonas, Serratia, Sphingomonas, Staphylococcus, Stenotrophomonas and Streptococcus. The prevalence and presence of the identified bacteria between healthy cows and RBCs differed significantly. Further studies are needed to determine the role of these bacteria in the uterus of HanWoo cattle with reproductive disorder.

Comparison of the Methods of Zona Pellucida Removal and Inner Cell Mass Isolation for the Generation of Parthenogenetic Embryonic Stem Cells in HanWoo Cattle

  • In general, zona pellucida (ZP) of the blastocyst has to be removed first, then either isolated the inner cell mass (ICM) or ZP-removed whole blastocyst, which is then cultured on the feeder layer to induce ICM outgrowth for the generation of embryonic stem cells (ESC). However, it is unclear whether ICM isolation before seeding on feeder layer is beneficial or not because the interaction between ICM and trophoblasts may affect cellular growth and/or pluripotency during the culture on the feeder. In the present study, two ZP removal methods (mechanically by splitting with a 28-gauge needle versus chemically by the treatment of acid-Tyrode's solution) and two ICM isolation methods (ZP-free whole blastocyst seeding versus mechanical isolation of ICM) were evaluated for the efficient isolation and culture of putative parthenogenetic bovine ESC. The number of maintained outgrown colonies was counted in each experimental group. As the result, mechanical removal of ZP with a needle and followed by whole ZP-free blastocyst seeding on feeder cells tended to attach more on the feeder layer and resulted in more outgrown colonies with its simple and less time-costing benefits. Currently we are generating ESC lines in HanWoo cattle by using this method for initial outgrowth of the parthenogenetic bovine blastocysts.

Epidemiological study of bovine neosporosis in Gyeonggi province

  • Neospora caninum (N caninum) is an intracellular protozoa parasite, and its infection is one of the important diseases because it can cause abortion in cattle. This study was conduct to gain epidemiological data for more effective control of the disease. Bloods were collected from 2,162 cattle on 90 farms from February to September 2006. Serums were tested for antibodies to N caninum using ELISA CherdCheck, IDEXX). The dairy breed presented a higher proportion of seropositive results than that of Hanwoo. The estimated proportion of N caninum-positive dairy farms was 71.1 %, but the overall seroprevalence was estimated as 22.8%. The seroprevalence of dairy cattle and Hanwoo were 31.0% and 4.9%, respectively. There was no significant difference in the seroprevalence by age, but the not-intensive managed farms had a high seroprevalence (OR=1.91, p-value<0.01). The antibody rate of cattle with dog(s) was greater than that of those without dogs (OR=2.13, p-value<0.01). There was a significant difference in abortion rate between seropositive cattle and seronegative ones (OR=6.2, p-value<0.01).

Application of AGNPS Water Quality Computer Simulation Model to a Cattle Grazing Pasture

  • This research compared the observed and model predicted results that include; runoff, sediment yield, and nutrient losses from a 2.71 ha cattle grazing pasture field in North Alabama. Application of water quality computer simulation models can inexpensively and quickly assess the impact of pasture management practices on water quality. AGNPS single storm based model was applied to the three pasture species; Bermudagrass, fescue, and Ryegrass. While comparing model predicted results with observed data, it showed that model can reasonably predict the runoff, sediment yield and nutrient losses from the watershed. Over-prediction and under-prediction by the model occurred during very high and low rainfall events, respectively. The study concluded that AGNPS model can be reasonably applied to assess the impacts of pasture management practices and chicken litter application on water quality.

  • In this study two stage investigation was used for seroprevalence of Neospora caninum in cattle between January, 2005 and November, 2007; first stage was to examine positiveness of the farms selected randomly, second was to test all individual cow in positive farms. A total of 850 sera were collected from 170 farms for the first stage, and positive rate of farm and head were 23.5% (40/170farms) and 7.5% (64/850heads), respectively. Seroprevalence of Holstein cattle was much higher than that of Hanwoo. In second stage positive rate of cow was 18.9% (246/1,303head), but seroprevalence of farm was very variable (1 - 90.5%). It was supposed that 3 farms with high positive rate had some risk factors to be infected; raising dogs in the same farm, location of hill-side where is easy to contact with wild animals.

The Safety of Brucella abortus strain RB51 in pregnant cattle (부루세라 RB51 백신의 임신소에 대한 안전성)

  • The safety of Brucella abortus strain RB51(SRB51) was investigated in dairy cows and Korean native cattle of 4~7th month of gestation. From experimentally inoculated cattle, 18 of 25 (72.0%) dairy cows, and 3 of 10 (30.0%) Korean native cattle were aborted or delivered premature fetus. There were no significant differences in the incidence of abortion depending on the inoculation route (intramuscular and subcutaneous) and dosages ($1{\times}10^9$, $2.8{\times}10^9$, and $4.0{\times}10^9$ CFU). The antibodies to the SRB51 were measured by a dot blot enzyme-linked immunosorbent assay. The highest titers to SRB51 were detected between 5~7 weeks after inoculation and the specitic antibody could be detected up to 28 weeks after inoculation. The SRB51 was isolated from amnio-allantoic fluid, bronchial lymph node, mammary gland, and supramammary lymph node in 5 of 25 dairy cows during 4 weeks after either abortion or delivery. Although SRB51 was isolated from 4 of 24 aborted fetus or normally delivered calves at parturition time, it was not isolated during 4 weeks afterward. Eleven of twentyfive dairy cows showed the endometritis and/or necrosis until 6 weeks after delivery, no lesions were seen at 8 weeks after delivery and uterus from control dairy cows. The results of present study revealed that SRB51 might induce the clinical signs of brucellosis in the pregnant cattle at 4~7th month of gestation.

Analysis of bee venom residues in milks of dairy cattle using UHPLC with newly developed pre-processing method (봉독 분석을 위한 전처리 방법 개발 및 이를 이용한 젖소 원유 중의 봉독 잔류물질 조사)

  • Bee venom has been used as to prevent and treat bovine mastitis as natural antimicrobial compounds in some dairy cattle farms in Korea. It is needed to determine the residual of bee venom in milks of dairy cattle treated with bee venom. Since bee venom is not approved as a raw material for animal drugs, the preprocessing method to detect bee venom residual in milk and the tolerance limit for its residue has not been established yet in Korea. Therefore, the purpose of this study was to develop pre-processing method not affecting major component of bee venom for detection of its residue in milks using ultra-high performance liauid chromatography (UHPLC). In addition, bee venom residue was also analyzed in milk samples of dairy cattle treated for mastitis with bee venom using UHPLC with the developed pre-processing method in this study. As a result, melittin, histamin and phospolipase A2, the major components of bee venom, were all detected by UHPLC with the pre-processing method developed in this study. The results of this study suggest that the pre-processing method developed in this study can be useful to detect bee venom residue in dairy cattle milk. We also found that no bee venom residues were detected in milk samples collected from dairy cattle treated with bee venom after 1 and 3 days, respectively.
